The JAK/STAT pathway contributes to control gene expression,cell growth,proliferation,differentiation,apoptosis and immunoregulation.Recent studies have found that the JAK/STAT pathway was closely related to the occurrence and development of lymphoid neoplasms.The study of JAK/STAT pathway could lead to new treatment strategies for lymphoid neoplasm treatment.

Objective To investigate the effect of integrated traditional Chinese and western medicine in the treatment of postoperative pain in patients with anorectal diseases. Methods From November 2015 to October 2017, 70 patients with anorectal diseases underwent surgical treatment in the First People 's Hospital of Yongkang were selected. The simple western medicine treatment was performed in 35 patients with anorectal diseases in the control group,and 35 patients in the observation group were given combined Chinese and western medicine. The total effective rate of the two groups was observed and calculated. The pain score at 2 h,6 h,12 h and 24 h after operation,the incidence rate of postoperative complications were observed. Results The total effective rate of the control group (80. 00％ ) was lower than that of the observation group (97. 14％ ),the difference was statistically significant(χ2 =5. 080,P < 0. 05). Conclusion The combination of traditional Chinese and western medicine is effective in the treatment of postoperative pain in patients with anorectal diseases,which can increase the effective rate and improve the quality of life of the patients, and it is worthy of clinical application.

Objective To study the relationship between intradialytic hypertension (IDH) and inflammatory factors in patients with maintenance hemodialysis.Methods Forty maintenance hemodialysis patients were involved,including 20 patients with IDH (IDH group) and 20 patients without IDH(non-IDH group) during hemodialysis.All of them had no sign of acute infections or other progressive disease.The levels of C-reactive protein (CRP),interleukin (IL)-1 β,IL-6,tumor necrosis factor (TNF)-α were measured by enzyme-linked immunosorbent assay (ELISA) before hemodialysis and 1,2,4 h after hemodialysis.The correlation of hypertension to CRP,IL-1 β,IL-6,TNF-α was analyzed.Results The levels of CRP,IL-1β,IL-6,TNF-α were increased in IDH group with the progress of hemodialysis,but were stable innon-IDH group.Logistic regression analysis indicated that there were positive correlations between CRP (r =0.3652),IL-1β (r =0.3261 ),IL-6(r =03156),TNF- α (r =0.3324) and blood pressure during hemodialysis (P ＜ 0.01 ).Conclusion The levels of CRP,IL-1β,TNF-α,IL-6 are positively related to the IDH in maintenance hemodialysis patients.

Objective To observe the level of serum hepcidin and the relationship of hepcidin with renal anemia and micro-inflammation state in peritoneal dialysis(PD) patients.Methods Serum hepcidin,interleukin-6(IL-6),soluble transferrin receptor (sTfR) and erythropoietin (EPO) were measured in 50 PD patients with anemia,30 PD patients without anemia and 40 cases of normal control by ELISA.The indexes of blood routine examination,biochemistry and iron metabolism were also detected at the same time.Results The level of hepcidin in PD patients was significantly higher than that in normal control[(103.65±43.6) μg/L vs (56.39±35.7) μg/L,P ＜ 0.05].Furthermore,the level of hepcidin in PD patients with anemia was higher than that in PD patients without anemia [(122.67±36.6) μg/L vs (83.65±26.4) μg/L,P ＜ 0.05].The results of correlation analysis showed that serum IL-6,sTfR,EPO and ferritin were positively correlated with hepcidin(R =0.821,0.742,0.711,0.531,all P ＜ 0.05),while creatinine clearance of residual kidney in 24 hours and hemoglobin were negatively correlated with hepcidin(R =-0.533,-0.685,all P ＜ 0.05).Conclusions The higher level of hepcidin in PD patients is related with the residual renal function and the micro-inflammatory state.The higher level of hepcidin may induce the iron metabolism imbalance,and then influence the state of renal anemia.The adjustment of hepcidin may provide clinical research value of improving renal anemia and micro-inflammatory state in PD patients.

Objective To investigate the genetic polymorphisms of 3 short tandem repeat (STR) loci D18S53, D18S59 and D18S488 on chromosome 18 in fetus of Tianjin Han population, and to provide basic data in the use of 3 STR lo-ci in the prenatal diagnosis of Edward syndrome (ES). Methods A total of 64 villus samples and 374 amniotic fluid sam-ples were collected from gravida in Tianjin Han population. QF-PCR and ABI PRISM 377 sequence were used in this study. The frequencies of the genotypes were tested with H-W equilibrium. Genetic analysis was performed to conclude some data of population genetics such as the frequency of the alleles, the heterozygosity of observation (Ho), the polymorphism informa-tion content (PIC), the probability of discrimination power (DP), and the probability of exclusion (PE). Results The 15, 13 and 15 alleles of D18S53, D18S59 and D18S488 were observed respectively. The frequencies of the genotypes were in good agreement with H-W equilibrium. The Ho of 3 STR loci were 0.797, 0.847 and 0.792. The PIC was 0.81, 0.75 and 0.73. The DP was 0.944, 0.901 and 0.881. The PE was 0.593, 0.689 and 0.585. Conclusion D18S53, D18S59 and D18S488 STR lo-ci were the favorable genetic markers of chromosome 18, which can be used in prenatal genetic diagnosis of ES.

