Objective:To study the functional status of the monocytes-derived dendritic cells (DCs) plused with HBsAg from patients with chronic hepatitis B and their capacity of inducing the HBV- special cytotoxic T lymphocytes (CTLs),and explore a new method of inducing the special anti-HBV cell-mediated immunity.Methods:Monocytes of patients were isolated from peripheral blood and incubated with GM-CSF+IL-4+TNF-? to induce DCs generation. Those DCs were pulsed with HBsAg to induce HBV- special DCs. The phenotype of DCs including CD1a,CD80,CD83,CD86,CD40 and HLA-DR was characterized by FCM and the stimulating reaction of allogenic T lymphocytes was detected by MTT assay. Objective To develop the severity score of pediatric hand foot and mouth disease (HFMD),guide health care providers in the early detection and intervention of critical ill cases,in order to improve the survival rate and reduce the incidence of complications and mortality.Methods Based on PCIS,the severity score of HFMD in children was designed by adding fever and nervous system damage symptoms,which was the common symptoms of severe cases.The data of the cases who admitted in April 2008 to November 2010 (group A,88 cases) was retrospectively analyzed to evaluate the validity and credibility of the score.The score were prospectively applied to all of the HFMD cases who admitted from May 2011 to October 2013(group B,118 cases),delivering corresponding treatment and care measures based on scoring result as well.At last the outcome of the two groups was compared.Results It exhibited quite good consistency between the scores and the clinical condition of patients with HFMD.The general information of two groups was not significantly different,but the survival rate,the incidence of neurological sequel and the mortality between two groups were significantly different by x2 test.Conclusions The score can objectively and accurately assess the severity of HFMD in children.Clinical application of this scoring method to assess the children with HFMD can identify critical ill cases as early as possible.Corresponding intervention delivering in time may improve the outcome.It is proved to have positive clinical value to increase the survival rate,decrease the incidence of neurological sequel and the mortality of critical ill HFMD cases,and is worthy of clinical application in pediatric department.

Objective To analysis the risk factors for elevated ALT level in patients with Non-alcoholic fatty liver diseases (NAFLD).Method Fifty-six (male 25,female 31) NAFLD patients with elevated ALT and 153 NAFLD patients (male 64,female 89) with normal ALT and 160 (male 80,female 80) health subjects were selected.ALT was detected by rate assay,total cholesterol (TC),triglycerides (TG),low-density lipoprotein cholesterols (LDL-C),high-density cholesterol (HDL-C) and uric acid (UA) were detected by enzymatic method.Free fatty acid (FFA) was detected by immunoturbidimetry assay.Spearman correlation was used to analysis the relation between ALT and TC,TG,LDL-C,HDL-C,UA and FFA.Logistic regression was used to analyses the risk factor for elevated ALT level.Results TC,TG,LDL-C,UA and FFA levels were significantly higher in NAFLD group (including normal ALT group and elevated ALT group) than those in health group no matter male and female,HDL-C was significantly lower in these two groups when compared to control group,but binary logistic regression and spearman analysis showed that only UA and FFA were the risk factors for elevated ALT level and significantly positively correlated with ALT level,abnormal UA and FFA for male were associated with 3.96-fold(95CI％ =1.40～ 11.22,P=0.010) and 3.27-fold (95 ％ CI =1.14 ～ 9.40,P =0.028) of increased risk of elevated ALT level respectinely,correlation coefficient were 0.40(P=0.000),0.29(P=0.006),Meanwhile Exp(B) of elevated ALT level for female with abnormal UA,FFA were 4.49 (95CI％=1.79～11.28,P=0.001) and 3.17 (95CI％ =1.25～8.04,P=0.015),correlation coefficient were 0.34 (P =0.000),0.27 (P =0.003).Conclusion Elevated ALT level was not associated with dyslipidemias,but related to UA and FFA.They can be used as candidate markers for predicting severity of liver damage.

Objective To know normal reference value of heart-type fatty acid-binding protein (H-FABP) in serum. Methods Serum H-FABP levels of 126 healthy individuals were measured by ELISA. Levels of MYO, cTnI and CK-MB were assayed in the same time as control. The normal threshold value was calculated as mean value?2s. Results The normal concentration of H-FABP in serum was 3 79?3 52?g/L and the difference in age and gender wasn’t significant. Conclusions The normal reference value of serum H-FABP is 0～10 83?g/L.

Objective: To study the effects of dental treatment and psychological treatment on daytime brycomania. Methods: 16 patients with daytime brycomania were treated with a series of measures, including eliminating dental nidi, application of bite plate on the front teeth so as to make barriers to abrasion information coming into the center occlusions, and administering medicine with psychological and mental treatment. Follow up was conducted for 6 months.Results: The symptom was relieved after one to two- week treatment; complete recover was achieved in all patients in 2 months and no recurrence was observed in 6 months.Conclusion: The study shows emotional and psychical disorders may be factors for daytime brycomania, psychological treatment is an important part for cure.

