1.Progress of single-cell sequencing in multiple myeloma

Multiple myeloma (MM) is a malignant hematological disorder caused by the abnormal proliferation of monoclonal plasma cells. Traditional high-throughput cell sequencing reflects the heterogeneity of cell population, while it ignores the genetic information in single-cell. The development of single-cell sequencing has enabled the access to cell-to-cell variation in tumor and microenvironment of bone marrow, providing a deep understanding of the pathogenic mechanisms of MM and guidance for clinical treatment. This paper reviews the latest progress of single-cell sequencing and its application in disease pathogenesis, monitoring of disease progression, immunotherapy and therapeutic responses of MM.

3.Analysis of Statistical Data Quality Under Hospital Information System

Objective To analyze current situation on the quality of statistical data under hospital information system in order to enhance data supervision and improve data quality.Methods The quality of statistical data in the first half year of 2008 was described,and the quality of the dates that dates of clinic service,dates of patients discharged from hospital and surgical dates were analyzed.Results The measures about how to ensure the authenticity and effectiveness of the data were introduced.Conclusion The high practical value is provided by utilizing "No.1 Military Medical Project" Information System of statistical data for manager to provide decision support.
4.Realization of Data Communion for SINNOWA-B300 Automatic Biochemistry Analyzer

Objective To share the data of Fully Automatic Biochemistry Analyzer SINNOWA -B300 with Hospital Information System (HIS). Methods The main characters and functions of the Data Pipeline of PowerBuilder were analyzed,and designed application program to connect Access Database of analyzer and Oracle Database of HIS. Results The analyzer can input automatically into Oracle Database of HIS. Conclusion The use of PowerBuilder data pipeline object combined with programmer achieves real-time data sharing,the simple and reliable technology can provide a technical reference for the development of LIS of hospitals.
5.The Effect of Chemotherapy Combined with Tumor Necrosis Factor and Thymosin on Advanced Liver Cancer

The effect of tumor necrosis factor (TNF) in combination with thymosin and chemotherapy on advanced primary liver cancer was observed. Eighty-nine advanced primary liver cancer cases were randomized into two groups: Patients in the control group were treated only with chemotheraputic drugs, and patients in the observing group were treated with TNF, thymosin and chemotheraputic drugs. The results showed that there were higher primary tumor regression rates and statistically reduced AFP in the observing group than control group. The rate of half year survival, one year survival and two years survival improved 28.6%, 18.0% and 5.1% than those of control group, respectively. We belived that this combined therapy could effectivelly improve the patients immunity, the quality of survival, and long the patients survival period.
6.Sixteen cases of H?rthle cell tumors of the thyroid:a clinical study

Objective Investigate the clinicopathological characteristics of h?rthle cell tumors of the thyroid. Methods Sixteen cases of HCT were studied by the clinicopathological study and immunohistochemical and review of the literature. Results There were 13 cases of h?rthle cell adenomas, 5 h?rthle cell follicular carcinomas. Immunostains showed that almost all benign and malignant HCT expressed TG, vimentin, S-100 protein. Conclusions HCT is a potential malignant tumor with a defined pathologic feature. It should be follow up for a longer time to their biological behaviors. To distinguish between the benign and the malignant depends on traditional encapsulation, and whether there is a blood vessel invasion or metastasis.
7.LPS induced macrophage priming effect on O_2~ production and its signal mechanisms

AIM: To investigate the effect of lipopolysaccharide (LPS) priming on macrophage(M?). METHODS: Macrophage cell line RAW264.7 were pretreated with or without LPS for 1 h, then challenged with PMA, or LPS, muramyl dipeptide(MDP), Zymosan, formyl-methionyl-leucyl-phenylalanine(FMLP) for 1 h . O 2 production in supernatants and intracellular free calcium([Ca 2+ ]i ) were measured, and changes in [Ca 2+ ]i and LPS induced O 2 production were compared. RESULTS: LPS pretreatment significantly increased O 2 production in RAW264.7 cells challenged with the stimuli, and in a certain extent, both O 2 production and increase of resting intracellular [Ca 2+ ]i were dose- and time-dependent on LPS pretreatment.Furthermore,the peak [Ca 2+ ]i was significantly higher in LPS pretreated groups than that of LPS unpretreated groups when challenged with PMA. Pretreatment with Ca 2+ inophore A23187 mimicked the LPS priming effects on O 2 production, but pretreatment with Ca 2+ chelator BAPTA and EGTA blocked this priming effect. CONCLUSION: LPS induced M? priming effect on O 2 production is dependent on elevation of resting intracellular [Ca 2+ ]i .
8.Protective effect of rosiglitazone against focal cerebral ischemia-reperfusion injury in rats

