2.Clinical analysis of Stenotrophomonas maltophilia septicemia in 15 patients with liver transplantation

Objective To investigate the related clinical factors and homology of strains in Stenotrophomonas maltophilia (S. Maltophilia) infections in 15 patients with liver transplantation. Methods Fifteen patients with S. Maltophilia infection from September to December 2006 were enrolled and their clinical data were collected. Minimal inhibitory concentrations (MICs) of 10 antimierobial agents against S. Maltophilia were determined by Etest strips. Antibiogram was carried out by resistance analysis assembly with WHONET 5 software. The genomic DNA of all the isolates was digested with Xbal and subjected to pulse-field gel electrophoresis (PFGE). Results All patients received mechanical ventilation during the treatment and had a history of long-term use of extended-spectrum β-lactamases and quinolones. MICs of 10 antimicrobial agents indicated that S. Maltophilia were susceptible to several antimicrobial agents including compound sulfamethoxazole and ciprofloxacin, but the best active agent against these resistant isolates was minocycline in vitro. The results of all 15 S. Maltophilia antibiograms were accordance with PFGE patterns. All 15 S. Maltophilia isolates were classified as 2 PFGE patterns: 9 for pattern A and 6 for pattern B. Conclusion Mechanical ventilation might be associated with the S. Maltophilia septicemia in patients with liver transplantation.
3.Docetaxel induced human uterine cervical carcinoma cell apoptosis and it's enhancement effect on radiosensitization

Objective To investigate therapeutic mechanism of radiation therapy associated with Doeetaxel on human uterine cervi-cal carcinoma. Method Hela cells were incubated with 0. 5 μmol/L, 1.01 μmol/Docetaxel for 24 hours, and were radiated with 0, 2, 4, 6, 8Gy X ray. The proliferation activity was detected with MTT method. Apoptosis was evaluated with TUNEL method. Expression of caspase-3 protein and the degradation fragment was measured with Western blot. Result Inhibitory effect of radiation therapy associated Docetaxel on Hela cells was significantly stronger than that of radiation group. Treated with 2 Gy and 6MV X ray radiation, inhibitory rate of Hela cells increased with the increment of associated Docetaxel doses (P<0.05). Inhibitory rate of Hela cells in radiation associated Docetaxel group was significantly increased compared with radiation group. Apoptosis index in radiation associated Docetaxel group 48 hours after treatment was significantly increased compared with radiation group or Docetaxel group with TUNEL method (P<0.05). Western blot results indies-ted that caspase-3 protein was degraded and small active fragments were produced. Conclusion The reason that Docetaxel enhanced Hela cell radiation sensitivity may probably he correlated to easpase-3 protein activation.
4.Best Operation Mode of AP-960 Full-automated ELISA System

Objective To compare the operation modes of AP-960 full automated ELISA system.Methods Two models were tested: beginning with adding specimens(model 1) and beginning with adding enzymes(model 2).The following factors were compared: the consumption of suckers,usage of time,result of test and the usual alarm during the test.Results The consumptions of suckers were 564 and 84 respectively.The needing time of the former mode was 125 minutes and that of the latter mode was 90 minutes.Conclusion The mode beginning with adding enzymes(model 2) is better for the current status of our clinical laboratory.
5.Relationship between the activated status of EGFR signal pathway and the sensitivity of colon carcinoma cells to gefitinib

Objective To evaluate the inhibitory effects of gefitinib on the growth of colon carcinoma cells with different degree of sensitivity,and the activated status of epidermal growth factor receptor(EGFR) associated signal pathway proteins.Methods The colon carcinoma cell lines(Lovo,HCT116 and HT29) were treated with 10?mol/L of gefitinib,and flow cytometry was employed to detect the cell cycle and apoptosis.Another portion of cells were treated with 50ng/ml of epidermal growth factor(EGF) or 5?mol/L or 10 ?mol/L of EGF+gefitinib,and Western blotting was used to determine the expressions of PTEN,EGFR,AKT and MAPK,as well as their corresponding phosphorylated proteins,p-EGFR,p-AKT and p-MAPK.Results After being treated with gefitinib,the G1 phase cells and apoptosis rate increased remarkably in Lovo,slightly in HT29,while no significant change was found in HCT116.With the treatment of EGF alone,the expressions of p-EGFR,p-AKT and p-MAPK increased significantly in Lovo and HT29 cells(P0.05).Conclusions The growth of colon carcinoma cells,which are resistant to gefitinib,is not dependent on EGFR,and the activated status of signal pathway of the gefitinib-resistant cells will not be inhibited as the EGFR is blocked.The present findings suggest that sensitivity of colon carcinoma cells to gefitinib relies on EGFR as well as the activation of its downstream signal pathway.
6.Relationship between the sensitivity of human colon carcinoma cells to gefitinib and the expressions of Akt and MAPK

