1.Let-7 and its function in lung cancer

Let-7 is found to be downregulated in lung cancer while many kinds of oncogenes expression always increase, and restoration of the let-7 to normal expression level inhibits lung cancer growth evidently.Thus, let-7 might serves as good targets or effective interference tools in exploration of anticancer therapeutics,and has a good prospect in applying it in the early diagnosis, prognosis and therapy in lung cancer.
2.The Regulatory Effects of Dexamethasone on the Acute Experimental Serum Sickness in Rabbits

This experiment takes the apute experimental serum sickness (AESS) in rabbits as an animal model to investigate the mechanism of the regulatory effects of Dexamethasone (DX) . The results show. DX doesn't inhibit the production of anti-BSA antibody but induces the tendency to increase the level of antibody in rabbits with DX administration on early days (EL, EH); immune complexes(BSA - anti-BSA IGg, CIC) in the rabbits of EL and EH are significantly increased comparing with the later days (LE, LH) and positive controls. Non-specific esterase stainings and the pathological changes of kidney sections are markedly lessened though the amount of anti - BSA antibody, circulating immune complex (CIC) and superoxides dismutase activity in the rabbits of LE and L H are the same as the positive control. The outcome of protecting kidneys of AESS rabbits from injury by DX administration . probadly results from the effects of that DX inhibits infiltration of monocytes into the glomeruli. The role of oxygen radicals in AESS needs further investigation.
3.Effects of acute hypervolemic hemodilution with HSH 40 on hemodynamics and fluid balance in patients antler general anesthesia

Objective To obeserve the effects of acute hypervolemic hemodilution(AHH) with hypertonic .sodium chloride hydroxyethyl starch 40(HSH 40) on hemodynamics and fluid balance in patients under general anesthesia.Methods Fifty patients undergoing radical surgery for gastral cancer under general anesthesia were randomly divided into 2 groups with 25 patients each.Acute hypervolemic bemodilution (AHH) was performed with HSH 40 6 ml/kg in group A or with hydroxyethyl statch(HES) 6 ml/kg in group,which was infused within 30 minuts.HR,MAP,CVP were recorded before(T_0),at 30 min (T_1),60 min (T_2) after infusionand and the end of operation (T_3).The amounts of bleeding,HSH 40 and HES and urine output were recorded as well.Results There were no significant diferences in HR and MAP between two groups at all time points.CVP was sighificantly higher at T_1-T_3 than that at To in two groups.The urine output was more in groups A than that in group B(P<0.05).Conclusion AHH with HSH 40 can effectively expand blood vlume and increase urine output in surgical patients under general anesthesia.
4.Enzyme-linked Immunosorbent Assay to Determine Specific Immune Complex in Rabbit circulation

A method, enzyme-linked immunosorbent assay, was introduced for the determination of antigen-specific immune complex (BSA-anti-BSA) in rabbit circulation. The antigen-specific immune complex in the circulation of twelve rabbits were determined by this method. The results showed that this method accords with the regular pattern of immune complex formation more closely than the antigen-nonspecific assay, PEG precipitation method. The procedure of this method is rather simple, its specificity is of high degree, and the amount of serum needed in test is small. This enzyme-linked immunosorbent assay is a better method to determine the circulating specific immune complex in rabbits.
5.Clinical investigation of surgery combined with the rehabilitation treatment for the patients with burn scar contracture on hand

Objective To investigate clinical effect of surgery combined with the rehabilitation treatment for the patients with burn scar contracture on hand,to provide a reference for clinical treatment.Methods 60 cases of hand burn scar contracture patients were selected,according to the digital table they were divided into control group and observation group(n =30 cases),which was underwent surgery in both groups.The control group was treated based on out -patient routine hand exercise methods,while a heated therapy,compression therapy and other profes-sional rehabilitation exercises were performed in the observation group.TAM in patients and ADL were compared. Results The observation group after treatment TAM score was (198.67 ±21.08)point,which was significantly higher than (144.43 ±16.65)point,the difference was statistically significant (t =8.476,P =0.000).Manicure, taking food,combing,brushing teeth,washing face,clothing,shoeing ADL scores in the observation group after treatment were (3.26 ±0.51)point,(3.56 ±0.55)point,(3.68 ±0.72)point,(3.38 ±0.68)point,(3.61 ±0.52)point, (3.55 ±0.61 )point and (2.57 ±0.47)point,which were improved significantly better than those of the control group,the differences were statistically significant (t =4.524,P =0.024;t =5.041,P =0.011;t =4.924,P =0.019;t =4.442,P =0.027;t =5.136,P =0.010;t =5.347,P =0.008).Conclusion Clinical effect of surgery combined with professional rehabilitation on hand burn scar contracture is good,and it is worthy of clinical application.
6.Pingyangmycin(bleomycin A5)-mediated electrochemotherapy on human prostate cancer cell line PC-3M in vitro

