1.Clinical observation on treatment of nonalcoholic fatty liver disease complicating hyperuricemia by Zhifang I Decoction

OBJECTIVE: To observe the therapeutic effect of Chinese herbal recipe Zhifang I Decoction on nonalcoholic fatty liver disease (NAFLD) complicating hyperuricemia (HUA). METHODS: Forty-six patients suffering from NAFLD complicating HUA were randomly divided into treatment group (25 cases) with Zhifang I and control group (21 cases) with Xuezhikang Capsule. One course of treatment was 8 weeks. The data were processed by SPSS 11.0 statistical package after 2 courses of treatment. RESULTS: The total effective rate of the treatment group was 80.00%, which surpassed the control group (71.43%) (P<0.01); Zhifang I could improve the image of B-mode ultrasonography and was better than Xuezhikang in ameliorating the clinical symptoms (P<0.05); Zhifang I could significantly decrease the serum uric acid (UA) (P<0.01), while Xuezhikang had no obvious therapeutic effect on it (P>0.05); Zhifang I was as good as Xuezhikang in recovering alanine aminotransferase (ALT), aspartate aminotransferase (AST),gamma-glutamyltransferase (gamma-GT), total cholesterol (TC) and triglyceride (TG) (P>0.05). CONCLUSION: Zhifang I has good effect in treating NAFLD complicating HUA.
2.Transfecting pcDNA3-TRAF4-DM-TRAF improves proliferation of human breast cancer cell MDA-MB-231

Objective_To investigate whether overexpression of TRAF4 in human breast cancer may have impact on its cell proliferation.Methods_This study has two groups, MDA-MB-231 transfected with pcDNA3-TRAF4-DM-TRAF or pcDNA3 .We detected the expression and localization of TRAF4 with immunofluorescence and western blot.We detected the expression of the phosphorylation level of p70S6K and S6 with westernblot.Flow cytometry was used to detecte cell cycle.MTT Assay was used to detected cell reproductive capacity.Results_TRAF4 local-ized in both cytoplasm and nuclei in MDA-MB-231.Nuclear expression of TRAF4 in nuclei was lower than that in cytoplasm (P<0.05).After pcDNA3-TRAF4-DM-TRAF was transfected into the cells, the expression of TRAF4 in nuclei was increased (P<0.05).The phosphorylation level of p70S6K and S6 significantly increaseced (P<0.05,P<0.01).More S phase cells were recorded(P<0.01) by FCM.The cell proliferation was promoted by MTT ( P<0.01) .Conclusions_The expression of TRAF4 in nuclei may play an important role in the cell prolif-eratuion by promoting p70S6K and S6 activation.
3.Identification of genes associated with gastric cancer prognosis from TCGA datasets

Objective To identify genes associated with prognosis or differentiated type in gastric cancer from fre quently mutated genes or highly-expressed genes,using large-scale genomic data from The Cancer Genome Atlas.Methods The somatic mutation data,RNAseqV2 data and clinical information were downloaded from the TCGA website.The frequency of deleterious somatic mutations for each gene was counted to select the frequently mutated genes.DESeq2 was used to analyze the gene expression data.Then survival analysis was performed on genes highlyexpressed in the tumor tissue.Kaplan-Meier curves were generated by R-survival package,and significance was evaluated by log-rank test.Results The frequency for pathogenic mutations in PIK3CA and APC was significantly discordant between different grades of gastric cancer.2 040 genes were up-regulated in tumor tissue,while 2 357 genes were down-regulated.Among the up-regulated genes,7 genes were associated with poor prognosis of gastric cancer and one was associated with better prognosis.Conclusions Genes associated with differentiation types or prognosis in gastric cancer are identified.The result may clue us future research on potential prognostic markers in clinical treatment.
4.Determination of Methylmercury and Ethylmercury in Aquatic Products by Gas Chromatography-Mass Spectrometry with Stable Isotope-Labelled Internal Standard

