1.A STUDY ON PERMISSIBLE TEMPERATURE FOR TRANSPORTATION OF CRYOPRESERVED PLATELETS DURING WAR

In order to explore the permissible range of temperatures for cryopreserved platelets, each of the eleven bags of platelets with 5% dimethyl sulfoxide was divided at random into eight aliquot paired groups labeled A, B, C, D, E, F, G, and H respectively. All of the groups were stored at -80℃ for one day. The control group A was stored at -80℃ throughout the experiment, and group B, C, D, E, F, G, H were replaced to the refrigerator at -80℃ after they were stored for 4 hours at -60℃, -50℃, -40℃, -30℃, -20℃, -10℃, and 0℃ respectively. All samples were thawed to test the phosphatidylserine expression, CD62p re expression, slide platelet aggregation test(SPAT), and activated plasma clotting test(aPCT). The above items were monitored in group A hourly when group A was thawed and stored at 22℃ for 4 hours. The results showed that the permissible storage temperature of cryopreserved platelet was under -40℃, and storage at temperature from -30℃ to 0℃ was detrimental for cryopreserved platelets. No significant damages were detected when the thawed platelets were stored at 22 with a gentle shaker.
2.Flow cytometry detection of CD62p reexpression by preserved platelets and it's value in evaluation of the quality of preserved platelets

Objective 1) To establish and optimize a flow cytometry method for detecting CD62p reexpression by preserved platelets.2) Daily determination of the CD62p expression by platelets stored at 22℃ to assess its value in evaluation of preserved platelet quality.Methods 1) Statistic methods including factorial experiment was applied to optimize the major conditions for sample management, and the feasible negative and positive control for FCM analysis of CD62p expression were check out.2) Thirteen donors' platelets including ten PRP and three apheresis concentrations were stored in a flatbed shaker at 22?2℃ for 12 days. The CD62p expression were detected daily. The correlation between CD62p expression and in vivo residual life span of 22℃ stored platelet were analyzed.Results 1) Satisfactory conditions for analysis CD62p anew expression by FCM were obtained. 2) The CD62p anew expression by stored platelets was correlated to the in vivo residual life span of the platelets with the correlation coefficients between 0.91 and 0.99.Conclusion 1) This FCM method is simple, sensitive, stable, and easy to carry. 2) Platelet CD62p expression can be used to monitor the function of stored platelets and has the potential in the study of new platelet preservation technique.

Objective To explore the weight for the training target of Applied nursing undergraduate. And find the degree of importance for the indexes of the target , thus highlighting theApplied training objective characteristics. Methods Developed by Bloom's teaching objectives classification theory and educational objectives theory to refined indexes, using Delphi method to establish the Applied nursing undergraduate training objectives. By the importance assignment from experts on each index, using analytic hierarchy process (AHP) method to construct the judgment matrix. Then calculate the weight of each index weight, and test its logical consistency. Results The weight of theattitudes and valueswas 0.56, andknowledgeandabilitywere 0.22. For the secondary indicators, in the values dimension,professional attitudeandideological qualityhad relatively high weight.Under the knowledge dimension, the professional knowledge had the highest weight; and for the ability dimension,communication skillsandapplicationswere heavily maximum. All the random consistency rates (CR) for the indexes were less than 0.10, so there was no logical confusion. ConclusionsAppliednursing undergraduate talents need to combine theory and practice quickly into clinical work. By weight setting, this study provides basis to further quantify and highlight the Applied nursing undergraduate training features, and provides the basis for the application-oriented undergraduate nursing personnel reforms.

Objective To develop the Applied nursing undergraduate students training framework objective, clear the entry of applied training target for undergraduate nursing students. Methods Through literature review and panel discussions, Delphi expert consultation questionnaires were preliminary designed. 23 experts were issued via email and paper questionnaires, conducted two expert advice and analysis. Results A total of 20 valid questionnaires were recovered after two consultation. The expert advice of good scientific reliability:experts had positive reply, and the authority level was 0.857, Kendall were 0.130-0.293, it showed a good level of coordination (P < 0.05). By two rounds of expert advice, three capacity index target level for Applied nursing undergraduate students training were gotten: attitudes, values and basic literacy; knowledge of the target; the ability. The secondary capacity index for target had 15 items, and the third capacity index had 63 items. Conclusions TheAppliednursing undergraduate students training objectives and targets were refined through Delphi which provide the basis for the Applied nursing undergraduate training model and curriculum reform.
5.Study regional homogeneity and low frequency amplitude of brain resting state functional magnetic resonance imaging in patients with depression

