1.The mechanisms of carbapenem resistance inKlebsiella pneumoniae

Klebsiella pneumoniae is well known as a causative agent of both community and nosocomial infections and is generally believed to be the cause of 10% of the nosocomial infection. Recently, with the widespread using of carbapenem, the isolation of carbapenem-resistant strains has been greatly increased, which bring great dififculties and challenges in clinical treat-ment. In this article, the progresses in the mechanisms of carbapenem, resistance inKlebsiella pneumoniae such as the acquisition of carbapenemases, hyperproduction of AmpC cephalosporinases or extended spectrum beta-lactamases (ESBLs) in combination with loss of the outer membrane protein, eflfux pump system, and bioiflm were reviewed.
2.A study on antiviral activity of defensins from neutrophil of human and rabbit against SARS coronavirus in vitro

Objective To determine the anti-SARS coronavirus (SARS-CoV) activity of defensins from human and rabbit, and explore the effects of defensin during the onset and progression of SARS. Methods Defensins were purified from neutrophil of human and rabbit respectively, namely HNP (human neutrophil protein) 1-3 and RNP (rabbit neutrophil protein) 1-5, by acid-urea-polyacrylamide gel electrophoresis (CAU-PAGE). The effect of defensin on Vero-E6 cell infected by SARS CoV BJ01 strain as well as the replication of virus were observed by micro-culture technique. Rusults The HNP 1-3 and RNP 1-5 separated by CAU-PAGE presented a single band with molecular weight of approximately 4000 dalton on 15% SDS-PAGE. HNP 1-3 was identified and validated by quantitative ELISA. The activity of anti-escherichia coli (ATCC25922 strain) of HNP 1-3 and RNP 1-5 was tested by MIC determination method, and the IC50 values obtained were 25~50?g/ml and 5~10?g/ml, respectively. In the antiviral experiment, HNP 1-3 and RNP 1-5 in the concentration from 25 to 100?g/ml were not able to prohibit cytopathic effect (CPE) of SARS-CoV and the replication of virus in Vero-E6. Conclution The natural defensins (HNP 1-3 and RNP 1-5) purified from neutrophils do not play any prohibitory or eliminative effects on SARS-CoV, indicating that that human and rabbit do not have natural immunity against SARS-CoV.
3.Effects of chitosan on hepato-mitochondria in fatty liver rats by stereologic analysis

Purpose The aim is to study the preventing effect and me chanism of chitosan on fatty liver rats.Methods The rats give n a lo w dose of carbon tetrachloride(CCl4) and fed a high-fat diet to induce fatty liver model were treated with chitosan in different concentrations.The extensio n of steatosis area and changes in mitochondrial ultrastructure in liver were es timated by a computer color image analysis system and a quantitative analysis(st ereology).Results When rats were treated with a mid dose of c hitosan [4g/(kg*d)],the degree of hepatocytic steatosis was markedly diminished, and the injured mitochondrial structure was improved compared with model group.Co nclusion It was suggested that chitosan might enhance the oxidative catabolism of lipid in fatty liver.

ObjectiveTo Investigate the state and needs of disabled veterans and support centre in our country, and explore the factors related with quality of life. Methods328 disabled veterans of 6 veterans' support centre were surveyed with questionaire or interview. Results and ConclusionThe overall psychological states of the disabled veterans are unacceptable, dysthymia, anxiety, loneliness, insomnia are the most common among them. The unreasonable disposition of aids tend to decreases their quality of life.
5.Study on the Improvement Effects and Mechanism of Polydatin on Renal Fibrosis in Rats

OBJECTIVE:To study the improvement effects of polydatin on renal fibrosis in rats and its mechanism. METH-ODS:50 rats was were randomly divided into sham operation group,model group,positive group(benazepril,5 mg/kg)and poly-datin high-dose and low-dose groups(100,50 mg/kg),with 10 rats in each group. Except for sham operation group,renal fibrosis model was induced by unilateral ureter obstruction. After modeling,administration groups were given relevant medicine intragastri-cally,and sham operation group and model group were given 0.5%sodium carboxymethyl cellulose solution once a day for consec-utive 4 weeks. The pathological change of renal tissue was scored. 24 h urinary protein and serum levels of urea nitrogen and creati-nine were determined,and the content of hydroxyproline,mRAN expression of TGF-β1 and FN were detected in renal tissue. RE-SULTS:Compared with sham operation group,pathological score,24 h urinary protein,serum levels of urea nitrogen and creati-nine,the content of hydroxyproline in renal tissue,mRNA expression of TGF-β1 and FN were all increased significantly (P<0.01). Compared with model group,24 h urinary protein,serum levels of urea nitrogen and creatinine and mRNA expression of FN in renal tissue decreased significantly in administation groups;the pathology scores,the content of hydroxyproline in renal tis-sue and mRNA expression of TGF-β1 of positive group and polydatin high-dose group were all decreased significantly(P<0.05 or P<0.01). CONCLUSIONS:Polydatin can prevent kidney fibrosis induced by unilateral ureteral obstruction,the mechanism of which may be associated with the mRNA expression down-regulation of TGF-β1 and FN in renal tissue.

AIM: To investigate the role of expression of peroxisome proliferator-activated receptor ?(PPAR ?) in pathogenesis of rat fatty liver.METHODS: The rats were treated with a low dose of carbon terachloride (CCl 4) and fed a high fat diet to produce fatty liver. We determined the concentrations of triglyceride (TG), total cholesterol (TC), free fatty acid (FFA) in liver and the alanine aminotransferase (ALT) activity, tumor necrosis factor-? Aim: To study the effects of chitosan on hepatic lipids in fatty liver rats. Methods: The rats given a low does of carbon tetrachloride (CCl4) and fed a high-fat diet were treated with chitosan in different doses. Results: When rats were treated with mid dose of chitosan (4mg? g~-1?d~-1), the contents of triglycerde (TG). total cholesterol (TC) in liver decreased significantly, the level of mitochondria malonal-dialdehyde (MDA) was markedly attenuated and the mitochondria membrane fluidity was restored, which compared with model group. Conclusion: Chitosan may decrease the hepatic lipids by reduced the lipid peroxida- tion and restored mitochondria membrane fluidity to improve the mitochondrial beta-oxida- tion.
