1.The value of CT angiography in the diagnosis of intracranial atherosclerosis

Objective To investigate the CT angiography (CTA) in the diagnosis of intracranial atherosclerosis.Methods Sixty-eight cases of suspected ischemic cerebrovascular disease patients were enrolled in this study,and all patients underwent CTA.Parallel image post-processing was done.CTA characteristics were observed and compared with digital subtraction angiography (DSA) results.Results A total of 376 lesions in 68 cases were examined,112 lesions were found in CTA; intracranial atherosclerotic plaques mainly distributed in the internal carotid artery (53 lesions),anterior cerebral,middle and posterior artery (39 lesions) and vertebral-basilar artery (20 lesions).In narrow artery segment,stenosis with calcified plaque was in 32 lesions,mostly in the middle cerebral artery,the cavernous segment of the internal carotid artery,clinoids segment and intracranial vertebral artery.For non-calcified plaque,CTA showed fuzzy.DSA showed 128 total lesions,and severe stenosis rate in CTA [9.31% (35/376)] and DSA [9.31% (35/376)] was consistent.Thirty-eight patients were correctly diagnosed.Conclusions CTA as a minimally invasive intracranial atherosclerosis diagnostic methods,consumes less time with low cost and high spatial resolution.It is suitable for clinical universal application.

3.Effects of Taxol on proliferation, apoptosis, and mRNA expression of α2,6-sialic acid and ST6Gal in cervical carcinoma cell line U14

AIM:To study the effect of Taxol on the proliferation, apoptosis, and mRNA expressions of α2,6-sialic acid (SA) and α2,6-sialyltransferase (ST6Gal) in mouse cervical cancer cell line U14.METHODS:After the U14 cells were treated with Taxol, the IC50 value of Taxol to U14 cells was detected by MTT assay.The expression of α2,6-SA and apoptosis-related factors (Bcl-2, Bax, caspase 8 and caspase 3), the apoptosis rate and cell cycle were determined by flow cytometry.The mRNA expression of ST6Gal1 and ST6Gal2 was detected by qPCR.RESULTS:As compared with control group, Taxol induced obvious U14 cell growth inhibition, reduced α2,6-SA expression, up-regulated Bax, down-regulated Bcl-2, decreased the ratio of Bcl-2/Bax, enhanced caspase 8 and caspase 3 activity, increased the apoptotic rate and cell proportions of Sub-G1 and S phases, and induced G2/M phase arrest.Taxol also down-regulated the mRNA expression of ST6Gal1, and slightly up-regulated the mRNA expression of ST6Gal2.CONCLUSION:α2,6-SA and ST6Gal are involved in the multiple effects of Taxol on modulation of the cell cycle and apoptosis in U14 cells.
4.MALAT1 reduces the sensitivity of EGFR-wild-type non-small cell lung cancer to gefitinib

Objective To investigate serum metastasis-associated lung adenocarcinoma transcript 1 (MALAT1) in non-small cell lung cancer (NSCLC) and observe gefitinib drug sensitivity before and after transfecting MALAT1-siRNA in epidermal growth factor receptor ( EGFR ) wild-type cell line A549.Discussing the relationship between MALAT 1 and epidermal growth factor receptor-tyrosine kinase inhibitor ( EGFR-TKI) drug resistance.Methods Serum MALAT1 was quantified by qRT-PCR in 90 cases of NSCLC, 30 cases of lung benign tumor and 30 cases of healthy people in the First Affiliated Hospital of Dalian Medical University during April to September in 2015.MALAT1 expression in cancer cells were detected in the cell lines A549, H1299, 95D and HCC827.The half maximal inhibitory concentration ( IC50) of gefitinib in cells before and after transfecting MALAT 1-siRNA were analyzed by cell proliferation toxicity test ( CCK).Flow cytometry technology was used to evaluate the apoptosis rates.One-way analysis of variance was statisticed between groups.Results Serum MALAT1 expression was increased in NSCLC compared with healthy control group ( NSCLC =1.88 ±2.51, Healthy control group =1, t=3.347, P<0.01).MALAT1 was over expressed in the EGFR wild-type cell lines compared with the EGFR mutant cell lines(HCC827=1, A549=4.44 ±1.05, 95D=3.55 ±0.63,H1299 =1.44 ±0.05).The IC50 value of gefitinib was declined 32.1% by silencing MALAT1.The results of flow cytometry showed an increasing apoptosis rate after silencing MALAT1 in A549 cells[NC=(9.20 ±1.85 )%, M=(10.24 ±3.43)%, NC+G=( 14.57 ±0.41 )%, M +G =( 21.48 ±2.47 )%].Conclusions Serum MALAT1 reflected the progression of NSCLC.Silencing MALAT1 resulted in enhancement in sensitivity to gefitinib in A549 cell line.It prompted the partly reversal of gefitinib resistance by MALAT1.
5.Clinical effect of percutaneous catheter drainage under the guidance of ultrasound in the treatment of hepatic abscess

