1.Comparative Study of Efficacy of Moxifloxaction and Levofloxaction in the Treatment of the Lower Respiratory Tract Infection

Objective:To evaluate the efficacy and safety of moxifloxaction and levofloxaction in the treatment of the lower respiratory tract inflection.Method:64 cases patients were randomly divided into two groups.The clinical trial group was treated with moxifloxaction 400mg once a day,orally for 7-10 days and the controlled group treated with levo- floxaction in 200mg twice a day for 7-10 days.Result:The effective rate of the moxifloxaction group(93.9%)was high- er than that of the levofloxaction group(71.8%),but not significantly.The recovery rate of the moxifloxaction group (71.8%)was significantly higher than that of the levofloxaction group(46.9%);the days of recovery of clinical symptoms were not significantly different in the two groups.There was no significant difference between the two groups in the bacterial eradication rate.Conclusion:Moxifloxaction is effective and safe in the treatment of the lower respiratory tract inflection.
2.Antiasthmatic action of ephedrine on guinea pig by transdermal administration in vivo

AIM:To study antiasthmatic action of ephedrine on guinea pig by transdermal administration in vivo. METHODS:The gelatinizer made from ephedrine was taken as the treatment to carry research for antiasthmatic effect and compared the difference between therapeatic and control drug. RESULTS:The ephedrine gelatinizet could prolonge the latent period and shock stage of asthmatic attack in sensitized guinea pigs. CONCLUSION:The results indicate that transdermal administration of ephedrine gelatinizer is reasonable
3.Pharmacokinetics of ephedrine gels and extractum ephedrae gels in guinea pig

AIM: To establish a method to determine the serum concentration of ephedrine,and to study its pharmacokinefics in guinea pig. METHODS: The serum concentration of ephedrine gels transdermal administration in guinea pig were determined by HPLC and pharmacokinetic parameters were calculated by computer.(RESULTS:)The calibration curve was linear in the range of 0.5~2 ?g with r=0.998 0.The limit of identification was 0.02 ?g/mL.The pharmacokinetics behavior of ephedrine showed one order absorption.The main parameters after adminstrition of ephedrine gels and extractum ephedrae gels were as follows:t_(1/2)k_a were 1.362 1 and(l.541 8),t_(1/2)k_e were 2.627 4 and 3.262 2 h,T_(max) were 3.124 3 and 3.440 9 h,C_(max) were 0.115 6 and 0.081 0 ?g/mL,AUC were(0.081 0) and 0.741 5 ?g?h/mL. CONCLUSION: Ephedrine shows a fast rate of absorption in guinea pig,serum concentration of ephedrine is stable and lasting potency,and indicates the reason of transdennal administration of Herba Ephedrae preparation.
4.EFFECTS OF FU-TZE "801" ON SURVIVAL AND MICROCIRCU- LATORY CHANGES IN BURN SHOCK RATS

The antishock properties of Fu-tze "801" , an active fraction isolated from the root of Aconite,were studied on burned rats. The rats were given 65-70% TB'SA second degree scalds. All animals died in a short time postburn. Using this rat model, it was found that the marked spasmodic rhythmic constriction in arterioles occurred and the microierculatory flow state showed pronounced derangements in the me-senteric microcirculation, and platelet aggregate increased in circulating blood.After treatment with Fu-tze "801" , the survival and the mirocirculation were improved in the scalded rats, these findings suggest that Fu-tze "801" affords the antishock action on rats after a major scald burn.
5.Study on transdermal permeability of ephedra extract in vitro

AIM: To study the transdermal permeability of ephedra extract. METHODS: With Valia Chien diffusion cell to the transdermal experiments are used to observe the effect of different factors on the transdermal permeability. RESULTS: Its transdermal resorption constant increased with the increase of the concentration of ephedrine and ephedra extract. pH value of the diffusioin medium could influence ephedrine permeability. CONCLUSION: The result can offer reference in preparation for Ephera (targeting drug delivery system) (TDS).
6.Study on the correlation between intercellular adhesion molecule-1 and lung cancer

