Objective To study whether peripheral blood (PB)CD34+ cell absolute count can reliably predict peripheral blood progenitor cell (PBPC) autograft CD34+ cell contents. Methods We examined the PB CD34+ cell count and PBPC CD34+ cell count with ProCOUNT by flow cytometry. Twenty-five collections were performed.The PBPC autograft parameters measured were the CD34+ cell?CFU-GM? CFU-E?BFU-E?CFU-MK and mononuclear cell (MNC) content. Objective: To investigate hyperthermia enhanced expression of heat shock protein70 (HSP70) in breast cancer drug sensitive cell line MCF7 and multi-drug resistant (MDR) cell line MCF7/ADR, so as to increase cytotoxic activity of immunologic effector cells against the target cells, and to provide an experimental basis for cell immunotherapy based on hyperthermia. Methods: The immunological effector cells were induced and expanded by cytokines. Breast cancer cell lines MCF7 and MCF7/ADR were treated at 42 ℃ for an hour. Then after being incubated for additional 4 hours, 24 hours and 48 hours, the cells were digested. Flow cytometry (FCM) was used to detect the expression of HSP70 on the target cells. MTT assay was employed to evaluate cell proliferation and the cytotoxic activity of immunologic effector cells against target cells. Results: The expressions of HSP70 in both the target cells had significant difference. The expression of HSP70 in MCF7/ADR cells was higher than in MCF7 cells before hyperthermia. After hyperthermia the expression of HSP70 increased by 6 folds in MCF7/ADR cells, and 22 folds in MCF7 cells and was higher than in MCF7/ADR cells at hour 4. The proliferation inhibition fraction of hyperthermia against MCF7 was significantly lower than that of MCF7/ADR. The cytotoxic activity of immunologic effector cells against MCF7 cells was lower than against MCF7/ADR cells before hyperthermia. After hyperthermia the cytotoxic activity of immunologic effector cells against MCF7 cells rose by 86.23%, and was higher than against MCF7/ADR cells (rose by 30.32%). Conclusion: The expression of HSP70 in breast cancer drug sensitive cell line MCF7 and MDR cell line MCF7/ADR were enhanced significantly by hyperthermia. Cytotoxic activity of immunologic effector cells against both the target cells were increased by hyperthermia. The differential expressions of HSP70 in breast cancer drug sensitive cell line MCF7 and MDR cell line MCF7/ADR affect the cytotoxic activity of immunologic effector cells.

Objective To study manganese superoxide dismutase (MnSOD) expression in colorectal cancer (CRC) cells and observe its relation to metastatic potential. To investigate the diagnostic performance of manganese-enhanced magnetic resonance imaging (MEMRI) for detecting metastatic potential of CRC. Methods High and low metastatic potential CRC cell lines SW620, HCT116, LoVo and SW480, DLD-1, HCT15, Caco-2, as well as normal colon mucosal cell CCD841 CoN were cultured. MnSOD expression level in cells was detected by western blot and the measurement was repeated for three times. HCT15, DLD-1, LoVo and SW620 cells were selected to perform in vitro MEMRI and subcutaneous xenografts were developed for subsequent in vivo MEMRI. MnCl2·4H2O solution was utilized as the contrast agent and T1 shortening was calculated. The differences of MnSOD expression level in cells, average T1 value shortening of cells and xenografts were separately compared by one-way ANOVA.Results The difference of MnSOD expression level in CRC cells were significant (P<0.05). All CRC cells have shown higher level of MnSOD expression than normal cell, except for Caco-2. The highest expression were shown in low metastatic potential cells (HCT15 and DLD-1). Whereas relatively low MnSOD expression was found both in high and low metastatic potential cells (LoVo, SW620, HCT116 and SW480). In vitro MEMRI demonstrated that the average T1 value shortening was greatest in high metastatic potential cells SW620 and LoVo [(289.33±0.57) and (268.45 ± 6.87) ms)], was significantly lower in low metastatic potential cells DLD-1 and HCT15, was lowest in CCD841 CoN [(65.12 ± 0.12)ms] (P<0.05). Twenty nine xenografts were successfully developed, high metastatic potential SW620 and LoVo tumors of 5 and 10 mm group showed greater T1 shortening, low metastatic potential DLD-1 and HCT15 tumors showed significantly less T1 shortening (P<0.05). However no significant difference was found in similar metastatic potential tumors (P>0.05). Conclusions MEMRI has the potential to noninvasively distinguish different metastatic potential CRC by revealing greater T 1 value shortening in more aggressive one. However the MnSOD expression in CRC cells is not corresponding to malignant potential.

