1.Observation of effect of DJL-1200B semiconductor laser machine in treatment of 145 children with secretory otitis media

Objective To observe the effect of DJL-1200B semiconductor laser machine in treatment of children with secretory otitis media.Methods 145 children with secretory otitis media treated in our department from March 2009 to March 2013 were selected as the observation group.116 cases with secretory otitis media from October 2004 to October 2008 were selected as the control group.Medical treatment was applied in both two groups,and laser treatment was added in observation group.The effect was evaluated after treatment for 7 days.Results The total effective rate of the observation group was 93.10%,which was significantly higher than 80.17% of the control group (x2 =51.361,P < 0.05).Conclusion The clinical effect of laser treatment added on drug treatment in the treatment of children with secretory otitis media is good,and it has high security and less pain,which is worthy of clinical application.
2.Study on the differences of nerve conduction of lower limbs in general population with different nationalities in Xining region

Objective To explore the differences of nerve conduction of lower limbs in general population with different nationalities in Xining region.Methods Physical examination and healthy volunteers in No.1 People's Hospital of Xining were collected.Two hundred and forty Hui people of them was Hui group.Two hundred and forty people with other ethnic minorities including Tibetan,Tu and Mongolian of them was mixed group.Two hundred and forty Han people of them was Han group.The nerve conduction of lower limbs in all the people were detected.And the differences were compared.Results The differences of nerve conduction of posterior tibial nerve among all the groups were not statistical significant (all P>0.05).The latent period of common peroneal nerve in Hui group was significantly longer than mixed group and Han group,the motor conduction velocity of which was significantly slower,the motor nerve action potential of which was significantly reduced (all P<0.05).The latent period of superficial peroneal nerve in Hui group was significantly longer than mixed group and Han group,the sensory conduction velocity of which was significantly slower,the sensory nerve action potential of which was significantly reduced (all P<0.05).The differences of nerve conduction of sural nerve among all the groups were not statistical significant (all P>0.05).The differences of nerve conduction of each group among different age periods were statistical significant(all P<0.05).Conclusions The nerve conduction of lower limbs in general Hui people in Xining region must be different with other general people.It must be noticed in clinical examination.
3.Changes of retinoic acid level in guinea pig sclera of early form deprivation myopia

Objective Researches showed that retinoic acid may be a signal molecular in the regulation of ocular axial length.Sclera is a target tissue of the pathogenesis of form deprivation myopia.Recently,guinea pigs are used to the study of the pathogenesis of form deprivation myopia.However,seldom studies are found in the change of retinoic acid level in the sclera of guinea pigs.This study was designed to investigate the role of retinoic acid of early form deprivation myopia in guinea pigs sclera.MethodsMonocular form deprivation models were established in 24 3-week-old guinea pigs by occluding the left eyes with facemask and randomly divided into form deprivation 15 days group and 24 days group.Other 6 normal guinea pigs were used as control group.The fellow eyes were used as internal control.Refraction was measured before and after experiment by using retinoscopy.Eyeballs were extracted immediately at the corresponding time points and ocular axial length was measured with outside micrometer.The level of retinoic acid in scleral samples was detected by high performance liquid chromatography.The use of animal followed the Regulations for the Administration of Affairs Concerning Experimental Animals by the State Science and Technology Commission.ResultsRefractive diptor reduced by -3.14±1.50 D in experimental eyes after form deprivation,showing statistically differences among before and various time points after form deprivation(F=23.053,P=0.000),and no significant difference was seen in reduce of refractive diptor in fellow eyes after form deprivation( -1.24±1.99 D)(F=1.506,P=0.240).The axial length was considerably increased in form deprivation eyes after experiment (F=14.551,P=0.000) but no evident change in fellow eys (F=3.612,P=0.060).The diopter and ocular axial length was significantly different between form deprivation eyes and fellow eyes (P>0.05).The expression of retinoic acid in sclera of form deprivation eyes was higher than fellow eyes in both form deprivation 15 days and 24 days(P<0.01).There was no significant difference in the level of retinoic acid between form deprivation 15 days and 24 days(P>0.05).ConclusionForm deprivation myopia can be induced in 15 days after monocular occlusion.Retinoic acid levels in guinea pig sclera increased significantly during early form deprivation.
4.Effect of "Dispersing Lung-qi and Relieving Asthma,Warming Yang to Induce Diuresis" Method on ADH of CPHD Patients during Acute Period with Peripheral Edema

