[Objective] To observe the effects of pine pollen on the changes of p16INK4a and p21WAF1 in 2BS cells.[Method]The 2BS cells were cultured from 28 population doubling(PD)and divided equally into the young control group,old control group,middle-dose pine pollen group(120mg/dl)and high-dose pine pollen group(240mg/dl)randomly.Regulate the phases of cell cycle by flow cytometry,and then the expression of p16INK4a,p21WAF1 mRNA were determined by semi-quantitative RT-PCR.[Result]The amount of cells in G1 phase was depressed in middle-dose pine pollen group(72.73%?1.76%)and high-dose pine pollen group(64.15%?0.87%)than in old control group(81.33%?3.43%)noticeablely(P

Objective To study the influence of siRNA targeting survivin on chemotherapy sensitivity of HCC cells.Methods siRNA eukaryotic expression vector was generated.After the vector stablely transfected into HepG2 cells,the effects of chemotherapy drugs on HCC cells were observed.Results The recombinant plasmid Psilence(+)-survivin was successfully constructed.Survivin mRNA expression inhibition ratio reached 73% detected by RT-PCR.MTT methods detected that siRNA treated HCC cells were affected by MCC,the survival rate of HepG2,HepG2/Silence(-) cells was inhibited only at 48 h(0.505?0.015) compared to control groups untreated with MCC(0.824?0.322)(P

Objective To study the influencing factors of pediatric in-hospital cardiopulmonary resuscitation(CPR).Methods It was a retrospective observational study.We studied a total of 281 children who suffered in-hospital cardiopulmonary arrest(CPA).The outcome was defined as sustained return of spon-taneous circulation ﹥20 min.Results A total of 281 patients met study entry criteria.In 129 children (45.9%),return of spontaneous circulation sustained ﹥20 min and 20 cases(8.8%)survived to hospital discharge.In the univariate analysis,gender,age,weight,time of CPA happened,first cardiac rhythm,pH, blood lactate had no obvious influence on the outcome.Underlying disease,the place of CPA,personnel fac-tors,airway support,the duration of CPR,doses of adrenaline,use of bicarbonate and blood glucose level were associated with outcome.Conclusion At present,the rate of successful CPR and discharge of hospital is still low.Respiratory disease has a higher survival rate.CPR ﹥20 min,use of bicarbonate and using adrenaline≥3 doses are associated with poor outcome and an increase in mortality.

Objective To investigate the clinical characteristics of endometrial carcinoma in young women,especially the normal weight women.Methods Seventy and five cases of endometrial carcinoma aged ≤ 40 years were treated in Obstetrics and Gynecology Hospital affiliated to Fudan University from January 2008 to December 2011.Among them,there are 68 cases of normal weight and obesity,who were divided into normal weight group (group A,32 cases) and obesity group(group B,36 cases) for comparisons and analysis.Results About 81.2％ of the patients were menstrual disorders in group A and 83.3％ in group B(P =0.822);The merge infertility accounted for 31.2％ and 19.4％ (P =0.279) respectively,and the oligomenorrhea accounted for 21.9％ and 30.6％ (P =0.418) respectively,the patients combined diabetes,high blood pressure or PCOS were 6.2％ and 44.4％ (P =0.001) respectively.According to International Federation of Gynecology and Ostetrics(FIGO) surgery-pathologic stage,two groups of stage Ⅰ were 68.8％ and 83.3％ (P =0.157),respectively.Stage Ⅱ-Ⅳ were 31.2％ and 16.7％ (P =0.157),respectively.Pathological type:8 (25.0％) cases were high-risk of pathology classification in group A,and Five cases (13.8％) in group B,others were endometrial adenocarcinoma.Operations were the main treatments.Five cases were progestogen therapied.After treatments,2 cases were full term pregnant,1 case is now pregnant.Conclusion The clinical manifestations of endometrial carcinoma in young women were mainly menstrual disorders.There were higher percentage infertilities and lower percentage diabetes,high blood pressure and PCOS in normal body weight patients.In normal body weight patients,pathology classification for middle-late of high-risk pattern was increasing.For patients in early stage,if have birth requirement,could be treated with the trial progesterone conservative.

Serum tumor biomarkers are significant for the early diagnosis and treatment,preoperative evaluation,prognosis prediction and relapse surveillance of endometrial carcinoma.Recently,serum CA125 and human epididymal secretory protein 4 are widely recognized as closely related with staging,metastasis and prognosis of endometrial cancer.In addition,the serum miRNAs are characterized by their features of high stability and specificity.Omics studies will be helpful for the general assessment of the initiation and progression of endometrial cancer.Insights into these two aspects will facilitate the identifications of new biomarkers with higher sensitivity and specificity for endometrial carcinoma.

Aberrant microRNA(miRNA) expression has been found in most of human cancers.miRNA-related regulation pathways,including the interaction between miRNA and protein-coding gene and the interaction between miRNA and IncRNA,have been shown correlated with the initiation and progression of human cancers.miRNA might be a therapeutic target in the treatment of malignancies.

Ezrin protein plays an important role in morphogenesis,motility,adhesion.Moreover,Ezrin protein participates in multiple steps of the process of tumor invasion and metastasis.In the progress of tumor metastasis,Ezrin protein is related to several signaling pathways such as Rho mediated signaling pathways,hepatocyte growth factor (HGF) pathways,Fas mediated cell apoptosis signaling pathways,phosphatidyl inositol 3-kinase (PI3K)-Akt pathways,and in which it plays different regulatory role.

MircoRNAs (miRNAs) are widely existing inorganisms, non-coding small RNA. MircoR-NAs play important role in cell proliferation, apoptosis, stress resistance and fat metabolism. In vitro studies indicate that miRNAs are important factors in platinum resistance of ovarian cancer cell lines. However, the pathway of platinum resistance of ovarian cancers is complex. Studies find miRNA expression play important roles in the sensitivity of platinum resistance of ovarian cancer cell lines , and can be used as an early alert for platinum resistance. Changing the expression of miRNA even can reverse the platinum resistance . But the regular role of miRNAs in target genes network is still unconfirmed. Thus, more studies about miRNA in vivo and animal tumor models are needed to identify the function of miRNAs in platinum resistance of ovarian cancer.

Experiments were carried on cultured neonatal myocardial cells of rat to study the effects of angiotensin Ⅰ(AngⅠ) and Ⅱ (AngⅡ) on the synthesis of DNA, RNA, protein and on the cardiac hypertrophy. It was shown that the incorporation of both ~3H-TdR and ~(14)C-uR increased over a set of time periods after addition of AngⅠ and AngⅡ to the medium. The mean protein content of the single myocardial cell was also enhanced, and the volume of myocardial cell was increased significantly after a continuous stimulation of AngⅠ and AngⅡ for 7 days. No stimulation effect was observed either in the synthesis of DNA, RNA, and protein or in the volume of myocardial cells after addition of AngⅠ together with the angiotensin converting enzyme inhibitor(ACEI, captopril). These results indicated that: (1). AngⅠ acts as a regulator by its indirect stimulation effect on the synthesis of DNA, RNA and protein of myocardial cells, since it must be changed into AngⅡ by the ACE present in myocardial cells. The ACE can be inhibited by the ACEI: (2) The hypertrophy of cultured neonatal myocardial cells may induced by a continuous stimulation of AngⅠ and AngⅡ.
