Objective To investigate the relationship between β3 - adrenergic receptor(β3 - AR)gene poly-morphism and the susceptibility of occurrence in children obesity. Methods The database of PubMed,Web of Science,Springer Link,Medlink,SinoMed,Wanfang,VIP and CNKI were systematically browsed online. The study re-ports regarding β3 - AR associated with children obesity from January 1995 to February 2014 were included in the re-search. The data were extracted,evaluated and heterogeneity tested by using software RevMan 5. 1. Results A total of 162 related literature were retrieved,and 9 of them were eligible with 1 494 obese children and 1 623 normal children were included. Allele T compared with C,the differences between the study group and the control group were statistical-ly significant(P < 0. 05),the findings from the same TC + CC compared to TT and TC compared with TT + CC were similar. Japanese research findings were similar to those of China,but the differences between the study group and the control group had no statistical significance(P ﹥ 0. 05)in Hungary. The sensitivity analysis in each study was stable and the funnel graphics were basically symmetric. The results of sensitivity analysis and publication bias diagnostics demonstrated the reliability and stability of the Meta analysis. Conclusions TC + CC/ TT,TC/ TT + CC genotype and allele T/ C are related to the occurrence of childhood obesity in China,Japan,but there is no significant correlation with childhood obesity in Hungary. There are regional difference between β3 - AR gene polymorphism and genetic susceptibility to children obesity.

Objective To evaluate the relationship between vitamin D receptor(VDR) (rs731236)single nucleotide polymorphisms and genetic susceptibility to overweight/obesity in school-age children of Han and Kazakh in Xinjiang Uygur Autonomous Region.Methods The VDR gene rs731236 genotypes were detected by using polymerase chain reaction-restriction fragment length polymorphism method in 250 overweight/obesity school-age children(127Han,123 Kazakh) and 255 healthy controls(131 Han,124 Kazakh) as well.Results (1)There were statistically significant differences in the frequencies of rs731236 genotypes and alleles between the healthy controls of Han and Kazakh (x2 =9.904,P =0.005;x2 =9.312,P =0.002).(2)There were statistically significant differences in the frequencies of TT/Tt + tt genotype and the alleles between the overweight/obesity cases and healthy controls of Kazakh (x2 =3.891,P =0.049 ; x2 =5.871,P =0.015).But the difference in frequencies of rs731236 genotypes and alleles between the overweight/obesity cases and controls of Han were not statistically significant (x2 =0.064,P =0.800 ; x2 =0.190,P =0.663).(3)There were statistically significant differences in the frequencies of TT/Tt + tt genotype and alleles between the overweight/obesity cases and healthy controls of male school-age children of Kazakh (x2 =5.25,P =0.02;x2 =4.57,P =0.03),but the differences in frequencies of rs731236 genotypes and alleles between the overweight/obesity cases and controls were not statistically significant in female school-age children of Kazakh (x2 =0.06,P =0.80 ; x2 =1.40,P =0.24).There was no statistically significant difference in the frequencies of TT/Tt + tt genotype and alleles between the overweight/obesity cases and healthy controls of female school-age children of Han (x2 =0.04,P =0.84 ; x2 =0.04,P =0.84),and there was no statistically significant difference in male school-age children of Han(x2 =0.05,P=0.82;x2 =0.18,P=0.67).Conclusions VDR gene(rs731236)polymorphism bears ethnic diffe-rence.The polymorphisms of VDR gene polymorphism rs731236 are not associated with the susceptibility to overweight/obesity in school-age children of Han,while it is associated with the susceptibility to overweight/obesity school-age children in Kazakh,especially in male school-age children.

BACKGROUND:Adipose stem cells can differentiate into bone cells, cartilage cells, smooth muscle cells, myocardial cells and nerve cells. Adipose-derived stem cells and mesenchymal stem cells have very similar biological properties, and they are also very close in terms of cellsurface marker expression profile. OBJECTIVE:To investigate the expression of human leukocyte antigen II in adipose-derived stem cells and its immunogenicity and immunomodulatory effect, and to explore the specific mechanism. METHODS:After isolation and culture, the adipose-derived stem cells were stimulated with interferon-γ, and the expression of human leukocyte antigen II was detected by flow cytometry, the expression of indoleamine 2,3-dioxygenase was determined by real-time PCR and western blot. Adipose-derived stem cells were used to stimulate lymphocytes, and the cellproliferation was observed. The lymphocytes were stimulated with phytohemagglutinin, and then 1×102-1×105 adipose-derived stem cells were added to inhibit the proliferation;meanwhile, adipose-derived stem cells, a third party, at a dose of 1×102-1×105 were added to the ongoing two-way mixed lymphocyte reaction, and the cellnumber was calculated as count per minute. RESULTS AND CONCLUSION:The expression of human leukocyte antigen II and indoleamine 2,3-dioxygenase was low in adipose-derived stem cells, but up-regulated with stimulation of interferon-γ. Adipose-derived stem cells could not stimulate lymphocyte proliferation, but could inhibit lymphocyte proliferation induced by phytohemagglutinin or al ogeneic lymphocytes, and interferon-γwas highly expressed in the supernatant of co-culture of adipose-derived stem cells and mixed lymphocytes. These results indicate that the adipose-derived stem cells have low immunogenicity and can inhibit cellproliferation of lymphocytes in vitro, and the addition of exogenous interferon-γcan enhance the immunosuppressive effects of adipose-derived stem cells.

