Renal Tubular Cell Apoptosis refers to activation,proliferation,distension,cast formation and cellular apoptosis of the tubular epithelium when the renal tubule undergoes hypoxia,poisoning,inflammatory reaction,plasma protein or amylaceum.This text summarized the researches on the prevention and control of lenal tubular cell apoptosts in renal interstitial fibrosis in the recent years.

Objective　To investigate the expression of endometrial leukaemia inhibitory factor（LIF） gene in patients with unexplained infertility. Methods　 By a quantitative reverse transcription-polymerase chain reaction (RT-PCR)，the expression of LIF gene on endometrium during mid-luteal phase was detected in 35 unexplained infertility (infertility group) cases and 20 infertile cases due to tubal obstruction or male factor (control group). Results　The level of LIF mRNA expression on endometrium during mid-luteal phase in infertility group was 0.448±0.239，significantly lower than those in the control group （1.093±0.761，P<0.01). Conclusions　Our findings suggested LIF might play an important role in the process of implantation. The decreased expression of LIF gene might be one of the major causes of unexplained infertility.

Objective To investigate the possible roles of Langerhans cells(LCs) and plasmacytoid dendritic cells(pDCs)in the pathogenesis of condyloma acuminatum(CA). Methods Tissue specimens were obtained from the lesions of 23 patients with CA and from perilesional normal skin of 13 of the 23 patients. The expressions and distribution of CD1a, CD2AP and CD123 were analyzed using the streptavidin-peroxidase (SP) immunohistochemical method. Statistical analysis was carried out to compare the density and percentage of CD1a + LCs, CD2AP + pDCs and CD123 +pDCs between lesional skin and perilesional normal skin. Results Most CD1a + LCs were observed in the prickle cell layer of the epidermis, and a few in the papillary layer of the dermis. There was no significant difference in the density or percentage of CD1a+ LCs in the epidermis or dermis between the lesional and perilesional skin specimens (all P > 0.05). CD2AP + pDCs and CD123 + pDCs were distributed mainly in the papillary layer of the dermis, and their density and percentage were significantly higher in the lesional than in the perilesional skin specimens (all P < 0.05). Conclusion Human papillomavirus infection may affect the quantity of pDCs but not LCs in local mucous membranes.

OBJECTIVE:To provide reference for the sustainable development of pharmaceutical enterprises in Guizhou prov-ince. METHODS:A questionnaire was conducted for 55 pharmaceutical enterprises in Guizhou province,the basic situation of en-terprises,familiarity of related directors to Good Manufacture Practice of Drugs(GMP)and relevant policy,the current situation of implementing the new version GMP were investigated and statistically analyzed,problems were found,and corresponding coun-termeasures were put forward. RESULTS & CONCLUSIONS:Totally 55 questionnaires were sent out,49 valid ones were received with effectively recovery of 85.1%. The results showed 43 enterprises(87.8%)had passed the GMP authentication;only 13 enter-prises(26.5%)directors were very familiar with the new version GMP. In terms of personnel management,the head of production management and quality management and the authorized person of quality and personnel had not yet met related requirements of the new version GMP fully;in terms of equipment and production management,production area transformation(clean areas,lounges, warehouses,water use) and air purification system design in most enterprises met related requirements of the new version GMP, 23 enterprises (46.9%) still can not conduct fully inspection to products and materials;in terms of document management,there were still some enterprises not meeting the new version GMP standards fully,enterprises'documents(health area layout,air purifi-cation layout,management procedures,operating procedures,etc.) of production site were imperfect. According to the investiga-tion,the main existing problems included lack of funds in implementing the new version GMP reform,not enough understanding or familiarity with the new version GMP,relevant personnel management not reaching the designated position,equipment and pro-duction management needing to be strengthened,document management systemic being poor,risk management being not sound, etc. It is suggested that government should give all forms of capital policy and strengthen the training of the new version GMP;en-terprises should attach great importance to the relevant personnel management,strengthen the equipment and production manage-ment,set up perfect document management system and a sound system of risk management.

At present,all parts of our country and the units are on their own to explore the ways and methods of public health physician training,and to develop a unified national public health standardized training system is particularly important.This study introduces the composition and practice of British public health specialist training and its access system,draws on the successful experience of this system and puts forward some suggestions such as paying attention to the cultivation of clinical basic knowledge,rationally using industry associations and societies and so on from the public health doctor training system,which provides the basis for promoting the establishment of a national public health physician training system.

