3.Establishment of a skin-infection model in immunosuppressive SD rats induced by cyclophosphamide

Objective To establish an effective and stable rat model of skin-infection for evaluating the therapeutic effects of topical anti-infection drugs.Methods SD rats were subcutaneously injected with cyclophosphamide to induce immunosuppression,and then surgical incisions were made on both sides of the spine.The rat model of skin-infection was established by applying methicillin-resistant Staphylococcus aureas suspension to the incisions for two consecutive days,and evaluated by analyzing infection status,pathological changes and healing time.Results The development of pyogenic infection was detected in all of the rats.Compared with the non-cyclophosphamide treated group,the cyclophosphamide treated group showed a more severe infection both from the visual inspection and the microscopic observation,moreover,its healing time was delayed more than 10 days.Conclusion The skin-infection model was successfully established in immunosuppressed rats induced by cyclophosphamide,which could be applicable to the efficacy evaluation of anti-infection drugs for external use on skin infection.
4.Epidemiological survey of human brucellosis in Wulanchabu City of Inner Mongolia in 2010

Objective To understand the morbidity of human brucellosis in Wulanchabu City of Inner Mongolia in order to provide a basis for development of prevention and control measures.Methods According to the requirements in Baseline Survey Programme of Human Brucellosis in Inner Mongolia Autonomous Regionbetween November and December in 2010,stratified cluster sampling method was used to selected three townships according to the conditions of brucellosis(mild,moderate and serious) in 11 flags(cities,counties,districts) of the city.Three villages were extracted from each township.There were at least 200 persons aged 10 and older were investigated in every township.At least 600 people were investigated in every flag(city,county,district).Respondents were investigated about their awareness rate of basic knowledge about brucellosis,risk factors and common sense of prevention and treatment of the disease by using health education questionnaire.Based on the principle of informed consent,we collected respondents venous blood for preliminary screening using Hu red tablets.Brucellosis was confirmed with standard tube agglutination test and positive result was confirmed if 1:100 antibody concentration was two + or more.Prevalence,false negative rate as well as different age,gender,occupational incidence was calculated based on the network reported results of brucellosis in 33 townships of Wulanchabu in 2010.Results We distributed 6998 questionnaires and 6763 questionnaires were effective.The witting rate of basic knowledge about brucellosis was 57.99%(15 687/27 052); the witting rate of risk factors about brucellosis was 44.33% (29 978/67 630); the witting rate for prevention and control of brucellosis was 41.66%(28 176/67 630),and total witting rate was 45.49%(73 841/162 312).The morbidity of brucellosis in Wulanchabu City was 91.39/10 000 (785/85 894) in 2010.Five hundred and ninety-one were serologically positive and the infection rate was 8.48% (591/69 972).Nine hundred and three cases of patients were diagnosed with brucellosis and the prevalence rate of brucellosis was 105.13/10 000 (903/85 894).Missing report 377 cases,and the false negative rate was 48.03% (377/785) in 2010.Men infection rate was 71.07%(420/3755) and women infection rate was 28.93%(171/3217).The infection rate[97.63%(577/591)] of people aged 31 and older was significantly higher than that [2.37%(14/591)] of the people aged 30 and younger.Infection rate of people engaged in aquaculture was 8.61% (582/669).The infection rate of people working in livestock processing industry was 2.91% (2/103).The infection rate of people working in animal by-product circulation was 2.85% (2/70).The Infection rate of people working in other industries was 4.59%(5/109).Conclusions In Wulanchabu City,the morbidity of brucellosis and missing report rate are both high.The infection rate of people engaged in aquaculture is high and man morbidity is high.The awareness rate about prevention and treatment knowledge of brucellosis is low.We should carry out health education to inhabitants in endemic areas to improve their self-protection awareness and reduce the incidence of brucellosis.
5.Regulatory effect of Skp2 on the expression and transactivation of the androgen receptor in the progression of castration-resistant prostate cancer.

