By observing the changes in LDH、AST、CHE、K、Na+ contents in supernatant of cultured rat pituitary cells after electromagnetic pulse (EMP) irradiation the injury mechanism to pituitary cells was explored. Pituitary cells of Wistar rat was cultured in 6-hole plates. The cells in five plates were irradiated with high field strength EMP ( electromagnetic pulse) for 5 times within 2 minutes. The electromagnetic pulse simulator provided a high electric field intensity of 60 KV/m, with rise time 20-nsec and pulse wide 30/?S. Contents of LDH, AST, CHE, K+ and Na+ in the supernatant were measured using reagent boxes (Beijing Zhongsheng High-tech Bioengineering Company) at Oh, 1h, 6h, 12h and 24h following irradiation, respeetively. All data were analyzed by statistical software Spss 8. 0. The results showed that LDH and CHE contents in the supernatant increased significantly at Oh after irradiation with EMP (P 0. 05). Our conclusion is EMP could injure membranes of pituitary cells.

By observing apoptosis and morphological changes in primary culture of neurons from cerebellar granular layer, the neurons were irradiated with EMP (electromagnetic pulses), the possible injury mechanism was explored. The changes in death and apoptosis of the neurons were detected by MTT and flow cytometry, respectively. The slides stained with HE and TUNNEL were examined with light and fluorescence microscopy. The results showed that not only death of the neurons occurred immediately, but also apoptosis appeared after irradiation with EMP. Our conclusion is EMP can promote necrosis and apoptosis of neurons from cerebellar granular layer at early stage, which may result from DNA injury induced by EMP.

AIM To prepare the hydroxypropyl-β cyclodextrin inclusion compound of volatile oil from Houttuyniae Herba.METHODS In the content determination of volatile oil by GC,2-undecanone was taken as a reference substance, and inclusion rate and drug loading were calculated. After the determination of optimal method for preparing the inclusion compound among techniques of mixing, grinding and ultrasonic methods.the preparation process was optimized by orthogonal test with ratio of volatile oil to hydroxypropyl-β cyclodextrin, inclusion temperature, mixing speed and inclusion time as influencing factors, and inclusion rate and drug loading as evaluation indices. The characterization was performed by TLC,infrared spectroscopy and differential scanning calorimetry. Then high temperature, high humidity and strong light tests were applied to investigating the inclusion compound stability.RESULTS Mixing method brought forth the maximal yield, inclusion rate and drug loading for the inclusion compound. And under the optimal conditions of 1 ∶ 25 for ratio of volatile oil to hydroxypropyl-β cyclodextrin,50 ℃ for inclusion temperature,420 r/min for mixing speed, and 80 min for inclusion time, the inclusion rate and drug loading were identified to be 77.35% and 4.48%,respectively.The obtained inclusion compound was found to be white powder with loose texture and significantly increased solubility, the in vitro accumulative release rate reached 80.85%.Deliquescence and agglomeration,as well as obviously decreased inclusion rate and drug loading, were observed at a relative humidity of more than 75%,despite the good stability at high temperature (60 ℃) and strong light (3 000 lx),and yet the rehydration character was good.CONCLUSION It is possible that the hydroxypropyl-β cyclodextrin inclusion compound of volatile oil from Houttuyniae Herba generates a new phase rather than a simple mixture, which should be kept dry and sealed at the time of storage.

Objective To explore the prognosis and its related factors of three-dimensional conformal radiotherapy (3D-CRT) for elderly patients with esophageal carcinoma. Methods From January 2001 to August 2007, 89 elderly patients aged 65 years or more with esophageal carcinoma were treated with 3D-CRT. And the local control rates, survival rates and its related prognostic factors were evaluated retrospectively, using SSPS11.5 software. Results The 1-, 2-, 3-and 4-year local recurrence-free survival rates were 65.2%, 43.0%, 27.3% and 22.3%, respectively. The 1-,2-, 3-and 4-year overall survival rates were 68.5%, 48.8%, 36.8% and 25.2%, respectively. And the median survival time was 21.5 months. With univariate analysis, the significant prognostic factors included the different diet before radiotherapy (χ~2=6.26, P=0. 012), T stage (χ~2=8. 80, P=0. 003), N stage (χ~2=4.33, P=0. 038), clinical stage (χ~2=7.88, P=0. 005), the largest diameter of tumor in CT scanning image (χ~2=10.88, P=0.004), recent efficacy (χ~2=5.28, P=0. 022) and chemotherapy (χ~2=5.49, P=0.019). And the factors which were not related with prognosis included gender (χ~2=0.74, P=0. 390), age (χ~2=1.89, P=0. 170), lesion length showed by esophageal angiography (χ~2=2. 38, P=0.123), lesion length showed by CT (χ~2=2.69, P=0.101),primary tumor site (χ~2=2.12, P=0. 146), coexistent disease (χ~2=0. 03, P=0. 874), exposure mode (χ~2=0.04, P=0.847) and radiation dose (χ~2=0. 24, P=0. 627). Multivariate analysis revealed that the different diet before treatment (P=0. 002), the clinical stage (P=0. 007) and chemotherapy (P=0. 011) were independent prognostic factors. Radiation induced esophagitis was observed in 20 cases in grade 0, 30 cases in grade 1, 32 cases in grade 2, 7 cases in grade 3. And acute radiation pneumonitis was observed in 89 patients, with 57 in grade 0, 20 in grade 1, 8 in grade 2, 2 in grade 3 and 2 in grade 5. Until the end date of follow-up, 60 patients had died: 29 cases died of local recurrence or uncontrolled disease, 11 cases died of metastasis, 5 cases died of local recurrence with metastasis, and 15 cases died of other reasons. Conclusions 3D-CRT can be considered as an effective and feasible approach for elderly patients with esophageal cancer. The 3D-CRT provides them with less obstruction of diet, early clinical stage and longer survival time. 3D-CRT combined with chemotherapy may improve survival rate.

Objective To investigate the relationship among Helicobacter pylori (Hp) infection, serum gastrin level and colorectal carcinoma. Methods Fasting serum samples from 47 consecutive patients with colorectal carcinoma and 30 demoographically matched colonoscopy-negative controls were detected for serum gastrin levels and IgG antibodies against Hp (Hp IgG) by enzyme linked immunosorbent assay. Some of them accepted 14C urease breath test and/or rapid urease test after undergoing endoscopy. Hp infection was affirmed when at least two of the three tests were positive. Results The Hp infection incidence was significantly higher in patients with eoloreetal carcinoma (61.7%, 29/47) as compared with controls (36.7%, 11/30) (P<0.05). The Hp infection incidence in distal distribution of carcinoma was higher than that in controls, and in female carcinoma was higher than that in female controls(P <0.05). Mean fasting serum gas-trin levels were significantly higher in patients with coloreetal carcinoma[(139.7±53.3 ) ng/L]than those in controls [(89.5±26.1 ) ng/L] (P < 0.01 ). There was a significant association between hypergastrinemia and distal distribution of carcinoma. Conclusion Increasing serum gastrin levels, higher Hp infection incidence has a positive relationship with colorectal carcinoma.
