3.Micro-sphere culture gathers cancer stem cell and its potential clinical application

Cancer stem cell hypothesis has become a new theory. According to this theory, cancer stem cells play a decisive role in car-cinogenesis, progression, and metastasis. This theory offered new ideas and strategies for cancer treatment. However, only a small fraction of cells in a tumor were cancer stem cells. Therefore, the identification and enrichment of these cells are significant challenges for numerous researchers conducting further study. Micro-sphere culture has been an effective tumor stem cell enrichment technolo-gy in recent years. The application of this technique makes the cancer stem cell research process easy. In addition, it makes the evalua-tion of tumor progression and prognosis of patients in clinical possible. This article will focus on the applications of this technology, and microsystem integration will be described in detail.
4.Study on theater nurse learning system based on PDCA theory

Objective To develop an informatized continuous education system for theater nurse based on PDCA theory.Methods The system was gifted with the functions of learning planning, learning execution, learning checking, data management and system management based on PDCA theory, informatized technology and other learning systems.Results The system behaved well in overall flow and configuration, man-machine interaction and application, which facilitated the nurses to learn with fragmented time.Conclusion The nursing continuous education is enhanced greatly with the system.
5.Effects of Fu Shen ke li on myocardial cell apoptosis and endoplasmic reticulum stress-related protein expressions in rats with adriamycin induced heart failure

Objective To explore the contribution of endoplasmic reticulum stress to cardiac myocyte apoptosis in mouse congestive heart failure induced by Doxorubicin.Methods Doxorubicin (DOX)was administered intraperitoneally to rats,2 mg/kg doses over 8 weeks.Echocardiographic and hemodynamics measurements were obtained at 8 weeks after treatment.Then left ventricular (LV) samples were collected.The successfully established rats were divided into 3 groups (Fu Shen Ke Li 40 mg/kg,60 mg/kg,and 80 mg/kg).After 5 weeks post the operation,an echocardiographic was performed on all the rats under narcotism,Western blot to test the B-cell lymphoma-2 (Bcl-2),myeloid cell leukemia-1 (Mcl-1),Bcl-2-Associated X (Bax),Caspase-3,C/EBP homologous protein (CHOP),and 78-kDa glucose-regulated protein(GRP78),and reverse transcription-polymerase chain reaction (RT-PCR) to test the mRNA expression levels of Mcl-1,and NOXA.Results The left ventricular end-diastolic diameter (LVEDD),left ventricular end systolic diameter (LVESD),left ventricular ejection fraction (LVEF) prompted modeling success The left ventricular end-diastolic diameter (LVEDD),left ventricular ejection fraction (LVEF),and fractional shortening (FS) have been improved in high and middle groups of Fu Shen Ke Li.Compared to the model group,higher expression levels of Bcl-2,and Mcl-1 were,lower expressions of Bax,NOXA,CHOP,and GRP78 were (P < O.05) in 3 groups of Fu Shen Ke Li.Compared to the model group,there were higher mRNA expression level of Mcl-1 was,lower mRNA expression of NOXA was (P < 0.05) in 3 groups of Fu Shen Ke Li.Conclusions Fu Shen Ke Li depresses endoplasmic reticulum stress,which can reduce the apoptosis of myocardial cells,and ultimately achieve the purpose of treatment of heart failure.
6.ROLE OF ENDOTHELIUM DYSFUNCTION IN EXPERIMENTAL VASOSPASM

To investigate the relationship between endothelium dysfunction and experimental vasospasm, twenty-eight New Zealand white rabbits were randomly allocated into three groups: sham operation control group(Group 1, n=8), balloon endothelial denudation + normal diet group (Group 2, n=8) and balloon endothelial denudation + hypercholesterol diet group (Group 3, n=12). Angiography was performed to detect the vasospasm induced by ergonovine and endothelium-dependent vasodilator response induced by acetylcholine before and immediately after denudation and 8 week later. Visible vasospasm was induced at the denuded sites in group 3, and endothelium dysfunction was also found in the same locations. Positive correlation was showed between vasospasm and endothelium dysfunction. Endothelium dysfunction resulted from balloon endothelial denudation and hypercholesterolemia may play an important role in the pathogenesis of experimental vasospasm.
7.Pay attention to digestive tract reconstruction after curative resection of gastric cancer.

