1.Neuroprotective effects of citicoline on 6-hydroxydopamine-treated mesencephalic dopaminergic neurons in primary culture

Objective To study the neuroprotective effects of citicoline(CC) on the toxicity induced by 6-OHDA towards dopaminergic mesencephalic neurons in primary culture-Parkinson′s disease(PD) model in vitro and its mechanism. Methods Mesencephalic neurons in culture were prepared from embryonic 15-day Wistar rats.Cultures were treated for 6,8,10 d with various concentrations of CC(2,1,0.1,0.01 and 0.001 mmol?L~(-1)).At 11th day,the cultures were co-treated with the toxin 6-OHDA(50 ?mol?L~(-1)) for 0.5 h,the cells were collected.Seven groups were categorized as follows:CC(2,1, 0.1,0.01 and 0.001 mmol?L~(-1)) +6-OHDA,control and(6-OHDA) group.The cell viability was evaluated with MTT assay.Intracellular free Ca~(2+),[Ca~(2+)]i,was labeled by using the fluorescent dye Fluo3-AM and detected by flow cytometer.By measuring the intracellular Rhodamine(123 fluorescence) density with flow cytometer,mitochondrial membrane potential(MMP) was evaluated.(Results Cultures) were treated with 2,1 and 0.1 mmol?L~(-1) CC,the viability of cell was increased(P
2.Clinical values of fast detection of hs-CRP in distinguishing between transudate and exudate

Objective To evaluate the clinical value of fast detection of high sensitivity C-reaction protein(hs-CRP)in distinguis-hing between transudate and exudate.Methods 100 cases of patients were enrolled in the investigation.The levels of TP,LDH, GLU in serum and pleural effusion respectively,as well as the level of hs-CRP in the pleural effusion.Results According to Light standard,82 cases of patients were diagnosed as transudate(transudate group)and 18 cases of patients were diagnosed as exudates (exudates group).The sensitivities of Rivalta test,TP(pleural effusion)/TP(serum)and LDH(pleural effusion)/LDH(serum) were 83.14%,80.49% and 79.27%,respectively.Hs-CRP showed 84.15% sensitivity and 27.78% specificity at a cutoff value of 5 mg/L.TP(pleural effusion)/TP(serum)and LDH(pleural effusion)/LDH(serum)had the highest specificities,and both were 100.00%.The levels of LDH(pleural effusion)/LDH(serum)and hs-CRP in transudate group were significantly higher than those in exu-dates group(P<0.05).Combined detection of hs-CRP,TP(pleural effusion)/TP(serum)and LDH(pleural effusion)/LDH(serum)had the highest specificity (100.00%).Conclusion The fast detection of hs-CRP in pleural effusion is sensitive,combining with other tests can prove its specificity.The fast assay of hs-CRP has special clinic value in the distinguish between exudate and transudate.

Objective Explore the effect lactobacillus on allergic rhinitis (AR) rat serum Il-4 ,Il-5 ,IFN-γ concentration and the influence of immune markers .Methods SD rat 60 were randomly divided into 4 groups ,each group of 10 only .Lactic acid bacteria group every day with lactobacillus living bacterium preparations irrigation stomach 3 weeks ,establish rats LuanBai protein sensitiza-tion model ,positive drug control group irrigation stomach loratadine ;Another 20 rats used for passive cutaneous anaphylaxis (PCA) ,inspection in the rat model preparation and treatment .Determination test group rats serum IL-4 ,IL-5 ,IFN-γ,the content of Eos in nasal secretion was detection .Results Model group IL-4 ,IL-5 content was significantly higher than normal control group (P<0 .05) ,IFN-γlevels were low ,lower than the normal control group(P<0 .05) .Lactic acid bacteria group IL-4 ,IL-5 content were significantly lower than the model group(P<0 .05) ,IFN-γcontent increased ,higher than the model group(P<0 .05) .The in-dex of lactobacillus group with positive drug compared difference was statistically significant .Conclusion Lactic acid bacteria can cut AR rats serum IL-4 ,IL-5 level ,increase IFN-γlevel ,adjust Th1/Th2 lymphocyte subsets balance ,reduce nasal mucosa allergic inflammation .
5.Clinical observation of laparosopy operation on the treatment of uterine appendages lump

Objective To evaluate the advantage of uterine appendages lump operation by laparoscopy and the choose of the patient. Methods 215 cases of ovrarian tumor operation and fallpian tubes operation by laparoscopy,50 cases of similar period and similar operation indication were chosen by laparoscopy or by abdorminial operation apiece, blood loss, postoperative analgesic rate, the time of using antibiotics, hospitalization day were compared. Results Uterine appendages lump operation by laparoscopy were all operated successfully, compared with abdorminial operation, the laparoscopy group had less blood loss,less drug after operation,less pain, shortened hospitalization day.Conclusion As long as choosing the appropriate cases before the operation, uterine appendages lump operation by laparoseopy has the advantage of minimal trauma, recovery fastly,less postoperative complication and safety.
6.Diagnostic value of combined detection of serum CEA, CA125, CA153 and CA199 in patients with lung cancer

