1.Explore and Analyse Professor Li Guixian’s Academic Idea of Treatment of Ulcerative Colitis

Objective] To analyse the academic thought of Professor Li Guixian, the famous veteran doctor of TCM and academic experience instructor of treating ulcerative colitis. [Methods] In-depth analyse the pathogenesis of ulcerative colitis, explore the academic connotation of Professor Li Guixian treatment of ulcerative colitis based on traditional Chinese medicine theory, according to the ancient classics. [Results] Professor Li Guixian puts forward the viewpoint that pathogenesis in the process of evolution of ulcerative colitis has a great relationship with the fundamental of spleen and stomach, symptoms of damp and heat-toxin, the outline of Qi and blood ,not only in line with the characteristics of the occurrence and prognosis of the disease but also has its theoretical basis ,combined with her many years of clinical experience. [Conclusion] The way of “invigorating spleen to help transport and transform the essence of water and grain, regulating the function of qi and blood, removing the muddle of damp and heat-toxin”of treating ulcerative colitis has clinical significance.
2.Investigation of Mental Health of College Freshmen by UPI in Medical Freshman

Objective: To get the information about freshmen's mental health and it relationship with sex, city and countryside difference, and to provide the reference for effective mental health education and consultation. Methods: The situation of 1351 freshmen's mental health was surveyed by using UPI. Results: ①11.91% of the freshmen had serious mental derangement, 26.15% of them had a little psychological problem. ②The distinct difference existed not only between male and female, but also between city and countryside. The mental health of male students surpassed the female students, the city students surpassed the countryside students. Conclusion: The situation of medical freshmen's mental health should not be neglected, and its influence factors are various. Mental health education and consultation should be pointed to the students.
3.Changes of expression levels of vascular endothelial growth factor and its receptors in diabetes rats with cerebral infarction

Objective To study the changes of expression levels of vascular endothelial growth factor(VEGF) and its receptors(FLT-1,FLK-1) mRNA in equal area of penumbra in diabetes rats with cerebral infarction(DMCI).Methods Middle cerebral artery occlusion(MACO) model was made in diabetes rats.The expression levels of VEGF and FLT-1,FLK-1 mRNA in equal area of penumbra were measured with situ hybridization in different times after MACO(1 h,3 h,6 h,12 h,24 h).The difference was compared between DMCI group and no diabetes cerebral infarction(NDMCI) group.Results The expression level of VEGF mRNA was lower in DMCI group than that of NDMCI group at every time point(P
4.Analysis on Utilization of Antibiotics and Non-steroidal Anti-inflammatory Drugs in 3 Hospitals of Guang’an from Jan. 2005 to Jan. 2008

OBJECTIVE: To evaluate application of antibiotics and non-steroidal anti-inflammatory drugs in 3 hospitals of Guang’an and find out the reason of unreasonable use of drug. METHODS: 120,000 prescriptions were randomly collected from inpatient and outpatient departments of 3 hospitals from Jan. 2005 to Jan. 2008, which included 20 000 pediatrics prescriptions. The DDDs and DUI of antibiotics and non-steroidal anti-inflammatory drugs were analyzed statistically. RESULTS: The DUI of 10 kinds of antibiotics and 3 kinds of non-steroidal anti-inflammatory drugs were above 1, which indicated that drug abuse occurred. CONCLUSION: Physicians’ prescribing behavior lack medical rationality and the situation of drug feeding hospital is still critical in primary hospitals.

6.Effects of Shugan granule on hepatic stellate cells activation and its trans-membrane signal transduction stimulated by transforming growth factor beta 1

BACKGROUND: Hepatic fibrosis is a reversible disease, interfering in course of disease promptly can decrease hepatic cirrhosis and fatal complication. Hepatic stellate cells (HSCs) is a key factor in pathogenesy of hepatic fibrosis.OBJECTIVE: To investigate the effects of Shugan granule on HSCs activation and trans-membrane signal transduction stimulated by transforming growth factor beta 1 (TGF-β1) in rats, and to explore the anti-fibrosis mechanism.DESIGN, TIME AND SETTING: Controlled observational trials based on cytology were performed in the Central Laboratory of Molecules, Luzhou Medical College between June 2008 and February 2009. MATERIALS: HSC-T6 cell line was purchased from Institute of Liver Diseases, Shanghai University of Traditional Chinese Medicine, its phenotype was the activated hepatic stellate cells. Shugan granule was offered by Drug Institute, Affiliated Hospital of Luzhou Medical College at a Batch No. 20071120.METHODS: The influence of different concentrations (0.01, 0.02, 0.04, 0.08 g/L) of Shugan granule on HSCs proliferation was determined by MTT. 0.01 g/L was defined as the dose of Shugan granule contributing no influence on cell proliferation. HSCs were cultured in a culture plate and then divided into 4 groups: control group without management, TGF-β1 group with 5 μg/L TGF-β1 solution in culture medium, Shugan granule group with 0.01 g/L Shugan granule in a culture medium, and TGF-β1 + Shugan granule group with 5 μg/L TGF-β1 solution and 0.01 g/L Shugan granule in culture medium. MAIN OUTCOME MEASURES: Morphological features of HSCs were detected by microscopic. Immunohistochemical method was used to detect the expression of Smad3 and Smad7 in HSCs. RT-PCR was applied to observe the HSCs activation and trans-membrane signal transduction. RESULTS: ①The cell morphology of TGF-β1 group was similar with that in the control group, and the extension was more obvious. In the TGF-β1 + Shugan granule group, the cell morphology was close to that in TGF-β1 group. There was no karyopyknosis or apoptosis observed in each group. ②Immunohistochemical method showed the expression of Smad3 and Smad7 in control groups were increased. TGF-β1 could slightly increase the expression of Smad3 and Smad7 (P < 0.05), while Shugan granule group and TGF-β1 + Shugan granule group increased the expression of Smad7 significantly, accounting for 1.99 times compared with control group (P < 0.01). ③RT-PCR result showed that Shugan granule could increase the expression of Smad7 (P < 0.05), but the expression of Smad3 was not regulated. 5 μg/L TGF-β1 could up-regulate the expression of Smad3 and Smad7 (P < 0.05). In the TGF-β1 + Shugan granule group, Smad7 expression was increased by 101% (P < 0.05), but Smad3 transcriptional level was not changed(P > 0.05). CONCLUSION: ①TGF-β can stimulate the gene expression of Smad3 and Smad7, it also obtain a balance of feedback regulation mechanism between R-Smads and I-Smads. ②Shugan granule may prevent and cure hepatic fibrosis through decreasing the proliferation of HSCs in a dose-dependent manner. ③Shugan granule can inhibit the TGF-β-Smad signaling pathway through increasing the expression of Smad7.
7.Malnutrition related factors in patients with gastric cancer

