Objective To determine an optimal method of in vivo endothelialization when homograft valve (HV) leaflets are treated with different methods. Methods Twenty five fresh HV leaflets were divided into five groups randomly. The leaflets of GroupⅠ were fresh HV ones without any treatment; GroupⅡwere cryopreserved in liquid nitrogen for 1 month; Group Ⅲ was treated by DOA; GroupⅣ was treated by 25 g?L -1 glutaraldehyde; Group Ⅴ was first treated by hypotonic/hypertonic, then with an enzyme-based solution. Each HV leaflet was trimmed to 2 mm one which was cultured with lymphocyte from the recipient. In order to compare the antigenicity of HV leaflets, 3H-TdR incorporation was observed after tissue-cell coculture and stimulous index (SI) was calculated. The HV leaflets of different groups were implanted into the sheep's femorofemoropopliteal for three months; then the following procedures were done. Immunohistochemistry of Ⅷ factor correlative antigen was used to identify the cells property; observation of growth condition of EC and superficial morphosis of HV leaflets was made with scanning election microscope (SEM). Results Multiple comparison showed that SI of Groups Ⅱ, Ⅲ, Ⅳ, and Ⅴ differed significantly from Group Ⅰ(P0.05), but Group Ⅲ, Ⅳ, and Ⅴ all had significant differences from GroupⅡ (P

AIM: To study the volatile oil of GanluXiaodu Granules GLXD (Talcum, Herba Artemisiae Scopariae, Radix Scutellariae etc.) and establish it's fingerprint. METHODS: By capillary gas chromatography. RESULTS: The fingerprints of volatile oil in Ganlu Xiaodu Granules were composed of 56 peaks, among which there were 17 characteristic peaks, having 86.97% of total area of the whole fingerprints peaks. The average RSD of retention time of characteristic peaks was 0.33%, that of peak-area was 2.76%. CONCLUSION: The fingerprints of volatile in Ganlu Xiaodu Granules can be used to control the quality.

Objective To evaluate the curative effect,adverse reactions and complications of transcatheter uterine arterial embolization (TUAE) therapy for leiomyoma of uterus with three different embolization agents: silk particles, pingyangmycin-lipiodol emulsion (PLE), polyvinyl alcohol (PVA).Methods One hundred and twenty-six patients of uterus leiomyoma were included, who undergoing TUAE treatment with silk particles(54 cases, silk particles group) ,PLE(40 cases,PLE group) and PVA(32 cases, PVA group) respectively.Adverse reactions and complications were recorded until 6-60 months after TUAE treatment.Measured the volume of the leiomyoma by ultrasonic inspection or enhanced computed tomography (CT).Results All clinical symptoms of three groups were relieved, and the volumes of uterus leiomyoma after TUAE treatment of 6,12,24 months in PVA group [(50.84 ± 30.77), (39.92 ± 25.72), (31.24 ± 30.60) cm~3]were less than those in PLE group[(76.24 ± 31.16), (58.21 ± 32.05), (52.48 ± 31.41 ) cm~3]and silk particles group[(69.22 ± 36.22), (68.31 ± 32.42), (68.01 ± 30.12) cm~3].The inferior belly and perineal region pain were most slight in silk particles group, but no significant difference in complications was observed in three groups.The symptom recurrence was 3 cases, 2 cases in silk particles group and 1 case in PIE group.Conclusions Clinical outcome of TUAE treatment with all the three embolization agents is acceptable.Adverse reactions and complications are associated with volume of the leiomyoma and the area being embolized.PVA is superior in prevention of complications and relapse of uterus leiomyoma after TUAE treatment.

OBJECTIVETo establish and experimental verification the mathematical model of the balance groups that is the steady-state of traditional Chinese medicine in extraction.

METHODUsing the entropy and genetic principles of statistics, and taking the coefficient of variation of GC fingerprint which is the naphtha of the Houttuynia cordata between strains in the same GAP place as a pivot to establish and verify the mathematical model was established of the balance groups that is the steady-state of traditional Chinese medicine in extraction.

RESULTA mathematical model that is suitable for the balance groups of the steady-state of traditional Chinese medicine and preparation in extraction, and the balance groups which is 29 683 strains (approximately 118.7 kg) were gained with the same origin of H. cordata as the model drug.

CONCLUSIONUnder the GAP of quality control model, controlling the stability of the quality through further using the Hardy-Weinberg balance groups of the H. cordata between strains, the new theory and experiment foundation is established for the steady-state of traditional Chinese medicine in extraction and quality control.

