1.Effects of acidosis on cardiac hemodynamics in aged rat heart after reperfusion injury

Objective:To observe the effects of prolonged transient acidosis on cardiac hemodynamics in aged rat heart after reperfusion injury.Methods:60 isolated SD rats hearts(≥18 months)perfused by Langendorff were randomly divided into 6 groups(n=10):(1)normal contro1(C)group;(2)reperfusion injury(I/R)group;(3)ischemic postconditioning(IPO)group;(4)acidosis(H+)group;(5)postconditioning+alkalotic(IPO+OH-)group;(6)alkalotic(OH-)group.The direct effects of prolonged transient acidosis on myocardiac contractility were observed by a langendorff apparatus through the hemodynamics parameters including heart rate(HR),left ventricular systolic pressure(LVSP),left ventricular end-diastolic pressure(LVEDP),and ?dp/dtmax.Results:The LVEDP were increased and HR,LVSP,?dp/dtmax were decreased in I/R group,IPO group,H+ group,IPO+OH-group and OH-group compared with those in C group(P
2.Effects of acidic reperfusion on ischemia-reperfusion injury in isolated rat teart: role of PDK/Akt signaling pathway

Objective To investigate the effects of acidic reperfusion on ischemia-reperfusion injury in isolated rat heart and the role of PI3K/Akt signaling pathway.Methods Seventy adult SD rats weishing 450-550 g were randomly divided into 7 groups(n=10 each):group Ⅰ ischemia-repedusion injury(X/R);groupⅡ ischemic post eonfioning(IPC);group Ⅲ acidic reperfusion(H+);group Ⅳ ischemie postconditioning+alkalotic nsperfusion(IPC+OH-);group Ⅴ alkalotic reperfusion(OH-);group Ⅵ acidic repeffusion+wortmannin (H+wort)and group ⅦVl wortmannin(wort).The animals were anesthetized with intraperitoneal(IP)urethane. Their chests were opened and hearts were excised and passively perfused in a Lsngendorff apparatus with KrebsHenseleit buffer(KHB)saturated with 95%O2-5%O2 at 37℃.The isolated hearts were made globally ischemic for 30 min followed by 120 rain reperfusion.Group II (IPC)WaS subjected to 6 cycles of 15 rain reperfusion and 15 min ischemia at the beginning of reperfusion. In group H+ and OH- (group Ⅲ ,Ⅴ ) the isolated hearts were perfused with KHB of pH 6.9 and 7.8 for 3 rain respectively at the beginning of reperfusion. In group IPC + OH( Ⅳ ) the isolated hearts were perfused with KHB of pH 7.8 for 3 rain followed by IPC. In group H + wort and wort ( Ⅵ,Ⅶ ) the isolated hearts were perfused with wortmannin 100 nmol/L in KHB of pH 6.9 and 7.4 for 3 min respectively at the beginning of reperfusion. LVEDP and ± dp/dtmax were measured before ischemia and at 30 min of reperfusion. Coronary effluent was collected for determination of NO concentration at 30 min of reperfusion. Lnfart size was measured at 120 rain of reperfusion. Results The LVEDP was decreased, ±dp/dt was increased, the infarct size was smaller and the NO concentration in coronary effluent was higher in group IPO and H+ compared with group I/R. Wortmannin abolished the protective effect of acidic reperfusion on isolated rat heart. Conclusion PI3K/Akt signaling pathway may be involved in the protective effects of acidic reperfusion on myocardium against ischemia-reperfusion injury.
3.Detection of free cancer cell CK-20 mRNA in peripheral blood in patients with gastric carcinoma

0.05)Conclusion There are some free cancer cells in the prepheral blood of some patients with gastric cancer before operation. Pulling and stimulating tumor in the operation may increase cancer cell dissemination.Simple ligation of peripheral veins around stomach could not completely prevent cancer cell dissemination in the peripheral blood.

The cancer stem cell hypothesis posits that cancer stem cells,which are rare among the tumor cell population and share many common properties with normal stem cells,and have been positively identified and successfully isolated from some cancers,are responsible for tumorigenesis and implicated in multistage cancer progression,particularly with respect to metastasis.A better understanding may provide insights into the development of cancer stem cell-targeting therapeutic strategies.The examines what is known regarding cancer stem cells' concept,comparison with normal stem cell,potential origins,assays for isolation and their roles in metastasis.
5.The Study on the Activity of Protein Kinase C in Platelets of Maternal Vein and Umbilical Blood in Pregnancy Induced Hypertension Syndrome Patients