Objective To investigate the efficacy of electrochemotherapy with pingyangmycin (bleomycin A5) for the treatment of human prostate cell line PC 3M in vitro. Methods The prostate cancer cells were treated by pingyangmycin alone, electric pulse alone and by electrochemotherapy, i.e. electric pulses were delivered after pingyangmycin was given. Cytotoxicity was examined by MTT assay and histological examination. Results The maximum nontoxic dose for pingyangmycin was 0.1 ?g/ml; The maximum voltage not showing cytotoxicity was 125 V/mm.The 50% inhibitory concentration was 100 times reduced in electrochemotherapy than pingyangmycin alone. Conclusions The cytotoxicity of pingyangmycin can be enhanced by electric impulses. Pingyangmycin may be a novel drug for electrochemotherapy.
7.Changes of cellular immunity in patients with hypertrophic sear

Objective To analyse the changes of T lymphocytes and its subset in patients with scar hypertrophy. Methods In 6 keloid, 14 hypertrophic scar patients and 10 normal control, peripheral blood T lymphocytes (PBTL) and its subset were analysed with flow cytometry (FCM). Lymphocytic proliferation responsed to Con A stimulation in vitro lymphocytic culture system were performed and lymphocytic proliferation activity were assayed by the measurement of 3H TdR incorporation. Results The percent of CD3 +,CD4 +,CD8 + and ratio of CD4 +/CD8 + in PBTL of keloid patients were 78.6 %, 49.7 %, 23.5 % and 2.4, and in hypertrophic scar patients group were 68.9 %, 47.6 %, 26.6 % and 2.2, in normal control group were 69.2 %, 37.2 %, 30.9 % and 1.9, respectively. Lymphocytic proliferation activity expressed as proliferation stimulation index were 17.4,15.9 and 8.7 respectively in keloid, hypertrophic scar and normal group. The level of CD4 +?CD4 +/CD8 + and lymphocytic proliferation stimulative index were all significantly higher in both keloid and hypertrophic scar patients than that in normal control. Conclusion Aberrant immunologic function may play an important role in the pathogenesis of scar.
8.Clinical characteristics and treatment of infective endocarditis in children

Objective To study the clinical characteristics, treatment and prognosis of infective endocarditis in children. Methords Clinical data from 83 patients of infective endocarditis admitted from 1998 to 2012 were retrospectively analyzed. Results In a total of 83 patients, there were 53 males and 30 females, and the average age was 6.8±4.6 years. The main clinical characteristics were fever (77.1%) and mild to moderate anemia (71.1%). The C-reaction protein (67.5%), erythrocyte sedimen-tation rate (60.2%), and white blood cell (47.0%) were elevated. Twenty (24.1%) patients had embolism. Blood culture was pos-itive in 56 (67.5%) cases with bacteria mainly being Gram-positive and Streptococcus and Staphylococcus accounted for 89.3%. Vancomycin and other sensitive antibiotics were effective. Neoplasm was detected in 68 cases (82%) by transthoracic echocar-diograerphy. Fifty-ifve (66.2%) patients underwent cardio surgery. Seven patients (8.4%) died. Conclusion In recent years, the distribution of pathogenic bacteria in infective endocarditis had changed. Streptococcus mitis and Staphylococcus aureus has become a major pathogens and need to be treated by vancomycin and other sensitive antibiotics. The detection rate of neoplasm is higher by echocardiography.
9.Effect of patient-controlled intravenous analgesia with dezocine combined with sufentanil on inflammatory response and pain after laparoscopic hepatectomy for hepatocellular carcinoma

