2.Effect of subretinal injection of retinal pigment epithelial cells for retinal pigment epithelium in mice

Objective To observe the effect of subretinal injection of retinal pigment epithelium (RPE) cells for RPE in mice.Methods A total of 30 postnatal day 7 C57BL/6J mice were randomly divided into normal mice group,OIR model group and OIR model cell transplanted group,10 mice in each group.The OIR model was induced in mice of OIR model group and OIR model cell transplanted group.The RPE cells were subretinal injected into the RPE of mice in OIR model cell transplanted group.At 20 days after the injection,the RPE thickness was evaluated by fluorescence microscope.The expression of RPE65,Bestrophin and zonula occludens-1 (ZO-1) were estimated by Western blot and real-time quantitative PCR (RT-PCR).Results The thickness of RPE in OIR model mice was thinner than that in normal mice;the thickness of RPE in OIR model cell transplantation mice was significantly thicker than that in the OIR model mice.The results of Western blot and RT-PCR indicated that the differences of protein (F=8.597,18.864,25.691) and mRNA expression (F=39.458,11.461,34.796) of RPE65,Bestrophin,ZO-1 were statistically significant between OIR model group and OIR model cell transplanted group (P<0.05).Conclusions Subretinal injection of RPE cells can promote RPE thickening.RPE65 and Bestrophin protein relative expression levels increased,ZO-1 protein relative expression levels reduced;mRNA expression levels of RPE65,Bestrophin and ZO-1 genes increased.
3.Effects of Different Mechanical Ventilation Parameters on Intra-Abdominal Pressure in COPD Patients

Objective To discuss the effect of different mechanical ventilation parameters on intra-abdominal pres-sure. Methods A total of 42 cases with mechanical ventilation suffering from chronic obstructive pulmonary diseases (COPD) and respiratory failure were selected for observing the effects of inspiratory platform time (Tplat), inspiratory time (Ti) and end-expiratory pressure (PEEP) on intra-abdominal pressure. Results In SIMV+PSV ventilation mode, changes of Tplat and Ti had no significant effects on intra-abdominal pressure (P>0.05). There were no significant differences be-tween SIMV+PEEP0 and base line values (P>0.05). Conclusion The alteration of peep has a great influence in intra-ab-dominal pressure, which suggests the importance of selecting proper mechanical ventilation parameters.
4.Changes of retinoic acid level in guinea pig sclera of early form deprivation myopia

Objective Researches showed that retinoic acid may be a signal molecular in the regulation of ocular axial length.Sclera is a target tissue of the pathogenesis of form deprivation myopia.Recently,guinea pigs are used to the study of the pathogenesis of form deprivation myopia.However,seldom studies are found in the change of retinoic acid level in the sclera of guinea pigs.This study was designed to investigate the role of retinoic acid of early form deprivation myopia in guinea pigs sclera.MethodsMonocular form deprivation models were established in 24 3-week-old guinea pigs by occluding the left eyes with facemask and randomly divided into form deprivation 15 days group and 24 days group.Other 6 normal guinea pigs were used as control group.The fellow eyes were used as internal control.Refraction was measured before and after experiment by using retinoscopy.Eyeballs were extracted immediately at the corresponding time points and ocular axial length was measured with outside micrometer.The level of retinoic acid in scleral samples was detected by high performance liquid chromatography.The use of animal followed the Regulations for the Administration of Affairs Concerning Experimental Animals by the State Science and Technology Commission.ResultsRefractive diptor reduced by -3.14±1.50 D in experimental eyes after form deprivation,showing statistically differences among before and various time points after form deprivation(F=23.053,P=0.000),and no significant difference was seen in reduce of refractive diptor in fellow eyes after form deprivation( -1.24±1.99 D)(F=1.506,P=0.240).The axial length was considerably increased in form deprivation eyes after experiment (F=14.551,P=0.000) but no evident change in fellow eys (F=3.612,P=0.060).The diopter and ocular axial length was significantly different between form deprivation eyes and fellow eyes (P>0.05).The expression of retinoic acid in sclera of form deprivation eyes was higher than fellow eyes in both form deprivation 15 days and 24 days(P<0.01).There was no significant difference in the level of retinoic acid between form deprivation 15 days and 24 days(P>0.05).ConclusionForm deprivation myopia can be induced in 15 days after monocular occlusion.Retinoic acid levels in guinea pig sclera increased significantly during early form deprivation.
5.Effects of Different PEEP on Abdominal Pressure and Liver in COPD Patients

Objective To research the impact of various PEEP parameters of mechanical ventilation on liver mor-phology and function and on hemodynamics. Methods Fifty patients of respiratory failure due to COPD using mechanical ventilation was observed by prospective, exoterical and self-control methods after their condition were stabilized. The varia-tion of abdominal pressure was measured with different PEEP of 0 cmH2O, 5 cmH2O and 10 cmH2O with mechanical ventila-tion, and measured when patients breathe autonomously with tracheostomy tube. The changes of liver morphology, hepatic por-tal vein diameter, hemodynamics and liver function were also measured on above circumstances. Results With PEEP value increases:the abdominal pressure (IAP) rise (P<0.01) while liver morphology, function and hepatic portal did not change markedly evidently (P>0.05). By contrast, blood flow of hepatic portal vein reduced with PEEP increase (P<0.05). Conclu-sion Higher PEEP values on mechanical ventilation will causes abdominal pressure increases and low blood flow of hepatic portal , but not obvious impact on portal vein diameter and liver function.
6.Effects of Exogenous Methionine on Arsenical Distribution in Female Mice Exposed to Sodium Arsenite through Drinking Water