Objective To investigate the relationship among the activity of protein kinase C (PKC) in platelets of maternal vein and umbilical blood , the pathophysiological changes of pregnancy induced hypertension (PIH) and fetal growth restriction (FGR) in PIH patients. Methods Activities of PKC in membrane and plasma of platelets from maternal vein and umbilical blood taken from 35 PIH patients and 20 normal pregnant women were measured with substrate phosphorylation method. Results No difference was shown in the PKC activities between the mild PIH patients and normal pregnant women in both maternal and cord blood.The PKC activities in moderate and severe PIH patients were significantly higher than those of the normal pregnant group.In normal pregnant women, the PKC activity in membrane and plasm of the platelets had no significant difference. 6.Proteomic analysis of alzheimer's disease Rats'serum

Objective Alzheimer's disease(AD) is a common and devastating disease and there is no readily available biomarker to aid diagnosis or to monitor disease progression. This study is to further understand the pathogenesis of Alzheimer's disease and seek new biomarkers for AD. Methods AD rats made by Aβ1-40 were assessed using the Morris water maze method. Sera of two groups were collected and albumin, IgG and sodium chloride were removed from the serum before concentration. Two-dimensional electrophoresis(2-DE) was performed on serum protein. Altered proteins were identified by matrix assisted laser desorption/ionization time of flight mass spectrometry(MALDI-TOF-MS). Results Compared with untreated samples, 10 altered proteins were found through 2-DE and all of them were down-regulated of which 2 were identified by MALDI-TOF-MS to be plasma retinol-binding protein precursor and complement component 4, gene 2 (C4b). Conclusion C4b may be the biomarker of AD and it may be helpful to understand the pathogenesis of AD.
7.Expression and Clinical Significance of Soluble E?cadherin before and after Operation in Breast Cancer Patients

Objective To detect the expression of serum soluble E?cadherin(sEC)before and after operation in breast cancer patients,and to ex?plore its clinical significance. Methods The levels of sEC in 42 patients with breast cancer and 18 patients with benign breast diseases were tested by enzyme?linked immunosorbent assay(ELISA)before and after operation as well as in 30 healthy controls. Its correlation with clinical and patho?logical characteristics was analyzed. Results The levels of sEC in breast cancer patients before operation were significantly higher than patients with benign breast diseases and controls,which declined significantly after operation. Moreover,the levels of sEC in those with high clinical grading and lymphatic metastasis were also higher than in the ones with low grading and negative lymphatic metastasis(P<0.05). Conclusion The levels of sEC in breast cancer patients before operation are specific and closely related with pathological grade and lymphatic metastasis,which may be a pre?dictive factor for prognosis of breast cancer.
8.Expression of phosphodiesterase 5 in clitoral corpus cavernosum and effect of icariin on cGMP levels in vitro

Objective To investigate the expression of phosphodiesterase 5 (PDE5) in clitoral corpus cavernosum and effect of icariin on cyclic GMP (cGMP) levels in vitro. Methods The presence of mRNA coding for PDE5 in clitoral corpus cavernosum was detected by means of RT PCR analysis.The effect of icariin with various concentrations on cGMP levels,which is the target of PDE5 in vivo,was measured by 125 I radioimmunoassay with sildenafil as positive control. EC 50 of icarrin and sildenafil was calculated. Results There was a lane around 800 bp on the agar electrophoresis of the production of RT PCR,which was as long as we designed according to cDNA sequence of PDE5.In the presence of SNP,icariin increased cGMP concentrations in rabbit clitoral corpus cavernosum with increasing dose. Conclusions The results support the proposal that icariin may influence cGMP level by NO cGMP signaling pathway as an inhibitor of PDE5.

10.Research on quality parameters of scutellariae radix (formula particles) using on-line NIR in pilot with different extraction process.

The on-line monitor for the changes in the content of baicalin in Scutellariae Radix formula particles during the extraction process was conducted by using near infrared spectroscopy (NIR). High performance liquid chromatography (HPLC) was used as a reference method. Kennard-Stone (KS) was used to divide sample sets, so as to compare different pretreatment methods. The synergy interval partial least squares (SiPLS) was used to screen out modeling wave band to establish partial least-squares models. The relative error method was applied to predict forecast set samples of Scutellariae Radix in three extraction phases. The results showed that the model established by Savitzky-Golay smoothing with 11 points (SG11 points) was the best, with the root mean square with cross validation (RMSECV), root mean square error of correction (RMSEC) and root mean square error of prediction (RMSEP) of baicalin of 0.092 7, 0.134 4 and 0.114 8, respectively, the determination coefficient R2 of greater than 0.99, and the relative error of baicalin content of less than 5%. This indicates that the on-line near infrared reflectance spectroscopy could be applied in on-line monitor and quality control of the extraction process of Scutellariae Radix formula particles.