Objective To explore the role of intercellular adhesion molecule-1(ICAM-1) in the metastasis of lung cancer.Methods The expression of ICAM-1 in the tissue and the serum ICAM-1(sICAM-1) level in the 52 cases of primary lung cancer were detected with immunohistochemical staining method(SP) and enzyme linked immunosorbent assay(ELISA),with 20 cases of adjacent non-cancerous tissue as the control.Results The positive rate of ICAM-1 expression in lung cancer was significantly higher than that of adjacent non-cancerous tissue.The expression of ICAM-1 in the cases with lymph node metastasis was higher than that of no lymph node metastasis.The expression of ICAM-1 in stage III+IV was higher than that of stage I+II,and that in adenocarcinoma was higher than that of squamous cell carcinoma.The level of sICAM-1 in patients with lymph node metastasis was higher than that without lymph node metastasis.The sICAM-1 level in patients of adenocarcinoma was higher than that of squamous cell carcinoma,and that in patients with positive expression of ICAM-1 in tissue was higher than that of negative expression of ICAM-1.Conclusion The expression of ICAM-1 may be associated with the metastasis in lung cancer.The sICAM-1 level reflects the expression of ICAM-1 in tissue and may be used as a tumor marker of metastasis and prognosis in lung cancer.
7.A mini-plate fixation for the treatment of children's ulna and radius distal unstable fractures: A 18-case analysis

OBJECTIVE: To observe the clinical effect of mini-plate fixation on the treatment of children unstable fractures of the distal ulna and radius. METHODS: A total of 18 children with unstable fracture of the distal ulna and radius (15 boys and 3 girls, aging 4.1-13.3 years, with the mean age of 9.8 years) were selected from Department of Orthopaedics, Wuhan Children and Women's Hospital, Tongji Medical College, Huazhong University of Science and Technology from August 2006 to September 2009. The fracture occurred on right side (n=9) and left side (n=9). All children were treated with mini-plate fixation which was instead of the traditional pinning fixation. Before operation, stainless steel or titanium plates of adult digital bone, metacarpal bone, and metatarsal bone were chosen according to fracture types and sites. The distance from fracture line to epiphyseal plate was measured to get ready the T-, L-, clover or straight plates. Joint function, degree of forearm rotation loss, bone healing, and complications were monitored postoperatively and within following up. RESULTS: All cases were followed up in out-patient for an average 26.4 months. An average 10°limitation was measured on forearm pronation function of all children, but the function was recovered by doing exercises which did not influence daily activities. Supination, flexion and extension of wrist joint were generally normal, and the fracture was generally healed. Complications including re-displacement, refracture, malunion, delayed union, bone bridge formation, vascular and nerve injury were not found. CONCLUSION: Patients who were not satisfied with the reduction, re-displacement, or old distal radius fracture needed an open reduction. Micro-plate was fixed to avoid epiphyseal injury of distal forearm and reduce postoperative complications. An ideal mini-plate may replace the traditional pinning fixation.
8.Diagnostic value of high frequency ultrasound on lesions of knee traumas

Objective To discuss the diagnostic value of high frequency ultrasound on lesions of knee traumas.Methods Ninty-eight cases with post-traumatic knee injuries were examined by high frequency ultrasound,and the characteristics of changing ultrasonogram were observed and comparing with surgical results and MR.Results Of the 98 cases with post-traumatic knee injuries,there were 57 cases with effusion and hematocele in knee joint,among which there were 41 cases with collateral ligaments and meniscus injuries,the diagnostic coincidence rate was 93.3% in tendon tear and complete tendon fracture(compared with MR),91.7% in partial and complete muscle tear(compared with MR),67.7% and 71.4% in collateral ligaments and meniscus injuries(compared with surgical results).Conclusions High frequency ultrasound can be used as a conventional imaging examination method for post-traumatic knee injuries.
9.Protective effect of flos puerariae flavonoid on adriamycin-induced toxic myocarditis