Objective To explore the effects of exogenous methionine on arsenical distribution in liver,blood and brain of mice exposed to sodium arsenite through drinking water. Methods The female Kunming mice were randomly divided into control group, the alone arsenic exposure group , the low level methionine intervention group, the moderate level methionine intervention group and the high level methionine intervention group, eight mice in each group . The mice in the experimental groups were exposed to sodium arsenite through drinking water at 50 mg/L arsenic for four consecutive weeks. And at the fourth week the 5 groups were treated intraperitoneally with saline solution (control and As group),100 mg/kg b.w,200 mg/kg b.w or 400 mg/kg b.w methionine,respectively . Twenty-four hours after cessation of methionine administration,mice were anaesthetized and rapidly dissected. The samples of blood,liver and brain were removed immediately for arsenic species analysis. Levels of inorganic arsenic (iAs),monomethylarsonic acid (MMA) and dimethylarsenic acid (DMA) were determined by HG-AAS method. Then total arsenic speciation( TAs), primary methylation ratio( PMR)and secondary methylation ratio( SMR)in each tissue were calculated. Objective To conduct a bibliometric analysis of pediatric literature in recent 5 years in Pubmed database,and to explore the focus issues of pediatrics.Methods Bibliographies from research literature of pediatrics in PubMed database from 2012 through 2016 were retrieved.The publication years,journals,the prolific authors and frequency of Mesh major topics were counted.MeSH heading/subheading matrix were formed.SPSS 19.0 statistical software was applied for clustering analysis.Results A total of 9375 articles were included.There were 45 MeSH heading/subheadings,which were clustered into 4 categories.Conclusion The research focuses in pediatrics are as follows:the clinical competence training and education research;the related pediatric issues from the perspective of social medicine;medical error prevention and the related legislation;the situation of Chinese pediatricians and doctor-patient relationship.
9.Expression and significance of SHP-2 in human papillomavirus infected cervical cancer.

This study investigated the expression and prognostic value of SHP-2 in cervical cancer caused by human papillomavirus (HPV) infection. Forty-five specimens from patients with cervical cancer (stage I-III), 32 specimens from patients with cervical intraepithelial neoplasia (CIN) (I, II) and 20 normal cervical samples from patients with hysteromyoma were collected in Department of Pathology for comparison. The expression levels of SHP-2 and IFN-β proteins were detected by using immunohistochemistry. The mRNA expression level of SHP-2 was detected by using quantitative real-time polymerase chain reaction (PCR). HPVs were detected by HPV GenoArray Test. The Spearman correlation was used to compare the expression level of SHP-2 in HPV infected cervical cancer vs non-HPV infected normal cervix. The level of SHP-2 protein expression in the cancer tissues (88.8%) was significantly higher than in CIN tissues (62.5%) and normal cervixes (45%) (P<0.05 and P<0.05, respectively). The SHP-2 mRNA levels in the cancer tissues were upregulated as compared with those in the normal cervixes (P<0.05). Twenty-one (46.7%) cervical cancers, 25 (78.1%) CINs and 17 (85%) normal cervixes showed IFN-β positive staining in cytoplasm. There was statistically significant difference in the expression rate of IFN-β between cervical cancer and normal cervix (χ (2)=8.378, P<0.05) as well as between cervical cancer and CIN (χ (2)=7.695, P<0.05). HPV16/18 infections could be found in normal cervixs (15%), CINs (68.7%) and cervical cancers (84.4%). There was a correlation between HPV infection and SHP-2 expression in cervical cancer (r (s)=0.653, P<0.05). SHP-2 may be a useful prognostic and diagnostic indicator for HPV infected cervical cancer. In cervical cancers, SHP-2 mRNA and protein overexpression was associated with IFN-β lower-expression.
10.Impact of health literacy on education retention of stroke patients

Objective To describe the health literacy level and to explore the effect of health literacy level on education retention of patients with stroke.Methods Totally 100 stroke survivors were selected as the subject by convenience sampling.They were evaluated by Short Test of Functional Health Literacy in Adults on admission,and they accepted 60-90 minutes education intervention during hospitalization.Evaluation retention were measured before discharge.Results The score of the overall health literacy was (15.60±1.36) points,among of them,low level was 37 cases,general level was 24 cases,high level was 39 cases.Age,education level and Mini-Mental Status Exam score had a significant effect on the health literacy of patients with stroke (x2/F=14.248,12.082,11.329,P< 0.05).After education intervention,the score of education retention was (6.69±2.00)points,and the score of education retention in low,general,high level health literacy were (5.08±2.06),(7.46±1.61),(7.51± 1.76) points,and there were significant differences(F=14.223,P < 0.05).Heahh literacy had a effect on education(P < 0.01).Conclusions The level of health literacy of stroke patients has the effect of improving the education retention.Improving and maintaining the health literacy of stroke patients should be the development direction of the prevention and treatment of stroke diseases.