Objective To evaluate the effectiveness of "dispersing lung-qi and relieving asthma, warming Yang to induce diuresis" method on CPHD patients during acute period with peripheral edema and observe the effect on ADH. Methods The CPHD patients during acute period with peripheral edema were randomly divided into treatment group and control group. The treatment group received Chinese medicine adding to regular western medicine, the principle is dispersing lung-qi and relieving asthma, resolving phlegm to stop coughing, warming Yang to induce diuresis. The control group received placebo adding to regular western medicine. Both of the groups were treated for 14 days. The effect observed indexes were as follows:general curative effect, integral of main symptoms of TCM, integral of western medical signs, serum electrolyte, etc. Mechanism index was about ADH. Results The treatment group had better effects than the control group on the general curative effect, reducing integral of symptoms and signs, rectifying electrolyte and acid-alkaline-balance disturbances, etc. The safety analysis of two groups had no significant abnormality. Meanwhile, the treatment group had more superiority than the control group on reducing serum ADH. Conclusion The comprehensive treatment of traditional Chinese and western medicine probably has more advantage in increasing the general effect, improving symptoms and signs, rectifying electrolyte and acid-alkaline-balance disturbances, etc. This therapeutic method can restrain secretion of ADH. It indicates that restraining secretion of ADH maybe part of the mechanism of using "dispersing lung-qi and relieving asthma, warming Yang to induce diuresis" method to treat CPHD patient during acute period with peripheral edema.
5.Protein microarray for simultaneous detection of pathogens in blood transfusion

Objective To study the diagnostic value of multiplex protein microarray and ELISA for human serum pathogens.Methods Totally 230 national standard samples were tested by multiplex protein microarray and ELISA respectively for the pathogens of HBV,HIV,HCV and treponema pallidum(TP).Results The matched pair test for numeration data showed ?2=0.5,P=0.125,indicating that no statistical significance between the two methods.Conclusion Multiplex protein microarray shows high specificity and sensitivity to the four pathogens so that it is worth extensive application.
6.Rapid detection of TSH and T_4 by microfluidic chip-based immunoassays

Objective To establish an immunoassay based on microfluidic chip for detection of TSH and total T4.Methods The direct-write laser micromachining technology was used to fabricate microfluidic chip,luminol-H2O2 as luminator.This method and Roche E2010 electrochemiluminescence immunoassay were used to examine TSH by double-antibody sandwich method and total T4 by competitive principle in 50 serum samples collected at our outpatient clinic.Results The detection was completed within 25 min.Ten microliter sample and reagent was required.The detection range of TSH was 0-50 ?IU/ml and its average intraassay and interassay precision was 4.7% and 4.6% respectively and its average recovery rate was 95.3%.The detection range of T4 was 9.375-300 ng/ml,and its average intraassay and interassay precision was 4.9% and 4.7% respectively and its average recovery rate was 91.27%.Conclusion Rapid detection of TSH and T4 by microfluidic chip-based immunoassays is convenient,stable and accurate and of less reagent and time consuming.
7.AN IMMUNOELECTRON MICROSCOPIC STUDY ON THE SYNAPTIC RELATIONSHIP BETWEEN DOPAMINERGIC AND GABAergic NEURONS IN THE BASOLATERALNUCLEUS OF THE RAT AMYGDALA