Objective To explore the abnormally functional brain regions of female patients with depression by resting state functional magnetic resonance imaging (fMRI),and analyze the correlation with the severity of depressive symptoms.Methods 32 female patients diagnosed with depressive disorder and 40 matched healthy controls completed resting state fMRI scans.The whole brain's regional homogeneity (ReHo) and amplitude of low-frequency fluctuation(ALFF) were calculated,and the correlation analysis be tween ReHo and ALFF of brain regions with significant difference and the severity of depressive symptoms was conducted.Results Compared with control group,the left precuneus/left cuneus (MNI:-18,-81,45),bilateral precentral gyrus (MNI:-58,-5,35 and 57,-6,33),left inferior parietal lobule (MNI:-42,-36,45) and right inferior temporal gyrus(MN1:60,-45,-18) (P<0.05,corrected by AlphaSim)in the case group showed significantly decreased ReHo,with statistical significance.Compared with control group,the left cuneus(MNI:-3,-87,30),right inferior temporal gyrus(MNI:60,-48,-18) and left precentral gyrus(MNI:-63,-3,26) (P<0.05,corrected by AlphasSim)in the case group showed significantly decreased ALFF.The ReHo in the right inferior temporal gyrus was negative correlated with the HAMD-17 total score and retarda tion factor(r=-0.484,P=0.017;r=-0.408,P=0.048),the ALFF in the right inferior temporal gyrus was positively correlated with weight factor(r=0.574,P=0.003),and negative correlated with the number of depressive episodes(r=-0.416,P=0.043).Conclusion Female with depression in resting state have several abnormally functional brain regions and the extent of damage is correlated with the severity of depressive symptoms.Combination of the two parameters may yield a more comprehensive pathophy-siological mechanism for depressive disorder.

Objective To investigate the changes of the levels of TNF and IL 8 in blood and in gastric mucosa and their mRNA expression after liver ischemia reperfusion (I/R). Methods 130 rats were randomly divided into 3 groups: (1) sham operation group(control group, n=10);(2)liver ischemia reperfusion group (n=60), including ischemia for 20min and 40min, and reperfusion for 1h, 24h and 72h respectively; (3) liver I/R rats treated with famotidine group (n=60). ALT and AST were detected after operation, pathomorphological changes of gastric mucosa were studied by light microscope and transmission electronic microscope, the concentrations of TNF and IL 8 in blood and gastric mucosa were measured by radioimmunoassay (RIA), TNF mRNA and IL 8 mRNA were detected by in situ hybridization. Objective To investigate the association between polymorphism of S1, S2 locus allele in ADAM 33 gene and chronic obstructive pulmonary disease (COPD) and lung function in Xinjiang Uygur population. Methods Blood sam?ples from 217 COPD patients and 218 healthy controls were collected. Samples of DNA was extracted, and S1, S2 single nu?cleotide polymorphism (ADAM 33) was detected by ABI SNaPshot SNP genotyping. Results There were no significant dif?ferences in the frequencies of S1 locus CC, CT, TT genotypes and C, T alleles between patient group and control group (P>0.05). There were no significant differences in the frequencies of S1 locus CC, CG, GG genotypes and C, G alleles between patient group and control group (P>0.05). In patient group, there were no significant differences in S1, S2 locus genotype and clinical indicators of lung function display, and in the FEV1%predicted and FEV1/FVC (P>0.05). Haplotype analysis showed that there were no significant differences in three kinds of haplotypes between patient group and control group ( P>0.05). Conclusion There is no significant difference in the polymorphism of S1, S2 locus allele in ADAM 33 gene and the susceptibility to COPD in Xinjiang Uygur population.
8.Comparative analysis of allogeneic bone pad and autogenous bone graft in anterior cervical interbody fusion

