BACKGROUND: Studies have proved that rotating magnetic field(RMF) can significantly increase bone mineral density(BMD) in rats, and such high BMD maintains for a long time which is independent of the actions of hormone.OBJECTIVE: To investigative the effect of RMF on bone calcium content and the correlation between bone-specific alkaline phosphatase(BAP) and deoxypyridinoline crosslinks (DPD) in rats.DESIGN: A randomized controlled study in rats.SETTING: A college of life sciences of a university.MATERIALS: The experiment was conducted in Shenzhen Key Lab of Microbiological Genetic Engineering from March 2004 to October 2004, using 90 healthy adult SD rats including 60 females weighing(259 ± 70) g and 30 males weighing (351 ± 104)g. The rats were randomly divided into 9 groups including 6 ovariotomy groups, namely female sham-operation group, normal calcium control group, low calcium control group, low calcium experimental group, normal calcium experimental group, and low calcium group with traditional Chinese medicine(TCM) treatment, besides a male control group,male low calcium experimental group, and male normal calcium experimental group, with 10 rats in each group.INTERVENTIONS: The female rats in each group, except for those in the sham-operation group, received ovariotomy. The rats in the ovariotomy control group had only ovariotomy. After metabolic exhaustion of the remnant estrogen on the 15th day, the rats in the ovariotomy experimental group were treated with RMF for 2 hours once a day for 15 days. The male rats, except for those in the control group, all bad RMF in identical manner. The rats were given food with normal calcium(containing 0.26% calcium), lower calcium (containing 0.1% calcium) and traditional Chinese medicine(powder of Fructus Psoraleae, Radix Astragali, epimedium herb, and herba cistanchis) .After RME treatment, the rats were fed for another 15 days before the femur was collected to measure the bone calcium content. Serum samples were also obtained for measuring the content of BAP and urine samples collected to measure the content of DPD.MAIN OUTCOME MEASURES: The bone calcium content of each group and serum BAP and urine DPD.RESULTS: Of the 90 rats used, 4 died during the experiment and 86 entered the result analysis. The bone calcium content was increased after RMF treatment, and the content in rats in the low calcium group with ovariotomy and TCM treatment and lower calcium control group with ovariotomy was(0.226± 0. 015) and(0. 206 ± 0. 015) g/g, respectively( t =4.63, P ＜ 0. 05). The bone calcium content in the male normal calcium experimental group and male control group was(0. 206 ±0. 031) and(0. 199 ±0. 014) g/g respectively ( t = 4.21, P ＜ 0.05 ) . After treatment with RMF, serum BAP content was increased and was(20. 52 ± 1.78) and(15.68 ±3.68) U/L in normal calcium experimental and control groups with ovariotomy, respectively( t=4.76, P ＜0.05), and(17.69±3.78) and (8.53±2.54) U/Lin the male low calcium experimental and male control groups, respectively( t= 4. 59, P ＜ 0.05). Urine DPD after RMF treatment was (86.97 ± 37.19)and(401.57 ±79. 34) nmol/L in the low calcium experimental group with TCM treatment and ovariotomy and the low calcium control group with ovariotomy, respectively( t =7.45, P ＜ 0.01), and(97.87 ±31.97) and (168.71 ±53.19) nmol/L in the male normal calcium experimental and male control groups respectively( t = 8.31, P ＜ 0.01 ).CONCLUSION: RMF can effectively increase bone calcium content in a short time( 15 days), and the effect was positively correlated with increased blood BAP and decreased urine DPD.

Objective To investigate the protective effect of Ca 2+ channal blocker(Verapamil)on the testis following testicular torsion in rats.Methods Forty healthy male Sprague-Danley rats were randomly divided equally into five groups:group A(sham operation),group B1(the left testis underwent 720? positive torsion,2 h later detorsion was performed),group B2(the process as group B1 except at 30 min before detorsion,intraperitoneal injection of physiological saline was performed),group B3(the process as group B1 except at 30 min before detorsion,intraperitoneal injection of 2 mg/kg Verapami was performed),group B4(the left testis underwent 720? positive torsion,and 2 h later the left testis was excised).Five days after operation,the bilateral testes(the right testis in group B4)were extracted and nitric oxide(NO),nitric oxide synthase activity(NOS),total antioxygen capacity(T-AOC)were measured.Results After detorsion,NO and NOS activity increased greatly,T-AOC decreased significantly in bilateral testes,but more obvious in the torsive testis;Verapamil could inhibit the changes.Conclusion The excessive production of NO and the decreased T-AOC may be the main causes of the contralateral testicular injury after unilateral testicular torsion.Verapamil can reduce the testicular injury in bilateral testes after testicular torsion/detorsion in rats.The resection of the torsive testis has protective effect on the contralateral one.

