1.Enteral nutrition support and nursing to batched severe burn patients caused by explosions

Objective To explore nursing points on the enteral nutrition support to batched patients with severe burn caused by explosion. Method An enteral nutrition support was carried out for 15 patients with severe burns admitted in our department and pertinent nursing was performed. Results Five patients died because of lack of autologous skin sources and complications of sepsis in 45~87 d after burn. Ten patients got a good recovery after enteral nutrition support, anti-infection treatment and nursing. They lived through the infection period, went into the recovery period. The hospital stay of these patients in ICU was 54~115 d with an average of (90.63 ± 30.37)d. Conclusion Early enteral nutrition support and nursing to the patients with severe burn can improve the nutritional status of patients effectively and promote the rehabilitation of patients.
2.Relationship between the number of circulating endothelial cells and blood lipid level in critically ill patients and their clinical significance

Objective To study the changes of the number of circulating endothelial cells (CEC) and blood lipid level in critically ill patients and their clinical significance. Methods The CECs were separated by using isopyknic centrifugation method and blood lipid level was determined in 35 critically ill patients and 20 healthy controls. CEC number and blood lipid level were compared between the two groups. And the correlation of CEC number and blood lipid level was analyzed. 3.Infusion of drug via portal vein for liver cirrhosis

Medication is still the main means for treating liver cirrhosis. However, micro-circulation obstacle in the liver and dynamic abnormality of portal vein flow suggest that liver protecting drugs via peripheral veins can hardly give full requirement for their effect. The authors reviewed a new method of percutaneous transhepatic portal vein implantation of port-catheter system for infusion of drugs in the treatment of liver cirrhosis, with theoretical basis and the outlook of application.
4.Levels of heme oxygenase-1 and carbon monoxide and its significance in the Mycoplasma pneumoniae infection in children

Objective To explore the heme oxygenase-1 (HO-1) content of carbon monoxide (CO) levels and clinical significance of Mycoplasma pneumoniae infection in children.Methods 219 cases of mycoplasma pneumonia from October 2013 to June 2015 in our hospital were divided into two groups,which were non-asthmatic group (156 cases) and wheezing group (63 cases),and the wheezing group were divided into two groups according to the presence or absence of hypoxemia and mild asthmatic group (39 cases) and severe asthmatic group (24 cases).HO-1 content in the blood was measured by enzyme-linked immunosorbent assay,using dual-wavelength spectrophotometric determination of hemoglobin oxygen percentage of broken (COHb%),analysis of the differences between the groups.Results Children with blood COHb content wheezing group were (2.59 ± 0.40)% and HO-1 levels were (1 813.24 ± 28.34)ng/L and non-respite group were significantly increased,it had significant differences between the two groups (P< 0.05).Group of children with severe wheezing COHb content were (3.63 ± 0.45)% and HO-1 levels were (2 594.34±23.94)ng/L and mild wheezing group were significantly increased,it had significant differences between the two groups (P<0.05).Positive correlation between blood COHb content and HO-1 levels were significantly (r=0.733,P<0.05).Conclusion Mycoplasma pneumoniae infection in children with wheezing blood COHb content and HO-1 levels were significantly increased,and with wheezing exacerbation and showed an increasing trend,suggesting that CO and HO-1 may be involved in Mycoplasma pneumoniae infection in children in the pathogenesis of wheezing,should lead to clinical attention.
5.Bromodomain4 inhibitor JQ1 inhibits the proliferation of Ph positive acute lymphocytic leukemia cells and its mechanism