BACKGROUND:The metastasis ways of colon cancer include blood metastasis, peritoneal metastasis and lymph node metastasis. Among them, the most common metastasis organ is the liver. It is necessary to establish a simple, suitable animal model to study the biological behavior of the colon cancer liver metastasis, the heterogeneity of cancer cells, and the mechanism of tumor metastasis. OBJECTIVE:To establish animal model of colon cancer liver metastasis with splenectomy injection method. METHODS:Human colon cancer celllines HT29 were cultured, and subcutaneously transplanted tumor was produced in BALB/c mice. Models of colon cancer liver metastases were established through spleen injection and excision of spleen method. The tumor growth and metastasis, the pathology of transplanted tumor, DNA content and cellcycle of the transplanted tumor were al determined. RESULTS AND CONCLUSION:In this study, we successful y established the model of colon cancer liver metastases through the spleen injection via the dorsal midline incision, the tumor formation rate reached 8/8 (100%). Observation of tissue slice showed that the transplanted tumor was adenocarcinoma I-II. Flow cytometry analysis results showed that DNA ploid was heteroploid DNA. This model provides a good technical platform for biology research for colon cancer metastasis.

OBJECTIVETo evaluate the effects of different treatments on the prognosis of patients with synchronous colorectal liver metastasis(CLM).

METHODSClinicopathological data of 121 patients with synchronous CLM in The First Affiliated Hospital of Xinjiang Medical University between January 2010 and December 2014 were retrospectively analyzed. According to the metastatic lesions, patients were divided into simple operation group(22 patients, receiving operation only), simple chemotherapy group(43 patients, receiving chemotherapy only), and combination group(56 patients, receiving chemotherapy based on operation). The prognosis of patients in three groups was investigated. Univariate and multivariate analyses were performed through the long-rank test and Cox model to evaluate the related factors affecting prognosis.

RESULTSThe median survival time was 10(3-39) months in simple operation group, 7(3-36) months in simple chemotherapy group, and 18 (4-66) months in combination group. The differences among groups were all statistically significant (all P<0.05). Extent of lymph node metastasis, number of liver metastatic lesion, size of liver metastatic lesion, resection edge extent of liver metastatic lesion, and treatment method were independent factors of synchronous CLM(all P<0.05).

CONCLUSIONCombination therapy of a variety of treatment can prolong the survival of patients with synchronous CLM.

Objective To investigate the expression levels and prognostic value of preoperative serum exosomes micro RNA(miR)-193a and micro RNA(miR)-208b in patients with gastric cancer(GC).Methods A total of 132 patients who underwent GC radical gastrectomy in the First Affiliated Hospital of Xinjiang Medical University from March 2018 to March 2020 were regarded as the GC group,while 132 healthy individuals who underwent physical examination were selected as the control group.Their clinical and pathological data were collected and compared.The relative expression levels of miR-193a and miR-208b in serum exosomes were detected using quantitative real-time polymerase chain reaction(qRT-PCR)method.Pearson method was used to analyze the correlation between miR-193a and miR-208b.The correlation between the expression of miR-193a and miR-208b in preoperative serum exosomes of GC patients and postoperative prognosis was analyzed using Kaplan-Meier method.Univariate and multivariate COX regression were applied to analyze the influencing factors of prognosis.Results The expression level of miR-208b in the serum exosomes of the GC group was higher than that of the control group(1.77±0.14 vs 1.02±0.01),while the expression level of miR-193a was lower than that of the control group(0.52±0.06 vs 1.01±0.01),and the differences were statistically significant(t=92.551,61.392,all P<0.05).The expression levels of miR-193a and miR-208b in GC patients before surgery were negatively correlated(r=-0.409,P<0.05).The low expression rate of miR-193a and high expression rate of miR-208b in patients with TNM stage Ⅰ+Ⅱ,no lymph node metastasis,and no distant metastasis were lower than those in patients with TNM stage Ⅲ+Ⅳ,lymph node metastasis,and distant metastasis,and the differences were statistically significant(χ2=5.008,4.397;7.142,4.688;4.407,5.189,all P<0.05).The 3-year cumulative survival rate of patients with low expression of miR-193a(30.43%)was lower than that of patients with high expression(60.32%)(χ2=17.861,P<0.001),while the 3-year cumulative survival rate of patients with high expression of miR-208b(27.14%)was lower than that of patients with low expression(64.52%)(χ2=16.340,P<0.001).The independent prognostic factors included serum levels of exosomes miR-193a(HR=0.493,95%CI:0.323～0.753)and miR-208b(HR=2.697,95%CI:1.382～5.262)(all P<0.05).Conclusion The preoperative serum miR-193a level in the exosomes was decreased and miR-208b level was increased,and their expression levels were related to the prognosis of patients undergoing GC radical gastrectomy.

