Objective To investigate the influence of the aberrant protein expression and mRNA transcription of transforming growth factor ? 1(TGF? 1) on the biological behavior of human bladder transitional cell carcinomas. Methods The expression of TGF? 1 was investigated in 74 specimens of TCCs by SP immunohistochemistry staining, and the level of TGF? 1 mRNA were determined in 43 cases of TCCs by the method of quantitative RT PCR. Objective Stress radiography provides an objective tool to measure posterior knee instability.This study was conducted to evaluate the intraobserver and interobserver reliability of measurements using Telos device to quantify posterior knee instability,compared wim KT-1000 and PDT test for consistency analysis.Methods From October 2008 to June 2009,68 stress radiographs in 34 patients with posterior knee instability were taken using Telos device.The amount of posterior displacement on the radiographs was measured independently by 2 surgeons on 2 separate occasions.Changes in intraclass correlation coefficients(ICCs)were examined to assess the intraobserver and interobserver reliability of the measurement,and were compared with those from KT-1000 and PDT test for consistency analysis.Results Intraobserver ICC was 0.992,while interobserver ICC Was 0.991.There was no significant difierence between Telos and KT-1000 in pair-t test.The data from Telos device was consistent with KT-1000.The coincident ratio Of PDT test was 20% while the posterior displacement of the tibia calculated on stress radiography was 5-10 mm.The coincident ratio of the PDT was 71.4% while the posterior displacement of the tibia calculatcd from stress radiography was 10-15 mm.Conclusion Using Telos device for stress radiograph provides a reproducible method to quantify posterior knee instability,and the consistency between Telos divece and KT-1000 was reliable.The coincident ratio of the PDT test with stress radiography increased when the posterior displacement of the tibia from stress view became more severe.

Objective To observe and analyse the levels of serum glucose,uric acid and lipid profile in bus drivers in Fuzhou City and provide the necessary information for the reasonable health management of bus drivers.Methods Serum from 3 877 bus drivers of Fuzhou City (3 522 males and 355 females)were collected and detected for the concentrations of serum glucose (GLU),uric acid (UA),cholesterol (CHOL),triglyceride (TRIG),high density lipoprotein cholesterol (HDL-C) and low density lipoprotein cholesterol (LDL-C).Statistical analysis was performed to compare the results.Results The levels and abnormal rates of GLU,UA,CHOL,TRIG and LDL-C of male drivers were significantly higher than that of fe-male drivers (P <0.05).The levels of GLU,CHOL,TRIG and LDL-C of male drivers increased along with the growth of age.And statistical differences were observed among the four groups (<30,31~40,41~50 and >50 years old).The level of UA of male drivers decreased with age while HDL-C invariable.Conclusion The levels of GLU,UA,CHOL,TRIG, HDL-C and LDL-C of male drivers varied and more severe than the female drivers.

Objective To explore expression of plexinA1 and Ki-67 in tissue of human brain glioma cells and their clinical significance.Methods 43 specimens from patients with brain glioma were collected.Immunohistochemical (IHC) staining was used for detecting the expression of tissue plexinA1 and Ki-67 in human glioma cells of 43 cases of patients with brain glioma.The positive expression rate of plexinA1 and Ki-67 among the different pathological grade tissues and their clinical significance were analyzed.So did correlation studies about plexinA1 and Ki-67.Results The positive expression rates of plexinA1 in Ⅰ-Ⅱ grade group (18 cases) and Ⅲ-Ⅳ grade group (25 cases) were 22.22 ％ (4/18) and 72.00 ％ (18/25) (P ＜ 0.05).The positive expression rates of Ki-67 in Ⅰ-Ⅱ grade group and Ⅲ-Ⅳ grade group were 16.67 ％ (3/18) and 56.00 ％ (14/25),respectively (P ＜ 0.05).PlxinA1 and Ki-67 expression in the tissue of human brain glioma were positively correlated (r =0.997,P ＜ 0.05).Conclusions The positive expression rate of plexinA1 is higher in high malignancy human glioma group than that in low malignancy group which has an important reference value in the estimation of prognosis for human glioma.PlexinA1 and Ki-67 maybe synergism in occurrence and development of glioma.