Objective To study the protective effect of different doses of rosiglitazone (RSG)against focal cerebral ischemia-reperfusion injury in rats.Methods Forty male sprague Dawley rats were randomly divided into the following groups:sham-operation,control,lowdose RSG(1 mg/kg·d),moderate-dose(2 mg/kg·d),high-dose(4 mg/kg·d)goups(n=8 in each grow).A rat model of middle cerebral artery occlusion for 2 hours and reperfusion for 22 hours were induced by the intraluminal suture method.An automatie biochemical analyzer was used to detect the blood glucose and lipid levels.A solid phase sandwich enzyme-linked immunosorbent assay was used to determine serum insulin levels,and the insulin sensitivity indexes were calculated.The effect of rosiglitazone on infarct volume and behavioristics was observed.Results Compared with the control group,the infarct volume was significantly reduced,and the neurological function scores were improved significantly in each RSG group(P<0.05 or P<0.01).The insulin sensitivity was significantly increased(P<0.05 or P<0.01),and hyperglycemia was reduced significantly after cerebral ischemia(P<0.05)and showed a dose-dependent manner.Conclusions RSG has obvious neuroprotective effect against cerebral ischemia/repeffusion injury in rats.Its mechanism may be associated with the increase of insulin sensitivity.
9.Hearing restoration in a deaf animal model with transplantation of adipose-derived mesenchymal stem cells from guinea pigs

BACKGROUND:Can adipose-derived mesenchymal stem cells be used to treat sensorineural deafness caused by hair celldegeneration and loss? OBJECTIVE:To investigate the effect of adipose-derived mesenchymal stem cells from guinea pigs transplanted via the scala tympani on the recovery from sensorineural hearing in an animal model. METHODS:Intraperitoneal injection of gentamicin was performed to prepare sensorineural deafness models in guinea pigs. Then, adipose-derived mesenchymal stem cells from guinea pig were transplanted via the scala tympani. After 1 and 3 weeks, auditory brainstem responses were detected to observe the hearing variation after adipose-derived mesenchymal stem celltransplantation. Meanwhile, the migration and distribution of EDU-labeled adipose-derived mesenchymal stem cells in the cochlea were traced. RESULTS AND CONCLUSION:After 1 and 3 weeks of implantation, the results from auditory brainstem response showed that the hearing was gradual y improved. At 1 week of implantation, most of the cells were distributed in the perilymph and some cells migrated to the Corti organ and attached to the basement membrane. At 3 weeks of implantation, cells migrated and attached to the basement membrane of Corti organ, furthermore, some cells migrated to the cochlear nerve. The longer the time of implantation, the less cellsurvived. The results indicate that adipose-derived mesenchymal stem cells from guinea pig that are transplanted via the scala tympani can directly migrate and survive ultimately to improve the hearing.
10.Comparative expression of MDR-1 and MRP in peripheral blood lymphocytes in lung cancer patients with chemotherapy treatment

Objective To determine the significance of MDR -1 and MRP mRNA expression in periph-eral blood lymphocytes in lung cancer patients with chemotherapy treatment .Methods Peripheral blood samples were taken from each lung cancer patient before chemotherapy and three weeks after the first chemotherapy cycle . The expressions of MDR-1 and MRP were tested for 39 lung cancer cases using RT -PCR technique comparing to 20 normal control cases .Results There was no significant difference for MDR -1 mRNA and MRP mRNA ex-isting in lung cancer cases and control group (P>0.05).MDR-1 and MRP mRNA expressions were increased after treatment of chemotherapy courses in almost all pathological types of lung cancer .Furthermore,the expres-sion level in small cell lung cancer after chemotherapy was significantly higher than that before .Conclusion Chemotherapy may induce the incidence rate of MDR -1mRNA and MRP mRNA expression in lung cancer .Ade-nocarcinoma and squamous cell carcinoma are mainly intrinsic MDR while small cell lung cancer is mainly ac -quired MDR to chemotherapy .Based on the positive expression rate of MDR -1 mRNA and MRP mRNA in pe-ripheral blood ,we may evaluate the results of chemotherapy in lung cancer patients easily and conveniently .