Objective To investigate the gait differences among patients with Alzheimer's disease (AD) and mild cognitive impairment(AD-MCI) and the person with normal cognitive function,and analyze the reasons for the abnormal gait.Methods Eighty patients were included and divided into 3 groups according to the cognitive status:noncognitive impairment (NCI group,n =30),patients with mild cognitive impairment (AD-MCI group,n =29),Alzheimer's disease (AD group,n =21).The mini-mental state examination (MMSE),the Montreal Cognitive Assessment (MoCA),Addenbrooke's Cognitive Examination (ACE-R),the connection test (TMT-A),Digit Span Test(DST),activities of daily living questionnaire(ADL) and associated gait and balance scale (5 times sit to stand test (FTSTT),3 m walking test (TUG),Berg balance scale) were used to evaluate all research objects in each group.Results There were significant differences in NCI group compared with AD-MCI group and AD group in terms of FTSTT,TUG,Berg balance Scale and leg speed((9.59±2.39) s vs.(13.71±4.65) s vs.(14.15±4.20) s,(7.70±1.58) s vs.(11.13±3.02) s vs.(11.35±4.43) s,(54.20±1.56) points vs.(48.17±4.93) points vs.(48.10±5.46) points,(82.12±22.79) cm/s vs.(57.49± 14.89) cm/s vs.(57.70±14.68) cm/s;P＜0.05).There were significant differences in NCI group compared with AD-MCI group and AD group in terms of MMSE,MoCA,ACE-R,TMT-A,DST((28.67±1.27) points vs.(26.76±2.59) points vs.(21.86±5.29) points,(26.03±2.58) points vs.(22.39±5.05) points vs.(16.90±5.05) points,(85.80±5.90) points vs.(78.03±7.58) points vs.(60.95± 13.99) points,(12.66±5.36) s vs.(18.99 ± 11.46) s vs.(30.49±26.98)s,(18.60±4.64) points vs.(15.94±4.76) points vs.(12.86±5.23) points;P ＜0.05).Conclusion Gait disorder might be an early sign for cognitive impairment.

Aim To investigate the effects of cell pro-liferation and activation in HSC-T6 cells by inhibiting the expression of EZH2 , and its partial relevant mech-anism. Methods By introducing the inhibitor DZNep in activated HSC-T6 cells stimulated by TGF-β1 , the protein expression levels of EZH2, p-ERK, p-AKT andα-SMA were detected by Western blot. The siRNA targeting EZH2 was designed and synthesized according to its nucleotide sequence, and their corresponding ex-pression vectors were constructed and transfected into HSC-T6 cells with LipofectamineTM 2000. The prolifer-ation of HSC-T6 cells was determined by MTT. And the protein expression levels of EZH2, p-ERK, p-AKT and α-SMA were measured by Western blot. Results By introducing the inhibitor DZNep in activated HSC-T6 cells stimulated by TGF-β1 , it effectively de-creased the protein levels of EZH2 and also the protein levels of p-ERK, p-AKT and α-SMA. By introducing EZH2-siRNA in activated HSC-T6 cells, it effectively inhibited the cell proliferation, and also the protein levels of EZH2, p-ERK, p-AKT andα-SMA. Conclu-sion Silencing EZH2 expression inhibits HSC-T6 cell proliferation and activation, and EZH2 may be a poten-tial therapeutic target gene for hepatic fibrosis.