Objective To evaluate the clinical value of percutaneous catheter drainage under the guidance of ultrasound in the treatment of hepatic abscess.Method Forty-eight cases who were clinical diagnosis of liver abscess by percutaneous catheter drainage was selected as the observation group.There were 12 cases of liver abscess diagnosed by parallel surgical drainage in the treatment as the control group.To observe the clinical effect,hospitalization time and complications of two groups of patients after treatment.Results The cure rate of observation group was significandy higher than that in control group:100% (48/48) vs.The temperature recovery time and hospitalization time of the observation group were significantly shorter than those in the control group:(1.5 ±0.3) d vs.(3.2 ±0.8) d,(9.3 ±2.3) d vs.(17.5 ±3.2) d.No complications occurred in the observation group,and there were 2 cases in the control group.Conclusions Percutaneous catheter drainage in the treatment of liver abscess has advantages of simple economy,less trauma,less complications and high cure rate.It has become the first choice for treatment of liver abscess.
6.The relationship among self-esteem, putting into perspective and social anxiety of college students

Objective To investigate the relationship between self-esteem and social anxiety,and the role of putting into perspective played in this relationship in university students.Methods Sample consisted university students of 376 females and 164 males,data were obtained on cognitive emotion regulation questionnaire,social anxiety subscale of the self-consciousness scale,and self-esteem scale.Results Cognitive emotion regulation strategies of putting into perspective is significantly and positively related to self-esteem(r=0.12,P< 0.01),it was also significantly and negatively related to social anxiety(r =-0.01,P < 0.05) ; self-esteem was significantly and negatively related to social anxiety(r =-0.35,P < 0.001).Cognitive emotion regulation strategies of putting into perspective played both a moderating(AR2 =0.124,P<0.05) and mediating role(F=30.717,P< 0.01) in the relationship between self-esteem and social anxiety.Conclusion Self-esteem is not only directly but also indirectly associated with social anxiety because of the moderation and mediation model of putting into perspective.
7.Constituents from Pholidota yunnanensis and their inhibitory effects on nitric oxide production

Objective To study the chemical constituents of the whole plant of Pholidota yunnanensis and to examine the inhibitory effects of the isolated compounds on nitric oxide(NO) production in activated rat macrophage-like cell line(RAW 264.7).Methods Various chromatographic techniques were used to separate and purify the constituents,and their structures were elucidated on the basis of spectroscopic analysis. The inhibitory effects of these compounds on NO production in RAW 264.7 cells of rats activated by lipopolysaccharide(LPS) and interferon-?(IFN-?) were tested in vitro by microplate ultraviolet-colorimetric method.Results Seven compounds were isolated and identified as trans-3,3′,5-trihydroxy-2′-methoxystilbene(Ⅰ),cis-3,3′-dihydroxy-5-methoxystilbene(Ⅱ),trans-3,3′, 5-trihydroxystilbene(Ⅲ),3,3′-dihydroxy-5-methoxybibenzyl(batatasin-Ⅲ,Ⅳ),3,4′-dihydroxy-3′,5-dimethoxybibenzyl(gigantol,Ⅴ),3,3′,5-trihydroxybibenzyl(Ⅵ),2,7-dihydroxy-4-methoxy-9,10-dihydrophenanthrene(coelonin,Ⅶ).Conclusion Compounds Ⅱ,Ⅳ,and Ⅵ show favorable inhibitory effects on NO production,compounds Ⅰ and Ⅱ are new compounds,and compounds Ⅲ-Ⅶ are isolated from this genus for the first time.
8.Application of the Luo’s fornix cup in laparoscopic total hysterectomy