Objective To investigate the Laparoscopic management for Nutcraker Syndrome(NCS) with resection of fibrous ring and placing extravascular stent.Methods This was a retrospective analysis of clinical data and treatment process of cases from March 2010 to February 2015 in urology department,affiliated hospital of Guizhou medical university.Five cases with NCS,4 males and 1 female;age were 28-40 years,mean age was 35 years,all cases were afflicted with gross hematuria and flank pain,the history of gross hematuria were 6-72 months.3 cases were afflicted with proteinuria.Duplex ultrasound scanning before surgery revealed the compressed left renal vein (LRV) between the aorta and the superior mesenteric artery(SMA),with peak velocity 110-132 cm/s,an average of 121 cm/s.The flow velocity of LRV in the renal hilum were 18-25 cm/s,an average of 21 cm/s.CT scanning showed that the stricture segment diameter of LRV were 1.2-2.5 mm,an average of 1.8 mm;and the max diameter of proximal dilatation of the LRV in renal hilum were 8.3-15.2 mm,an average of 10.1 mm.The ratio between the dilated segment inner diameter and the stricture segment were 3.4-9.5.Bleeding from the left ureteral orifice was detected by cystoscopy in 3 cases.5 cases were treated by resection of fibrous ring and placing extravascular stent with Laparoscopic management,and the average length of extravascular stent was 4.0 cm.Results The operation was successful in the 5 cases.The average operation time was 83 min.The average blood loss was 65 ml.Hematuria gradually reduce 5-6 days and resolved 7-20 days after surgery in 5 patients.Proteinuria was disappeared successful 2 weeks after surgery in 3 patients.There was no recurrence at 8-24 months' follow-up.3 days after surgery Doppler ultrasound showed the stricture segment diameter of LRV were 3.8-5.6 mm,an average of 4.9 mm;the ratio between the dilated segment inner diameter and the stricture segment decreased were 1.1-2.0,an average of 1.6;the peak velocity of compressed LRV were 25-45 cm/s,an average of 34 cm/s.6 months after surgery,CTA result showed no LRV compression in the aortomesenteric region;the max diameter of LRV in renal hilum were 7.9-9.8 mm and 6.0-8.8 mm in the aortomesenteric region of LRV.Conclusion Etiology of NCS exist a fibrous ring around the left renal vein outflow of the inferior vena cava besides the commonly anatomic extrinsic compression on the LRV as it crosses between the superior mesenteric artery and the aorta.The Laparoscopic management for NCS with resection of fibrous ring and placing extravascular stent is an effective minimally invasive treatment.

Aim To study the antitumor effect of cispl-atin ( DDP) chemotherapy promoted by Taohong Siwu Decoction (TSD) on mice with lung adenocarcinoma mice. Methods Lewis lung carcinoma cell line was used to make homologous lung adenocarcinoma trans¬plantation mouse model. Normal control, Model, TSD, DDP, TSD + DDP groups were set up. The change of transplanted tumor volume after administration was observed, the weight of transplanted tumor was weighed, the expression of Ki67 in transplanted tumor tissue was detected by immunohistochemistry, TUNEL was detected by fluorescence staining, Bcl-2, Bax, cleaved Caspase-3 and cleaved Caspase-9 were detected by immunoblotting, and the content of D-dirtier in plasma was measured by ELISA. Results DDP plus TSD significantly inhibited the growth of transplanted tumor. Ki67 expression in tumor tissue was lower than that in DDP group (28. 3% ±3. 1% vs 40. 3% ±2.1% ). The combined use of TSD and DDP significantly promoted the apoptosis level of transplanted tumor. The positive rate of TUNEL was significantly higher than that of DDP group (41. 0% ±3.0% vs 30.7% ± 4.5%). Bax, cleaved Caspase-3 and cleaved Caspase-9 expressions in tumor tissue were also higher than those of DDP group, while the expression of Bcl-2 was significantly lower than that of DDP group. Moreover, we found a significant interaction between TSD and DDP on the expression of four apoptotic proteins ( P < 0.05 ) .