A method for the determination of methylmercury ( MeHg ) and ethylmercury ( EtHg ) in aquatic products was developed using gas chromatography-mass spectrometry with stable isotope-labelled internal standard. After ultrasonication assisted hydrochloric acid extraction, MeHg and EtHg in samples were extracted into toluene under the presence of sodium chloride and then back-extracted into cysteine aqueous solution. The MeHg and EtHg were released from their complexes with cysteine by adding cupric ions, and then derived with sodium tetraphenylborate. Under the optimal chromatographic conditions, MeHgPh and EtHgPh, the resulting derivatives, were separated completely on a DB-5MS capillary column and detected by electron impact ionization mass spectrometry in the selective ion monitoring ( SIM) mode, and quantified by a stable isotope dilution method using the d3-methylmercury as internal standard. The calibration curves were linear in the range of 1-500 μg/L of MeHg and EtHg. Concentration of 0. 828 mg Hg/kg with relative standard deviation ( RSD ) of 3 . 2% ( n=6 ) was obtained for MeHg in GBW 10029 . This was in good agreement with the certified values of (0. 84±0. 03) mg Hg/kg. The average recoveries were 94%-101% and 81%-104% for MeHg and EtHg spiked in aquatic samples, with RSDs of 1. 9%-4. 7% and 3. 1%-8. 2%(n=6), respectively. The limits of detection (S/N=3) of the two targets were 0. 1-0. 3μg/kg. This method was sensitive, accurate and could meet the demand of the determination of methylmercury and ethylmercury in aquatic products.
5.Study on the effect of perindopril and benazepril on nude mice bearing esophageal squamous cell carcinoma

ObjectiveTo investigate whether there was inhibitory effect of angiotensin-Ⅰ converting enzyme inhibitors (ACEI) on esophageal carcinoma cell line.Methods The highest expression of vascular endothelial growth factor (VEGF) at mRNA level was screened in esophageal squamous cell carcinoma cell lines KYSE30,TE-1,EC109 and EC9706 by reverse transcriptionpolymerase chain reaction (RT-PCR) and which was transplanted and established nude mice xenografts model.After tumor formed,the nude mice were evenly divided into control group,perindopril group and benazepril group (n=6) of which 0.9% saline,4 mg/kg perindopril and 6 mg/kg benazepril was given respectively to nude mice in each group.The tumor size and the tumor inhibitory rate were measured.The expression of CD31 in xenograft tumor was tested by immunohistochemistry and the microvessel density (MVD) was assessed.ResultsThe highest expression of VEGF at mRNA level was in EC9706 cell line.At second and third week,there was no significant difference in xenograft tumor size between each group. At fourth and fifth week,there was no significant difference in xenograft tumor size between perindopril group and control group,and the tumor inhibitory rate was 24.6 % and 21.1 %.There was significant difference between benazepril group and control group (t=-2.450 and -3.120,P=0.035 and 0.008),and the tumor inhibitory rate was 33.1% and 45.4%.After treated with perindopril and benazepril,the expression of CD31 in xenograft tumor of nude mice decreased.The MVD of benazepril group was significantly lower than that of control group (10.98±1.18 vs 13.98 ± 1.76; t =-3.732,P =0.002),and there was no significant difference between perindopril group (12.41±1.15) and control group (t=-2.053,P=0.07).ConclusionsBenazepril (6 mg/kg) could significantly inhibit the growth of EC9706 cell line formed xenograft tumor of nude mice.One of the possible tumor inhibitory mechanisms was inhibiting new vessel forming in tumor.Perindopril (4 mg/kg) may have the similar effect.
6.Clinical efficacy of single span video-assisted thoracoscope surgery for elderly spontaneous pneumothorax

Objective To compare the clinical efficacy between single span video-assisted thoracoscope (VATS) surgery and traditional VATS surgery for spontaneous pneumothorax in elderly patients.Methods A total of 88 elderly patients diagnosed as spontaneous pneumothorax by chest X-ray and CT were selected and randomly divided into single span VATS group and traditional VATS group (n=44,each).The bleeding volume,duration of operation,hospitalization time,time of postoperative analgesic use,chest tube indwelling time,postoperative complications and adverse reactions were observed.All patients were follow-up for 1-2 years to record the recurrence rate and death rate.Results All patients were received VATS surgery successfully.Their clinical symptoms disappeared and X-ray showed that pneumothorax disappeared.There were significant differences in bleeding volume,time of postoperative analgesic use,hospitalization time between single span VATS group and traditional VATS group [(46.7±18.8)ml vs.(46.7±18.8)ml,(1.7±0.9)d vs.(2.4± 1.7)d,(6.3±1.7)d vs.(9.1±2.1)d,t=3.04,2.18,5.59,respectively,all P<0.05].The duration of operation and chest tube indwelling were shorter in single span VATS group than in traditional VATS group (both P<0.05).Patients with chest pain and incision numb after operation were less in single span VATS group than in traditional VATS group (11 cases vs.26 cases,7 cases vs.21 cases,t=10.50,10.26,both P<0.05).There were no significant differences in recurrence rate and death rate between the two groups.Conclusions Single span VATS surgery is an safe and effective treatment for spontaneous pneumothorax in elderly patients,which has a better postoperative recovery and fewer adverse reactions.