Objective To explore the relationship between dopaminergic axon terminal and GABAergic neurons and explore the neuroanatomic mechanism of their effects in schizophrenia. Methods The co-location and the association of dopaminergic axon terminal and GABAergic neurons in the basolateral nucleus (BL) of rat amygdala were examined by using double labeling immunoelectron microscopic techniques. Dopaminergic axon terminal and GABAergic neurons were labeled with the antidopamine (anti-DA) and the anti-glutamic acid decarboxylase (anti-GAD) antibodies respectively. Results 43% of the DA-input synapses was observed to relate directly or indirectly to GAD-immunoreactive(IR) dendritic structures(DA/GAD), which includes single (38%), convergent (30%), serial (20%), and axoaxonic (12%) contact types.And 57% of the DA-input synapses was found to associate with unlabeled elements (DA/UE), which includes unlabeled perikarya (10%), dendrites (82%) and axons (8%). All of the synapses that show DA-IR terminals profiles were found to be symmetric (inhibitory) synapses.Conclusion These results suggest that the dopaminergic system in the BL of rat amygdala controls the mediations of the GABAergic interneurons via symmetric synapses. In addition, the dopaminergic axon terminal associates with the glutamatergic projection neurons and exerts influence on its activity.
8.An Investigation on the Mental Health of Heroin Addicts

Objective:To study the mental health status of heroin addicts and their need for psychotherapy after detoxification. Methods:The mental health of 83 heroin addicts and 70 normal subjects were assessed by SCL-90, Social Support Rating Scale , EPQ, SAS and SDS. Results:As compared to normal controls, heroin additions were found to score higher on various mental health measures, including SCL-90 total and factor scores, the N score of EPQ, as well as total scores on SAS and SDS. The social support in heroin addicts scored lower than normal subjects. Conclusion:Heroin addicts have severe psychological problems, which warrant long-term psychological rehabilitation after detoxification.
9.Effects of endogenous carbon monoxide/nitric oxide on cholecystokinin octapeptide reversed vascular hyporeactivity in endotoxemic rats

AIM: To explore the effects of cellular signal transduction pathways of heme oxygenase-1(HO-1)-carbon monoxide (CO)-cyclic GMP (cGMP) and nitric oxide synthase (NOS)-nitric oxide (NO)-cGMP on cholecystokinin octapeptide (CCK-8) reversed vascular hyporeactivity in endotoxemic rats. METHODS: According to the treatments given in vivo , rats were devided into four groups: control; lipopolysaccharide (LPS); CCK and CCK+LPS. Using isolated vascular ring tension detecting technique, thoracic aortic rings (TARs) were prepared and accumulation of contractive responses to phenylephrine (PE) were measured under which the TARs were incubated with Hemin (He, donor of CO), Zinc-protoporphyrin-IX (ZnPP-IX, selective inhibitor of HO-1), L-arginine (L-Arg, substrate of NOS), aminoguanidine (AG, selective inhibitor of iNOS), N ?-nitro-L-arginine (L-NNA, inhibitor of NOS) or methylene blue (MB, inhibitor of guanylyl cyclase), respectively. RESULTS: CCK-8 alone did not affect vascular tension. Injection of LPS induced the hyporeactivity of the TARs and was reversed by pretreatment of CCK-8. In LPS and CCK+LPS groups, the hyporeactivity was partly reversed by incubation of TARs with ZnPP-IX or AG, and restored to normal by incubation of TARs with L-NNA or MB. Incubation of TARs with He or L-Arg showed to make the vascular hyporeactivity worse in different degree. CONCLUSIONS: CCK-8 alone did not affect the activity of HO-1 and iNOS but influenced the activity of these enzymes induced by LPS, which lead to reduced CO/NO production, decreased the content of cGMP and plays its important role in reversing vascular hyporeactivity in endotoxemic rats.
10.A study on correlation between risk factors of cerebral vascular disease(CVD) and cognitive deficit associated with cerebral infarction

Objective To study the correlation between risk factors of CVD and cognitive deficit associated with cerebral infarction. Methods Cognitive function was tested in 101 cases of cerebral infarction using HDS-R at the end of 3 weeks and 6 months after onset of the stroke. Meanwhile, the data of risk factors and relative information of cerebral vascular disease were collected by using the scale of complications and inquiring the past history. The risk factors of CVD, such as neurological deficit, diabetes, hypertension, obesity, fever, heart diseases, smoking, overdrinking were compared between the cognitive deficit group and non-cognitive deficit group. Logistic regression analysis was adopted for data analysis. Results Of the 101 cases , 65 had cognitive deficit at the end of 3 weeks and 53 at the end of 6 months after onset of stroke. Age and score of scale of complications between cognitive deficit group and non cognitive deficit group were significantly different(P
