1.Changes of 5-HT & 5-HIAA Levels in Spinal Cord after Compression Injury

Changes of neurotransmitter 5-hydroxytryptamine(5-HT) and its metabolic product 5-hydroxyinaoleacetic acid(5-HIAA) in the spinal cord after comprssion injury in 10 dogs were studied. The spinal cords were compressed for 1 hour with a rubber balloon filled with 0.45ml water when the animals were under pento-barbital anesthesia and dissected after the sacrifice of the animals.The 5-HT and 5-HIAA contents in injured segments, the segments just above and below the injured part, and normal segments of the spinal cord were determined with the fluorometric method. It was found that the 5-HT levels in the segments just above and below the injury were markedly elevated, and the 5-HIAA levels in the injured segments were significantly lowered. It implies that the metabolism of the serotonergic nervous system in the injured segments is inhibited, while that in the segments abvoe or below the injury is enhanced. It is concluded that the serotonergic nervous system of the spinal cord can be involved in the pathophysiology of spinal cord injury.
2.Effect of Pentobarbital on Monoamine Levels in Ischemic Brain

The effect of pentobarbital on cerebral protection and brain monoamine levels during cerebral ischemia were studied in 60 Mongolian gerbils. The animals were randomly divided into three groups. Pentobarbital(n = 20)and control(n = 20) groups which underwent left common carotid artery (CCA) ligation received 60 mg/kg of Pentobarbitalum natricum and equal volume of normal saline respectively at 30 min prior to operation. Sham-operated group (n=20)was treated as same as the control group with exception of not ligating CCA. The heurological deficits were evaluated and brain monoariiines measured at the third nour after operation.It was found that the stroke index was reduced, neurological deficits improved and brain norepinephrine(NE), dopamine(DA)and 5-hydroxytrytamine(5-HT) elevated to normal levels in pentobarbital-treated animals. 4.Research progress in nerve growth factor and immunopathology of rheumatoid arthritis

Rheumatoid arthritis (RA) is a common autoimmune disease characterized by chronic inflammation and aggressive arthritis. The basic pathological changes of RA include intra-articular hyperplasia synovitis and extra-articular vasculitis, symmetrical joint cavity effusion and stenosis, and the formation of pannus leading to the destruction of articular cartilage or joint accessory structures. In the course of RA, swelling and pain of the affected joints occur, causing joint deformities, joint stiffness, and joint dysfunction, and eventually disability. In recent years, significant progress has been made in the study of the relationship between nerve growth factor (NGF) and RA immunopathology. The results of existing studies have shown that the level of NGF in the synovial fluid of RA patients is elevated, suggesting that NGF plays an important role in immune inflammation-mediated pain behavior. In addition, nerve growth factor precursors (including proNGF and proBDNF, etc.) can promote cell apoptosis and inflammation. Among them, the levels of proBDNF and its receptors have significant changes in the blood of RA patients. Therefore, it can be inferred that proNGF and proBDNF may become new targets for RA treatment. According to the latest international domestic research results, in this paper the research progress of NGF and RA in immunopathology, immune inflammatory response, pain behavior, etc. were briefly introduced, and the potential application value of NGF in the treatment of RA was summarized.
5.Bone marrow mesenchymal stem cells cultured by umbilical cord blood serum versus adult autoserum in vitro

BACKGROUND:Studies concerning bone marrow mesenchymal stem cells (BMSCs) cultured by umbilical cord serum in vitro were hot topic to avoid the heterogenous serum rejection during BMSC culture and improve culture efficiency of BMSCs.OBJECTIVE:To compare biological feature of BMSCs by the umbilical cord serum and adult autoserum in vitro.DESIGN,TIME AND SETTING:The opening experiment was performed at the Laboratory of Cell Biology,Xiangya Second Hospital,Central South University,China from October 2006 to June 2008.MATERIALS:Sixteen bone marrow samples were provided by Xiangya Second Hospital,Central South University and Xiangtan Central Hospital of Hunan Province.METHODS:BMSCs were obtained from 16 fresh adult bone marrow by gradient centrifugation with Ficoll Paque,cultured with DMEM/F12 with 10% umbilical cord blood serum and 10% adult autoserum.The fourth passage was used in this study.The surface antigens of BMSCs were detected by flow cytometry.BMSCs could differentiate into ostenblasts and adipocytes under culture in conditioned medium for osteogeneais and adipogenesis and the differentiated BMSCs were identified by morphological observation,immunophenotype and immunochemical staining.MAIN OUTCOME MEASURES:The following parameters were measured:morphological observation;cell cycle and cell count;identification of surface antigen;observation of osteoblasts and adipocytea induced from BMSCs by immunochemical staining.RESULTS:The quantity and velocity of BMSCs in umbilical curd blood serum was better than those in adult autoserum (P < 0.05)Only few cells were proliferating in both medium,BMSCs in S phase in umbilical cord blood serum was more than those in adult autoserum (P < 0.05).The positive rate of CD29,CD73 and CD105 on BMSCs in umbilical cord serum (above 90%) was higher than those in adult autoserum (P < 0.05),and the positive rates of CD31,CD34 and CD45 were lower than 2%,and the positive rate of CD31 was lower than those in adult autoserum (P < 0.05).The positive rate of BMSCs differentiated into osteoblasts and adipocytes in umbilical cord blood serum was also higher than these in adult autoserum (P < 0.05).CONCLUSION:Purity and differentiated potency of BMSCs in umbilical cord blood serum are better than those in the adult autoserum in vitro.The umbilical cord serum is more adapt to clinical application.
6.The study of precusor of Brain-drived neurotrophic factor inhibits proliferative acyivities of oligodendrocite precusor cells after spinal cord injury