Modern wars result in a significant increase in the incidence of burn injury,and a lot of soldiers with extensive deep burn will occur intensively in a short time.At present,allogeneic skin graft is a principal method to treat extensive burn,but allogeneic skin source is in extremely short supply.Therefore,this review concentrates on the source of allogeneic skin,the necessity to establish allogeneic skin bank for preparedness against war,the present existing problems and main solutions to related problems.
8.Effects of Irbesartan on Expression of MMP-2,TIMP-2 and Ⅳ-C in Kidney of Diabetic Rats

OBJECTIVE:To investigate the expression of matrix metallopro teinase-2(MMP-2),tissue inhibitor of metalloproteinase-2(TIMP-2) and collagen Ⅳ(Ⅳ-C) in kidney of diabetic rats and the intervention effect of irbesartan.METHODS:30 male SD rats were randomly divided into three groups:normal control group(NC group),diabetes mellitus group(DM group) and diabetes mellitus+50 mg?kg-1 irbesartan group(DI group).DM and DI group were injected with streptozocin intraperitoneally to reduce diabetic model.After 8 weeks the expression of MMP-2,TIMP-2 and Ⅳ-C in kidney tissure were determined by immunohistochemistry method(SP) and RT-PCR.RESULTS:Compared with NC group,DM group showed the expression of protein and mRNA of MMP-2 significantly reduced while that of TIMP-2 and Ⅳ-C increased(P
9.Determination of Residual Solvents in α-Ketophenylalanine Calcium by Gas Chromatography

To develop a method for the determination of residual solvents inα-ketophenylalanine calcium by capillary gas chromatography. Methods:The residual solvents were separated on a DB-624(30 m × 0. 32 mm, 0. 25 μm) capillary chromato-graphic column with temperature programming. The column temperature was maintained at 40℃ for 1 min,and then raised to 180℃at a rate of 10℃·min-1 and maintained for 2 min. N2 was used as the carrier gas, and FID was used as the detector with temperature of 250 ℃. The injector temperature was 200 ℃ and the split ratio was 10∶1, and direct injection was adopted. Methanol, ethanol, ethyl acetate and tetrahydrofuran in α-ketoleucine calcium were detected using an external standard method. Results:The four solvents were separated completely. There was a good linear relationship between the peak area and the concentration of each solvent ( r=0. 997 2-0. 999 5). The average recovery of the four solvents was 95. 47%-100. 26%(RSD≤4. 7%, n=9). Conclusion:The method is rap-id, simple, accurate and sensitive, and can be used in the determination of residual solvents in α-ketophenylalanine calcium.
10.Determination of Tetrodotoxin in Human Urine and Plasma after Immunoaffinity Column Clean-up Using Ultra-Performanc Hydrophilic Interaction Chromatography Coupled with Triple Quadrupole Mass Spectrometry

Anultra-performancehydrophilicinteractionliquidchromatography-triplequadrupolemass spectrometric ( UPLC-MS/MS) method was developed for the determination of tetrodotoxin ( TTX) in human urine and plasma. After the sample was cleaned-up and concentrated by immunoaffinity column, the separation of the TTX was carried out on an Acquity UPLC BEH amide column (100 mm×2. 1 mm, 1. 7 μm) with gradient elution using mobile phases of 0. 1% ( V/V) formic acid in water and acetonitrile. The analyte was detected by positive electrospray ionization mass spectrometry in the multiple reaction monitoring ( MRM) mode, and quantified by external solvent standard calibration. The measuring ranges of TTX in urine and plasma were 0. 05-400 μg/L. The average recoveries were 92%-95% and 91%-96% for TTX respectively spiked in urine and plasma with relative standard deviations of 3 . 3%-7 . 2% and 3 . 9%-7 . 8% ( n=5 ) . The limits of detection (LOD, S/N=3) and limits of quantitation (LOQ, S/N=10) of TTX were 0. 02 μg/L and 0. 05μg/L for urine and plasma, respectively. This method is suitable for the detection of TTX in urine and plasma for both forensic and clinical purposes.
