1.Pharmacokinetics of ephedrine gels and extractum ephedrae gels in guinea pig

AIM: To establish a method to determine the serum concentration of ephedrine,and to study its pharmacokinefics in guinea pig. METHODS: The serum concentration of ephedrine gels transdermal administration in guinea pig were determined by HPLC and pharmacokinetic parameters were calculated by computer.(RESULTS:)The calibration curve was linear in the range of 0.5~2 ?g with r=0.998 0.The limit of identification was 0.02 ?g/mL.The pharmacokinetics behavior of ephedrine showed one order absorption.The main parameters after adminstrition of ephedrine gels and extractum ephedrae gels were as follows:t_(1/2)k_a were 1.362 1 and(l.541 8),t_(1/2)k_e were 2.627 4 and 3.262 2 h,T_(max) were 3.124 3 and 3.440 9 h,C_(max) were 0.115 6 and 0.081 0 ?g/mL,AUC were(0.081 0) and 0.741 5 ?g?h/mL. CONCLUSION: Ephedrine shows a fast rate of absorption in guinea pig,serum concentration of ephedrine is stable and lasting potency,and indicates the reason of transdennal administration of Herba Ephedrae preparation.
2.Establishment and evaluation of DAS-ELISA for detecting bovine respiratory syncytial virus

In order to develop a double antibody sandwich assay (DAS-ELISA) for detecting bovine respiratory syncytial virus (BRSV),New Zealand white rabbits and BALB/c mice were immunized with the purified G protein as an antigen to prepare anti-G protein polyclonal and monoclonal antibodies.The antibody concentration and reaction conditions of DAS-ELISA were optimized by square titration,and its sensitivity,specificity,and coincidence rate were validated.Five hybridoma were stably secreting Mab which subclass belonged to IgG1κ.Western blot and IFA test showed that PcAb and Mab could react specifically with G protein and BRSV.The PcAb and Mab as the capture antibody and detection antibody respectively,and their optimal working concentrations were determined to be 2.5 μg/mL and 10μg/mL,the critical value 0.22 and the detection limit of 1.43 μg/mL,batch,inter-assay coefficient of variation less than 10 %.The DAS-ELISA had no cross-reaction with several pathogens which often caused bovine respiratory disease.When 45 nasal swabs of clinical samples were simultaneously detected by the DAS-ELISA and RT-PCR,the sensitivity,specificity and coincidence rate were 92.0 %,100 %,95.6 %,respectively.It' s indicated that the established DAS-ELISA detection method can be used to detect a large number of clinical samples.It was the foundation of monitoring and quick diagnosis for BRSV.
3.Records and reflections of teaching assistant's participation in the teaching of authentication of TCM

Teaching assistant was introduced to solve the outstanding issues in the teaching of authentication of TCM, such as irrationality for teaching subjects, separated theory and practice, incomplete evaluation system. With the help of teaching assistant, the new generation was able to play the advantages on Web-based course and WeChat subscription, and the students are encouraged to apply new media flexibly. Practice teaching was introduced into class to increase the combination of theory and practice. Teaching assistant played an important role in evaluation mechanism standardization and specialization, and forming the basis for formative evaluation. Updating the teaching mode, forming the strategy of autonomous learning and interactive teaching, we are hoping to improve the teaching quality of authentication of TCM, improve students' interest in specialized courses, and to provide reference for better teaching assistants' working and selection.
4.Antiasthmatic action of ephedrine on guinea pig by transdermal administration in vivo

AIM:To study antiasthmatic action of ephedrine on guinea pig by transdermal administration in vivo. METHODS:The gelatinizer made from ephedrine was taken as the treatment to carry research for antiasthmatic effect and compared the difference between therapeatic and control drug. RESULTS:The ephedrine gelatinizet could prolonge the latent period and shock stage of asthmatic attack in sensitized guinea pigs. CONCLUSION:The results indicate that transdermal administration of ephedrine gelatinizer is reasonable
5.Discussion on Factors Influencing Quality of Coronary Angiography with Dual Source CT

Purpose To study the factors influencing quality of coronary angiography with dual source CT. Methods 50 cases undergoing examination on coronary artery with dual source CT are involved. The images are reviewed retrospectively. Results Satisfying images are obtained after three -dimensional process in 47 of all. Conclusion It is important for accurate diagnosis to summarize the influencing factors and improve the quality of images of coronary angiography with dual source CT.
6.The Quality Control of Leukodepleted Red Cell

