Objective To explore the relationship between TSLC-1 protein expression and the progression and carcinogenesis of human papillary thyroid carcinogenesis. Methods Expressions of TSLC-1 protein in 20 pieces of benign thyroid lesions and 40 pieces of papillary thyroid carcinoma tissues were examined by immunohistochemistry. Results The expression of TSLC-1 in papillary thyroid carcinoma (9/40) was lower than that in benign thyroid lesions (22.50% versus 90.00%, P < 0.05). Expression of TSLC-1 was significantly associated with lymph node metastasis and TNM in papillary thyroid carcinoma (P < 0.05). Conclusions Expression of TSLC-1 is closely related with carcinogenesis , lymph node metastasis , and clinical stage in papillary thyroid carcinoma.

Objective To investigate the effect of sevoflurane postconditioning on the renal injury induced by hind limb ischemia-reperfusion (I/R) in rats. Methods Twenty-four healthy male Sprague-Dawley rats, weighing 200-250 g,were randomly assigned into 3 groups (n =8 each):sham operation group (group S),group I/R and sevoflurane group (group Spo).Limb ischemia was induced by occlusion of bilateral hind limbs for 4 h followed by 6 h reperfusion. In group Spo,sevoflurane was inhaled for 6 h at the end-tidal concentration of 2.5 % before reperfusion, while pure oxygen was inhaled instead of sevoflurane in groups S and I/R.Blood samples were taken from the inferior vena cava at 6 h of reperfusion to determine the serum blood urea nitrogen (BUN) and creatinine (Cr) concentrations. The rats were then sacrificed and the kidney was removed for determination of superoxide dismutase (SOD) activity and contents of malondialdehyde (MDA),tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) and for microscopic examination. Results The levels of BUN,Cr,MDA,TNF-α and IL-6 were significantly higher, the SOD content was significantly lower (P < 0.05),and the pathological damage was severer in groups I/R and Spo than in group S.Compared with group I/R,the levels of BUN,Cr,MDA,TNF-α and IL-6 were significantly decreased, the SOD content was significantly increased (P < 0.05),and the pathological damage was attenuated in group S.Conclusion Sevoflurane postconditioning can reduce the renal injury induced by hind limb I/R in rats, and the reduction of oxygen radical release and inhibition of inflammatory response may be involved in the mechanism.

Purpose To explore the relationship between tumor suppressor in lung cancer 1 (TSLC1) protein expression and the carci-nogenesis and progression of human gastric carcinoma. Methods Expression of TSLC1 protein in 20 normal gastric mucosa, 30 intra-epithelial neoplasias ( IN) and 50 gastric cancers was examined by immunohistochemistry. Results The expression of TSLC1 in gas-tric cancer was 14. 00% which was lower than that in IN (46. 67%) and normal gastric mucosa (95. 00%, P<0. 05). TSLC1 ex-pression in high-grade IN was lower than that in low-grade IN and normal gastric mucosa (P<0. 05). TSLC1 expression in high-grade IN and gastric cancer was of no significant difference ( P>0. 05 ) . Expression of TSLC1 was significantly associated with lymph node metastasis and TNM in gastric cancer (P<0. 05). Conclusion The expression of TSLC1 is closely related to carcinogenesis, lymph node metastasis and clinical stage in gastric cancer, which suggest that TSLC1 may be a new target for the prevention and treatment of gastric cancer.

To prokaryotic express prokaryotically and to purify the efaA protein from Enterococus faecalis so as to provide the basis for the further study on the pathogenesis and clinical sero-diagnosis of endocarditis caused by E.faecalis, efaA gene of E.faecalis was amplified by PCR, the PCR-amplified product was digested with restriction enzymes and cloned into prokaryotic vector pET32a to construct the recombinant plasmid pET30a/efaA. This recombinant plasmid was confirmed by double enzyme digestion with BamhI and Xhol and then subjected to sequencing, and transformed to E.coli BL21 (DE3). Expression of the fusion protein was induced by IPTG, and analyzed by SDS-PAGE and Western blotting. The recombinant fusion protein was purified by His-binding affinity chromatography. It was shown that efaA gene of 943 bp in size was amplified from Enterococus faecalis and the recombinant plasmid pET30a,/ efaA was successfully constructed and expressed in E.coli BL21. The purified product was found to be 34 kDa in molecular weight as demonstrated by SDS-PAGE and Western blotting. It is evident that the efaA protein of E.faecalis can be successfully expressed and purified.

Objective To study the application value of chest low-dose spiral CT for the screening of lung cancer in physical examinees . Methods The data of physical examinees that underwent chest low-dose CT for screening of lung cancer from January 2013 to January 2016 was analyzed, the index included the correlation between age and sex with the detection rate of lung nodules and the detection rate of lung cancer, pathological types and clinical stages of lung cancer and so on. Results 9762 physical examinees underwent chest low-dose spiral CT, the detection rates of pulmonary nodules and lung cancer were 58.16%(5678/9762) and 0.34 % (33/9762) respectively. The detection rate of lung cancer increased with the age, the highest detection rate [1.13 % (8/709)] of lung cancer was found in 80 years old and above, the rate in 60-69 years old group was 0.36%(6/1641). In 33 cases of lung cancer, there were 22 cases in stage Ⅰ, 5 cases in stageⅢ, and 6 cases in stage Ⅳ. Early diagnosis rate was 66.67 % (22/33). Compared with the conventional thoracic spiral CT, the low-dose chest CT had loner radiation dose (P<0.05), and the image can meet the diagnostic requirement. Conclusions Chest low-dose spiral CT has great advantages in the detection of pulmonary nodules and early stage lung cancer. The application of low-dose CT in physical examinees, especially in high-risk groups of lung cancer, can improve the positive rate of lung cancer screening.