BACKGROUND: Multiple sclerosis(MS) is a chronic autoimmune disease induced by the interaction between genetic and environmental factors. Its pathogen and the mechanism of the relapse and remission m the course of the disease are still unknown. Most of the MS research centers are looking for the pathogenic polypeptide epitope in proteolipid protein(PLP), myelin sheath basic protein (MBP) and oligodendrocyte glycoprotein (MOG) OBJECTIVE: To compare the proliferation of T cell lines(TCL) in MS induced by myelin sheath and delipidated myelin sheath towards 11 components of myelin sheath to mainly search the possible pathogenic polypeptide epitope in PLP, and investigate the possible effects of abnormal dcgrease in myelin sheath.DESIGN: A case-controlled trial.SETTING: Department of neurology in a hospital of a university.PARTICIPANTS: Mononuclear cells(MNC) of 16 MS cases(clinical relapsing-remitting type, patients did not receive any immunosuppresant for at least 3 months when their peripheral blood samples were taken) and 12 HLA-DR15 healthy volunteers were furnished by Dr. Trotter JL of MS Research Center of Washington University from the cell database.INTERVENTIONS: MS-TCL and normal TCL were induced twice by stimulation with myelin sheath and delipidated myelin sheath in vitro by cell culture in vitro. TCL proliferation was tested by 11 antigens including PLP,MBP, M87-106, P30-49, P40-60, P89-106, P95-117, P117-137,P139-151, P178-191, and P185-206.MAIN OUTCOME MEASURES: Difference of scintillation counting in every minute of every well, and the stimulative index of each well were calculated, and the mean wells with positive proliferation of TCL towards each antigen were confirmed as well.RESULTS: The general specific proliferation towards myelin sheath antigens was bigger in MS group than control group 5.49 ±5.31 to 3.10 ± 3. 17, and delipidated myelin sheath-induced TCL was bigger than myelin sheath-induced one 5. 49 ± 5.31 to 3.41 ± 4. 83 . Delipidated myelin sheath significantly changed the immune responses of MS group,especially the changes of responses towards P30-49, P40-60, P89-106,P117-137, P139-161, and P185-206 were significant compared with that the control group only responded to two polypeptides, which indicated that the antigen epitope of MBP, PLP, M87-106, P95-117, P40-60, and P185-206 might have significance in the triggering of MS autoimmune responses.CONCLUSION: TCL induced by MS myelin sheath has different proliferation towards antigen components of myelin sheath from control group. Delipidated myelin sheath significantly increases TCL proliferation in MS group, which suggests that if MS patients developed abnormal degrease in myelin sheath, TCL would produce autoimmune response towards self-myelin sheath, MBP, PLP and its polypeptide segments all can trigger MS or aggravate the state of the illness. Our finding supports the hypothesis of MS autoimmune pathogenic mechanism.

Objective To investigate the clinical significance of anti-β2 glycoprotein Ⅰ(anti-β2GP Ⅰ)antibodies in patients with systemic lupus erythematosus(SLE)and to assess their association with lupus thrombocytopenia.Methods Anti-β2GP Ⅰ antibodies were tested in 108 SLE patients(including 37thrombocytopenia patients),30 patients with rheumatoid arthritis(RA)and 30 healthy individuals by enzyme-linked immunosorbent assay(ELISA).The clinical features and laboratory findings were collected,and the associations of anti-β2GP Ⅰ antibody with laboratory findings and disease activity were analyzed.Results Sensitivities of anti-β2GP Ⅰ antibody were 19.44%(21/108)in SLE and 10%(3/30)in RA,respectively.The specificity of anti-β2GP Ⅰ antibody was 95.0% in SLE.The anti-β2GP Ⅰ antibody titers of the SLE group were significantly higher than normal group(P ＜ 0.05).There was no significant correlation between anti-β2GP Ⅰ antibodies and thrombocytopenia(r =-0.028,P ＞0.05).Anti-β2GP Ⅰ antibody was positively correlated to anticardiolipin antibody(r = 0.566,P ＜ 0.01).No associations were found between anti-β2GP Ⅰ antibody and other clinical and laboratory features.There was no statistical correlation between the level of anti-β2GP Ⅰ antibody and SLEDAI scores.Conclusions Anti-β2GP Ⅰ antibody had high specificity in SLE.There was no significant correlation between anti-β2GP Ⅰ antibodies and lupus thrombocytopenia.

Objective To study the role of mesenchymal stem cells (MSC) in an animal model combining ischemia-reperfusion with 85％ liver resection.Methods Eight-week-old male SD rats received BM-MSC by tail vein and then underwent 30-min ischemia followed by 85％ liver resection.The survival rate was monitored for 7 days after surgery.Liver regeneration was assessed on day 2 after hepatectomy.Liver damage,liver cell apoptosis,and cytokine expression in the first 24 h after hepatectomy were also assessed.Results BM-MSC mostly homed to the spleen.Transplantation significantly inhibited myeloperoxidase [(19.9 ± 6.0) mg/g vs.(41.4 ± 10.2) mg/g] and downregulated proinflammatory cytokines.BM-MSC significantly reduced the ALT and AST levels [AST (1 475 ± 275) IU/L vs.(2 550 ± 441) IU/L,P ＜ 0.05;ALT (738 ± 101) IU/L vs.(1 113 ± 268) IU/L,P ＜ 0.05].The attenuation of liver injury was also verified histologically 24 h after surgery.Liver cell apoptosis was markedly reduced.Moreover,BM-MSC infusion significantly promoted remnant liver regeneration.As a result,the survival rate was improved by BM-MSC treatment in this model (95％ vs 70％,P ＜ 0.05).Conclusion In an animal model combining ischemia-reperfusion with 85％ liver resection,BM-MSC infusion attenuated liver injury and promoted hepatocyte regeneration,resulting in improved survival rate.
