Immunoreactive substance P (Ir-SP) level and substance P-like activity (SP-LI)in the spinal cord were determined with radioimmunoassay and immunohistochemistry in rats after they were given an intraperitoneal injection of morphine(7.5mg/kg)or electroacupunctured(3V and 3Hz)on the "Jiaji" point. Objective To investigate the clinical efficacy of DPP-4 inhibitors in the treatment of newly diagnosed type 2 diabetes mellitus and its effect on blood pressure and body weight.Methods A total of 100 newly diagnosed elderly patients with type 2 diabetes mellitus were enrolled in this study from July 2013 to June 2015 in this hospital.They were divided into observation group and control group(n=50).The control group was treated with placebo on the basis of diet control and exercise.The observation group was treated with sitagliptin on the basis of diet control and exercise.Fasting plasma glucose(FPG),2 h postprandial blood glucose(2 h PG) and glycated albumin (GA) were measured before and after treatment,and the incidence rate of hypoglycemia,blood pressure,body weight and other adverse reactions were monitored.Results After treatment,two groups of patients,condition were significantly improved,FPG,2 h PG,GA changes in observation group were significantly better than that of control group,the difference were statistically significant(P<0.05).There was no significant difference in blood pressure,body weight and body mass index before and after treatment,and also between the two groups(P>0.05).The incidence of adverse reactions in the observation group and the control group were 10.0% and 8.0%,The incidence of adverse reactions in the two groups was not statistically significant(P>0.05).Conclusion The clinical efficacy of DPP-4 inhibitor siglitazide in the treatment of newly diagnosed elderly patients with type 2 diabetes mellitus is good,and it is very useful in the treatment of these patients.
3.Relationship between NO changes and LTP production in rat hippocampus in vitro

Objective To observe the time course of the changes in nitric oxide (NO) content and the activity of NO synthase (NOS) during hippocampal long term potentiation (LTP) in vitro . Methods The production and maintenance of LTP were detected by using extracelluar electrophysiological recording. NO content and NOS activity were determined by biochemical reaction. The expression of NOS mRNA was detected by in situ hybridization. Results Conditioning stimulation for 10 min induced LTP production and significant increases in NO content, NOS activity and expression of NOS mRNA. However, at 60 min after conditioning stimulation, LTP remained stably but NO content and NOS activity returned to the pre conditioning stimulation level. Moreover, the NOS mRNA was overexpressed at the early stage of LTP production. Conclusion Significant NO changes may occur at the early stage of LTP formation.
4.Pathogenic Bacteria Distribution and Drug Resistance in Inpatients of Burn Department

OBJECTIVE To analyze the pathogenic bacteria distribution and drug resistance in inpatients of burn department,and instruct clinical application of antibiotics reasonably.METHODS Pathogenic bacteria were isolated from inpatients of burn department from 2003 to 2006.K-B slip diffusion method was taken to carry out the sensitive test.Rate of drug resistance of the pathogenic bacteria was analyzed.RESULTS Totally 1325 strains were isolated,among them 846 strains were Gram-negative bacteria,464 strains were Gram-positive ones,and 15 strains were fungi.The percentage of these three groups was 63.85%,35.02% and 1.13%,respectively.The main strains of the Gram-negative bacteria were Pseudomonas aeruginosa,Escherichia coli,Acinetobacter baumannii,Enterobacter cloacae and Klebsiella pneumoniae,and that of the Gram-positive bacteria were Staphylococcus aureus,Enterococcus and Staphylococcus epidermidis.The major strain of fungi was Candida albicans.The pathogenic bacteria tested showed high drug resistance.The detection rate of the meticillin-resistanct S.aureus(MRSA) was 82.12%.The detection rate of the ESBLs from the K.pneumoniae and the E.coli was 60.17% and 41.89%.CONCLUSIONS It was showed that the major pathogenic bacteria infected of the inpatients of burn department are Gram-negative bacteria.The pathogenic bacteria are the multidrug-resistant ones.Enhanced monitoring on pathogenic bacteria distribution and drug resistance analyses among inpatients of burn department could benefit for the guide of clinical rational administration and depression of multidrug-resistant bacteria.