Objective · To study the expression of microRNA-143 (miR-143) in human colon cancer and adjacent normal tissues, and to explore its effect on the function of colon cancer cells and downstream target genes. Methods · Lentiviral expression vector of miR-143 was designed and used to stably transfect colon cancer cell lines HCT116 and RKO. Real-time PCR was used to detect the expression of miR-143 in colon cancer, adjacent normal tissues, HCT116 cells and RKO cells infected with lentivirus. The apoptosis ratio of HCT116 cells and RKO cells was detected by Annexin V-APC single staining and flow cytometry. The migration ability of HCT116 cells and RKO cells was detected by Transwell method. The expression of FOSL2 protein was detected by Western blotting assay. Results · The expression of miR-143 in colon cancer tissue was significantly lower than that in adjacent mucosal tissue (0.339±0.454 vs 1.003±0.003) (U=16.000, Z=-4.231, P=0.000). By up-regulation of miR-143, apoptosis rates were significantly higher in HCT116 and RKO cells than those in the negative controls (P=0.000). The count of migrating HCT116 cells in up-regulated group was significantly lower than that in the negative control group (P=0.000). The count of migrating RKO cells in down-regulated group was significantly higher than that in the negative control (P=0.003). By down-regulation of miR-143, the expression of FOSL2 protein was increased. Conclusion · The expression of miR-143 was significantly decreased in colon cancer tissues. Up-regulation of miR-143 can promote the apoptosis rate in both cell lines, and inhibit the migration ability of HCT116 cells. Down-regulation of miR-143 could promote the migration ability of RKO cells. By inhibiting miR-143, the expression of FOSL2 protein in colonic cancer cells would be increased.

Methods\ Histochemical and immunohistochemical staining techniques were used,and immunostained sections were observed under a light microscopy. Results\ NPY positive neuron and nerve fiber,CGRP positive nerve fiber existed in adrenal cortex.NADPH d positive neuron and nerve fiber, c fos positive neuron and nerve fiber,CGRP positive nerve fiber,SP positive nerve fiber were detected in adrenal medulla.NADPH d positive cell,NPY positive cell,CGRP positive cell,SP positive cell were found among the adrenal chromaffin cells.The zona glomerulosa,zone fasciculata and zona reticularis were stained strongly by NADPH d in cortex. Conclusion\ The adrenal gland was innervated by NO ergic,NPY ergic,CGRP ergic,SP ergic, c fos ergic nerves.The adrenal parenchyma,especially chromaffin cells can produce NO,NPY,CGRP,SP which regulate adrenal endocrine by itself.The nervous control of adrenal function and neuroendocrine function of adrenal gland is more complex than hitherto believed.\;

Objective Our purpose is to explore the occurrence and distribution of glutamate and metabotropic glutamate receptor in earthworm, P. aspergillum and to analyze their function significance. Methods By employing immunocytochemistry tchnique, the positive cells of glutamate and metabotropic glutamate receptor, mGluR1, mGluR2/3, mGluR4 in earthworm, P. aspergillum were investigated under a light microscope. Results It was found that glutamate positive cells existed in cerebroganglion, subpharyngeal ganglion, ventral ganglion, neurons between muscle cells,intestinal epithelium and epidermis, that thin and dense glutamate positive fibers distributed in pharyngeal nerve cycle, subpharyngeal ganglion, ventral neurochain, that mGluR1 or mGluR4 positive neurons were detected in cerebroganglion and mGluR2/3 positive neurons weren't observed in earthworm. Conclusion During glutamate positive neurons, some possess remarkable processes and matured appearance from which glutamate positive fibers in pharyngeal nerve cycle, subpharyngeal ganglion, ventral neurochain derive, another have few branches, unmatured appearance in which glutamate doesn't serve as neurotransmitter acting to mGluR1 or mGluR4 of neurons in earthworm cerebroganglion.