Objective To investigate the dynamic images of traumatic penumbra(TP)in a rat model of traumatic brain injury (TBI)treated with AQP4-RNAi using multimodal MRI(MM-MRI)at 7.0 Tesla.Methods A rat model of TBI was established by the improved Feeney's method.MRI scans were performed including T2WI,DWI,ADC and SWI.The pathological changes in penumbra area were observed.All statistical analyses were performed using SPSS 21.0 software package.Results Over time rs-T2WI,rs-DWI and rs-SWI in TBI group were gradually increased.The r-ADC began to increase at one hour after trauma and reached the peak at 6 h. Then it began to fall and reached the bottom at 12 h.At each time point,the difference was statistically significant(P<0.05).Compared with RNA interference group,the rs-T2WI and rs-DWI in TBI group were decreased significantly at 6 h and 12 h(P<0.05),the r-ADC was not decreased significantly at 1 h(P>0.05)and decreased significantly at 6 h and 12 h(P<0.05).At each time point,there was no significant difference in rs-SWI(P>0.05).The area of mismatch between rs-SWI and rs-DWI was the most obvious at 6 h and 12 h,and the area of mismatch was decreased in size after treatment with AQP4-RNAi.There was no significant difference between TBI group and placebo group(P>0.05).In two groups,similar pathological changes were observed,which was depicted as vasogenic edema predominantly at 1 h and mixed edema at 6 h and 12 h. In RNA interference group,intracellular edema was markedly reduced at 6 h and 12 h,and the vasogenic edema was relieved to some extent at 12 h.Conclusion The treatment with APQ4-RNAi markedly alleviates cerebral edema.MM-MRI can reflect its pathological changes.The area of mismatch between SWI and DWI can prompt early detection of traumatic penumbra,which may provide useful information for clinical treatment.

OBJECTIVETo explore the once sampling quantitation of Houttuynia cordata through its DNA polymorphic bands that carried information entropy, from other form that the expression of traditional Chinese medicine polymorphism, genetic polymorphism, of traditional Chinese medicine.

METHODThe technique of inter simple sequence repeat (ISSR) was applied to analyze genetic polymorphism of H. cordata samples from the same GAP producing area, the DNA genetic bands were transformed its into the information entropy, and the minimum once sampling quantitation with the mathematical mode was measured.

RESULTOne hundred and thirty-four DNA bands were obtained by using 9 screened ISSR primers to amplify from 46 strains DNA samples of H. cordata from the same GAP, the information entropy was H=0.365 6-0.978 6, and RSD was 14.75%. The once sampling quantitation was W=11.22 kg (863 strains).

CONCLUSIONThe "once minimum sampling quantitation" were calculated from the angle of the genetic polymorphism of H. cordata, and a great differences between this volume and the amount from the angle of fingerprint were found.

Objective To investigate the anti-oxidative effects of alprostadil on contrast-induced nephropathy(CIN)after percuteous coronary intervention (PCI)in patients with chronic kidney disease(CKD).Methods A total of 200 patients with CKD were enrolled in our hospital.According to the random number table was divided into alprostadil 100 cases,100 cases of conventional treatment group.The levels of serum creatinine (Scr),creatinine clearance (eGFR),serum cystatin C (ScysC)and 8-hydroxy-deoxyguanine (8-OHdG)were observed before and after operation at 72 h and 7 d after operation.Results The incidence of CIN in the alprostadil group was significantly lower than that in the conventional treatment group (6% vs 12%,P<0.05).There was no significant difference in the level of Scr,eGFR,ScysC and 8-OHdG between the alprostadil group and the conventional treatment group (P>0.05).The level of Scr in the alprostadil group was significantly lower than that in the conventional treatment group at 72 h and 7 d after operation.The level of eGFR was significantly higher than that of the conventional treatment group (P<0.05).The levels of ScysC and 8-OHdG in the two groups were significantly higher than those before operation at 72 h and 7 d(P>0.05).The levels of ScysC and 8-OHdG in the alprostadil group were significantly lower than those in the conventional treatment group at 72 h and 7 d after PCI(P<0.05).Conclusion Alprostadil may improve the oxidative stress in patients with CKD and provide a preventive effect on CIN.

OBJECTIVE: To explore the clinical effects on primary dysmenorrhea treated with moxibustion at Shenque (CV 8) and warm needling at Guanyuan (CV 4) and Sanyinjiao (SP 6). METHODS: A total of 120 patients with primary dysmenorrhea were randomized into an observation group and a control group, 60 cases in each one. In the control group, the warm needling technique was used at Guanyuan (CV 4) and Sanyinjiao (SP 6). In the observation group, besides the same treatment as the control group, moxibustion was added at Shenque (CV 8). The treatment was given for 4 menstrual cycles consecutively. Before and after treatment, the score of the severity and the score of the total frequency in the retrospective scale of dysmenorrhea symptoms as well as the score of the visual analog scale (VAS) were recorded and compared in the patients between the two groups. Additionally, the safety of the two therapeutic methods was evaluated. RESULTS: After treatment, the score of severity and the score of total frequency as well as VAS score of menstrual pain were all reduced as compared with those before treatment in the patients of the two groups (all <0.05). The scores in the observation group were more obvious as compared with the control group (all <0.05). Regarding the safety evaluation, the difference was not significant between the two groups (>0.05). CONCLUSION The combined treatment of moxibustion at Shenque (CV 8) with the warm needling technique at Guanyuan (CV 4) and Sanyinjiao (SP 6) achieves the better clinical effects on primary dysmenorrhea as compared with the simple application of the warm needling technique at Guanyuan (CV 4) and Sanyinjiao (SP 6). This therapy is safety in clinical practice.