Objective To investigate the protective effect of flos puerariae flavonoid on adriamycin (ADR)-induced toxic myocarditis and its mechanisms from morphological,biochemical and molecular levels.Methods 96 healthy Kunming male mice were randomly divided into 6 groups:normal control group,ADR model control group,ADR+low dose of flos puerariae flavonoid group(50 mg/kg),ADR +middle dose of flos puerariae flavonoid group(100 mg/kg),ADR +high dose of flos puerariae flavonoid group(200 mg/kg),and Vit E positive control group(40 mg/kg),16 in each group.The drugs were orally administered for consecutive 15 d and the model of toxic myocarditis was induced by intraperitoneal injection of ADR(3 mg/kg)in mice from day 2,one time every other day,for 7 times Colorimetry was used to measure the changes of marker enzymes about myocardial injury and inducible nitric oxide synthase(iNOS)activity in serum and tissue;immunohistochemical method was adopted to detecte the expression of myocardial apoptosis related proteins Bax and bcl-2;HE staining was conducted to observe the pathological changes of cardiac structure.Results Compared with normal control group,ADR(3 mg/kg,ip,7 times)induced the elevation of serum creatine kinase (CK),lactate dehydrogenase (LDH),aspartate transaminase(GOT)and iNOS activity increased significantly in mice(P<0.01).Meanwhile myocardial superoxide dismutase(SOD)activity decreased, and the malondialdehyde(MDA)content increased(P<0.01).Myocardial cell apoptosis in mice increased significantly,and the apoptosis rate was(40.5 ± 5.2)%;the expressions of Bax and Bcl-2 were significantly increased(P<0.01),However,the Bcl-2/Bax ratio decreased.The flos puerariae flavonoid (50,100,200 mg/kg,ig,15 d)and Vit E positive control group could reverse the changes induced by ADR,decrease serum CK,LDH,GOT and iNOS activities,increased myocardial SOD activity,lower MDA content and the expression of bax protein,and elevated Bcl-2/Bax ratio,in a dose-dependent manner.Light microscopy confirmed that flos puerariae flavonoid significantly alleviated the changes of myocardial microstructure.Conclusion ADR could induce myocardial cell apoptosis and lead toxic myocarditis in experimental mice.The flos puerariae flavonoid has protective effect on ADR-induced myocardial injury and the mechanism may be related to elevating myocardial SOD activity and anti-lipid peroxidation,inhibiting the expression of Bax protein and adriamycin-induced cardiomyocyte apoptosis.
10.Relationship between hemolymph phenol oxidase and melanization of Plasmodium yoelii oocysts in Anopheles dirus

Objective To explore the relationship between hemolymph phenol oxidase and the melanization of Plasmodium yoelii oocysts in Anopheles dirus. Methods An Anopheles dirus-Plasmodium yoelii system was used Anopheles dirus were divided into 3 groups, that is, non-blood-fedding (N), normal-blood-fedding (B) and infected-blood-fedding (I). The activities of MPO and o-DPO in hemolymph from 3 groups were determined with native polyacrylamide gel electrophoresis (PAGE) and density scanning at 5, 7, 11 and 15 d after blood feeding. Results Both MPO and o-DPO activity were significantly higher in group I than group N and B (P<0.05). But with the melanization of Plasmodium yoelii oocysts, both MPO and o-DPO activity in group I were decreased in comparison with group N, especially on the 15 th day after infected-blood feeding. MPO and o-DPO activity in group B were significantly stronger than those of group N. Conclusion Blood feeding and infection of Plasmodium yoelii both can activate the cascade. The heamolymph phenol oxidase may play an important role in the melanization of Plasmodium yoelii oocysts in Anopheles dirus.
