1.Awareness of knowledge about iron deficiency anemia in patients with it

Objective To investigate awareness of knowledge about iron deficiency anemia (IDA)in patients with it.Methods Patients diagnosed as IDA at their initial visits to Affiliated Hospital of Qingdao University Medical School were randomly selected for the study during January 2008 to January 2009 with self-designed questionnaire to investigate their awareness of knowledge about IDA.Results Among 199 patients with IDA investigated, their awareness was lower in general, five percent about its etiologies, 34.2 percent about its clinical manifestations, 18.1 percent about its hazard to health, and two percent about food abundant of iron.For preventive measures of IDA, 50.7 percent (101/199) of them knew that daily diet should be diversified and 43.7 percent (87/199) knew that meals should be prepared with ferric cooking pot.Awareness of knowledge about other preventive measures of IDA was lower in them.No significant difference in awareness of IDA-related knowledge between them at varied ages was found, except for knowledge about etiology and hazard of IDA.Awareness of etiology of IDA differed between them at varied ages (P <0.05), and about 75.3 percent (64/85)of patients with IDA aged 30 -45 years were caused by chronic blood loss.There was no significant difference in awareness of IDA-related knowledge between patients with IDA caused by varied etiologies (P > 0.05).Conclusions In general, awareness of IDA-related knowledge was lower in patients with IDA.It is necessary to strengthen its popularization, with varied focuses in preventive measures for IDA patients with different etiologies.

A survey was conducted among 121 health care managers and health workers in community health care institutions with questionnaire and interview.Data on health care conditions in the community and health needs of elderly people were collected and analyzed using principle component analysis method.The priority order of 17 community health care projects for elderly people was ranked with this analysis method and the development strategy of community health service was suggested.

5.Mining drug-disease relationships:a recommendation system

Aim Drug repositioning is to find new indications for existing drugs,however,potential drug-disease relationships are often hidden in millions of unknown relationship.With the analyzing of medical big data,we predict the potential drug-dis-ease relationships.Methods Based on the assumption that similar drugs tend to have similar indications,we applied a rec-ommendation-based strategy to drug repositioning.First,we col-lected the information of known drug-disease therapeutic effect, side effect,drug characters and disease characters;second,we calculated the drug-drug similarity measurements and disease-disease similarity measurements;last,we used a collaborative filtering (CF)method to predict unknown drug-disease relation-ships based on the known drug-disease relationships by integra-ting the similarity measurements,and built a ranking list of pre-diction results.Results The experiments demonstrated that a-mong the TOP 500 of the list,1 2.8% were supported by clinical experiments or review,and 20% were supported by model or-ganism or cell experiments.Conclusion Compared to the clas-sification model and random sampling results,the CF model can effectively reduce the false positives.
6.Comparison of single-bundle and double-bundle autografts in anterior cruciate ligament reconstruction

BACKGROUND:Different weaving methods of autologous tendon lead to various treatment efficacies on cruciate ligament rupture, but the preferred method is stil controversial.
OBJECTIVE:To comparatively analyze the clinical efficacy of single-bundle and double-bundle autografts on anterior cruciate ligament reconstruction.
METHODS:A retrospective analysis of 48 cases of anterior cruciate ligament rupture was performed. According to the composition of graft beam, these 48 patients were divided into two groups:double-bundle group (n=24) and single-bundle group (n=24). Al surgical patients underwent autologous anterior cruciate ligament reconstruction by arthroscopy, and were fol owed up for at least 6 months. The function of knee joint after operation was comprehensively analyzed through IKDC and Lysholm scores.
RESULTS AND CONCLUSION:Al of the patients (n=48) exhibited no joint complications postoperatively, such as intra-articular infection, joint effusion and incision inflammation. The IKDC scores and Lysholm scores in the double-bundle group were better than those in the single-bundle group, but there was no statistical y significant difference (P>0.05). This study demonstrated that single-bundle and double-bundle autografts both have good curative effects on anterior cruciate ligament reconstruction by arthroscopy. But in contrast, the curative effects of double-bundle autografts are better.
7.Mutation of Notch1 gene and its protein expression in T-cell lymphoma

