1.The inhibitory effect of endostatin on human gastric carcinoma cells in an athymic mouse model and the impact on expression levels of Bcl-2 and Bax

Objective To investigate the inhibitory effect and the mechanism of endostatin on human gastric carcinoma model in nude mice. Methods Athymic mice were injected by in vitro-cultured SGC-7901 human carcinoma cell strain to observe the inhibitory effect of endostatin. Real time fluorescent quantitative PCR and immunohistochemistry stain were used to quantify the expression levels of Bcl-2 and Bax genes both on mRNA and protein levels. Results The average tumor volume in endostatin group was statistically smaller than control group (P<0.05). The inhibitory rate on the 7~(th), 10~(th), 13~(th), 16~(th) and 19th day was 26.2%, 47.7%, 38.9%, 45.8% and 51.7%, respectively. The results of real time fluorescent quantitative PCR demonstrated that mRNA expression of Bcl-2 in endostatin group was obviously lower than it in control group (P<0.05), however, the expression level of Bax had no statistical difference between these two groups (P>0.05). The results of immonohistochemistry were consistent with PCR. Conclusions Endostatin can inhibit the proliferation of SGC-7901 human gastric carcinoma in athymic mouse model, and the mechanism involves the suppression of Bcl-2 expression.
2.Cost-Effectiveness Analysis of3Kinds of Traditional Chinese Medicinal Injectable Preparations in the Treatment of Children's Fever Caused by Exogenous Pathogens

OBJECTIVE:To evaluate the cost-effectiveness of3kinds of traditional Chinese medicinal injectable prepa?rations in the treatment of children's fever caused by exogenous pathogens METHODS:44cases were assigned to receive Qin_ gkailing injection(Group A),Shuanghuanglian for injection(Group B)and heartleaf houttuynia herb injection(Group C),re?spectively.The curative effects of the3groups were observed and the cost-effectiveness analysis were conducted as well.RESULTS:The costs for the3groups were28.68yuan,45.00yuan and59.40yuan,respectively;the total effective rates were80.00%,86.67%and85.71%,respectively;the cost-effectiveness ratios of the3were35.85,51.92and69.30,re?spectively;the incremental cost-effectiveness ratios of Group B and C were244.68and538.00,respectively as compared with Group A.CONCLUSION:Qingkailing injection(Group A)is the preferred option in the treatment of children's fever caused by exogenous pathogens.
3.Determination of Intracellular Ara-CTP Level by RP-HPLC

OBJECTIVE:To establish a simple and rapid ion-pair-RP-HPLC method for the determination of intracellular concentration of ara-CTP.METHODS:Mononuclearcells were separated from the whole blood,centrifugated and injected for determination on Symmetry C18 column with the mobile phase consisted of 0.1 mol?L-1 KDP buffer solution and 0.01 mol?L-1 tetrabutylammonium hydrogen sulfate(pH=2.7)at a flow rate of 1.0 mL?min-1 with the UV wavelength set at 278 nm.RESULTS:The linear range of ara-CTP was 0.35-9.12 ?g?mL-1(r=0.998 6)with its detection limit at 0.35 ?g?mL-1.The recoveries of ara-CTP at low,middle and high concentrations,and the intra-day and inter-day RSDs were all up to the standards.CONCLUSION:The established method is simple,rapid,stable,reproducible and applicable for the monitoring of intracellular concentration of ara-CTP.
4.Determination of Syringin and Syringaresinol in Ciwujia Injection by RP-HPLC

Objective To establish the determination method of syringin and syringaresinol in Ciwujia Injection with RP-HPLC. Methods C18 ODS was used as a stationary phase. The mobile phase consists of water and acetonitrile. The gradient condition was 0→20 min, A:90%→50%. The flow rate was 1.0 mL/min. The detective wavelength was set at 220 nm. Results The linear range of syringin was 0.03~0.16 ?g (r =0.999 7, n=5) and the linear range of Syringaresinol was 0.02~0.13 ?g (r =0.999 3, n=5). The average recovery of syringin was 99.34% and RSD=0.58%. The average recovery of Syringaresinol was 99.22% and RSD=0.71%. Conclusions The method was simple, convenient and accurate, and can be used for content determination of syringin and syringaresinol in Giwujia injection.
5.Determination of Ara-C in plasma and Ara-CTP in leukemic cells after intravenous infusion of high-dose Ara-C in patients with acute leukemia and analysis of influence factors