With the development of laparoscopic technology, surgeon's requirements for precise anatomy are gradually increasing. Laparoscopic system has great advantages in the selection of anatomical level, the protection of nerve and blood vessel in surgery. 4K laparoscopic system can provide high-definition vision of operative field, which is conducive to precise anatomical operation. The authors share the practical team cooperation experience of laparoscope left hemi-colectomy for colon cancer in order to provide references for surgical colleagues.

With the development of laparoscopic techniques, the requirement of fine dissection in surgeons is also growing. Laparoscopic technique has the advantages in the choice of anatomical level and nerve vascular protection for surgery. 4K laparoscopy provides more clear field of vision and more detailed anatomic prerequisites. Combined with surgical examples, the authors share experiences in protection of plant nerve during 4K laparoscopic radical resection of colorectal cancer.

OBJECTIVE To explore the risk signal of ixazomib and provide a reference for clinically rational drug use. METHODS The Open Vigil 2.1 online tool was used to extract the data of adverse drug events （ADE） reported by the database of FDA adverse event reporting system （FAERS） from the launch of ixazomib in America （November 20th， 2015） to the latest update of the Open Vigil website （March 31st， 2023）. The data were mined by using the proportional reporting ratio （PRR） and Bayesian confidence propagation neural network （BCPNN） of the proportional imbalance method. The signals were coded by system organ class （SOC） and preferred term （PT） according to MedDRA v25.1. RESULTS A total of 13 841 ADE reports with ixazomib as the “primary subject” were extracted， involving slightly more male patients （49.53%）， and most of them were 65 years old and above （72.48%）； the reports came from 57 countries/regions， mainly America （52.90%）. A total of 186 positive signals were excavated， with 51 high-intensity， 99 medium-intensity， and 36 low-intensity signals， involving 19 SOCs. The top 50 PT in frequency and signal intensity of PRR included neuropathy peripheral （414 cases， high-intensity signal）， platelet count decreased （379 cases， high-intensity signal）， thrombocytopenia （360 cases， high-intensity signal）， cytopenia （75 cases， high-intensity signal） and neurological symptoms （41 cases， high-intensity signal）. SOC involved included nervous system disorders， investigations， and blood and lymphatic system disorders. ADE occurred most frequently in gastrointestinal diseases （2 588 cases）， including diarrhea （1 077 cases， high-intensity signal）， nausea （737 cases， medium-intensity signal）， vomiting （459 cases， medium-intensity signal）， constipation （275 cases， medium-intensity signal）， and so on. The positive signals of infections and infestations contained the largest number of PTs， and most of them were not recorded in the drug instruction， including 12 high-intensity signals （1 030 cases） and 30 medium-intensity signals （627 cases）， which were mainly distributed in lung infection， upper respiratory infection， gastrointestinal infection， sepsis， herpes zoster and so on. The signals of cardiac amyloidosis （7 cases， high-intensity signal） and acute coronary syndrome （14 cases， high-intensity signal） of cardiac disorders and renal dysfunction （91 cases， medium-intensity signal） of renal and urinary disorders were all strong and had not been recorded in the drug instruction. CONCLUSIONS In addition to routine attention to the common ADE of ixazomib in gastrointestinal diseases，nervous system disorders and blood and lymphatic system disorders， clinical attention should also be paid to various infections that may occur during the treatment of patients， and the occurrence of cardiovascular toxicity and renal dysfunction should be monitored.

OBJECTIVETo understand the threshold of Breteau Index (BI) on Dengue fever outbreak in Guangzhou.

METHODSDengue cases from Guangzhou during 2006 to 2012 in the National Infectious Disease Report and Management System were collected and described. Receiver-operating characteristic (ROC) curve was used to judge the threshold of BI on the outbreaks of Dengue fever.

RESULTSA total of 1 038 local cases were reported from 2006 to 2012 in Guangzhou city, with a total of 71 outbreaks and 259 sporadic episodes. Data from the ROC curve analysis showed that the highest Youden index that BI predicting Dengue fever outbreaks or sporadic were 6.4 and 9.5, respectively. When using BI = 6.4 in predicting the sporadic case of Dengue, sensitivity and specificity were 67.8% , 79.1% , respectively. When using BI = 9.5 in predicting the outbreaks of Dengue, sensitivity and specificity were 81.7%, 90.9%, respectively.

CONCLUSIONBoth BI = 5.0 and BI = 9.5 showed effects on predicting the nature of sporadic or outbreak on Dengue, suggesting that the threshold need to be monitored, according to the purpose of control and availability of manpower, in order to get better sensitivity and specificity.