Objective To investigate the relationship between the sensitivity of colon carcinoma cells to epidermal growth factor receptor(EGFR) inhibitor,gefitinib,and the downstream proteins of EGFR.Methods The expressions of EGFR proteins of 6 colon carcinoma cell lines(Lovo,HCT116,HT29,LS174T,SW480 and SW620) were determined with immunocytochemistry staining.The inhibitory effects of gefitinib on the growth of colon carcinoma cells were assessed by MTT,and the expression levels of Akt and MAPK as well as their phosphorylated forms(p-Akt and p-MAPK) were assessed by Western blotting.Results EGFR protein expressed in all the Lovo,HCT116,HT29,LS174T and SW480 cells,and the highest expression was found in Lovo cells,but not in SW620 cells.Lovo cells showed the highest,HT29 and SW480 cells showed moderate,sensitivity to gefitinib,while the others showed more or less resistance to gefitinib.No significant difference was found between the growth inhibition and IC50 values among the 6 cell lines despite of being treated with fetal bovine serum or EGF.Akt protein existed in all the cell lines without notable difference.Lovo and SW620 cells showed the least of p-Akt expression(P

Objective: Docetaxel as a newly semi-synthesized anti-tumor drug,exhibits significant cyrotoxic activity and enhanced sensitivity for radiotherapy.The aim of this study was to investigate the correlation of the apoptosis of human Hela cell line with the expressions of TRAIL and its receptors induced by the combination therapy of Docetaxel with radiation.Methods: We treated the Hela line by Docetaxel,radiation,and combination therapy of Docetaxel with radiation,respectively,detected the apoptosis of the Hela cells using Hoechst 33342 staining and the TUNEL method,and observed the changes in the mRNA expressions of TRAIL and its receptors TRAIL-R in the three groups by RT-PCR.Results: Compared with the Docetaxel and the radiation groups,the apoptosis of the Hela cells was increased significantly in the combination therapy group,and so were the mRNA expressions of TRAIL and its receptors TRIAL-R1 and TRAIL-R2.Conclusion: The combination therapy of Docetaxel with radiation increased the apoptosis of the Hela cell line,which might be mediated by the up-regulation of mRNA expressions of TRAIL and its receptors TRAIL-R1 and TRAIL-R2.
9.The diagnostic value of heart fat acid binding protein in myocardial damage of children with hand-foot-mouth disease

Objective To study the diagnostic value of heart fat acid binding protein (h‐FABP) in myocardial damage of children with hand‐foot‐mouth disease (HFMD) .Methods From February 2012 to December 2014 ,100 children with HFMD were chosen as study objects .All children study were divided into 2 sub‐groups according to the severity of disease:71 in ordinary HFMD sub‐group ,29 in severe HFMD sub‐group .At the same time ,100 healthy children were chosen as control group .The routine blood test , rate of abnormal electrocardiography ,rate of abnormal cTnI and rate of abnormal h‐FABP were compared among all children .The cTnI and h‐FABP at different time were compared between ordinary HFMD sub‐group and severe HFMD sub‐group .Results The WBC ,RBC and L had significant difference among different groups/sub‐groups(P<0 .05) ,the difference of PLT had no statistical significance(P>0 .05) .In ordinary HFMD sub‐group ,rate of abnormal electrocardiography was 19 .72% (14/71) ,rate of abnormal cTnI was 4 .23% (3/71)and rate of abnormal h‐FABP was16 .39% (10/71);in severe HFMD group ,rate of abnormal electrocardio‐graphy was 72 .41% (21/29) ,rate of abnormal cTnI was 82 .76% (23/29) and rate of abnormal h‐FABP was 82 .96% (23/29);in control group ,rate of abnormal electrocardiography was 1 .00% (1/100) ,rate of abnormal cTnI was 2 .00% (2/100)and rate of ab‐normal h‐FABP was 0 .00% (0/100) ,the difference had statistical significance(P<0 .05) .The cTnI and h‐FABP at different time had significant difference between ordinary HFMD sub‐group and severe HFMD sub‐group(P<0 .05) .Conclusion Heart fat acid binding protein (h‐FABP) can reflect the early myocardial damage in children with hand‐foot‐mouth disease .

Objective To investigate the feature of Th1 cytokines induced by Mtb-specific antigen in patients with refractory pulmonary tuberculosis.Methods 28 patients with refractory pulmonary tuberculosis,67 patients with first-treated pulmonary tuberculosis,and 25 healthy controls with positive T.spot (LTBI group) were enrolled.IFN-γ,IL-2 and TNF-α in supernatants from PBMCs stimulated with ESAT-6 and CFP-10 were analyzed with Bender Flowcytomix on flow cytometry.Results The levels of the three cytokines were utmost high in patients with first-treated pulmonary tuberculosis.The lowest level of IFN-γ and IL-2 were induced in patients with refractory pulmonary tuberculosis,and were significantly lower than the LTBI group(Mann-Whitney U = 105.5,162.5,P < 0.01).Conclusions The immunotherapy with IFN-γ and IL-2 may play a role in treatment for refractory pulmonary tuberculosis but not for most of first-treated pulmonary tuberculosis.