Objective To observe the effect of bromodomain4 (brd4) inhibitor JQ1 on proliferation inhibition and apoptosis of Ph positive acute lymphocytic leukemia (Ph+ ALL) cells, and to explore the influence on the expression of brd4 and its downstream genes (myc and p53), and the reverse effect on bcr-abl.Methods Different concentrations of JQ1 were used on SUP-B15 cells.The proliferation inhibition rate was detected by MTT, the apoptosis rate was determined by flow cytometry (FCM), and the expressions of bcr-abl mRNA, brd4 mRNA, myc mRNA and p53 mRNA were detected by real-time fluorescent quantitative PCR (RT-PCR).Results Different concentrations of JQ1 inhibited SUP-B15 cells proliferation and induced cell apoptosis.The apoptosis rate was significantly increased compared with that in control group with a time-dose dependent manner.Median inhibitory concentration at 72 h was 1.0 pmol/L.At the same time, JQ1 decreased the transcription levels of bcr-abl mRNA, brd4 mRNA and myc mRNA, and increased the transcription level of p53 mRNA.Conclusions As a brd4 inhibitor, JQ1 can decrease the expression of brd4 to affect the expression of its downstream genes myc and p53, meanwhile, it can change the over expression of bcr-abl to suppress the proliferation of Ph+ ALL cells and induce apoptosis.
6.The association between 8-hydroxy-deoxyguanosine and Graves' ophthalmopathy

To investigate the role played by 8-hydroxy-deoxyguanosine (8-OHdG) in patients with Graves'ophthalmopathy(GO),and the association between 8-OHdG and the disease activity of GO.Serum levels of 8-OHdG were tested in 48 cases with GO and 30 cases with Graves' disease(GD) without ophthalmopathy.30 healthy subjects participated in this study as controls.8-OHdG was detected by enzyme-linked immunosorbent assay (ELISA).Compared with the controls,the mean level of 8-OHdG in GO patients and GD patients was increased significantly [(2.98±1.33),(2.07±1.30) vs(0.72±0.93)ng/ml,P<0.05].Mean 8-OHdG level in GO patients was markedly higher when compared to GD patients(P<0.05).Pearson regression analysis showed that elevated levels of 8-OHdG were significantly correlated with the Clinical Activity Score of GO (r =0.54,P<0.01).The serum level of 8-OHdG in GO patients was raised obviously,it implies that oxidative stress plays an important role in the pathogenesis of GO,and the serum level of 8-OHdG might act as an activity index of GO.
7.Change in serum level of 8-hydroxy-deoxyguanosine during corticosteroid treatment in patients with active Graves' ophthalmopathy

The change in serum level of 8-hydroxy-deoxyguanosine (8-OHdG),an oxidative stress biomarker,in patients with active Graves' ophthalmopathy (GO) during corticosteroid treatment was observed.The serum level of 8-OHdG was significantly increased in patients with active GO as compared with that of normal controls and patients with Graves' disease (P < 0.05).After systemic corticosteroid treatment,patients with GO showed significantly lowered 8-OHdG level as compared with that before treatment and patients with Graves' disease.These changes in serum 8-OHdG level were accompanied by decreases in clinical activity score (P < 0.05) during corticosteroid treatment.Oxidative stress may play a role in the pathogenesis of GO.Serum 8-OHdG level may be used as an objective and quantitative parameter in patients with GO during immunosuppressive treatment.
8.Icarrin induces apoptosis of the thyroid carcinoma cell line B-CPAP via promotion of reactive oxygen species

