Colorectal carcinoma ( CRC ) is one of the malignant tumors with the highest morbidity and mortality rates in the world.The invasion and metastasis of CRC are the major reasons for treatment failure and death .It was believed that CXCR 4 was the exclusive receptor for CXCL12.However, recent studies have identified CXCR7 as a second receptor for CXCL12.Both chemokine ax-es of CXCL12/CXCR4 and CXCL12/CXCR7 closely correlate with the proliferation , migration, adhesion of tumor cells and the forma-tion of tumor-associated vessels in CRC , which may become new significant targets for anti-cancer and anti-metastatic treatment of CRC.

Objective To establish the determination method of syringin and syringaresinol in Ciwujia Injection with RP-HPLC. Methods C18 ODS was used as a stationary phase. The mobile phase consists of water and acetonitrile. The gradient condition was 0→20 min, A:90%→50%. The flow rate was 1.0 mL/min. The detective wavelength was set at 220 nm. Results The linear range of syringin was 0.03～0.16 ?g (r =0.999 7, n=5) and the linear range of Syringaresinol was 0.02～0.13 ?g (r =0.999 3, n=5). The average recovery of syringin was 99.34% and RSD=0.58%. The average recovery of Syringaresinol was 99.22% and RSD=0.71%. Conclusions The method was simple, convenient and accurate, and can be used for content determination of syringin and syringaresinol in Giwujia injection.

Objective To explore the relation between prostaglandin E2(PGE2) and the mechanosensitivity of osteoblasts and to analyze its action mechanism in cellular level .Methods Osteoblasts cell line MC3T3-E1 cells were performed the pre-treatment by using 16 ,16-dimethyl prostaglandin E2(dmPGE2) .Then the regulating effect of PGE2 on intracellular calcium ion signal was ob-served;the activator agent and inhibitor of the protein kinase A (PKA) signal path were adopted to detect the participating effect of the PKA signal path in the regulating process .Results Compared with the blank treatment group ,dmPGE2 pre-treatment signifi-cantly increased the calcium ion signal intensity induced by hypotonic swelling stimulation in MC 3T3-E1 cells .This effect of dmPGE2 was mimicked by 8-bromo-cAMP(8br-cAMP) ,an activator of PKA pathway ;but blocked by PKI ,an inhibitor of PKA pathway .Conclusion dmPGE2 is able to up-regulate the calcium response induced by hypotonic swelling stimulation by activating PKA pathway ,which provides an important cellular mechanism for explaining PGE2′s anabolic effect in bone tissue .

AIM: Previous studies paid more attention to the effect of fibroblast immigration in normal surrounding tissue or blood on wound healing. This study explored the morphological character of fibroblasts after heat injury to support the new operation of autogenous epidermis overlapping denatured dermis. METHODS: The experiment was performed at the State Key Laboratory of Trauma, Burn and Combined Injury in the Third Military Medical University of Chinese PLA from April 2006 to April 2007. The dermal fibroblast from the infant prepuce disposed after surgery was cultured in vitro, and divided into 2 groups. In normal group, the dermal fibroblasts were put in water bath at 37 ℃ for 30 seconds; the cells in experimental group were stimulated at different temperatures (50, 51, 52, and 53 ℃) for different time (30, 60, 90, and 180 seconds). Cell survival was detected by MTT; morphology of injured cells were observed under inverted microscope and transmission electron microscope at 3 hours, 1, 2, 3, 4, 5, and 7 days; cell cycle was detected by flow cytometry. OBJECTIVE:To evaluate the cost-effectiveness of3kinds of traditional Chinese medicinal injectable prepa?rations in the treatment of children's fever caused by exogenous pathogens METHODS:44cases were assigned to receive Qin_ gkailing injection(Group A),Shuanghuanglian for injection(Group B)and heartleaf houttuynia herb injection(Group C),re?spectively.The curative effects of the3groups were observed and the cost-effectiveness analysis were conducted as well.RESULTS:The costs for the3groups were28.68yuan,45.00yuan and59.40yuan,respectively;the total effective rates were80.00%,86.67%and85.71%,respectively;the cost-effectiveness ratios of the3were35.85,51.92and69.30,re?spectively;the incremental cost-effectiveness ratios of Group B and C were244.68and538.00,respectively as compared with Group A.CONCLUSION:Qingkailing injection(Group A)is the preferred option in the treatment of children's fever caused by exogenous pathogens.

Objective To investigate the expressions of microRNA-21,microRNA-143,and microRNA-145 in the sera of patients with nasopharyngeal carcinoma (NPC),and their assessment vales in the recurrence,metastasis,and prognosis of NPC patients.Methods From January 2012 to January 2014,80 NPC patients in our hospital Department of Internal Medicine and Head and Neck Surgery was used as tumor group,80 cases of healthy volunteers as control.The real-time fluorescence quantitative polymerase chain reaction (RT-PCR) was used to detect the expression levels of microRNA-21,microRNA-143,and microRNA-145 in the sera.Results Expression of microRNA-21 in NPC patients was significantly higher than that in healthy control group with statistically significant difference (P ＜ 0.05).The expressions of microRNA-143 and microRNA-145 in NPC patients was significantly lower than those in healthy control group with significant difference (P ＜ 0.05).At the same time,those microRNAs were significantly associated with tissue differentiation,invasion,and metastasis.Conclusions Increased microRNA-21 expression level in NPC patients,and decreased expression of microRNA-143 and microRNA-145 in NPC patients play an important role in differentiation,invasion,and metastasis in the development process of NPC.microRNAs can be used as a new index in the auxiliary diagnosis of NPC and the evaluation of recurrence,metastasis,and prognosis evaluation.

