Chinese Journal of Biologicals 2024;37(10):1153-1160+1166
Objective To compare the production of Coxsackievirus A10(CV-A10) in Vero cells cultured in WAVE bioreactor, Basket bioreactor and NBS bioreactor. Methods Vero cells were cultured in 2 L WAVE bioreactor, 15 L Basket bioreactor and 40 L NBS bioreactor filled with pre-irradiated fibracel disk, fibracel disk and Cytodex 1 microcarriers, respectively,to prepare CV-A10. The adherence efficiency, nutrient metabolites and growth characteristics of Vero cells cultured in different types of bioreactors, as well as the virus titers and antigen yields of the prepared CV-A10, were compared. Then, the CVA10 production process in a 2 L WAVE bioreactor was scaled-up to a 50 L WAVE bioreactor, and the virus titers and antigen yields of CV-A10 prepared in different scale WAVE bioreactors were compared. Results The adherence efficiency of Vero cells on fibracel disk was much higher than that on microcarriers, and the content variation curves of glucose and lactate in different types of bioreactors were consistent. The WAVE bioreactor, Basket bioreactor and NBS bioreactor could all be used for high density culture of Vero cells, while the WAVE bioreactor and Basket bioreactor with fibracel disk could achieve much higher cell density than the NBS bioreactor with Cytodex 1 microcarriers. CV-A10 could be efficiently prepared in different types of bioreactors and there was no significant difference in the virus titers and antigen yields of the prepared CV-A10. Vero cells and CV-A10 were successfully scaled up in the 50 L WAVE bioreactor, and there was no significant difference in the virus titers and antigen yields of different scale WAVE bioreactors. Conclusion The WAVE bioreactor, Basket bioreactor and NBS bioreactor can all be used for the stable culture of Vero cells and the efficient preparation of CV-A10.
4.Optimization of culture process and characterization of critical quality attributes of Coxsackievirus A16
Chinese Journal of Biologicals 2025;38(04):385-391+398
Objective To optimize the culture process of Coxsackievirus A16(CV-A16) and compare the different quality attributes of CV-A16 harvest solution in order to determine the critical quality attributes(CQAs), and lay a foundation for the development of the preparation process and quality control method of CV-A16 vaccine. Methods Using Vero cells as matrix, virus titer and mature complete viral particle antigen content(1H5 antigen) as evaluation indicators, the cell growth phase(mid-log phase, late-log phase, plateau phase), MOI(0. 01, 0. 001, 0. 000 1), culture medium type(serum-free M199, DMEM,50% M199 + 50% DMEM medium), serum concentration of culture medium(serum-free, 1%, and 2% newborn bovine serum) and culture temperature(33, 35, and 37 ℃) for CV-A16 inoculation were optimized, and the optimized process was further verified in T25 cell bottle and 10-layer cell factory. Correlation analysis was performed between virus titer, antigen contents of mature complete viral particles(1H5 and 20E8 antigens), and neutralizing epitope antigen content(3H4 antigen)with immunogenicity to identify critical quality attributes(CQAs). Results The optimum culture conditions of CV-A16 were determined by single factor experiment and enlarged scale culture. Vero cells were inoculated at a MOI of 0. 001 at the end of logarithmic phase and cultured at 33 ℃ in serum-free M199 medium. The antigen contents(1H5 and 20E8 antigens) of mature complete viral particles obtained from CV-A16 harvested solution under four culture conditions(serum-free DMEM culture medium at 33 ℃ for 6 d, serum-free M199 culture medium at 33 ℃ for 6 d, serum-free DMEM culture medium at37 ℃ for 4 d, serum-free M199 culture medium at 37 ℃ for 4 d) were quite different, and the trend of change was different from that of neutralizing antibody titer. The virus titer and neutralizing epitope antigen content(3H4 antigen) of CV-A16harvested solution obtained under the four process conditions changed little, and the trend of change was the same as that of neutralizing antibody titer. Conclusion Based on the quality attribute of immunogenicity(neutralizing antibody titer), the virus titer and neutralizing epitope antigen content(3H4 antigen) can be classified as the CQA of CV-A16 virus harvest solution, and the antigen contents of mature complete viral particles(1H5 and 20E8 antigens) can be used for the analysis of virus particles in the quality control process of CV-A16 production.