Background and purpose:Icariin (ICA) is the important active flavonoids extracted from Berberidaceaeepimedium. It has been shown to be effective in suppressing cancers including lung cancer and gastric cancer. Thus, it is expected to be developed for cancer treatment. However, there were few studies on icariin as a promising anticancer drug for the treatment of thyroid cancer. The mechanisms underlying anticancer effects of ICA in thyroid cancer are rarely reported. This study was to explore the proliferation and apoptosis, intracellular ROS and antioxidant enzyme systems of the thyroid carcinoma cell line B-CPAP treated with different concentrations of ICA. It aimed to explore the mechanism underlying anticancer effects of ICA, and to determine whether it is concentration- or time-dependent manner.Methods:The proliferation of B-CPAP cell line treated with different concentrations of ICA was detected by cell counting kit-8 (CCK-8). The cell apoptosis and intracellular ROS were observed by flow cytometry. The expression of intracellular superoxide dismutase and intracellular malondialdehyde were measured by SOD detection kit and MDA assay kit, respectively. Bcl-2 and γ-HA2X were detected by Western blot.Results:ICA reduced B-CPAP cell activity, increased the rate of apoptosis in a dose- and time-dependent manner after 48 h (P<0.01). The ROS of ICA 50 mg/L and 200 mg/L groups were (2.12±0.14)-fold and (2.41±0.12)-fold of the control group, respectively. ICA promoted accumulation of malondialdehyde, and reduced antioxidant enzyme SOD activity. The SOD activity was decreased by (9.35±1.45)% (ICA 50 mg/L group) and (21.5±1.52)% (ICA 200 mg/L group) compared with the control group, respectively. The anti-apoptotic protein Bcl-2 in ICA 200 mg/L group was decreased by (13.64±1.71)%compared with the control group.Conclusion:Icariin inhibited activity of thyroid cancer B-CPAP cells in a dose- and time-dependent manner. It plays an important role in promoting intracellular ROS expression, inhibiting superoxide dismutase expression and decreasing Bcl-2, which leads to irreversible damage to the cell, thereby inducing apoptosis.
9.CHANGES ON EXPRESSION OF bcl-2,p53 AND CADHERIN IN ADULT HAMSTER SPERMATOGENIC CELLS DURING DES INDUCING ABNORMAL SPERMATOGENESIS

Objective To prove the mechanism of diethylstibestrol(DES) induced abnormal spermatogenesis,this study on the changes of proto-oncogenes bcl-2 and p53 proteins as well as cadherin adherens molecules expression in adult male hamster spermatogenic cells. Methods Adult male golden hamsters were treated daily olive oil vehicle either alone(control)or containing 1 mg/kg body weight of DES (experiment)by subcutaneous injection for 7 days.The changes of testes histological morphology were observed under light and electron microscope.The expressions of bcl-2 and p53 and cadherin in the testes seminiferous epithelium were detected by immunohistochemistry. Results DES induced adult hamster testes abnormal spermatogenesis.As compared to the control group animals,in DES-treated group animals the expression of both bcl-2 and p53 significantly increased in spermatogenic cells,especially in spermatocytes and round spermatids.However,the expression of cadherin adherens molecules obviously decreased.Conclusion It is one of reasons of DES inducing abnormal spermatogenesis that DES can result in increasing in the expression of proto-oncogenes bcl-2 and p53,and reducing cadherin adherens molecules expression.
10.Effects of four Chinese herb extracts on cell proliferation in IEC-6 cells of rats and their glucose absorption

Objective To study the effects of Chinese herb extracts on cell proliferation and glucose absorption in intestinal epithelial cell line(IEC-6 cells) of rats.Methods The extracts of four Chinese herbs,including Herba Agastaches(HA),Rhizoma Atractylodis(RA), Cortex Phellodendri(CP),and Gypsum Fibrosum(GF),were made.Their suitable concentration on the cell proliferation in IEC-6 cells was determined by MTT method,and glucose absorption and the activity of Na+,K+-ATPase in IEC-6 cells were assayed.A method of real time PCR was applied to the determination of SGLT1 and GLUT2 mRNA expression in the cells.Results Chinese herb extracts treatment altered the cell proliferation in a dose-dependent manner.Moreover,RA volatile oil(50 ?g/mL) and CP alkaloid(10 ?g/mL) treatment increased glucose absorption and the activity of Na+,K+-ATPase genes(P0.05).Conclusion The three extracts treatment,including RA volatile oil,CP aklaloid,and HA volatile oil,could increase glucose absorption and the expression of glucose transport carrier genes,but their regulative mechanism are not totally the same.