OBJECTIVETo determine the expression of Skp2 in different prostate cancer (PCa) cell lines and tissues, and explore its influence on the androgen receptor (AR) signaling pathway and development of castration-resistant prostate cancer (CRPC).
METHODSThe expression levels of Skp2 and AR in different PCa cell lines were detected by Western blot. After knockdown of Skp2 in the C4-2 and 22RV1 cells transfected with shRNA, the expressions of AR and P27 were determined and the activity of ARR3-Luc measured by dual-luciferase reporter gene assay following treatment with dihydrotestosterone (DHT). The expressions of AR and Skp2 in human naïve PCa or CRPC specimens were detected by immunohistochemical staining followed by analysis of their differences and correlation.
RESULTSThe Skp2 protein expression level was significantly higher in the C4-2 or 22RV1 cells than in the LNCaP cells. DHT treatment increased the expression of Skp2 in the C4-2 cells, but knock-down of Skp2 significantly up-regulated the expression of the well-known downstream protein P27 and down-regulated that of AR. Consistently, DHT treatment increased the activity of ARR3-Luc, while knockdown of Skp2 remarkably decreased it in the C4-2 and 22RV1 cells (P < 0.05). In addition, significantly higher expressions of Skp2 and AR were observed in the CRPC than in the naïve specimens (P < 0.05), with a positive correlation between the two proteins (r = 0.658 1, P < 0.05).
CONCLUSIONSkp2 can enhance the expression and transcription activity of the AR protein in CRPC cells or tissues and is promising to be a critical molecular therapeutic target.

6.Can post-activation potentiation induced by high-intensity dynamic and static kinetotherapies enhance muscle explosive power?

BACKGROUND: Increasing evidence has shown that high-intensity warming up, including dynamic and static kinetotherapies, can improve the performance of muscle explosive power. However, the training scheme to achieve the best effect remains unclear. OBJECTIVE: To discuss whether the high-intensity dynamic and static kinetotherapies can enhance post-activation potentiation of the upper and lower limbs, so as to fully understand the high-intensity warming up, and thus promote the training efficacy. METHODS: Ovid Medline, Elsevier SDOL, PubMed, and CBJ databases were retrieved for the articles addressing the post-activation potentiation of high-intensity dynamic and static kinetotherapies published before 2016. The articles were classified in accordance with Oxford Centre for Evidence-based Medicine Levels of Evidence. RESULTS AND CONCLUSION: The possible mechanisms of the post-activation potentiation include the phosphorylation of myosin regulation light chain, the increase in motor unit recruitment as well as the change of pinnate angle. The resistance exercise at an appropriate mode, including dynamic, static, and dynamic combined with static, significantly induces post-activation potentiation, and improves the explosive force. Post-activation potentiation is impacted by muscle contraction type, amount of exercise, exercise intensity, induction time, and history of the subjects (training experience, absolute muscle force, and muscle fiber composition). High-intensity stimulus of post-activation potentiation may be accompanied by the generation of fatigue, and the inappropriate recovery time may result in the imperfect performance of post-activation potentiation.
7.Clinical and pathologic features of renal mucinous tubular and spindle cell carcinoma.

OBJECTIVETo investigate the clinical and pathological features of the mucinous tubular and spindle cell carcinoma (MTSCC) of the kidney.
METHODSSeven cases of MTSCC were analyzed by gross examination and light microscopy. Immunostaining was performed to detect the expression of CD10, CK7, CK18, CK19, Villin, EMA, P504S and vimentin. The literature on this tumor was reviewed to discuss the histological features of MTSCC and its clinical behavior.
RESULTSThree of 7 cases were male and the other 4 were female. The mean age of the patients was 48.2 years old, with a range from 39 to 61 years. All the patients presented no symptom and their tumors were found by health examination. Tumors averaged 5.5 cm in greatest dimension (range from 4.0 cm to 9.0 cm). The tumors were well-circumscribed without capsules, and the cut surfaces were solid and soft with white-tan color. By light microscopy, tumors were composed of tightly packed, small, elongated tubules with transitions to spindle cell components. Five cases had mucinous stroma. Clear cell clusters, focal sarcomatoid differentiation, papillations and foamy macrophages were seen in several cases. Immunohistochemically, all 7 cases showed positive for CK7, five of 5 cases positive for EMA, CK18 and P504S, four of 5 cases positive for CK19, but heterogeneous for CD10, villin and vimentin expression. No evidence of local recurrence or distant metastases was identified in the 5 patients with follow-up information.
CONCLUSIONSThe mucinous tubular and spindle cell carcinoma is a low-grade and polymorphic neoplasm. The morphology of these tumors may not be uniform with a wide histological spectrum. The tumors can be tubular predominant or spindle cells predominant with scant to abundant mucinous stroma, which coupled with the presence of other unusual features such as clear cells, papillations, foamy macrophages, necrosis and sarcomatoid differentiation. Immunohistochemically, MTSCC can express the markers from the proximal convoluted tubules to collecting tubules.

