1.Expressions of estrogen receptor, progesterone receptor, human epidermal growth factor receptor 2 and relationships of these expressions with clinical pathology in breast cancer patients aged 70 years old and over

Objective To investigate the expressions of estrogen receptor (ER), progesterone receptor (PR), human epidermal growth factor receptor 2 (HER-2) and relationships of these expressions with the clinicopathology in breast cancer patients aged 70 years old and over. Methods The expressions of ER, PR and HER-2 were detected by immunohistochemical staining from 124 elderly breast cancer patients, and the relationships of these expressions with pathological classification, tumor size, lymph node metastasis and TNM stage were analyzed. Results Among 124 patients, the positive expression rate of ER and PR was 75.0% (93/124) and 71.0% (88/124),respectively. They were not associated with pathological classification, tumor size, lymph node metastasis or TNM stage (all P>0. 05). The positive expression rate of Her-2(+++) and Her-2 (++) was 11.3% (14/124) and 24.2% (30/124), respectively. And the expression of Her-2 was significantly associated with tumor size, lymph node metastasis and TNM stage (P<0. 05) but not with pathological classification (P>0. 05). Conclusions The positive expressions of ER and PR are not correlated with the clinicopathology in elderly breast cancer patients. Her-2 overexpression is positively associated with tumor size, lymph node metastasis and TNM stage.
2.Clinical observation on treating severe neonatal diaper rash with hip bath and oxygen blowing

Objective This research is to observe the treatment effects of hip bath and oxygen blowing on severe neonatal diaper rash.Methods 289 neonates with severe diaper rash were randomly divided into three groups.91 cases in the flat tube group,102 cases in the round tube group and 96 cases in the control group.Three groups of neonates were cleaned on the hips and perineuma after poops and then dried with wet tissues and Mupirocin Ointment.The control group was treated with the above-mentioned method.The oxygen blowing groups were treated with hip bath of 1:5000 chameleon solution twice a day.Blowing the hips with oxygen five minutes every time after hip bath.The method of blowing oxygen was that oxygen humidifying containers as normal oxygen aspiration facilities was not filled with water,whose oxygen flowing volume standed at 10 L/min and whose tube blew at the afflicted parts until being dry.The oxygen blower held the oxygen exit and blowed at the afflicted parts in the round tube group and the oxygen blower flattened the oxygen exit and blew at the afflicted parts in the fiat tube group.The treatment effects will be compared among the three groups four days later.Results The cure period of the round tube group was obviously shorter than that of the control group,and the cure period of the fiat tube group was remarkably shorter than that of the round tube group.The total effective rate in the round tube group was obviously higher than that of the control group and the total effective rate in the fiat tube group was obviously higher than that in the round tube group.The difference had a statistical significance.Conclusions The treatment effects for the severe neonatal diaper rash with hip bath and oxygen blowing are remarkable and the oxygen blowing effects with fiat tubes are better than those with round tubes.
4.Performance of Automatic Blood Cell Analysis Parameters for Differential Diagnosis of Myelodysplastic Syndromes and Aplastic Anemia

Objective To evaluate the performance of automated blood cell analysis parameters for differential diagnosis of myelodysplastic syndromes (MDS) and aplastic anemia (AA).Methods Data of automatic blood cell analysis parameters at diagnosis of confirmed patients with MDS and AA from December 2002 to February 2011 in Peking University Shenzhen Hospital were retrospectively reviewed.Results 33 cases of MDS and 36 cases of AA were recruited in this study.Based on the evaluable data,mean corpuscular hemoglobin concentration (MCHC) (328.58 ± 17.24 g/L vs 342.47±18.75 g/L,n=33/36) was significantly lower (P=0.002 1),while monocyte percentage (MONO%) (11.48±9.99 vs 6.94±2.50,n=32/34),platelet distribution width (PDW%) (13.51±4.24 vs 10.62±3.68,n=20/22) and platelet hematocrit (PCT%)(0.11 ±0.10 vs 0.04±0.07,n=11/15) were markedly higer (all P<0.05) in patients with MDS than that of AA.No significantly differences for other blood cell analysis parameters were seen between patients with MDS and AA.Under the condition of best cut-off value,areas under the ROC curve of MCHC,MONO%,PDW and PCT were 0.706 (95% confidence interval:0.584~0.809),0.666 (0.540~0.778),0.668 (0.506~0.805) and 0.745 (0.538~0.894) respectively.MONO% and MCHC had high specificities (97.06% and 88.89%) and positive predictive values (93.3% and 80.0%) for differential diagnosis of MDS from AA.Conclusion MONO% and MCHC may be used as simple indicators for differential diagnosis of MDS and AA.
5.Analysis on pedestrian traffic injury among aged 0-14 years children in Guangzhou, China.