Objective:Through the analysis of the influence indicators of average hospitalization and comparison of differences of these indicators among different clinical departments in a hospital, to evaluate the medical quality and management level of the hospital and provide scientific reference for improving the hospital management efficiency and benefits.Methods:Gender, age and the type of medical insurance were analyzed using the chi square test. The length of hospital stay through 2009 to 2013 were analyzed using F test for patients of different insurance type and using t test for patients from different departments. All tests were completed using SPSS18.0.Results: The average hospital stay was decreasing year by year. Compared with the overall average length of stay, the patients who pay free were stay longer in hospital except for a small number of departments of internal medicine. Besides, the severe cases also prolonged the average length of stay in hospital.Conclusion: To effectively control the average length of stay, the Physicians Examination in Three Levels should be strictly implemented and the information management system should be used scientifically. Reasonable measures should be taken to solve the existing problems.

BACKGROUND:Matrix extracel ular phosphoglycoprotein phosphorylated extracel ular matrix glycoprotein (MEPE) gene plays an important role in bone mineralization and absorption as wel as the balance of osteoblasts and osteoclasts. Studies on the function and regulatory mechanism of MEPE can provide new ideas for the treatment of osteoporosis. OBJECTIVE:To analyze the regulatory effects of transcription factor Runx2 on MEPE promoter in mouse preosteoblasts, thereby preliminarily studying the Runx2 effects in the process of bone formation and development. METHODS:First of al , the Runx2 eukaryotic expression vector was built according to the gene sequence of Runx2 in Genebank;then the dual luciferase reporter assay was employed to analyze the effects of Runx2 on transcription activity of MEPE promoters with different lengths in order to determine the promoter region in which Runx2 has significant effect. Afterwards, the effects of Runx2 on transcipition activity of MEPE gene promoter which induced by three MAPK signaling pathway inhibitors were investigated. Final y, real-time PCR was used to analyze the expression activity of MEPE gene promoter regulated by Runx2. RESULTS AND CONCLUSION:We successful y constructed the Runx2 eukaryotic expression vector. Dual luciferase reporter assay showed that Runx2 could increase the transcription activity of MEPE gene promoter in preosteoblasts, and the fragment area in which Runx2 exhibited the more significant up-regulatory effectiveness was (-300 to+66)366 bp. Runx2 could increase the transcription activity of MEPE gene promoter by activating the MAPK single pathway. The real-time PCR verified that Runx2 increased the expression activity of MEPE gene promoter. These findings indicate that Runx2 can regulate the express of MEPE gene promoter by the MAPK single pathway, in order to build the basis for exploring the process of bone formation and development.

Objective To investigate the clinic application of micro-implant anchorage in the treatment of maxil-lary protrusion malocclusion. Methods Twenty-two patients,aged 18 to 25 years old,with maxillary protrusion were divid-ed into two groups:experimental group and control group with 11 patients in each group. All patients were treated with ex-traction. Micro-screw palatal implant was used in the cases of experimental group as orthodontic anchorage ,and traditional anchorage composed of extraoral arch used in the cases of control. The cephalometric films were measured before and after treatment. Statistical methods were utilized to analyze the morphological changes of facial profile and hard tissues in both groups. Results The values of U1-NA(mm:3.08±1.18 vs 8.15±3.05) and U1-SN(101.90°±3.50° vs 117.90°±6.05°) were sig-nificantly decreased after treatment compared with those before treatment in the experimental group ( P<0.01). The value of U1-L1(123.98°±5.78°vs 103.89°±8.95°) was significantly increased after treatment (P<0.01). In control group, the values of U1-NA (mm:5.01±1.34 vs 9.12±2.13) and U1-SN(101.90°±3.97° vs 114.87°±7.69°)were significantly decreased after treat-ment. The values of U1-L1(126.01°±3.12°vs 112.98°±5.98°) and U6-PtPNS(mm:21.45±2.43 vs 18.36±2.19)were significant-ly increased after treatment (P<0.05). The value of U1-L1(19.48°±8.90° vs 13.01°±5.90°) was significantly changed in exper-imental group than that of control group, but the value of U6-PtPNS(mm:0.90±0.29 vs 3.78±0.12)was significantly changed in control than that of experimental group (P<0.01). Conclusion The maxillary protrusion malocclusion with micro-im-plant anchorage can be used as treatment for patients with maxillary protrusion that needs strong anchorage.

Objective To evaluate the orthodontic effect of surgery on mixed dentition period class Ⅲ malocclusion and to investigate the effective therapeutic method of the skeletal class Ⅲ malocclusion.Methods A total of 20 patients aged 8.5 to 9.1 years with early mixed dentition class Ⅲ skeletal malocclusion.The treatment was divided into two phases.During the first phase,the entire stuffy group was treated with a bonded maxillary expansion and protraction.During the second phase,the entire study group received multiple loop edgewise arch wire (MEAW) treatment until an ideal occlusion relationship achieved.Comparison of the pre-and post-orthodontic treatment cephalometric radiographs had been done after maxillary protraction and MEAW were applied for patients.Results In the 20 patients after orthodontic treatment,Class Ⅲ skeletal facial profile was improved,the retraction of anterior teeth was obvious,and there were significant changes in dentition; the profile of soft tissue and satisfactory occlusion relationship were significantly improved,as compared with the changes between pre-treatment and post-treatment.Conclusions Skeletal Ⅲ malocclusion in early mixed dentition can be successfully treated.The results can be satisfied with improved class Ⅲ skeletal and molar relationship,high-angle and profile of soft tissue.
