1.Treatment of Graves Hyperthyroidism by Jiakangling Capsule Combined with Reduction of 131I: an Efficacy Observation.

OBJECTIVETo observe the clinical efficacy of Jiakangling Capsule (JC) combined with reduction of 1311 in treatment of Graves hyperthyroidism.
METHODSTotally 387 Graves hyperthyroidism patients were randomly assigned to the treatment group (200 cases) and the control group (187 cases). Patients in the treatment group took JC combined with reduction of 131I. The 131I dosage per gram of thyroid tissue was 50-80 microCi. They additionally took JC one week after taking 1311 for one consecutive month. Patients in the control group took 131 routinely as one disposable treatment. The 131I dosage per gram of thyroid tissue was 70-120 microCi, without using JC or other anti-thyroid drugs. All patients were reexamined after 24-month treatment. Whether hyperthyroidism was cured, incurred, or permanent was observed. Efficacies of thyroglobulin antibody (TGAb) and thyroid microsome antibody (TMAb) were compared between the two groups.
RESULTSCompared with the control group, the incurred ratio increased in the treatment group [3.2% (6/187) vs. 16.0% (32/200), P < 0.01], the incurred ratio of strong positive TGAb and TMAb patients increased [3.5% (2/57) vs. 27.1% (16/59), P < 0.01], the permanent hypothyroidism ratio decreased [21.1% (12/57) vs. 3.4% (2/59), P < 0.05 ].
CONCLUSIONJC combined with reduction of 1311 was superior in treating Graves hyperthyroidism induced permanent hypothyroidism than routine 1311 treatment, especially for strong positive TGAb and TMAb patients.

4.RNF213 gene polymorphisms and susceptibility of intracranial vascular stenosis disease: a meta-analysis

Objective To investigate the correlation between RNF213 gene polymorphisms (rs112735431,rs138130613,and rs148731719) and the susceptibility of intracranial vascular stenosis disease.Methods The literature of studies on the correlation between RNF213 gene polymorphisms and intracranial vascular stenosis disease was collected according to the related databases.Using the Stata 12.0 software and selecting suitable genetic model,the heterogeneity was analyzed and the pooled odds ratio (OR) and its 95% confidence interval (CI) were calculated.Results A total of 12 articles were included after screening.The result of meta-analysis showed that the rs112735431 polymorphism had a significant correlation with the susceptibility of moyamoya disease (MMD) in all genetic models,especially the most significant dominant model (AA + GA genotype vs.GG genotype:OR 101.46,95% CI 59.41-173.27;P <0.001),at the same time,the polymorphism of this site also had significant correlation with the nonMMD intracranial large artery stenosis/occlusion (AA + GA genotype vs.GG genotype:OR 13.82,95% CI 4.48-42.61;P< 0.001);the rs138130613 polymorphism had significant correlation with the susceptibility of MMD in Chinese population (OR 5.01,95% CI 1.57-15.98;P=0.006);and no correlation between the rs148731719 polymorphism and the susceptibility of MMD was observed.Conclusions The RNF213 gene rs112735431 polymorphism is a susceptible factor of MMD,at the same time,the polymorphism of this site is also associated with the formation of non-MMD intracranial large artery stenosis.Systematic study on the molecular function of RNF213 may have important significance for diagnosis and treatment of such vascular stenosis diseases.
5.The construction of adenovirus vector carrying human tissue inhibitor of metalloproteinase-2 gene

Objective To construct an adenoviral vector carrying human tissue inhibitor of metalloproteinase-2 (TIMP-2) gene for gene therapy. Methods A recombinant adenovirus (AdhTIMP-2) containing human TIMP-2 cDNA fragment was generated by homologous recombination in BJ5183 bacteria. Recombinant plasmids were screened by alteration of antibiotic. The adenovirus vector was then packaged and amplified in 293 cells. The expression of TIMP-2 was detected by the techniques of Western blot and RT-PCR. Results The recombinant adenoviral vector carrying human TIMP-2 was constructed. The titer was 4?10 11pfu/ml after purification. The expression of TIMP-2 gene in 293 cells was detected by RT-PCR. After the 293 cells were transfected with AdhTIMP-2 24 hours, TIMP-2 protein could be detected in the medium by Western blot. Conclusions The recombinant adenoviral vector carrying human TIMP-2 is successfully constructed and paved the way for further application in vascular disease gene therapy.
6.Upregulatory effect of 17?-estradiol on estrogen receptor?expression in MG63 cells and human osteoblast-like cells

The effect of 17?-estradiol ( 17?-E2 ) on expression of estrogen receptor ?( ER?) was observed in cultured MC63 cells and human osteoblast-like (HOB) cells. The results show that the expression of ER* is in a dose-dependent manner with 17p-E2 (0-1?10-6 mol/L) in MC63 cells and has an optimal 17?-E2 concentration (1?10-8 mol/L) in HOB cells resulting in maximum expression of ERp protein.
7.Study on the construction of standard D12S375 allelic ladder and its genetic polymorphism in Chinese Han, Hui and Wei populations.

s:To resolve the problem of the accuracy and standardization of STR PCR typing in forensic science practice,we have designed a new method to produce standard D12S375 allelic ladder.Seven different PCR amplified D12S375 allelic fragments were isolated from the gel,eluted into the distilled water and reamplified by PCR.The purified allelic fragments were then blunt end subcloned individually into the pUC plasmid vectors and transfected into competent E.coli DH5? TM cells.The sequencing results confirmed that the size and the constructure of the inserts were correct.The recombinant plasmids DNA with 7 inserts were then used as templates for reamplification to generate D12S375 standard ladder, with which the genetic polymorphisms of D12S375 locus in Chinese Han population in Chengdu,Hui population in Gansu and Wei population in Xinjiang were studied.
9.Interventive effect of Angongniuhuang injection on inflammatory response after brain injury in rats

Objective To observe the influence of Angongniuhuang injection on the level of in- terleukin-1?(IL-1?),intercellular adhesion molecule-1 (ICAM-1),serum protein S100B and neuron specific enolase (NSE) after brain injury to explore its protective effect on the injured brain tissues. Methods Brain contusion model was made in rats by Feeney's method.Then,the levels of IL-1?and ICAM-1 in the brain tissues and the levels of serum protein S100B and NSE in serum were measured by ELISA method at different time points.Results The level of IL-1?and ICAM-1 in brain tissues and that of S100B protein and NSE in serum in treatment group were lower than that in control group 6-48 hours after injury (P<0.05).Conclusion Angongniuhuang injection can alleviate inflammatory re- sponse after brain injury and protect effectively brain tissues.
10.CT-Guided Biopsy of the Retroperitoneal Lymph Nodes

Objective To discuss the value of CT guided biopsy of the retroperitoneal lymph nodes for diagnosis.MethodsTotally 18 patients with enlarged retroperitoneal lymph nodes underwent CT-guided biopsy in our hospital between June 2006 and June 2007.Results The results of CT were identified by the pathological examination in 94.4%(17/18) of the patients.The operation time of the biopsy was 20 to 40 minutes.CT scan was performed for 3 to 6 times(mean,4.1) during the procedure.No severe complications occurred in this series.Conclusion CT-guided biopsy of enlarged retroperitoneal lymph nodes is a safe and effective diagnostic method.