Objective To investigate the clinical efficacy of atorvastatin in patients with lacunar infarctions (LI) and influence on cerebrovascular reactivity(CVR). Methods 60 patients with LI were randomly divided into observation group and control group. The patients of control group were given aspirin treatment, patients of observation group were given atorvastatin combination with aspirin therapy. The breath-holding index(BHI) before and after treatment was calculated. After continuous treatment for 3 months the clinical efficacy was evaluated. Results The total effective rate of the observation group was 90. 0% ,it was significantly higher than that of the control group(73.3% ),compared with the control group the difference was significant( x2 = 10. 428 ,P < 0. 05). The bilateral BHI of observation group significantly increased, compared with per-treatment and control group the difference was significant ( t =9. 455,6. 135 ,all P < 0. 05 ) ,but the bilateral BHI of control group had no significant difference compared with pertreatment ( P ＞ 0. 05 ). Conclusion Atorvastatin could effectively improve CVR in patients with LI. Therefore, atorvastatin had great significance for the prevention of stroke.

OBJECTIVE: To study the effects of Salvia miltiorrhiza on serum levels of cytokines including interleukin-6 (IL-6), interleukin-8 (IL-8) and tumor necrosis factor-alpha (TNF-alpha) in patients with severe acute pancreatitis (SAP). METHODS: Thirty-six SAP patients were randomly divided into Salvia miltiorrhiza-treated group and non-Salvia miltiorrhiza-treated group with eighteen patients in each group. Fourteen age-matched healthy volunteers were assigned to control group. Serum levels of IL-6, IL-8, and TNF-alpha were tested within 24 hours of admission and 7 days after admission. RESULTS: Compared with those of the healthy volunteers, serum levels of the three cytokines of the SAP patients were significantly elevated (P<0.05). Serum levels of the three cytokines were significantly decreased in both Salvia miltiorrhiza-treated group and non-Salvia miltiorrhiza-treated group after 7-day treatment (P<0.05). The declining of the serum levels of the three cytokines in Salvia miltiorrhiza-treated group was more obvious than that in non-Salvia miltiorrhiza-treated group. CONCLUSION: Salvia miltiorrhiza can treat SAP patients by reducing the serum levels of IL-6, IL-8, and TNF-alpha.

Objective To investigate the clinical application value of serum gastric function detection in early gastric cancer screening.Methods Forty-two cases of gastric cancer,75 cases of benign gastric diseases and 44 individuals undergoing normal physical examination in our hospital from December 2015 to December 2015 were selected as the research subjects,performed the gastroscopic examiantion,simultaneously conducted the microscopic biopsy,and extracted blood for conducting the related indexes detection of gastric function,including pepsinogen Ⅰ (PG Ⅰ),pepsinogen Ⅱ (PG Ⅱ),PG Ⅰ/PG Ⅱ,gastrin 17 (G17) and helicobactor pylori(HP).Serum gastric function situation in the research subjects of 4 groups was studied.Results PG Ⅰ and PG Ⅰ/PG Ⅱ expressions from low to high were gastric cancer group,atrophic gastritis group,gastric ulcer group,superficial gastritis group and normal group respectively,the pairwise comparison had statistical difference (P<0.05);G17 in the normal group was lower that that in gastric ulcer group,atrophic gastritis group and gastric cancer group G17 in the gastric ulcer group was lower than that in the gastric cancer group,the differences were statistically significant (P<0.05).The HP positive rate in the normal group was lower than that in the superficial gastritis group,the superficial gastritis group was lower than the gastric ulcer group,the gastric ulcer group was lower than the atrophic gastritis group,and the atrophic gastritis group was lower the gastric cancer group,the differences had statistical significance (P<0.05).Conclusion Serum gastric function detection can better distinguish gastric benign lesions from malignant lesions,and can serve as an important indicator of early screening,which is easy to accept with low cost,for the patients with positive results,the gastroscopic examination should be conducted in order to improve the early diagnostic rate.