Objective To explore the clinical value of the Luo’s fornix cup in laparoscopic total hysterectomy. Methods A total of 7 patients with indications for hysterectomy from October 2004 to February 2005 was given laparoscopic total hysterectomy. During the operation, the Luo’s fornix cup was used for the orientation and dilatation of the vaginal fornix. Then the anterior vaginal fornix was circlewise incised by using a harmonic hook to extract the Luo’s fornix cup. Afterwards the posterior fornix was opened to remove the uterus by way of the vagina. And finally the vaginal stump was closed. Results All the operations were successfully completed. The operation time was 90~250 min. With the use of the Luo’s fornix cup, the operator could easily identify the top of the vaginal fornix, and avoid the damage of important neighbouring organs. No injuries of bladder, ureter, and rectum were noted. Conclusions The Luo’s fornix cup is a rational accessory instrument in laparoscopic hysterectomy, being worthy of recommendation.
9.Curettage as endometrial pretreatment before microwave endometrial ablation:A clinical and experimental study

Objective To investigate the value of thorough uterine curettage in microwave endometrial ablation(MEA).Methods Preoperative uterine curettage was conducted in 126 women with abnormal uterine bleeding(AUB)scheduled for MEA from June 2001 to June 2005.At the same period,another 12 patients with AUB underwent hysterectomy.And before the operation,uterine curettage and subsequent MEA were conducted.Then the uterine specimens were histopathologically and histochemically examined for evaluating the tissue damages.Results No intraoperative complications occurred in the 126 patients.Postoperatively,amenorrhea was observed in 78 patients(61.9%),normal menstruation was achieved in 46 patients(36.5%),and small amount of irregular vaginal bleeding was found in 2 patients(1.6%),the overall satisfaction rate being 98.4%(124/126).Postoperative complications included endometritis in 10 patients,hematometra in 2 patients,and dysmenorrhoea leading to hysterectomy in 2 patients.Follow-up observations were conducted for 6~28 months(22?4 months)in 126 patients,116 of whom were followed for more than 2 years.Follow-up findings showed satisfactory surgical effects.After MEA,the injury depth was 4.2~4.9 mm in the uterine fundus,4.1~5.7 mm in the anterior wall,4.7~6.6 mm in the posterior wall,and 4.0~4.7 mm in the uterine horn.Conclusions MEA with preoperative thorough uterine curettage can completely destruct the endometrium.The method offers good efficacy,high satisfactory rate,and low risk of tissue injury.
10.Pathogenesis of brain damage in infant rats induced by corticosteroids

Objective To explore the possible pathogenesis of brain damage in rats induced by prednisone or corticotrophin. Methods The doses of prednisone or corticotrophin were determined by a primary experiment to obtain corresponding plasma cortisol or corticosterone level as same as that in sick children after drug taking. Then 192 healthy infant (at the age of 7 d) and 192 adult (at the age of 2 months) male SD rats were divided into 4 groups as infant prednisone group, adult prednisone group, infant corticotrophin group and adult corticotrophin group (n=96 in each). then every group was further subdivided into 12 subgroups (n=8 in each). The subgroups were divided according to the dose (therapeutic or low doses), the course [short (10 d) or long (3 weeks)], the sacrificed time (24 h or 4 weeks after withdrawal), and their corresponding controls. Serum neuron-specific enolase (NSE) concentration was quantified by ELISA. Expressions of apoptosis-related proteins, Bax and Bcl-2 in the brain were detected by immunohistochemical assays. Neuronal apoptosis was detected by TUNEL. Results Our primary experiment indicated that the therapeutic dose was 4 mg?kg-1?d-1 or 150 U?m-2?d-1 for prednisone or corticotrophin, and the small dose was 2 mg?kg-1?d-1 or 38 U?m-2?d-1 for them. In infant rats treated with prednisone or corticotrophin at therapeutic-dose for short or long term, their serum NSE concentration were increased significantly by 50.6% to 103.2%. And serum NSE was increased by 38.3% to 60.3% in infant after low-dose treatment for long term. Over-expression of Bax protein (P