Objective To investigate the effect of the expression of Notch1 protein and the mutation of Notch1 gene in.T-cell lymphoma (TCL).Methods Immunohistochemistry was used to detect the expression of Notch1 protein,and PCR amplification and DNA sequencing were used to detect the mutation of Notch1 gene in the 26th and 27th HD domain and the 34th PEST domain in 30 cases.10 cases of reactive hyperplasia tissues of lymph node were as the control.Results The positive rates of Notch1 protein expression and Notch1 gene mutation were 70.0 % (21/30) and 56.7 % (17/30).8 cases of Notch1 mutations were detected in the HD domain,6 cases in the PEST domain,and 3 case in both HD and PEST domains.Inscrtion,deletion,nonsense mutation and missense mutation were included in Notch 1 mutations.Conclusion Notch1 gene mutation may play an important role in the expression of Notch1 protein.The occur of TCL is related to the expression of Notch1 protein and the mutation of Notch1 gene.
8.Three-dimensional PCR-based screening of Chinese fine wool merino sheep BAC library.

For rapid screening, we constructed two levels pools (primary and secondary pools) of the bacterial artificial chromosome (BAC) library of Chinese fine wool merino sheep. The primary pools were based on the individual 384-well microtiter plate and were prepared with a three-dimensional pooling scheme. Three dimension (plate, row and column) pools were made for each. The secondary pools were based on the entire BAC library. We developed a PCR based strategy to identify positive BACs from sheep BAC library. First, we analyzed secondary pools DNAs, according to the result, we analyzed correlative primary pools. It was one-step screening (66 PCR reactions) that we could screen a single positive clone from 74 000 BACs by our method, or three-step screening (less than 100 PCR reactions) could screen more clones. By one-step screening (66 PCR reactions), we screened successfully a positive clone 373D13 with polymorphism marker BF94-1.

9.Ultrastructural study of alveolar type Ⅱ cells in young rats with lipopolysaccharide-induced acute lung injury

Objective Acute lung injury (ALI) and acute respiratory distress syndrome (ARDS) are common and life-threatening disease in children with mortality as high as 40%-70%. Alveolar type Ⅱ cells (ATII cells),characterized by the presence of lamellar bodies (LBs),synthesize and secret surfactant proteins (SPs),which contribute significantly to surfactant homeostasis and pulmonary immunity.The functions of ATⅡ cells including pulmonary surfactant production are autocratically dominated by the structural integrity of ATII cells.Our study is focused on the ultrastructural alterations of AT Ⅱ cells in rats with lipopolysaccharide(LPS)-induced ALI.Methods Rat ALI models were established by intraperitoneal injection of LPS (4 mg/kg).0.9 % NS with same amount was given in the normal control group.The rats were randomly chosen and sacrificed at 24, 48 and 72 hrs after LPS injection (8 rats at each time point).Lung samples (1 mm3 of the size) were obtained from the lower parts of left lungs and fixed with 2.5% glutaraldehyde for the transmission electron microscope examination.Results The microvilli around ATII cells disappeared and the number of LBs increased at 24 hrs after LPS administration.LBs rearranged like a ring around the nuclei.It was commonly seen that two nuclei were present in one AT Ⅱ cell.Vacuole-like deformity prominently occurred in cytoplasm at 48 hrs.Giant LBs presented at the same time.The shapes of nuclei were irregular and some of the borders were unclear at 48 and 72 hrs.The remnant of ruptured LBs scattered in cytoplasm at 72 hrs.The number of LBs reduced obviously.Karyolysis occurred in some of the nuclei.Conclusions The ALI-related alterations of ATII cells characterized by the changes of LBs,nuclei,and nucleoli were time-dependent. ATII cell injury was serious at 48 and 72 hrs.This may lead to the insufficiency of pulmonary surfactant synthesis and unstability of pulmonary homeostasis,which contributed to to the pathogenesis of acute lung injury.
10.Studies on identification and secondary metabolites of endophytic fungi strain E8 from Curcuma wenyujin.

OBJECTIVETo identify the endophyte strain E8 with high activity from Curcuma wenyujin and study its secondary metabolites.
METHODThe strain E8 was identified by morphological observation and ITS sequence analysis. Manifold chromatographic methods were used to separate and purify the chemical constituents of fermentation broth from strain E8, and their structures were identified by physiochemical properties and spectral data.
RESULTThe strain E8 belongs to P. oxalicum. Four compounds were isolated from the fermentation broth of this strain and elucidated as chrysophanol, emodin, secalonic acid A and beta-sitosterol.
CONCLUSIONThe endophyte P. oxalicum was isolated from medicinal plant Curcuma wenyujin for the first time. Four compounds were first isolated from endophytic fungus in C. wenyujin. Thus, microbial fermentation is a new access for these compounds production.