OBJECTIVETo describe the characteristics of child pedestrian traffic injuries among aged 0-14 years old in Guangzhou city and to provide information on road, vehicle and human injuries in pedestrian.
METHODSA database of all motor vehicle crashes involving child pedestrian traffic injury cases over a five-year period (2000-2004), was from the police department.
RESULTSThe annual pedestrian traffic injury rates on incidence, mortality and case fatality were 17.63/ 1,100,000, 1.78/ 100,000 and 10.07% respectively. Both incidence rate and mortality peaked in the 5-9 age group were 26.80/ 100,000 and 2.57/ 100,000, with case fatality peaked in the 0-4 age group (13.29%). For males, the incidence (20.98/ 100,000) and mortality (2.00/ 100,000) were higher than that in females (13.83/ 100,000, and 1.52/ 100,000). The main injured parts of body would include multi-areas (40.34%), head (26.75%) and lower limbs (21.53%). 10.07% of the cases died and 66.18% and 23.75% of them having had severe and slight injures. The major causes were due to drivers (57.50%) and pedestrians (33.47%) faults with top three responsible vehicles were cars (40.14%), two-wheeled motorcycles (34.38%) and vans (22.15%).
CONCLUSIONData from pediatric pedestrian injury poses a threat to children and adolescents, data suggesting that it is important to change behaviors of both drivers and pedestrians.

6.Background chloride currents in fetal human nasopharyngeal epithelial cells.

To characterize the background current in fetal human nasopharyngeal epithelial cells and clarify its relationship with volume activated Cl(-) currents (I(Cl,vol)), whole-cell patch clamp and cell imaging techniques were employed. Under isotonic conditions, a background current [(5.9+/-2.1) pA/pF at +80 mV, n=21] was detected. The current presented a weak outward rectification and a negligible time-dependent inactivation. The current-voltage relationship showed that the reversal potential of the background current [(-0.73+/-1.7) mV, n=21] was close to the calculated equilibrium potential for Cl(-)(-0.9 mV). Application of extracellular hypertonic stimulation (440 mOsmol/L) suppressed the current by (59.6+/-7.1)% and the inhibition was reversible after returned to isotonic conditions. Bathing the cells in hypotonic solution (160 mOsmol/L) induced a volume-sensitive Cl(-) current. The Cl(-) channel blockers, tamoxifen (20 micromol/L) and 5-nitro-2-(3-phenylpropylamino) benzoic acid (NPPB) (100 micromol/L), inhibited the background current by (74.0+/-5.2)% (P<0.01, n=5) and (60.9+/-8.9)% (P<0.01, n=6) at +80 mV and increased basal cell volume by (107.7+/-2.9)% (P<0.01, n=25) and (104.4+/-2.4)% (P<0.01, n=19), respectively. The data indicate that Cl(-) current is an important component of the background current in fetal human nasopharyngeal epithelial cells. The background Cl(-) current is involved in volume activated Cl(-) current and basal cell volume regulation.

7.Regional genotyping and epidemiological characteristics regarding Yersinia pestis isolates in Shaanxi Province, China

We typed Yersinia pestis isolated from plague foci of Shaanxi Province using different region (DFR) and analyzed epidemiological characteristics.Twenty-three DFRs primers and PMT1 (plasmid) primer were used to verify the DFR genomovars and 48 Yersinia pestis were involved to analyze DFR profiles and epidemiological characteristics.In the same year,the genotypes of Yersinia pestis isolated from different infected vector and animals were basically the same.Three genomovars named Genomovar 11,17,and 20 were verified in 48 Yersinia pestis strains in Shaanxi Province.The main genotypes were different in different epidemic years.In 1987-1988 and 2000-2001 years,genomovar 17 was major genomovar and genomovar 20 in 2006 year.In conclusion,the dominant genotypes were different in different epidemic years.As time goes on,DFR genomovars of Yersinia pestis undergone the evolution of gene deletion,which changes genomovar 17 into genomovar 20.
8.Retrospective analysis for cancer screening among 898 elderly male patients