Objective To explore the quality control method of leucodepleted red cell concentrate (LDRC) preparation ,to choice an optimal processing , in order to raise the quality of LDRC,so as to ensure the efficiency and safty for the preparation application.Methods 62 bag red cells concentrates suspensin(each 150ml,preparation from 200ml whole blood) were investigated.By linear regression multianalyzed the effects of some factors on efficiency for removal of leukocytes from red cell concentrates.Free hemoglobin value(n=8) was detected before and after leukodepleted filtration.1% samples were tested and bacterial cultured,and observed the clinical adverse effects after LDRC tranfusion.Results ⑴The plasma and platelet in red cell contentrate before filtration,the time of filtration had influence significantly on the result of leukocyte depletion.⑵The percentage of leukodepleted red cell in which residual leukocyte less than 5?10 6/unit and 1?10 6/unit were 100% and 46.9%. The percentage of leukodepleted red cell in which the leukocyte removal efficiency more than 2log(99%)and 3log(99.9%) were 100% and 56.1% .After controlling the time of filtration,the percentage of leukodepleted red cell in which residual leukocyte less than 1?10 6/unit was up from 46.9% to 90.91% and the percentage of leukodepleted red cell in which the leukocyte removal efficiency more than 3log(99.9%)/bag was up from 56.1% to 78.1%.⑶Red blood cell recovery rate was (90.48?4.73)%. There was no significant change of free hemoglublin before and after filtration.⑷No bacteria were detected by germiculture.⑸Evident side effects was no seen after transfusion.Conclusions During the course of removal of leukocytes from red cell concentrates by filtration, the sustained quality control is necessary for improving the quality of leukodepleted red cell.So enforcing aseptic manipulation rigorously and controlling the components of blood,the time of blood storage and filtration appropriately are quite important.
7.EVALUATION OF PRESTORAGE LEUCODEPLETION IN MASSIVE BLOOD TRANSFUSION

The patients received massive blood transfusion in General Hospital of the Chinese Pepole's Liberation Army between 1999to 2001were reviewed. The following items of leucodepleted blood transfusion (LDBT)group and routing blood transfusion(RBT) group at pretransfusion, the first day, third day, and seventh day after blood transfusion were recorded and compared: glutamic pyruvic transaminase(GPT), glutamic oxaloacetic transaminase(GOT), lactate dehydrogenase(LDH), total bilirubin(TB), direct bilirubin(DB), alkalescence phosphorylase(ALP), total protein(TP), albumin(ALB), glucose (GLU), Ca 2+ , total CO 2, pH and febrile nonhemolytic transfusion reactions(FNHTR). The results showed that compared with RBT group, GPT in LDBT group on the third day after blood transfusion was significantly lower ( P 0 05). 8.The effect of parecoxib sodium for preemptive analgesia on nasal endoscopic surgery.

OBJECTIVE:
To evaluate the perioperative analgesic effects of parecoxib sodium in patients undergoing nasal endoscopic surgery.
METHOD:
In the randomized, double blind, controlled study, 120 patients undergoing septoplasty were divided into 3 groups (n = 40): A group received parecoxib at a dose of 40 mg by muscle injection 30 min before the operation followed by saline at the same volume every 24 h for 48 h; B group received parecoxib at a dose of 40 mg by muscle injection 30 min before the operation followed by 40 mg every 24 h for 48 h; C group received an equal volume of 0.9% saline at the same time points. Patients were assessed with respect to pain score (VAS), rescue analgesia requirement and the side effects during the operation as well as at 3, 24, 48 h after the surgery.
RESULT:
Intra-operative as well as the postoperative pain scores were less in the A group and B group than in the.control group. Compared with group A, group B had significantly lower VAS score at 24 h after the operation, however there were no significant difference on other time points. Fewer participants of both the A and B groups required rescue medication after operation.
CONCLUSION
Administration of parecoxib can provide ideal analgesic effects without serious adverse side effects at the perioperative period for patients who received nasal endoscopic operation. Intramuscular parecoxib (40 mg 30 min before the operation followed 40 mg qd for 48 h) designed as preoperative analgesia mode resulted in sufficient perioperative analgesia that deserves popularization in the clinical works.

9.Study on transdermal permeability of ephedra extract in vitro

AIM: To study the transdermal permeability of ephedra extract. METHODS: With Valia Chien diffusion cell to the transdermal experiments are used to observe the effect of different factors on the transdermal permeability. RESULTS: Its transdermal resorption constant increased with the increase of the concentration of ephedrine and ephedra extract. pH value of the diffusioin medium could influence ephedrine permeability. CONCLUSION: The result can offer reference in preparation for Ephera (targeting drug delivery system) (TDS).
10.Study on the correlation between intercellular adhesion molecule-1 and lung cancer

Objective To explore the role of intercellular adhesion molecule-1(ICAM-1) in the metastasis of lung cancer.Methods The expression of ICAM-1 in the tissue and the serum ICAM-1(sICAM-1) level in the 52 cases of primary lung cancer were detected with immunohistochemical staining method(SP) and enzyme linked immunosorbent assay(ELISA),with 20 cases of adjacent non-cancerous tissue as the control.Results The positive rate of ICAM-1 expression in lung cancer was significantly higher than that of adjacent non-cancerous tissue.The expression of ICAM-1 in the cases with lymph node metastasis was higher than that of no lymph node metastasis.The expression of ICAM-1 in stage III+IV was higher than that of stage I+II,and that in adenocarcinoma was higher than that of squamous cell carcinoma.The level of sICAM-1 in patients with lymph node metastasis was higher than that without lymph node metastasis.The sICAM-1 level in patients of adenocarcinoma was higher than that of squamous cell carcinoma,and that in patients with positive expression of ICAM-1 in tissue was higher than that of negative expression of ICAM-1.Conclusion The expression of ICAM-1 may be associated with the metastasis in lung cancer.The sICAM-1 level reflects the expression of ICAM-1 in tissue and may be used as a tumor marker of metastasis and prognosis in lung cancer.