Objective To determine the plasma and intracellular concentration of Ara-C by the RPHPLC method and analyse the influence factors and the relationship between the concentration and drug dose.Methods Mononuclear cells and serum of 75 patients with acute leukemia were extracted after the first intravenous infusion of different administration dosage of Ara-C (0.5, 1.0, 2.0 g/m2), and analysed with different chromatographic conditions by RP-HPLC. Results The linear range of Ara-CTP was 0.28-18.96 μg/ml (r =0.998), and the detection limit was 0.28 μg/ml. The detection limit of Ara-C and Ara-U in plasma was 0.0157 μg/ml and 1.034 μg/mnl respectively. In 27 samples preserved for more than 1.5 years, 11 (40.7 %)cases of the plasma concentration of Ara-C were below the detection limit. In 36 samples of mononuclear cell count below 1.5×106/ml, 15 cases (41.7 %) of intracellular concentration of Ara-CTP were below the detection limit. The plasma concentration of Ara-U and intracellular concentration of Ara-CTP were increased with administration dosage of Ara-C increased, and the plasma concentration of Ara-C was not increased. The intracellular concentration of Ara-CTP in old patients over 40 years was tend to in crease with age.Conclusion The RP-HPLC method is simple, rapid, stable, reproducible and applicable for the monitoring of the plasma concentration of Ara-C and intracellular concentration of Ara-CTP. In 0.5-2.0 g/m2 dose range of Ara-C, the plasma concentration of Ara-U and intracellular concentration of Ara-CTP was increased with administration dosage of Ara-C increased.

Objective Our retrospective study is aimed to analyze the correlative clinical features of be-tween expressions of miR -135b and miR-141 in colorectal cancer.Methods Clinical data and specimens of patients with colorectal carcinoma at our hospital from 2013 to 2014 were retrospectively collected .The expres-sions of miR-135 b and miR-141 between colon cancer and matched normal colonic tissues were compared .The correlation between the expressions of miR -135 b and miR-141 in colorectal carcinoma and the clinical charac-teristics were discussed .Results A total of 42 patients was retrospective analyzed .The expressions of miR -135b,miR-141 in the colon cancer tissues were obviously higher than that in the normal colonic tissues .The differences of the expression of miR -141 in the colon cancer tissues of different Dukes stages ,degree of differen-tiation,lymph node metastasis and infiltration depth were all significant .The expressions of miR -135 b and miR-141 displayed a positive linear correlation .Conclusion The expressions of miR-135b,miR-141 in colorec-tal cancer are significantly increased ,and the expressions display a remarkable correlation with the Dukes stages , degree of differentiation ,lymph node metastasis and infiltration depth of tumor .The expressions of miR-135 b and miR-141 demonstrate a positive correlation .
7.NUP210 and MicroRNA-22 Modulate Fasto Elicit HeLa Cell Cycle Arrest

