Breast cancer is the most frequent malignant tumors in women,The p53 gene is found to be one of the most closely associated tumor-suppressor genes in human malignant tumors.Some studies have shown an association between polymorphisms at the forth exon (codon 72) and human malignancies recently.At present, the domestic investigations concerned the p53 polymorphisms mainly in lung cancer, uterocervical cancer,esophagus cancer,ovarian cancers,but the studies in breast cancer are rare in domestic.The correlation between the genotypes of p53 codon 72 polymorphism and breast cancer were reported.

3.EVALUATION ON THE EXTERNAL QUALITY ASSESSMENT OF NEWBORN SCREENING FOR INHERITED METABOLIC DISEASE LABORATORY

Objective To evaluate our results of external quality assessment (EQA) program of the newborn screening laboratory for 6 years, summarize the problems in the screening measurement and improve the quality of measurement.Methods The phenylalanine (Phe) and thyrotropin (TSH) values in dried blood spot specimens on filter paper distributed by national center were measured by fluorometric method and time -resolved fluorescence im-munoassay ( DELFIA ) respectively.The results sent by national center were analyzed .Results In the 90 specimens of 18 batches, the bias range of Phe and TSH measurement was wide (-62.50%~66.67%and-66.67%~54.31%respectively) and their average values were 8.62%and 10.18% respectively;85 quantitative results of Phe and 84 quantitative results of TSH measurement were in the quality control , accounting for 94.44%and 93.33%respective-ly.The qualitative analysis of 90 Phe and TSH specimens all conformed to the expected results , accounting for 100%.The result of every batch was up to 80%.Conclusion Our neonatal screening laboratory is in control and it is useful to evaluate the measurement competence , find out the problems in the Phe and TSH measurement and re-solve them in time through participated in the national EQA in neonatal screening laboratory , so as to improve the quality of measurement and ensure the accuracy and reliability of results .
4.SEDATION AND HYPNOSIS EFFECT OF JIAOTAI PILLS ON RAT INSOMNIA IN PCPA RAT MODEL

Objective To discuss the sedative and hypnotic effect of Jiaotai Pills on insomnia in PCPA rat model .Methods The PCPA ( PCPA acid) rats insomnia model was established, and the rats were divided into the model group, high, middle and low dosage of Jiaotai Pills group, and diazepam positive control group.The high performance liquid detective method was adopted to observe the effect of Jiaotai Pills on rat insomnia and hypothalamus neurotransmitters 5-HT and GABA.Results Compared with the model group, there existed significant difference in the insomnia state of rats and the central neurotransmitters 5 -HT in the high and middle dosages of Jiaotai Pill group and diazepam group.However, in the comparison of the high and middle dosages of Jiaotai Pill group and the model group, there was no significant difference in GABA in the central nervous system .Conclusion Jiaotai Pills has sedative and hypnotic efficacy.Its mechanism may be related to the effects of central neurotransmitter 5 -HT.Its relationship with GABA in the central nervous system is not obvious.The research has significant influence on the further development of Jiaotai Pills.
5.Application of the severity score of hand foot and mouth disease in children

Objective To develop the severity score of pediatric hand foot and mouth disease (HFMD),guide health care providers in the early detection and intervention of critical ill cases,in order to improve the survival rate and reduce the incidence of complications and mortality.Methods Based on PCIS,the severity score of HFMD in children was designed by adding fever and nervous system damage symptoms,which was the common symptoms of severe cases.The data of the cases who admitted in April 2008 to November 2010 (group A,88 cases) was retrospectively analyzed to evaluate the validity and credibility of the score.The score were prospectively applied to all of the HFMD cases who admitted from May 2011 to October 2013(group B,118 cases),delivering corresponding treatment and care measures based on scoring result as well.At last the outcome of the two groups was compared.Results It exhibited quite good consistency between the scores and the clinical condition of patients with HFMD.The general information of two groups was not significantly different,but the survival rate,the incidence of neurological sequel and the mortality between two groups were significantly different by x2 test.Conclusions The score can objectively and accurately assess the severity of HFMD in children.Clinical application of this scoring method to assess the children with HFMD can identify critical ill cases as early as possible.Corresponding intervention delivering in time may improve the outcome.It is proved to have positive clinical value to increase the survival rate,decrease the incidence of neurological sequel and the mortality of critical ill HFMD cases,and is worthy of clinical application in pediatric department.
6.Chromosome abnormality analysis of 7 036 chorionic villi inspontaneous miscarriage cases by multiplex ligation-dependent probe amplification