Objective To explore the effect of pre-filled catheter flushing device on the sealing of venous indwelling needle. Methods One hundred and sixteen patients with venous indwelling needle were assigned into observation group, in which pre-filled catheter flushing device was used to seal the needle tube and another 109 with venous indwelling needle as control group, in which the needle tube was sealed with 50U/mL heparin saline.The two groups were compared in terms of blockage of the tube, phlebitis, catheter-associated infection, indwelling time of the needle and time for nurses to seal it.Results The rates of blockage of catheter, phlebitis and catheter-associated infection of the observation group were all lower than those of the control group.The indwelling time of the needle was significantly longer than that in the control group.The time for nurses to seal the catheter was significantly shorter than that in the control group(all P<0.05). Conclusion Pre-filled catheter flushing device can reduce complication rate and improve working efficiency of nurses.

Objective To evaluate the expression of CD4+CD25+CD127low regulatory T cells(Tregs) in the peripheral blood of the patients with NHL and discuss the relationship between expression of Tregs and prognostics factors of NHL.Methods 58 NHL patients was enrolled for analyzing.Among of them,23 were new patients without any treatment,35 patients received 3 to 8 cycles of chemotherapy,and 16 were normal controls.The expression of CD4+CD25+CD127low Tregs was detected by Flow Cytometry and the proportions of Tregs were analyzed among the groups of the patients before or after chemotherapy and the normal controls.Bivariate Correlation was used to analyze the correlation among the expression of Tregs in the peripheral blood and prognostic factors,such as age,sex,PS state,LDH,?2-MG,IPI,clinical stage and pathology category of the new patients.Results The proportion of Tregs in the peripheral blood of B cell lymphoma(B-NHL) group(3.89?2.49)% was significantly higher than that of normal control(2.52%?0.88%)(P=0.005).In B-NHL,Tregs was signifantly higher in the aggressive NHL(DLBCL,MCL) than that of nonaggressive HHL(FL,MZL)(P=0.002).In T cell lymphoma,Tregs was increased markedly in the highly aggressive T-NHL(T-LBL,NK/TCL) as compared with aggressive T-NHL(ALCL,PTCL)(P=0.006).Tregs expression was significantly higher in newly diagnosed NHL patints than that of normal control(P=0.004) and post-chemotherapy group(P=0.034).The expression of Tregs in peripheral blood was significantly correlated with ?2-MG,LDH and IPI of the patients in newly diagnosed patients group(P=0.003,0.003 and 0.038),whereas was not significantly correlated with the age,sex,PS state,LDH,clinical stage and pathology category of the patients.Conclusion The expression of CD4+CD25+CD127low Tregs in peripheral blood of B-NHL group was significantly higher than that of the patiens post-chemotherapy and normal control,which means that it may be an useful prognostic factor for NHL patients.

Objective To investigate the effects of the microbubbles combined with different mechanical index ultrasound irradiation on ultrastmcture and migration of colon cancer cells.Methods The experimental study was conducted.Colon cancer cells inn vitro (Lovo ceils) were cultured and divided into 4 groups,ceils in the A group were not treated and cells in the B,C and D groups were treated by microbubbles combined with different mechanical index ultrasound irradiation (mechanical index were 0.20,0.80 and 1.45).The changes of ultrastructure and migration of cells were observed using laser scanning confocal microscope and MilliceIl-PCF cell culturechamber method,respectively.Measurement data with normal distribution were represented as (x)±s.Comparisons among groups were analyzed by the one-way ANOVA.Pairwise comparisons were done by the t test.Results (1)Effects of the microbubbles combined with different mechanical index ultrasound irradiation on ultrastructure of Lovo cells:Lovo cells in the A group showed big nucleus,less plasma,regular arrangement,jagged-like or more irregular varicosity surrounding nucleus,twisted euchmmatin,expansion of nucleus cisternal space,homogeneous distribution and normal development of granular soil and clear mitochondrial ridge-like structures.Lovo cells in the B group showed big nucleus with regular arrangement,obvious nucleolus margination,endoplasmic reticulum dilatation,normal development of mitochondrion and clear mitochondrial ridge-like structures.Lovo cells in the C group showed broadening nucleus space,abnormal nucleus with karyopycnosis,chromatin condensation,a few remaining or obvious dilatation of rough endoplasmic reticulum,typically consisting of different fragments or bubbles,cytoplasmic vacuoles changes and decreasing mitochondrial ridge-like structures.Lovo cells in the D group showed big and irregular nucleus,nucleolus margination,obvious endoplasmic reticulum dilatation,fewer mitochondrion with extended cell area and swelling shape,rare mitochondrial ridge-like structures with disordered or broken arrangement,even disappearing.(2) Effects of the microbubbles combined with different mechanical index ultrasound irradiation on migration of Lovo cells:Millicell-PCF cell culture chamber method showed that number of migration of Lovo cells were respectively 63±7,61±4,21±3 and 19±5 in the A,B,C and D groups,with a statistically significant difference (F=55.040,P＜0.05).There were no statistically significant difference in migration of Lovo cells between group A and B (t =1.571,P＞0.05) and between group C and D (t =2.013,P＞0.05).There were statistically significant differences in migration of Lovo cells between group A and C or D (t=7.861,10.652,P＜0.05) and between group B and group C or D (t=7.161,10.453,P＜0.05).Conclusion Microbubbles combined with high mechanical index ultrasound irradiation can make the ultrastructural alterations in the cancer cells,resulting in tumor cell degeneration and death,ultimately inhibit tumor cell migration and metastasis.