Several reconstruction techniques are available after gastrectomy. Roux-en-Y reconstruction following distal gastrectomy is the reference in terms of long-term functional and endoscopic outcomes. It is the preferred reconstruction for benign lesions and early gastric cancer. In patients with advanced gastric cancer, BillrothII (reconstruction is an acceptable alternative. After total gastrectomy, Roux-en-Y reconstruction is the simplest solution, with satisfactory functional outcome. Addition of a jejunal reservoir seems to improve long-term outcome after total gastrectomy and could be of benefit to patients with good prognosis. After distal or total gastrectomy, hand-sewn anastomoses should be preferred because of lower costs. Mechanical sutures can facilitate transhiatal esophagojejunostomy. After proximal gastrectomy, esophago-gastric anastomosis is the basic reconstruction method. Gastric remnant is made into gastric tube in the operation. The effect of pyloroplasty remains controversial, and further study is needed to improve the quality of life after operation.

9.Isolation and biological characteristics of umbilical cord mesenchymal stem cells

BACKGROUND:Cord tissues,which immune system is relatively non-development with low expression of MHC,rely on the protection of the mother's immune system.Accordingly,studies underlying biological characteristics of umbilical cord mesenchymal stem cells (MSCs) arouse more and more attention.OBJECTIVE:To isolate,separate,and culture MSCs from the tissues of umbilical cord,and to explore the ability of multi-directional differentiation.DESIGN,TIME AND SETTING:The in vitro cytology experiment was performed at the Laboratory of Tissue Engineering of Sichuan University from October 2006 to May 2007.MATERIALS:The umbilical cord of healthy neonate with 37-40 weeks gestational age was obtained from delivery room of the Second West China Hospital,Sichuan University.METHODS:Pieces of umbilical cord were dissected along its long axis,vessel was pulled away,then cord was tied together with a suture creating "loops",then it was placed into collagenase solution,after 6-8 hours,suspended cells obtained from the loops suspension were centrifuged and cultured with adherent method,pass,aged when single cell-derived colonies were appeared.Finally,the 5th generation of cells was induced differentiation towards osteoblasts and lipoblasts.MAIN OUTCOME MEASURES:Morphology,growth,surface antigen expression,as well as the multiple differentiation potential of umbilical cord MSCs.RESULTS:The adherent cells obtained from umbilical cord after removal vessels showed short rod-like or fusiform shape,which can be proliferated and form single cell-derived colonies.Cells isolated from the umbilical cord high expressed integrin markers (CD29,CD51,CD71) but not hematopoietic lineage markers (CD34,CD45 and HLA-DR).These colonies can differentiate into osteoblasts that produce mineralized matrices,stained by alizarin red and alkaline phosphatase,and differentiate adipocytes that accumulate lipid vacuoles and are demonstrated by morphology,stained by oil red.CONCLUSION:Umbilical cord tissue contains a high number of MSC-like elements forming colonies that may be successfully expanded in culture.The proliferation colonies present matrix ceil immunophenotypes,and can differentiate into osteobiasts and adipocytes.
10.Effect of ferulic acid on cholesterol efflux in macrophage foam cell formation and potential mechanism.

The formation of macrophage-derived foam cells is a typical feature of atherosclerosis (AS). Reverse cholesterol efflux (RCT) is one of important factors for the formation of macrophage foam cells. In this study, macrophage form cells were induced by oxidized low density lipoprotein (ox-LDL) and then treated with different concentrations of ferulic acid, so as to observe the effect of ferulic acid on the intracellular lipid metabolism in the ox-LDL-induced macrophage foam cell formation, the cholesterol efflux and the mRNA expression and protein levels of ATP binding cassette transporter A1 (ABCA1) and ATP binding cassette transporter G1 (ABCG1) that mediate cholesterol efflux, and discuss the potential mechanism of ferulic acid in resisting AS. According to the findings, compared with the control group, the ox-LDL-treated group showed significant increase in intracellular lipid content, especially for the cholesterol content; whereas the intracellular lipid accumulation markedly decreased, after the treatment with ferulic acid. The data also demonstrated that the mRNA and protein expressions of ABCA1 and ABCG1 significantly increased after macrophage foam cells were treated with different concentrations of ferulic acid. In summary, ferulic acid may show the anti-atherosclerosis effect by increasing the surface ABCA1 and ABCG1 expressions of macrophage form cells and promoting cholesterol efflux.