ObjectiveTo retrospectively determine the prevalence of Ramp lesion (the peripheral attachment of the posterior horn of the medial meniscus) of the posterior horn of the medial meniscus with concurrent anterior cruciate ligament(ACL) ruptures and analyze the relevant risk factors.MethodsFrom April 2002 to October 2007,868 consecutive knees were diagnosed as ACL injury and received arthroscopic surgery for ACL reconstruction(ACLR).All the patients were verified for the tears of Ramp area under arthroscopy.The prevalence of Ramp lesion was evaluated respectively.Then all cases were divided into different groups depending on the time interval from ACL injury to ACLR (TFI) and other relevant risk factors as age and gender.ResultsAmong 868 knees underwent surgery for ACLR,144 knees were diagnosed as Ramp lesion.The average age was 24.7 years,113 males,31 females.The mean TFI to ACLR was 29.3months.The prevalence of Ramp lesion was 16.59％,which was analyzed as logarithmic correlation with TFI.Patients younger than 30-year old and male patients had significant higher prevalence of Ramp lesion.ConclusionRamp lesion is a common meniscus injury that can occur at the time of ACL rupture or as a result of knee laxity associated with ACL insufficiency.The prevalence of Ramp lesion is 16.59％ which increases with time until 24 months after initial injury.Patients younger than 30-year old and male patients have higher prevalence of Ramp lesion.

Objective To investigate the expression of Notchl and NUMB in gastric cancer and explore the relationship between two genes and clinicopathologic features. Methods The mRNA and protein expressions of Notchl and NUMB in the gastric cancer tissues and normal gastric tissues were determined by realtime PCR and Western blotting. Results The mRNA expression of Notchl in gastric cancer tissues was 1.67times that of Notchl in normal gastric tissues. The increase of the mRNA expression of Notchl was correlated with tumor differentiation and lymph node metastatis of the patients ( P < 0. 05 ). The mRNA expression of NUMB in gastric cancer tissues was 0.597 times that of Notchl in normal tissues. The decrease of the mRNA expression of NUMB was correlated with tumor differentiation of the patients (P < 0.05). The expression of Notchl was negatively correlated to that of NUMB ( r = - 0.459, P < 0.05 ). The protein expression of Notchl in gastric cancer tissues was (0. 348 ±0. 133) , which was significantly higher than (0. 208 ±0. 140) in normal gastric tissues (P < 0.05 ). The protein expression of NUMB in gastric cancer tissues was (0.490 ±0.440), which was significantly lower than 0.746 ±0.390 in normal gastric tissues (P<0.05).Conclusion Up-regulation of Notchl and down-regulation of NUMB may play an important role in the occurrence and development of gastric cancer.

Objective:To express and purify recombinant human collagen type Ⅲ and evaluate its properties.Methods:The recombinant genetic engineering strain pET30a(+)-1880/pACYCDuet-hy726/bL21(DE3) was constructed to stably co-express recombinant human type Ⅲ collagen (rhCol) and prolyl hydroxylase. rhCol was prepared and purified by E. coli high-density fermentation, salting out and column chromatography protein purification technology. Sodium dodecyl sulfate-polyacrylamide gel electrophoresis was used to determine the purity of rhCol. The N-terminal amino acid sequences of rhCol were determined by automatic protein polypeptide sequencing instrument. The hydroxyproline content of rhCol was determined by ultraviolet spectrophotometry. The cellular compatibility of rhCol was evaluated by MTT assay. Results:The final wet weight of high-density fermentation was about 200 g/L. The expression level was about 3 g/L. The purity of rhCol by affinity chromatography was over 95%. The results showed that the hydroxyproline content of rhCol was 11.44%, and the rhCol products have good water solubility and cell compatibility.Conclusions:RhCol can be widely applied to the field of skin care and biomedicine as an excellent biological material.

Objective To explore the clinical characteristics and treatment of infants with cytomegalovirus(CMV)pneumonia.Methods The clinical and auxiliary examination data of 24 patients with CMV pneumonia,which were diagnosed by detection of serum CMV-IgM used enzyme immunodot technique.All patients were treated with ganciclovir (GCV).Results The most common clinical manifestations of CMV pneumonia were cough,breathlessness,fever,crackles and wheezing rale.Chest X-ray findings predominately revealed increased and thickened bilateral lung markings and patchy areas of increased opacity in both lungs.Sixteen cases were cured,8 cases improved.Conclusions Clinical manifestations of infant CMV pneumonia lack specificity.The presence of serum CMV-IgM antibody is a laboratory diagnostic evidence.GCV is the best choice of drug in treatment of infants with CMV pneumonia.