Objective:To explore the effect of radiotherapy on the FHIT protein expression and cell apoptosis of cervical squamous carcinoma and discuss the relationship between FHIT protein expression and cell apoptosis. Methods:Expression of FHIT protein was measured by immunohistochemical method and cell apoptosis was detected by TdT-mediated dUTP terminal nick end labeling (TUNEL) staining in 50 cases of squamous cell cervical carcinoma at ⅡB-ⅢB stages before, during (Dt 10 Gy and Dt 30 Gy), and after radiotherapy. Results:Of the 50 patients, the positive rates of the expression of FHIT protein was 56% at Dt 10 Gy, 68% at Dt 30 Gy, and 84% after radiotherapy, which were significantly increased compared with that before radiotherapy (36%, P<0.05). The positive rates of cell apoptosis was 52% at Dt 10 Gy, 64% at Dt 30 Gy and 78% after radiotherapy, which were significantly elevated compared with that before radiotherapy (28%, P<0.05). In the process of radiotherapy, cell apoptosis was positively related to the expression of FHIT protein (P<0.05). Conclusion:Radiotherapy reinforces the expression of FHIT protein and induces apoptosis cocurrently. FHIT protein has regulatory effects in cell apoptosis induced by radiotherapy.

Background Randomized controlled trial (RCT) can offer high quality of evidence for the medical decisions.Consolidated standards of reporting trials (CONSORT) statement,an international criteria for RCT report,is a guideline and methodology of RCT report.However,the assessment and application of RCT according to CONSORT are dissatisfactory in China.Objective This study was to evaluate the reporting quality of ophthalmic diseases related RCTs published in SCI journals from mainland China and offer a good methodology of RCT report.Methods The RCT literature were searched on PubMed database according to Ophthalmology, Eye and all of their inferior subjective terms,and the published papers were limited in mainland China and before March,2013.Included papers were evaluated independently and checked by two searchers.The methodology and results of all retrieved papers were stratifiedly analyzed by the year of CONSORT published and revised.Results Sixty eligible RCTs were assessed,and the intervention methods were primarily surgery trials (34/60).Forty-four papers presented the comparison study of two groups with the median sample size of 76.0.Only 22％ (13/60) trial reports gave the clear identification of RCT in the title.Majority of papers described trial design,eligibility criteria for participants,detail of interventions and outcome measures.Twelve pieces of articles reported how sample size was determined,and 41 pieces referred settings and locations,but only 6 pieces recorded interim analyses and stopping guidelines.After CONSORT 2010 Statement was published,the proportion of trial reports that included details of the randomization had little improvement (RR=1.06,95％ CI:0.62-1.82).Only 42％ trial reports (25/60) offered a description about blinding method.Conclusions RCT reports related to ophthalmic diseases from mainland China in SCI journals are still flawed with certain shortcomings.Following the guidance of the CONSORT statement can improve the quality of ophthalmic diseases related RCT from mainland China.

AIM: To investigate the effect of oral clarithromycin on serum concentrations of aminophylline at steady state and its pharmacokinetics in New Zealand rabbits. METHODS: The serum concentration of aminophylline was determined by FPIA in rabbits given(po) aminophylline or aminophylline combined with clarithomycin. The experiment was divided into 2 stages: (Ⅰ)the subjects only received a four day course of oral aminophylline until steady state;(Ⅱ)aminophylline and clarithromycin were coadministrated from the d 5 to d 10. The dose of aminophylline was 30 mg?kg -1 and the dose of clarithromycin was 50 mg?kg -1 for each rabbit. The two series of pharmacokineticis parameters were tested by statistic analysis. RESAULTS: There was no significant variation in pharmacokinetic parameters between two stages. CONCLUSION: It is not necessary to change the therapeutic dose of aminophylline when the drug is taken in combination with clarithromycin.

Objective To track superparamagnetic iron oxide (SPIO)-labeled pancreatic islet cells in rats using 3.0T magnetic resonance imaging (MRI), and to detect the survival and rejection of grafts after transplantation. Methods Twenty male Wistar rats and 5 male Lewis rats were included in the study. SPIO-labeled pancreatic islet cells were tracked using a GE 3.0T Signa Excite MRI scanner with an animal coil. The images of SPIO-labeled islet cells in rats after transplantation were compared with those of the unlabeled ones. FSE T2WI sequence and GRE T2*WI sequence were used for the detection. The sensitivity of images for detection of grafts was also compared. SPIO-labeled pancreatic islet cells isolated from Wistar and Lewis rats were transplanted into the liver of Wistar rats. Afterwards, the survival and rejection of islet cells were observed sequentially in these two growps. The rats in the syngeneic group were sacrificed 3 months post-transplantation, while the rats in the allogeneic group were sacrificed 3 weeks post-transplantation. MRI of the grafts were correlated with the pathological results. Results SPIO-labeled pancreatic islet cells were seen on MRI as distinct homogenous, hypointense spots in the liver. GRE T2*WI were more sensitive to the detection of SPIO-labeled islet cells than FSE T2WI. The relative count of hypointense spots in the syngeneic group were (90.03±9.52)%, (92.87±18.21)% and (86.25±24.81)%, respectively at 1 week, 2 weeks and 3 weeks after transplantation, while the relative count in the allogeneic group were (41.40±15.41)%, (33.41±14.01)% and (23.58±16.78)%, respectively. The difference between these counts was statistically significant (P<0.01). Iron particles were detected only in the SPIO-labeled cells. Three months post-transplantation, the grafts were found well-preserved in the liver of the rats of the syngeneic group, while only a few grafts were found in that of the allogeneic group. Conclusions MRI can be used to track SPIO-labeled islet cells in vivo, and has significant value in detecting the survival and rejection of grafts after transplantation in rats.