BACKGROUND:Surgical treatment is often required for fractures of the cervical vertebrae. Anterior interbody fusion technology is stil the main method for the treatment of cervical degeneration or traumatic instability. Here, the self-made oval al ogeneic bone pad can adapt to different height and width of the intervertebral space, in line with the physiological shape of the intervertebral space. OBJECTIVE:By comparison with autogenous iliac crest bone, to evaluate various types of self-designed al ogeneic bone pads on anterior cervical interbody fusion. METHODS:From January 2009 to December 2013, 58 patients with cervical disc herniation were enrol ed and subjected to cervical discectomy and anterior cervical interbody fusion. According to different bone grafts, these patients were divided into al ogeneic bone pad and autogenous iliac bone groups. The course of disease was 12 to 24 months. The postoperative effect was measured by Japanese Orthopaedic Association (JOA) score, cervical fusion rate, fusion time, operative time, blood loss and rejection rate. RESULTS AND CONCLUSION:At 6 months postoperatively, the JOA score of two groups had no significant difference at 6 months after treatment (P>0.05);the cervical fusion rates were 83.7%and 87.8%, respectively, in the al ogeneic bone pad and autogenous iliac bone groups, with no significant difference (P>0.05). Fusion time was higher in the al ogeneic bone pad than in the autogenous iliac bone group (P<0.05). The internal fixators in the two groups were firmed without loosening, and there was no rejection during the fol ow-up. Compared with the autogenous iliac crest bone, anterior cervical interbody fusion with al ogeneic bone pad can achieve satisfactory effects, which can be better for intervertebral fusion and cannot induce pain due to bone cutting.
9.The application value of MSCT in diagnosis of acute appendicitis

Objective To discuss the application value of MSCT in the diagnosis of acute appendicitis.Methods 30 patients with appendicitis were enrolled as a research group,meanwhile 30 healthy adults without any diseases were as a control group.Some MSCT findings were observed or measured including diameter and position of appendix,fluid and gas density in appendix cavity, periappendicural fat density and peritoneal thickening,ascites and so on.Results The outside diameter of acute appendicitis appen-dix was (9.78±2.33)mm on average,whereas the diameter of normal adults appendix was (5.05±0.53)mm,exhibiting a signifi-cant difference (t=24.85,P <0.01).Rough periappendicural fat with increased density in 25 patients (83.33%)with the CT value of about -40 to 20 HU,exudation and peritoneal thickening around the appendix in 13 (43.33%),thickening and edematous appen-dix walls in 29 (96.67%),fecal stone in the appendix cavity with high density in 9(30.00%),the appendicitis in combination with intestinal obstruction in 1 (3.33%),and the appendicitis with peritoneal thickening in 80.00% and with peritoneal effusion in 23.33%were found in the study.Conclusion Full abdominal MSCT can help to the diagnosis of acute appendicitis,and some MSCT findings can be useful for the diagnosis including the appendix diameter≥6 mm,appendix cavity effusion,lumen stone,peripheral increased adipose density and peritoneal effusion.
10.Expression of Growth Associated Protein 43 in Inferior Colliculus of Rats at Different Times after Impulse Noise Exposur

Objective To investigate the effect of impulse noise expose on the expression of growth associated protein 43(Gap-43) in inferior colliculus in rat.Methods SPF grade Male SD rats were randomly divided into 5 groups.The normal control group received noise exposure.The model groups received an averange impulse noise exposure of 156 dB SPL with a pulse duration of 0.23 ms, once for 6 s, for 50 times.Auditory brainstem responses (ABRs) were measured before and 3,7,14, and 28 d after noise exposure with tone pips of 2,4,8,16, and 32 kHz, from 20 to 110 dB SPL.Bilateral inferior colliculus of rats in the model groups was collected and treated by immunohistochemical staining.Gap-43 expression of rats in different groups was measured by determining the gray value of inferior immunohistochemical images.Results After noise exposure, ABRs threshold in the model groups were significantly higher than those of in the normal group (P<0.05 or P<0.01).ABRs threshold at 14 and 28 days after noise exposure were significantly lower than 3 days after impulse exposure (P<0.05).Expression of Gap-43 in inferior colliculus was significantly up-regulated in the noise exposed groups compared with the normal group (P<0.05 or P<0.01).Expression of Gap-43 was significantly down-regulated 28 days after noise expose compared with 3 days after noise expose(P<0.05).Conclusion Impulse noise exposure leads to significant elevation of ABR thresholds and up-regulation of Gap-43 expression in inferior colliculus.Impulse noise exposure may induce auditory cortex prominent remodeling.