Objective To investigate the distribution,treatment and prognosis of cancer among elderly male patients.Methods Clinical data of 898 male subjects aged 60-105 years with a median age of 82 years who participated in cancer screening in gerontology center of Beijing Hospital from January 1995 to January 2015 were collected in this retrospective study.Low-dose computed tomography,gastroenterological endoscope,alpha fetoprotein plus abdominal ultrasonography,and prostate specific antigen were used to screen lung cancer,gastroenteric cancer,liver cancer,and prostate cancer respectively.The incidences of malignant tumors,treatments and efficacies were collected during the study period.Results The 136 cases(85.0%)were diagnosed as malignant tumors by screening and 24 cases(15.0 %)by clinical symptoms.Three of the most common cancers were lung cancer(46 cases,28.8 %),prostate cancer(33 cases,20.6 %),and colorectal cancer(15 cases,9.4 %).The median age of cancer diagnosis was 73 years.The prevalence of cancer was higher in males aged over 70 years than in males aged under 70 years(18.1% or 112/620 vs.7.2%,20/278,P < 0.01).The median survival time after cancer diagnosis was 5 years (range of 1-20 years).Compared with patients diagnosed by clinical symptoms,patients diagnosed by screening had higher early detection rate of cancer(97.8% or 133/136 vs.54.2% or 13/24,P <0.01),higher definitive therapy rate(79.4% or 108/136 vs.58.3% or 14/24,P<0.05),and lower recurrence rate(6.5% or 7/108 vs.28.6% or 4/14,P<0.01).Conclusions The positive rate of cancer screening is high in males over 70 years old.Elderly males could benefit from early diagnosis and treatment by cancer screening.
9.Preparation of monoclonal antibodies against prion proteins with full-length hamster PrP.

OBJECTIVETo prepare the PrP specific monoclonal antibodies (mAbs) that can be used for the detection of mammalian prions and study of pathogenesis of prion diseases.
METHODSSeveral BALB/c mice were immunized with recombinant hamster prion protein (HaPrP). Three hybridoma cell lines designated as B7, B9, and B10, secreting monoclonal antibodies against HaPrP, were established by hybridoma technique. The mAbs reactivities were evaluated with ELISA, Western blot, and immunohistochemistry.
RESULTSThe mAbs produced by these cell lines reacted well with different recombinant hamster PrP proteins. Western blot analyses showed that mAbs B7 and B9 reacted with PrPSc from the scrapie-infected animals after proteinase K digestion with three glycosylated forms. The mAbs exhibited cross-reactivity with various PrPC from several other mammalian species, including humans and cattles. Immunohistochemistry assays confirmed that mAbs B7 and B9 could recognize not only extracellular but also intracellular PrPsSc.
CONCLUSIONThe mAbs of prion protein are successfully generated by hybridoma technique and can be applied for the diagnosis of prion associated diseases.

10.Activation of chloride current and decrease of cell volume by ATP in nasopharyngeal carcinoma cells.

Whole-cell patch clamp and cell volume measurement techniques were used to investigate the ATP-activated chloride current and the ATP effect on cell volume in nasopharyngeal carcinoma cells. Extracellular application of ATP in micromolar concentrations activated a current with the properties of modest outward rectification and negligible time-dependent inactivation in a dose-dependent manner. The current reversed at a potential [(-0.05+/-0.03) mV] close to the Cl- equilibrium potential (-0.9 mV). Substitution of Cl- with gluconate in the extracellular solution decreased the ATP-activated current and shifted the reversal potential positively. NPPB, one of the chloride channel blockers, inhibited the current by (81.03+/-9.36)%. The current was also depressed by the P2Y purinoceptor antagonist, reactive blue 2, by (67.39+/-5.06)%. ATP (50 micromol/L) decreased the cell volume under the isotonic condition. Depletion of extracellular and intracellular Cl- abolished the ATP effect on cell volume. The results suggest that extracellular ATP of micromolar scales can induce a chloride current associated with cell volume regulation by activation of chloride channel through binding to purinoceptor P2Y.