Objective To evaluate the clinical value of combined detection of serum CEA, CA125,CA153 and CA199 in lung cancer diagnostec. Methods The serum level of CEA,CA125,CA153 and CA199 in 58 patients with lung cancer,38 patients with benign lung diseases and 40 healthy persons were determinated by Chemiluminesent immunoassay. The data were analyzed to evaluate the diagnostic value of the combination use of four markers. Results The serum levels of CEA,CA125,CA153 and CA199 in lung cancer group were 5.6 (1.0-619. 0) μg/L,124. 5 (5.3 - 994. 8) U/ml,14.4(3. 1 - 800. 0) U/ml and 21.8(2.0 - 1200.0) U/ml respectively ;The serum levels of CEA, CA125, CA153 and CA199 in benign lung diseases were 1.9 (0. 6 -14.4) μg/L, 17. 7 ( 1.8 - 303.6 ) U/ml, 6. 8 ( 2. 4 - 20. 1 ) U/ml and 9. 3 ( 2. 0 - 82. 6 ) U/ml respectively. The serum levels of CEA, CA125, CA153 and CA 199 in healthy controls were 1.4 (0. 6 - 5.0 ) μg/L, 9.6 (4. 4 -36. 5 ) U/ml,6. 6 (3.8 - 19. 9) U/ml and 6. 4 (2.0 - 58. 1 ) U/ml respectively. The levels of markers in the lung cancer group were significantly higher than those in patients with benign lung disease and healthy controllers (Ps < 0. 01 ). The sensitivity of combined measurement was 94. 8%, significantly higher than a single measurement ( CEA 48. 3% ,CA125 75.9% ,CA153 20.7% ,CA199 31.0% ,Ps <0. 01 ). Conclusion The combined detection of CEA, CA125, CA153 and CA199 can significantly improve detection sensitivity of lung cancer and might provide valuable laboratory proof for early diagnosis of lung cancer.
7.P-selectin and Platelet activating factor in acute patients with deep venous thrombosis

Objective To evaluate the status of P-selectin and platelet activating factor(PAF) in vivo in patients with deep venous thrombosis(DVT) and to observe their changes under interneving of medicines.Methods P-selectin and PAF of fourty patients and twenty normal subjects were fluorescence labled with corresponding monoclonal antibodies by flow cytometry( FCM ) and immunologic method respectively. Results P-selectin and PAF in patients with DVT were higher than that in normal subjects in early period of the disease, and they significantly decreased in different time after patients were treated. P-selectin was significantly different between patients who received sodium ozagrel treatment and those who not (P <0.05 ), but PAF was similar( P > 0. 05 )after fourteen days. One month later, P-selectin and D-dimer in DVT patients were lower than before. However, the positive rate of P-selectin of DVT was still higher than normal subjects. Conclusions The platelet is activated in vivo in patients with DVT, so does fibrinolysis. Sodium ozagrel can decrease activity of platelet. P-selectin and PAF may be used diagnostic markers. Post-discharge patients are still at high-risk and must be regularly followed-up.
9.Endoplasmic reticulum stress induced by Thapsigargin in vascular smooth muscle cells of rat coronary artery

AIM:To establish the endoplasmic reticulum stress ( ERS) cell model in vascular smooth muscle cells ( VSMCs) of Sprague-Dawley (SD) rats.METHODS:Under sterile condition, the coronary arteries were isolated from SD rats .The primary VSMCs were cultured by tissue-sticking method , and observed the basic morphological characteristics under optical microscope .The marker proteins of VSMCs including α-smooth muscle actin (α-SMA) and smooth muscle myosin heavy chain ( SM-MHC) were identified by immuno-fluorescence technique .VSMCs were treated with thapsigargin (0.5, 1 and 2 μmol/L) for 24 h, and the expression levels of binding immunoglobulin protein (BiP) and C/EBP homologus protein (CHOP), the marker molecules of ERS, were detected using Western blotting.RESULTS:VSMCs climbed out from coronary artery tissues after about six days , and the cells had a nice state and formed the VSMC-like typical peak valley.The results of immunofluorescence technique show that the marker proteins of VSMCs ,α-SMA and SM-MHC were expressed significantly .The results of Western blotting show that the protein expression levels of BiP and CHOP were increased by thapsigargin in a dose-dependent manner .CONCLUSION:VSMCs can be successfully cultured by tissue-sticking method and built the ERS model induced by thapsigargin .
10.Expression and significance of fascin in ductal intraepithelial neoplasia and invasive ductal carcinoma

Objective_To study the significance of fascin protein expression in ductal intraepithelial neoplasia ( DIN) and invasive ductal carcinoma ( IDC) .Methods_Thirty cases of usual ductal hyperplasia ( UDH) , 30 ca-ses of flat epithelial atypia (FEA,DIN1a), 15 cases of atypical ductal hyperplasia (ADH,DIN1b), 2 cases of DCIS grade1(DIN1c),10 cases of DCIS grade2(DIN2)and 18 cases of DCIS grade3(DIN3)were immunohisto-chemically investigated using monoclonal antifascin antibody and compared with 120 cases of invasive ductal carci-noma (IDC).Results_Fascin protein expression was not found in normal breast tissue, UDH, FEA (DIN1a), ADH(DIN1b),DCIS grade1(DIN1c)and DCIS grade2(DIN2),but only seen in 2 of 18 cases of DCIS grade3 (DIN3)(11.1%), and 18 out of 120 cases of IDC (15%).Fascin protein expression was correlated with ER neg-ative( P<0.05) , PR negative( P<0.05) , tumor grade( P<0.05) and axillary lymph node status( P<0.05) , but not correlated with age,location,tumor sizeand HER2 expression.Conclusions_Fascin protein expression seems to be a late event, usually present in carcinoma.Targetting the fascin pathway may be a noval therapeutic strategy of mammary carcinoma.