Malnutrition is one of the most important factors for the prognosis of patients with gastric cancer,but the mechanism of malnutrition is unclear at present. These patients are at risk of nutritional deple-tion from anorexia,dysphagia,and may be associated with the presence of two or more of the following cyto-kines with metabolic activity such as TNF-α,IL-1,IL-6,interferon,leptin,nuclear factor-κB and so on, which are secreted by the host-tumor interaction. The cytokines constitute a sophisticated regulatory network. Therefore,the study of the relationship between malnutrition and related factors in patients with gastric cancer may improve the nutritional status and prognosis of these patients.
8.Effects of Different Mechanical Ventilation Parameters on Intra-Abdominal Pressure in COPD Patients

Objective To discuss the effect of different mechanical ventilation parameters on intra-abdominal pres-sure. Methods A total of 42 cases with mechanical ventilation suffering from chronic obstructive pulmonary diseases (COPD) and respiratory failure were selected for observing the effects of inspiratory platform time (Tplat), inspiratory time (Ti) and end-expiratory pressure (PEEP) on intra-abdominal pressure. Results In SIMV+PSV ventilation mode, changes of Tplat and Ti had no significant effects on intra-abdominal pressure (P>0.05). There were no significant differences be-tween SIMV+PEEP0 and base line values (P>0.05). Conclusion The alteration of peep has a great influence in intra-ab-dominal pressure, which suggests the importance of selecting proper mechanical ventilation parameters.
9.Effects of Different PEEP on Abdominal Pressure and Liver in COPD Patients

Objective To research the impact of various PEEP parameters of mechanical ventilation on liver mor-phology and function and on hemodynamics. Methods Fifty patients of respiratory failure due to COPD using mechanical ventilation was observed by prospective, exoterical and self-control methods after their condition were stabilized. The varia-tion of abdominal pressure was measured with different PEEP of 0 cmH2O, 5 cmH2O and 10 cmH2O with mechanical ventila-tion, and measured when patients breathe autonomously with tracheostomy tube. The changes of liver morphology, hepatic por-tal vein diameter, hemodynamics and liver function were also measured on above circumstances. Results With PEEP value increases:the abdominal pressure (IAP) rise (P<0.01) while liver morphology, function and hepatic portal did not change markedly evidently (P>0.05). By contrast, blood flow of hepatic portal vein reduced with PEEP increase (P<0.05). Conclu-sion Higher PEEP values on mechanical ventilation will causes abdominal pressure increases and low blood flow of hepatic portal , but not obvious impact on portal vein diameter and liver function.
10.Large-scale expansion of human umbilical cord mesenchymal stem cells in human platelet lysate as a substitute of fetal bovine serum

BACKGROUND:Classic media and fetal bovine serum are commonly used in the culture of umbilical cord mesenchymal stem cells, but the potential risk of serum culture limits its clinical application. OBJECTIVE:To use human platelet lysate alternative to fetal bovine serum for large-scale production of mesenchymal stem cells for therapeutic applications. METHODS:Human platelet lysate was prepared by repeated freezing and thawing, centrifugation, filtration, and concentration. Human umbilical cord mesenchymal stem cells were cultured in Iscove’s Modified Dulbecco’s Medium containing 5%concentrated platelet lysate (experimental group) or 10%fetal bovine serum (control group). After separation and enzymatic digestion, human umbilical cord mesenchymal stem cells were subcultured at a concentration of 3 000/cm2 up to the fifth generation. Then, cellmorphology and diameter, immune phenotype, osteogenic and adipogenic differentiation, and cloning efficiency were detected and compared between two groups. RESULTS AND CONCLUSION:Human umbilical cord mesenchymal stem cells cultivated in human platelet lysate-supplemented media showed a smal er size and more elongated morphology than those in fetal bovine serum-supplemented media. Colony forming unit-fibroblast analyses further showed no significant differences in colony efficiency. Human umbilical cord mesenchymal stem cells cultivated in human platelet lysate-supplemented media showed an increase of proliferation capacity;whereas, similar immunophenotypes remained in the two groups. In vitro assays revealed intact differentiation potential. Moreover, human umbilical cord mesenchymal stem cells cultivated in human platelet lysate-supplemented media showed a significantly higher capacity to differentiate towards osteocytes, indicating human platelet lysate is an alternative to fetal bovine serum for low-density production of mesenchymal stem cells for therapeutic applications.