Chinese Journal of Primary Medicine and Pharmacy 2008;15(6):987-989
Objective To investigate the easier and more effective moduses of artificial respiration to the neonatal continuous chest compression eardiopulmonary resuscitation. Methods The experience of the treatment on 66 inpatient neonates(with 84 vices cardiac arrest) by continuous chest compression cardiopulmonary resuscitation was summarized. Based on different moduses of artificial respiration matched with mask-gasbag pressure breathing or tracheal intubation pressure breathing to cardiopulmonary resuscitation(CPR), and according to the principles of therandomized block design,sixty-six neonates in cardiac arrest were randomly divided into two groups of A and B. Fun-damental therapeutics in these two groups were alike. A group(38 cases with 53 vices cardiac arrest) was with mask-gasbag pressure breathing. B group(28 cases with 31 vices cardiac arrest) was with tracheal intubation gasbag pres-sure breathing. Time of cardiac arrest (Tca), time of cardiac restore independent rhythm(Tr), Time of cardiopul-monary resuscitation completed(Tc), achievement ratio of cardiopulmonary resuscitation, and 24 hours survival rateof these two groups were compared. Results Compared with B group, there was no significant deviation of Pca, Tr,Tc, the successful rate of CPR and 24 hours survival rate in A group. Comparing A group to B group, the Tea[ (0.99±0.75)rain vs (0.92±0.69)min, P = 0.69];Tr[(3.58±2.15)rain vs (3.66±2.01)min, P = 0.87];Tc [(23.28±9.26)min vs (23.73±9.51)min,P=0.84];suecessful rate of CPR [88.68% vs 83.87% ,P>0.05];24h survival rate [84.21% vs 82.14 %, P > 0.05 ]. Conclusion The mask-gasbag pressure breathing was an easy, safe and effective artificial respiration method for the neonatal continuous chest compression cardiopulmonary resusci-tation. Only in a few eases with airway resistance heightening was the modus of tracheal intubation gasbag pressure breathing applied.
6.Research of correlative factors between the ultrasonography of trauma-caused deep venous free-floating ;thrombus of lower limbs and the prognosis
Chinese Journal of Ultrasonography 2015;(6):496-499
Objective To analyze the color Doppler ultrasonography of trauma-caused deep venous free-floating thrombus(FFT)of lower limbs, discuss the relations between sonography and the prognosis. Methods Clinical data and color Doppler ultrasound examination results of 130 cases with trauma-caused deep venous FFT of lower limbs were retrospectively analysed.Univariate analysis was conducted to identify the statistically significant factors of patients' prognosis,non-conditional logistic regression analysis was performed to analyse the independent prognostic factors of FFT.Results The non-conditional logistic regression analysis showed that the position of FFT,thrombosis edge definition, floating degree and the volume of detouring blood around the thrombus were independent factors of patients'prognosis(P <0.05). The FFT prognosis of iliofemoral vein was more likely to be fixed than the FFT prognosis of popliteal vein. The FFT prognosis with blurring edges was more likely to be fixed. The more obvious the floating degree of FFT,the higher chance that the FFT prognosis is recanalized.More detouring blood flow around FFT indicated more possibilities that the prognosis could be recanalized. Conclusions The color Doppler ultrasound is able to evaluate the relations between the deep venous free-floating thrombus of lower limbs and prognosis of the patients, which is beneficial for early detection and early treatment,as well as reducing mortality and disability resulted from deep venous thrombus during the orthopaedic perioperative period.

Chinese Journal of Tissue Engineering Research 2013;(31):5705-5710
BACKGROUND:Lumbar spinal stenosis can severely affect the activities and functions of the lower limbs, and even cause paralysis. OBJECTIVE:To evaluate the clinical effect of intervertebral bone graft with internal fixation for the treatment of lumbar spinal stenosis. METHODS:The lumbar spinal stenosis patients treated with posterior decompression and intervertebral bone graft with internal fixation were fol owed-up. The effect of intervertebral bone graft with internal fixation for the treatment of lumbar spinal stenosis was evaluated through assessing the clinical symptoms and improvement of signs, recovery of neurological function, as wel as intervertebral bone graft and fusion and the occurrence of complications. The effects of posterior decompression and intervertebral bone graft with internal fixation and simple decompression for the treatment of lumbar spinal stenosis were compared, in order to further determine the treatment of intervertebral bone graft with internal fixation. RESUTLS AND CONCLUSION:The fol ow-up results of the lumbar spinal stenosis patients treated with posterior decompression and intervertebral bone graft with internal fixation showed that the symptoms of low back pain, partial limb sensory disturbances and intermittent claudication were improved significantly;the neurological recovery score could be up to 90%or above. Radiographic examination showed good intervertebral bone graft and fusion with less complication of nerves, blood vessels and dural injury. Comparative study showed that the excel ent and good rate of posterior decompression and intervertebral bone graft with internal fixation can reach to 95.2%, while the excel ent and good rate of simple decompression was 23.8%, indicating that the effect of posterior decompression and intervertebral bone graft with internal fixation was significantly better than that of simple decompression.