Purpose:
Cervical cancer is one of the most fatal diseases among women in under-developed countries. To improve cervical cancertreatment, discovery of new targets is needed. In this study, we investigated the expression of NUP210, miR-22, and Fas in cervicalcancer tissues and their functions in cell cycle regulation.
Materials and Methods:
We detected and compared the expression levels of NUP210, miR-22, and Fas in cervical cancer tissueswith paired normal tissues using immunohistochemistry, Western blot, and real-time quantitative polymerase chain reaction.NUP210 was knocked down in HeLa cells via lentivirus, followed by cell cycle and proliferation analysis. Using a luciferase reporterassay, we explored the link between miR-22 and NUP210. We overexpressed miR-22 in HeLa cells and analyzed cell cycle and proliferationfunction. We then overexpressed miR-22 in NUP210 knockdown cells to explore the connection between Fas and miR-22-NUP210 signaling.
Results:
We found that NUP210 was overexpressed in cervical cancer patients. Knocking down NUP210 restored cell apoptosisand proliferation. We confirmed miR-22 as a regulator of NUP210 and verified that miR-22 was inhibited in cervical cancer development.We also found that restoring miR-22 expression could induce cell apoptosis. Finally, we found that miR-22-regulated expressionof NUP210 could alter Fas expression and, in turn, elicit cell cycle arrest and proliferation.
Conclusion
miR-22 in cervical cancer is downregulated, resulting in NUP210 overexpression and inhibition of Fas-induced cellapoptosis.
8.Acute myeloid leukemia with NPM-MLF1 fusion gene: report of one case and review of literature

Objective To investigate the characteristics of NPM-MLF1 fusion gene in acute myeloid leukemia (AML). Methods The data of one AML patient with NPM-MLF1 fusion gene was analyzed,and literatures were reviewed. Results A female patient was diagnosed as AML M6. In the course of the disease, 2 hematologic relapsed, and 2 recurrences were associated with NPM-MLF1 fusion gene positive. After inductive treatment, hematologic complete remission was achieved, and NPM-MLF1 fusion genes were all negative. Survival time surpassed 6 years when the chemotherapy was performed alone. Conclusion The incidence of NPM-MLF1 fusion gene in AML is low. It is necessary to collect more clinical data to judge whether an independent disease type or not.

Health service is an important part of the integrated development of the Yangtze River Delta. Taking the cooperation practice between Shanghai Tenth People′s Hospital and Suzhou Yinshanhu Hospital as an example, this article introduced the multi-agent cooperation mode of the loose medical alliance including the government, urban hospitals and cross provincial grassroots medical institutions. Among them, the local government provided policy, fund guarantee and guidance, the urban hospital exported management ideas, medicine talents and technologies, and the primary hospital conducted dual training by inviting in and going out to achieve double growth. Through the high gap cooperation between tertiary hospital and primary hospital, Yinshanhu hospital had been comprehensively developed. The loose medical alliance with multi subject coordination and cross region could give full play to the advantages of the loose healthcare alliance mode, achieve multi-win, and have reference significance for promoting the regional integration of medical and health services in the Yangtze River Delta.
10. Characteristics and prognosis in adult acute myeloid leukemia patients with MLL gene rearrangements

Objective:
To analyze the clinical and laboratory characteristics, and prognosis of adult acute myeloid leukemia (AML) patients with MLL gene rearrangements.
Methods:
The medical records of 92 adult AML patients with MLL gene rearrangements from January 2010 to December 2016 were retrospectively analyzed.
Results:
92 cases (6.5%) with MLL gene rearrangements were identified in 1 417 adult AML (Non-M3) patients, the median age of the patients was 35.5 years (15 to 64 years old) with an equal sex ratio, the median WBC were 21.00(0.42-404.76)×109/L, and 78 patients (84.8%) were acute monoblastic leukemia according to FAB classification. Eleven common partner genes were detected in 32 patients, 9 cases (28.1%) were MLL/AF9(+), 5 cases (15.6%) were MLL/AF6(+), 5 cases (15.6%) were MLL/ELL(+), 2 cases (6.3%) were MLL/AF10(+), 1 case (3.1%) was MLL/SETP6(+), and the remaining 10 patients’ partner genes weren’t identified. Of 92 patients, 83 cases with a median follow-up of 10.3 (0.3-74.0) months were included for the prognosis analysis, the complete remission (CR) rate was 85.5% (71/83), the median overall survival (OS) and relapse free survival (RFS) were 15.4 and 13.1 months, respectively. Two-year OS and RFS were 36.6% and 29.5%, respectively. Of 31 patients underwent allogeneic hematopoietic stem-cell transplantation (allo-HSCT), two-year OS and RFS for patients received and non-received allo-HSCT were 57.9% and 21.4%, 52.7% and 14.9%, respectively (
