Objective To analyze the expression stability of the three widely used reference genes β-glueuronidase (GUSB), glycera]dehydes-3-phosphate dehydrogenase (GAPDH), β2-microglobulin (β2-M) in Chinese lung cancer tissue specimens and normal lung tissue specimens. Methods Gene expression wasmeasured by quantitative real time PCR and expression stability was analyzed with two widely used softwares genorm and normfinder. Results The intra-and inter-group difference of GAPDH is maximum (The intra and inter-group s is 1.07 and 0.93 respectively, |△ Ct|=2.01±1.06; P =0.000). The mean of these three genes' Ct value is the most stable one analyzed by the two softwares. But the t test showed that the mean of Ct value of GUSB and β2-M is the unique combination that had the minimum intra-and inter-group difference, with no statistically significant differences between normal and malignant samples (The intra-and inter-group s is 0.53 and 0.79 respective]y, |△Ct|= 0.73±0.53; P =0.053). Conclusion It is inappropriate to normalize data derived from lung tissue specimens using one of these three housekeeping genes alone. Among the different combinations of these three genes, the mean of the Ct values of GUSB and β2-M is the best choice as the internal control of lung tissue specimens.

Objective To explore the effect of 25% insulin lispro( Mix 75/25) intensive treatment on plasma glucose and pancreatic function in newly diagnosed type 2 diabets. Methods Sixty patients with newly diagnosed type 2 diabetes were treated by 2 months 25% insulin lispro(Mix 75/25) treatment. The level of fasting plasma glucose (FPG) ,2 hours postprandial glucose (2hPG), glycosylated hemoglobin A1c (HbA1c), the fasting insulin and Cpeptide, insulin resistance index (HOMA-A), insulin secretion index (HOMA-B), insulin sensitivity index (ISI) and the ratio of fasting insulin to fasting glucose(FINS/FPG) were compared before and after 25% insulin lispro(Mix 75/25) treatment. Results After the treatment,FPG,2hPG,HbA1c and HOMA-A were significantly decreased( all P<0. 01 ) ;the fasting insulin and C-peptide,FINS/FPG, HOMA-B and ISI greatly increased( all P<0. 01 ). Conclusion 25% insulin lispro( Mix 75/25) intensive treatment can effectively improve plasma glucose and pancreatic function in newly diagnosed type 2 diabetes.

Objective To observe the changes in NO content and NOS activity of serum and the testis in Wistar rats with type I diabetes mellitus. Methods 63 rats were randomly divided into control group(C) and diabetes group(D). Diabetes was induced by intraperitoneal injection of Alloxan. Rats with diabetes were observed for 1 week(D 1), 2 weeks(D 2), 3 weeks(D 3), 4 weeks(D 4), 5 weeks(D 5), 6 weeks(D 6), 7 weeks(D 7) and 8 weeks(D 8), respectively, and they were sacrificed at the end of each observation period. Serum was obtained and testis homogenates were prepared, and the levels of NO content and NOS activity of them were determined respectively. Results Compared diabetes group with control group, NO contents and NOS activity of serum from diabetic rats were higher, then NOS activity began to decline to approach the control level. In the testis homogenates, NO content dropped slightly after injection of Alloxan, while NOS activity showed no significant change. Thereafter, both NO content and NOS activity of testis homogenates were increased. Conclusion There are changes in NO content and NOS activity of serum and testis of rats after diabetes are induced with Alloxan.

Objective To observe the protective effects of mycelium of cultured Cordyceps sinensis on testis in diabetic Wistar rats. Methods 21 rats were divided into control group(C), diabetes group(D) and treatment group (T) at random. Diabetic model was reproduced by intraperitoneal injection of alloxan. After having been treated with Jinshuibao capsule for 8 weeks, serum levels of glucose, insulin, testosterone and levels of high energy phosphate compound, MDA, SOD, GSH-Px, and ATPases in the testis were determined respectively. Results Compared with control group, testicular mitochondria showed lipid peroxidase injury, with disturbed energy metabolism, and the activity of ATPases was lowered. All the changes in serum and testes were improved when Jinshuibao capsule was given. Conclusion Mycelium of cultured Cordyceps sinensis might have protective effects on the testis in diabetic Wistar rats.

Objective To determine the causative organisms and antimicrobial susceptibility of community- and hospital-acquired pneumonia (CAP and HAP) in type 2 diabetes in Fujian Provincial Hospital. Methods The data of becteria spectrum and their drug susceptibility in patients with type 2 diabetes complicated by pulmonary infection were retrospectively analyzed in January 1995 to October 2006. Results The isolated bacteria of sputum culture of 494 cases included 73 gram-positive cocci(16.7%),139 gram negative bacilli(31.9%)and 224 fungus(51.4%).G+ cocci mainly included staphylococci,G-bacilli mainly included Klebsiella pneumoniae,Bowman immovability bacillus and Pseudomonas aeruginosa. In both CAP and HAP,fungus and G- bacilli were the dominant pathogens. The pathogenic bacteria were resistant to multi-antibiotics,and the resistant rates from HAP patients was higher than those from CAP. Of the bacterial strains isolated from blood culture,G-bacilli constituted 87.5%. Conclusions Fungus and G-bacilli were the dominant pathogens.Phlegm culture and drug sensitive test are helpful for reasonable use of antibiotics for patients with type 2 diabetes complicated by pulmonary infection in clinical practice.