Objective To observe the effect of patient-controlled intravenous analgesia (PCIA)with dezocine combined with sufentanil on inflammatory response and pain after laparoscopic hepatectomy for hepatocellular carcinoma.Methods Sixty patients (43 males,17 females,aged 18-60 years,ASA grade Ⅰ or Ⅱ) scheduled for laparoscopic hepatectomy for hepatocellular carcinoma were divided into sufentanil group (group S) and dezocine+sufentanil group (group DS) according to the random number table,n=30 each.Patients in group S were given 100 ml normal saline containing sufentanil 2.0 μg/kg and tropisetron 5 mg.Patients in group DS were given 100 ml normal saline containing sufentanil 2.0 μg/kg,dezocine 0.5 mg/kg and tropisetron 5 mg.VAS scores and numeric sedation scale (NSS) scores were recorded at 4,24,48 h after operation and patients' satisfaction scores were recorded at 48 h after operation.The levels of serum tumor necrosis factor-α (TNF-α),interleukin-2 (IL-2),interleukin-6 (IL-6) in blood samples harvested before induction of anesthesia and 0,4,24 and 48 h after operation were measured by ELISA.The times of efficient injection and incidence of adverse effect within 48 h after operation were recored.Results Compared with group S,the VAS scores in group DS were decreased significantly while the satisfaction of patients to analgesia were increased significantly at 4,24,48 h after operation (P<0.05).There were no obvious differences in NSS scores between two groups.Compared with before induction of anesthesia,the concentrations of TNF-α and IL-6 were increased significantly while the concentrations of IL-2 was decreased significantly in both groups at 4,24,48 h after operation (P<0.05).Compared with group S,the concentrations of TNF-α and IL-6 were decreased significantly while the concentrations of IL-2 was increased significantly in group DS at 24,48 h after operation (P<0.05).The times of efficient injection in group DS were less than that in group S significantly within 48 h after operation [(2.0±0.7) times vs.(7.2±1.3) times] (P<0.05).There were no obvious differences in adverse effects between two groups.Conclusion PCIA with dezocine 0.5 mg/kg combined with sufentanil 2.0 μg/kg can alleviate the inflammatory response to some extent in patients after laparoscopic hepatectomy for hepatocellular carcinoma,and it can offer a safe and effective analgesic effect.
10.Universal primer V3 coupled with multiplex PCR for the pathogen detection of infective endocarditis

Objective To investigate the pathogen of 21 infective endocarditis (IE) cases treated with operation in Shanghai Children's Medical Center from 2007 to 2010.Methods Blood culture,vegetation culture and vegetation PCR assay(target gene to the conserved region V3 in 16SrRNA gene) were detected in 21 IE patients; multiplex PCR amplification of staphylococci for methicillin-resistant staphylococcus was performed.Results Of 21 IE cases,20 cases were detected positive by vegetation PCR with the detection rate of 95.2%,12 IE cases were detected positive by blood culture with the detection rate of 57.1%,2 IE cases were detected positive by vegetation culture with the detection rate of 9.5%.The difference of the positive rates of the three methods was statistically significant (P < 0.0001).The vegetation PCR of one case was actinobacillus actinomycetemcomitans,while the blood culture was haemolysis pasteurell which was inconsistent with the vegetation PCR result.Howerver,the PCR result of colony obtained by blood culture was consistent with vegetation PCR that was confirmed as actinobacillus actinomycetemcomitans.The endocardium PCR results of 11 IE cases were consistent with the results of blood culture.MecA gene was detected by multiplex PCR,which could identify methicillin-resistant staphylococcus quickly,sensitively and accurately and could also effectively identify methicillin resistant staphylococcus aureus,when coupled with femA gene detection,thus glycopeptides antibiotic could be prescribed promptly.All the 21 patients recovered and discharged without infection recurrence in the follow-up.Conclusion Universal primer V3 coupled with multiplex PCR can improve vegetation pathogen detection rate of IE patients and is minimally influenced by antibiotic therapy.Multiplex PCR can be applied for etiological diagnosis of IE patients with indication of surgery and negative blood culture or difficult diagnosis,contributing to post-surgery antibiotics selection and improvement of recovery rate of IE patients.
