Recent scientific achievements in cell and developmental biology have provided unprecedented opportunities for advances in biomedical research. The demonstration that fully differentiated cells can reverse their gene expression profile to that of pluripotent cells, and the successful derivation and culture of human embryonic stem cells (ESCs) have fuelled hopes for applications in regenerative medicine. Ethical issues concerning the use of cloned human embryos for the derivation of stem cells have stimulated the search for alternative methods for reversing differentiated cells into pluripotent cells. The present state of these reprogramming technologies will be reviewed and their relative success will be discussed.
2.The moderating effect of psychological resilience between family structure and children's peer relationship

ObjectiveTo compare children's resilience and peer relationship in divorced and complete family,and explore the moderating effect of psychological resilience between family structure and peer relationship.Methods208 children from divorced family and 465 children from complete family were surveyed.Results(1)There were significant differences in children's resilience(divorced family (3.58±0.73),complete family(3.83±0.59),t=-2.765,P=0.006) and peer relationship(divorced family (3.47±0.72),complete family(3.73±0.59),t=-2.069,P=0.042) between complete and divorced family.(2) Family structure had remarkable negative correlation with resilience and peer relationship(r1=-0.19,r2=-0.20),and resilience had positive correlation with peer relationship(r=0.43).(3)The moderating effect of resilience between family structure and peer relationship was significant(β=0.641,P<0.05).ConclusionParent divorce has negative influence on children's resilience and peer relationship.Furthermore,the relationship of family structure and peer relationship is mediated by resilience.The negative influence of parent divorce on peer relationship is not significant among children with high resilience,while it is significant on children with low resilience.
3.Preliminary Study on the Thinking Mode of Nephrology Clinical Pharmacists Reviewing Medical Orders

OBJECTIVE:To investigate the thoughts and ways of clinical pharmacist in nephrology department to review medi-cal orders,and to provide reference for rational drug use in nephrology department. METHODS:Based on specific cases of nephrolo-gy department,the thoughts and ways of clinical pharmacist in nephrology department to review medical orders were investigated from several aspects,including whether the therapeutic drugs had covered all the problems,drug duplication,drug selection,usage and dosage and course of treatments,drug interaction and compatibility. RESULTS & CONCLUSIONS:By establishing rigorous and systematic review thinking mode,based on their professional specialty,clinical pharmacists of nephrology department can sort the reviewing thought,fill in the gaps of medical orders reviewing and enhance the ability of participating in the clinical practice.
4.Effects of NLP workshop on resilience of children from divorced family

Objective To investigate the intervention effects of NLP workshop on resilience of children from divorced family.Methods A pre/post test and subsequent follow-up design between experimental group and control group were performed.Thirty-one children from divorced family with low resilience were intervened by NLP workshop,while the other thirty-one children with low resilience were chosen as the control group.Results There was no significant difference between experimental group and control group before the intervention on resilience.But after the intervention,the two groups achieved statistical significant difference on resilience(experimental group (3:68±0.37),control group(2.99±0.18),t=9.60,P=0.00).Furthermore,the results of follow-up showed that the effect of NLP workshop still existed after six months (total average score of resilience before intervention (2.96±0.17),subsequent follow-ups (3.69±0.35),t=10.40,P=0.00).Conclusion NLP workshop can enhance the level of resilience of children from divorced family.Effect of NLP workshop not only has short-term effect,but also has some long-term effect.
5.Survey on the pressures from night shifts and the countermeasures

Objective To investigate the pressures from night shifts among nurses from pediatrics department and find the countermeasures.Methods A self-designed questionnaire was used to investigate the pressures from night shifts among 112 nurses from pediatrics department as well as the reasons.Results 110 of the nurses(98.2%) assumed that the pressures from night shifts was intensive.The main reasons for the pressures came out of heavy workload,shortage of staff and stuff and the unsettled balance between job and family.Conclusion Such measures as reasonable distributions of nurses,abound supply of stuff and reduced working links may be effective for the alleviation of the pressures on nurses at night shifts,motivated activeness in them and then the enhancement of nursing quality.
6.Cri-du-chat syndrome: a case report and literature review

