1.Oral inoculation with Lactobacillus live vaccine expressing hCG? induces an anti-hCG? antibody response in reproductive tract and the sera of in different strains of mouse
Chinese Journal of Immunology 2001;0(07):-
Objective:To induce an anti-hCG? antibody response in reproductive tract and the peripheral by oral inoculation with a recombinant alive Lactobacilli expressing hCG? in different strains of mouse. Methods:The 108,109,1010 Lb.hCG?, a recombinant Lactobacillus expressing hCG?, were inoculated orally into 6-8-week-old female BALB/c and C57BL/6 mice. The immunized mice were all boosted with the same dose at interval of 3 weeks. An indirect ELISA was used to determine anti-hCG? IgG and IgA antibodies in vaginal lavage and serum that were got at the end of 2-8 weeks after the primary immunization. The B and T lymphocytes of spleen, uterus and vagina were prepared respectively, and their proliferation was analyzed by flow cytometry. The numbers of the cells secreting anti-hCG? IgG or IgA antibody were detected by ELISpot. Results:Inoculation of 109 or 1010 alive Lb.hCG? could induce similar response while 108 induce a lower attitude of response. The antiserum of either 109 or 1010 Lb. hCG? immunization after booster could neutralized more than 100 ng/ml of hCG antigen both in BALB/c and C57BL/6 mice. The B cell proliferation of the reproductive mucosa was more significantly higher than that of spleen in BALB/c mice(P

China Oncology 1998;0(04):-
Breast cancer metastasis is the main cause of tumor death and poor long-term effect in clinical treatment. In the process of metastasis, chemokine may play the main role. In the breast cancer cell, some chemokine and receptor were expressed excessively. It has been shown that through the interaction between chemokine and correspondence receptor, phagocyte and lymphocyte infiltration stimulated, tumor cell invasion into normal tissue with angiogenesis, thus the metastasis was formed. This article reviews the mechanisms of chemokine receptor CXCR4 and chemokine RANTES ,IL-8 MCP-1 in breast cancer metastasis.

ObjectiveTo compare the influence of laparoscopic and abdominal hysterectomy on cellular immune function. Methods There were 28 patientsreceived laparoscopic hysterectomy (laparoscopic group) and 28 patients treated with abdominal hysterectomy (abdominal group). The surgical effects and the changes of preoperative and postoperative T lymphocyte subsets of two groups were compared.ResultsThe intraoperative blood loss, anal exhaust time, hospital stay and postoperative morbidity of the laparoscopic group were significantly better than those of the laparotomy group[ (84.7 ± 21.7) ml vs.( 108.0 ± 23.8) ml,(19.3 ±4.1) h vs.(23.8 ±3.8) h,(5.12 ± 1.14) d vs.(7.81 ±2.27) d,7.1% (2/28) vs.17.9%(5/28),P < 0.05 ].CD4 and CD8 in the 1st day after operation of two groups were significantly lower than that before surgery respectively [ laparoscopic group:(38.41 ± 5.52)% vs.( 40.72 ± 6.46)%,(24.41 ± 3.78 )% vs.(26.33 ± 4.17)% ;abdominal group:(38.41 ± 4.97)% vs.(40.13 ± 6.12)%,(24.41 ±6.32)% vs.(26.25 ±4.56)%,P < 0.05 ]; but CD4 and CD8 in the 3rd day after operation in the laparoscopic group[ (40.15 ±6.29)%,(27.23 ± 5.12)%] almost returned to normal level before surgery, while CD4 and CD8 in the 3rd day after operation in the abdominal group [ (36.15 ± 5.12)%,(23.15 ± 4.87 )%] still had significant differences compared with that before surgery(P < 0.05).ConclusionsLaparoscopic hysterectomy with less blood loss, rapid postoperative recovery, fewer complications and less impact on cellular immune function,is superior to abdominal hysterectomy. Therefore,it deserves promotion and wide application.

Objective To optimize the technology conditions of ultrasonic-enzyme-assisted extraction for sanguinarine from Zanthoxylum nitidum. Methods Extraction rate of sanguinarine determined by HPLC was served as an index. The applicability of the extraction solvent added with acid and enzymatic hydrolysis pretreatment to the ultrasonic-enzyme-assisted extraction of Zanthoxylum nitidum was investigated.Ultrasonic power, extraction frequency and solvent volume were optimized by orthogonal experiment. Finally, ultrasonic extraction time was optimized in dynamic process. Results The optimal process was as follows:Zanthoxylum nitidum powder was extracted 3 times by ultrasonic-wave (250 W) with 40% ethanol (0.2%hydrochloride) as solvent (extracted for 15 rmin with 6-fold solvent at the first time, then extracted for 12 min with 3-fold solvent at the second and the third time, respectively). The extraction rate of sanguinarine was 88.6%.Conclusion The process is economic,efficient,energy-and time-saving,and provides experimental basis for industrial production of sanguinarine.

Chinese Journal of Endocrinology and Metabolism 2010;26(9):770-773
Objective To explore the difference involved in the activation of inflammation pathway and the plasma level of inflammatory factors in the subjects with different sorts of insulin sensitivity. Methods The study was carried out in 38 women, consisting of obesity (n = 22 ) and control (n = 16 ) groups according to body mass index. The insulin sensitivity was assessed by homeostasis model assessment of insulin resistance (HOMAIR). Plasma concentrations of interleukin-6 (II-6) and IL-1β were determined by enzyme immunoassay. Western blot analysis was used to examine total protein expression and phosphorylation levels of IκB kinase (IKK) ,inhibitor of nuclear factor-κB ( IκB ) in peripheral blood leukcocytes. Electrophoretic mobility shift assay (EMSA)was used to detect the binding activity of NFκB. Results The levels of fasting plasma insulin[62.2 ( 20.0-127. 0) pmol/L vs 19. 15 ( 14. 2-47. 8 ) pmol/L, P<0. 01], HOMA-IR[2. 32 ( 0. 76-5.49 ) vs 0.70(0.53-1.7),P<0.0l], HbA1 C[(5.42±0. 45 ) % vs ( 5.08 ±0. 38) %, P<0. 05], triglyceride[( 1.75 ±0. 68 vs 1.22 ±0. 58 )mmol/L, P<0. 05], plasma IL-6[3. 15 (0. 03-22. 2) pg/ml vs 1.26 (0. 74-6.06 ) pg/ml, P<0. 01], and IL-1 β[6. 53 ( 0. 84-36 ) pg/ml vs 3. 16( 1.48-8. 86 ) pg/ml, P<0. 01]in obesity group were significantly higher than those in control group. Compared with control group, the levels of IKKo, IKKβ expression and IκBα serine phosphorylation in obesity group were markedly increased, while the expression of IκBα was significantly reduced. Accompanied with the degradation of IκBα protein, the binding activity of NFκB in obesity group was significantly increased. Conclusions The plasma levels of IL-6 and IL-1β were significantly raised in obesity group. The activation of IKK-IκB-NFκB pathway is closely associated with the genesis and development of insulin resistance in obese subjects.