Objective To observe whether immature Brain Derived Neurotrophic Factor (proBDNF)can affect the activities of OPCs in the fields of cell proliferation and migration after SCI,and to investigate the relationship between proBDNF and p75NTR signal pathway on OPCs.Methods OLN-93 cell line was cultured and maintained for in vitro experiments.Immunofluorescence were used to check the expression of endogenous proBDNF,p75NTR and sortilin on OPCs.MTT assay was used to illustrate the inhibitory effect of proBDNF.The effects of anti-proBDNF was also observed by BrdU staining to find a probably signal pathway for proBDNF on OPCs.The Sprague-Dawley rats were administered for T9 spinal cord injury animal model.BBB score was applied to observe the situation of functional recovery after treated by anti-proBDNF.BrdU staining was managed to observe the situation of OPCs proliferation and migration after SCI.Results Endogenous proBDNF inhibited proliferation and migration of OPCs after SCI.BrdU staining showed that population of proliferative OPCs in lesion site of spinal cord was less in proBDNF in treated group than that in control group and anti-proBDNF group.While anti-proBDNF could inhibit proBDNF specifically and might induced a better functional recovery which was illustrated by BBB scores.The in vitro experiments found the inhibitory effect of proBDNF is dose-dependent and can be neutralized by anti-proBDNF properly.Moreover,the expression levels of p75NTR and sortilin are down regulated by proBDNF antibody treated group.This indicated that proBDNF may inhibit OPCs via p75NTR pathway.Conclusion Endogenous proBDNF can inhibit cell proliferation of OPCs after SCI and can be neutralized by specific antibodies of proBDNF.This kind of detrimental effect may be induced by p75NTR-sortilin pathway.Furthermore,proBDNF antibody treatment is effective to block proBDNF and promote the functional recovery.
7.EFFECT OF ANISODINE ON BRAIN MONOAMINE

The effect of anisodine on brain monoamine was studied in 30 rats. The brain monoamine levels of caudate nucleus, hippocampus, diencephalon, brain stem and cerebral cortex were estimated by fluoro-metric method in drug-treated rats and saline-treated controls. Only the NE level of the brain stem was significantly increased in anisodine-treated animals 48 hours after injection. There were no significant differences in the dopamine values between anisodine-treated rats and saline-treated controls, while the 5HT and 5HIAA levels were significantly increased in anisodine-treated animals. The relation of brain monoamines to learning and memory is discussed. The increased 5HT and 5HIAA levels caused by anisodine may play a role in the impairment of memory.

Objective To observe the safety and effectiveness of high-dose chemotherapy(HDT) supported by autologous peripheral blood hematopoietic stem cells transplantation(APBHSCT) in the patients of terminal or relapsed malignant tumors.Methods The patients achieved CR or PR after 4 cycles of conventional salvage chemotherapy were divided into two groups randomly.26 patients in the research group were adapted APBHSC+HDC,50 patients in the control group were adapted 2~4 cycles of routine chemotherapy.Results In the research group,hematopoiesis was reconstructed in all patients,8 out of 17 patients who achieved PR after 4 cycles of conventional salvage chemotherapy turned into CR after APBHSC+HDC(CR rate 47.1%).In the control group,33 patients who achieved PR after 4 cycles of conventional salvage chemotherapy were still PR after 2~4 cycles of routine chemotherapy.The median survival period in the research group was 11 months,which was longer than the one in the control group.The survival rates of 2,3 and 4 years in the research group were evidently higher than the ones in the control group(P

ObjectiveTo compare the safety and efficacy of ligature between traditional ligation way in gastrectomy.Methods60 patients with gastrointestinal cancioma were selected and randomly divided into two groups,30cases of ligature(treatment group),30 cases of ligation (traditional group).ResultsThe operative time,blood loss,postoperative hospital stay,complications and treatment costs was compared between the two groups.Compared with the traditional group,there were shorter treatment time,reduced bleeding,shorter hospital stay,fewer complications,reduced treatment costs in treatment group.ConclusionLigature gastrectomy is safe and effective.