A neonatal girl with overextended knees admitted to NICU of our hospital was diagnosed as cri du chat (cat cry) syndrome.We collected 34 cases of cri du chat reported in journals since 2000,the clinical features of total 35 cases were retrospectively analyzed.Among 35 cases 12 were boys and 23 girls.The most common clinical manifestations were characteristic face features(100%),difficult feeding(100%) and typical sound of cry(94%).The main complains at hospital visit were typical cry,difficult feeding and cyanosis in the neonatal period,while in childhood period were recurrent respiratory infection,developmental retardation and other abnormalities.Most cases were diagnosed in the neonatal phase,while 85.3% were in the first year.The diagnosis was based on karyotype analysis; chromosome 5 short arm deletion (5P-) was the most significant genetic variation and clinical features were associated with the position of deletion.
7.Identification and application of monoclonal antibodies against aspergillus fumigatus

Objective:To produce monoclonal antibodies(McAb) against aspergillus fumigatus and to establish rapid assay for the measurement of aspergillus fumigatus antigen.Methods:Recombinant galactomannoprotein of aspergillus fumigatus(AFMP1) was used to immune BALB/c mice.Monoclonal antibodies against AFMP1 were produced from hybridoma.Results:Three hybridomas producing antibodies against AFMP1 were obtained.IgG isotypes of three McAb were IgG1,IgG2a and IgG2b.The affinity constants(K) were 1.2?10 10 ,4.56?10 9 and 1.81?10 10 mol/L.The antibodies were proved to be specific for aspergillus fumigatus by Western blot and recognized different epitopes on AFMP1 by the additivity assay.An sandwich enzyme-linked immunosorbent assay(ELISA) to detect AFMP1 was established to produce standard curve which showed linearity between 0.1~60.0 ng/ml with a sensitivity of 0.1 ng/ml.Conclusion:These results show three hybridomas producing high specificity and affinity monoclonal antibodies against AFMP1 and can provide for rapid assay for the measurement of aspergillus fumigatus antigen.
8.Determination of Amantadine Hydrochloride in Pediatric Paracetamol and Amanatadine Hydrochloride Granules by GC

Objective:To establish a GC method for determining the content of amantadine hydrochloride in pediatric paracetamol and amanatadine hydrochloride granules. Methods:The ingredients were separated on an Agilent DB-5 quartz capillary column (30 m ×0.25 mm, 0.25 μm), the carrier gas was N2,and the flow rate was 1 ml·min-1. The column temperature was maintained at 130℃, the injection temperature was 250℃,the detection temperature was 280℃ with an FID as the detector , the injection volume was 2 μl, and the split ratio was 20∶1. Results:A good linear relationship was obtained between the peak area and the concentration of amantadine hydrochloride within the range from 0.103 2 to 6.456 0 mg·ml-1(r =0.999 9). The average recovery was 100.18%(RSD=0. 11%, n=6). Conclusion:The method is specific, accurate, reliable and reproducible, which can be used in the quality control of pediatric paracetamol and amanatadine hydrochloride granules.
9.Rapid Identification of Staphylococci in Positive Blood Culture Bottles by PCR

OBJECTIVE To establish a method of polymerase chain reaction(PCR) for detecting staphylococci in positive blood culture bottles.METHODS Genomic DNA in 493 positive blood culture bottles was extracted by guanidine hydrochloride and benzenemethanol,then genes 16S rRNA,ssa and mecA were amplified by PCR to identify staphylococci.Finally,the results of PCR were compared with that of traditional method.RESULTS To compare with traditional method,as the golden standard the sensitivity and specificity of PCR method were 98.6% and 100.0%,respectively,the detection could be finished in four hours.Method of PCR was better than traditional method in detecting meticillin-resistant staphylococci.CONCLUSIONS The PCR-based assay is simple,rapid,sensitive and specific,it can be used to detect staphylococci in positive blood culture bottles.
10.Transfection of human hyperplasia suppressor gene and hyperthermia have a synergistically apoptotic effect on colon cancer cell HT-29 in vitro

Objective:To investigate the suppression effect and apoptosis of the colon cell line HT-29 treated with adenovirus-expressed human hyperplasia suppressor gene(hHSG)and hyperthermia.Methods:Replication-defective recombinant adenovirus containing hHSG was constructed,and then infected into HT-29 cell line to detect its overexpression by western blot.The synergic effect with hyperthermia was observed,cell survival was assessed using Vi-CELL TMXR Cell Viability Analyzer,and cell apoptosis and cell cycle distribution were determined by flow cytometry.Results:After the cells were treated with 100 MOI(multiplicity of infection)Ad5-hHSG and heated at 42 ℃ for 1 h,the number of viable cells decreased sharply,compared with single treatment with Ad5-hHSG or 42 ℃ for 1 h(P