Objective To investigate chromosomal aneuploidy of chorionic villior tissue from embryo arrestor spontaneous miscarriage by the method of multiplex ligation-dependent probe amplification(MLPA), and to provide genetic etiology data for recurrent spontaneous abortion(RSA) and embryo arrest.Methods The clinical retrospective analysis was used during November 2013 to October 2016 in Shenzhen Maternity and Child Healthcare Hospital.The genomic DNA of 7 036 chorionic villi or tissue samples from embryo arrestor pregnancy loss were extracted by resin and Proteinase K, following detected by multiplex ligation-dependent probe amplification, and then analyzed by descriptive and frequency statistic using SPSS17.0 software.Results Totally 2 984 samples (42.41%) with chromosomal aneuploidy were detected, of which, the top five were trisomy 16(826)11.74%, monosomy X (Turner,401)5.70%, trisomy 22(247)3.51%, trisomy 13(149)2.12% and trisomy 21(144)2.05%.Aneuploidy 19 or 17 were rare,just 1 and 3 cases respectively.The aneuploidy of chromosome 1 was not discovered yet.There were 161 samples (2.28%) for segmental deletion and/or duplication cases.Besides of the monosomy X(Turner,401), monsomy 21 was also high frequency(17,0.24%).In addition, 56 samples(0.80%)were double trisomic,while 2 samples(0.03%)were tris-trisomy.Thirteen samples(0.18%)were heterosome trisomy,and five samples(0.07%)were Turner combined trisomy.Fifteen samples(0.21%) were unbalanced translocation of the arm,while 20 samples(0.28%) were nonhomologous chromosome translocation.Conclusions Trisome and monosome were the majority of chromosome abnormality (above 90%), as well as the main cause of spontaneous miscarriage.Meanwhile the chromosome aneuploidy (including segment deletion or duplication) could be screened by MLPA quickly, thoroughly and efficiently, further more discover the carrier of balanced translocation in couples and provide valuable genetic information for next pregnancy.
7.Simultaneous Determination of Bavachin and Bakuchiol in Psorslea corylifolia by HPLC

OBJECTIVE:To establish a method for content determination of bavachin and bakuchiol in Psoralea corylifolia.METHODS:HPLC method was adopted.The determination was performed on Shim-Pack VP-ODS column with mobile phase consisted of acetonitrile-water (gradient elution) at the flow rate of 1.0 mL/min.The detection wavelength was set at 246 nm,the column temperature was 30 ℃,and the sample size was 10 μL.RESULTS:The linear ranges of bavachin and bakuchiol were 48.9-489 ng(r=0.999 8),784-7 840 ng(r=0.999 9).RSDs of precision,stability and reproducibility tests were all lower than 2.0%.Average recoveries ranged 95.30%-99.63% (RSD=1.45%,n=9),96.89%-100.82% (RSD=1.36%,n=9).CONCLUSIONS:The method is simple,accurate,stable and reproducible,and can be used for the simultaneous determination of bavachin and bakuchiol in P corylifolia.
8.Determination of Gastrodin in Qianglitianmaduzhong Capsule by HPLC

OBJECTIVE: To develop an HPLC method for the determination of Gastrodin in Qianglitianmaduzhong capsule. METHODS: The samples were separated on Diamond C18 column (250 mm?4.6 mm,5?m).The mobile phase consisted of acetonitrile - 0.1% phosphoric acid(2∶98) at a flow rate of 1.0 mL?min-1.The UV detection wave length was set at 210 nm and the column was kept at room temperature. RESULTS: The linear range of Gastrodin was 0.126 4~3.160 0 ?g(r=0.999 8), and the average recovery was 98.30%(RSD=0.8%,n=6).CONCLUSION: The method is suitable for the determination of gastrodin in Qianglitianmaduzhong capsule.

PURPOSEObservation of clinical efficacy of idarubicin in the treatment of patients with both newly-diagnosed and relapsed/refractory acute leukemia. 10.A Contrastive Analysis on Clinic Manifestation and TCD and MRI Results in Transient Ischemic Attack of Internal Carotid Artery System

Objective To study the diagnostic value of TCD and MRI in transient ischemic attack (TIA) of internal carotid artery system.Methods The clinic data was analyzed to contrast the relation of the clinic manifestation and the TCD and MRI results of 66 patients suffered from TIA of the internal carotid artery system.then chi-square test was adopted.Results In TIA interval, extensive hemodynamics changes were occurred in the intracranial arteries among 60 patients(90 9%), and 22 patients(33 3) presented different degrees of stricture on the corresponding symptom side, MRI test showed lacunar infarct or small infarct foci among 50 patients(75 8%), and ischemic leukoenporosis among 35 patients(53 5%). But the foci took a two-side distribution and were relatively symmetrical. No significant correlation existed between the symptom and the foci,and the positive rate of TCD did not show remarkable difference between the symptoms and the foci, and the positive ratio of TCD did not show remarkable difference whether the MRI infarct foci existed or not. Meanwhile, there were also no notable distinction between the stricture change of arteries in the corresponding symptom side with the infarct foci and that of arteries in the corresponding symptom side without the infarct foci.Conclusions TCD can find the change of arteries related in TIA of the internal carotid artery system, and can be used as routine and primary screening. While MRI may bear no special clinic significance to TIA of the internal carotid artery system, but it has localization meaning to CWS, a clinical subtype of TIA.
