1.The role of mesenchymal stem cells in cancer drug resistance
Cancer Research and Clinic 2013;25(12):850-852
Mesenchymal stem cells (MSC) possess an innate ability for self-renewal and can differentiate into multiple cell types.MSC have been used in cell-based therapy in various disease conditions including tumors. However,as MSC therapy gained popularity among practitioners and rescarchers, there have been reports on thre adverse effects of MSC especially in the context of tumor modulation and malignant transformation.Recent studies have shown that MSC also play an important role in the resistance to various anti-cancer drugs. This review provides an overview of the functional properties of MSC in cancer drug resistance.

Objective To explore the expression and clinical significance of secretagogne (SCGN) in neuroendocrinal tumors. Methods Immunohistochemistry was applied for detecting expression of SCGN,chromogranin A (CgA),synaptophysin (Syn) and neuronspecific enolase (NSE) in 45 cases of neuroendocrinal tumors. The difference between the expression of SCGN and the remaining indicators were compared. And different expression profiles of SCGN between groups categorized by sex,age and degree of differentiation were also compared. Results The positive expression rate of SCGN,CgA,Syn and NSE in neuroendocrinal tumors was 95.56% (43/45),100.00% (45/45),46.67% (21/45) and 88.89% (40/45) respectively, and the differences among them were statistically significant(P < 0.01).Expression of SCGN was not correlated with the patient's sex,age and degree of differentiation (P> 0.05).Conclusions Most cases of neuroendocrinal tumors are positive for SCGN and SCGN is more sensitive than CgA.Syn is the most sensitive one for diagnosis of neuroendocrinal tumors.SCGN can be seen as a novel neuroendocrinal marker in the clinical diagnosis of neuroendocrinal tumors.

These six aspects are discussed, including perfect management institutions, the development of management measures, strict quality control, the different management and update optimization, and the establishment of the reward and punishment systems. It aims to strengthen management, improve the management of field medical equipment and maintain in good condition. Update optimization in practice and quality improvement of field medical equipment are required for suitable rescue, treatment needs in peacetime and wartime.

Objective To construct the bicistronic retroviral vectors containing GFP gene and human survivin gene(wildtype and T34A mutant). Methods Survivin fragments were amplified from eukaryotic expression vectors by PCR, then cloned into pMIG retroviral vectors. The constructed recombinant was identified by double digestion with restriction enzymes. The constructed retroviral vectors was transfected into Phoenix E package cells under mediation of liposome. The expression of mRNA was examined by RT-PCR and the transcription of gene was determined by FACS analysis of GFP. Results The target fragments were successfully bound to pMIG vector. After the vectors were transfected into NIH3T3 cells, the expression of mRNA was confirmed by RT-PCR and the expression of GFP was confirmed by FACS analysis. Conclusion Human survivin gene wildtype and T34A mutant retroviral vectors have been constructed successfully, which can provide strong molecular tools for further studies and a novel approach for selective cancer gene therapy.

Allyamine or sodium vinyl sulfate was added into the solutions of acrylamide and N,N′-methylenebisacrylamide to make mixed monomer solutions. These solutions were respectively introduced in methacrylsiloxane-modifidied capillaries. In situ polymerization was initiated by thermally induced radicals. A polymer layer was chemically bonded to the inner wall, resulting in valently modified capillary columns. By altering the addition amounts of allyamine or sodium vinyl sulfate, differently charged polymeric layers were obtained. Five columns were prepared. The result indicated that columns modified with polymers containing amino groups were advantageous over those containing sulphonic acid groups or none. Further comparison revealed that the columns modified with a solution containing 0.15 mol/L acrylamide, 0.03 mol/L N,N′-methylenebisacrylamide and more than 1 mol/L allylamine exhibited good resolution. It is convenient to adjust electroosmotic flows(EOFs) in the columns by this way. In the meantime unspecific protein adsorption was efficiently depressed. When subjected) to zone electrophoresis of proteins from chicken egg white, the column showed high resolution and good reproducibility.

Objeetive To explore the situation of rotavirus infection and extraintestinal organe damage in children in Yueqing city. Methods Two hundred and eighty-seven cases with acute rotavirus gastroenteritis in our hospital were analyzed for prospective study from October 2011 to January 2013 by stool tests. Results Rotavirus infection was found to be more common in autumn and winter. There were 223 cases (17.7%) got extraintestinal organe damage,175 cases (60.80%) got myocardial lesion, and 78 cases (27.18%) got respiratory infection. At the same time, there were 51 cases (17.77%) and 21 cases (7.31%) got liver function lesion and convulsion respectively.Among the metabolic acidosis(48 cases),39 cases were combined with myocardial lesion. While non metabolic acidosis were 239 cases (81.25%),and 136 cases were combined with myocardial lesion. Therefore, the myocardial lesion was significant correlation with metabolic acidosis (P < 0.01).Iron deficiency anemia was 123 cases and combined with 15 cases (12.19%) convulsion, while the convulsion prevalence rates of non iron deficiency anemia was 6 cases (3.65 %).There was a statistically significant difference between the convulsion prevalence rates of iron deficiency anemia and that of non iron deficiency anemia (P < 0.01).Conclusion Rotavirus diarrhea can lead to extraintestinal organe damage, and the clinical doctors should pay attention to them.

To study whether the late-acting co-stimulatory molecules ICOS can suppress the apoptosis and sustain the survival and proliferation of T cells through the survivin pathway, ICOS signals deficient T-cells were infected with adenovirus carried survivin gene, other T-cells were given ICOS co-stimulatory signals, then infected with adenovirus carried dominant-negative mutant survivin gene. Apoptosis and proliferation were determined by TUNEL and CCK-8 respectively. The results show that engagement of ICOS signal increased the expression level of survivin significantly. Survivin can sustain co-stimulatory deficient T cells survival and suppress the apoptosis. Mutant survivin inhibits ICOS signal positive T cells survival and increase its apoptosis. Late-acting co-stimulatory molecules ICOS can suppress the apoptosis and sustain the survival of T cells through the survivin pathway.