8.Study of X-irradiation to enhance the functional and structural recovery of the injured spinal cord of rat.

OBJECTIVETo explore whether X-irradiation can enhance the functional and structural recovery of the injured spinal cord of rats.
METHODSSeventy Sprague-Dawley rats received spinal cord injury by clip compression at the T2 level were randomly divided into two groups. The experimental group received X-irradiation at 14 days after injury, the control group did not receive X-irradiation. The functional tests were performed at day 3, 7, 14, 21, 28, 35, and 42 after irradiation including open field movement, inclined plane and pain withdrawal test. All injured rats were sacrificed at 43 days after injury and the injured spinal cords were taken out for histological tests.
RESULTSSixty-two rats met the experimental requirements among 70 injured rats, 32 rats in experimental group and 30 rats in control group. Statistically significant difference was achieved between two groups in open field movement and inclined plane (P < 0.01), but not for the pain withdrawal test. The edema and necrosis area of injured spinal cords of experimental group were less than those in control group, and the number of axons of experimental group were more than those in control group.
CONCLUSIONSX-irradiation can enhance the functional recovery by improving and restoring structural integrity of the injured spinal cord.

9.Expression and switching of TH1/TH2 type cytokines gene in human gliomas.

OBJECTIVETo study the expression and switching of Th1/Th2 cytokines gene in human gliomas and its effects on occurring and developing of human gliomas.
METHODSInterleukin (IL)-2 and interferon-gamma represent Th1 type cytokines. IL-4, IL-6, IL-10, and IL-13 represent Th2 type cytokines. The gene expressions of Thl/Th2 cytokines in human glioma cells, glioma infiltrating lymphocytes, and glioma cell lines were detected by reverse transcription polymerase chain reaction (RT-PCR). The biological activity of cytokines in the supernatant of glioma cell lines was assayed by enzyme-linked immunosorbent assay (ELISA).
RESULTSThe total positive rates of Th1 and Th2 type cytokines gene in human glioma cells were 14.77% and 75%. The total positive rates of Th1 and Th2 type cytokines gene in glioma infiltrating lymphocytes were 22.73% and 68.17%. There was obviously predominant expression of Th2 type cytokines in human glioma tissues, glioma infiltrating lymphocytes, and glioma cell lines. There was no unbalanced expression of Th1/Th2 cytokines in normal brain tissues.
CONCLUSIONThere is a predominant expression of Th2 type cytokines in human glioma cells. The switching of Thl/Th2 cytokines gene may play an important role in the occurring and developing of human gliomas.

10.Early complications after simultaneous pancreas-kidney transplantation.

OBJECTIVETo explore the early complications after simultaneous pancreas-kidney transplantation (SPK).
METHODSThe clinical data of 20 patients who underwent SPK in our center from September 2002 to September 2007 were retrospectively analyzed.
RESULTSThe complications after SPK included hematuria (n = 4), abdominal bleeding (n = 4), abdominal infections (n = 6), lung infections (n = 5), urinary infection (n = 1), poor wound healing (n = 3), abdominal distension (n = 1), and acute cardial infarction (n = 1).
CONCLUSIONSInfection and bleeding are the most common early complications after SPK. Urinary infection and metabolic acidosis are common in BD mode.
