Objective:To find out the impacting factors in uncertainty by analyzing the measurement uncertainty in the determina-tion of irbesartan tablets by HPLC and provide the evidence for the measurement evaluation. Methods:A mathematic model for calcu-lating uncertainty was established, and every component in the uncertainty was assessed. Results:The expanded standard uncertainty for the HPLC determination was 2. 4%. Conclusion:The measurement uncertainty of the experiment is mainly caused by the measur-ing method.

Objective To investigate the effect of alternating shift on general well-being of nurses,and analyze its relationships with related factors. Methods Alternating shift nurses from general hospi-tals above courty-level in Liaocheng city who were engaged in clinical work were selected by duster sam-pling.Questionnnires were sent out and later gathered. The investigation tools included general well-being schedule(GWB) ,marital quality inventory, social support inventory, Eysenck personality questionnaire were used in survey. Results (1)There was difference in the score of general well-being between nurses with different ages and professional titles(P<0.01). (2)There was significant difference between GWB and oth-er factors such as marital quality,EPQ (psychoticism, neuroticism and introversion and extroversion), so-cial support(P<0.01). (3)Multivariable linear regression which took general well-being as dependent vari-able, the variables which entered the equation were neuroticism, family relationship,support utilization,psychoticism, objective support, introversion and extroversion, marriage satisfaction,arrangement of econ-omy. Conclusions (1)The older, the higher of professional tltles,the stronger general well-being of nurses. (2)Sabjective well-being and marital quality, social support, lying defense were positively corre-lated; neuroticism, psychoticism were negatively correlated. (3)Neuroticism, family relationship,support and utilization, psychoticism, objective Support, introversion and extroversion, marriage satisfaction,ar-rangement of economy can affect general well-being,among which neuroticism had great influence.

Objective To discuss the effect of night- shift work on marital quality, and analyze the relations between the marital quality and related factors in order to explore the method to tackle the marital crisis of night- shift nurses. Methods 500 nurses were selected in general hospitals of Liaoeheng above county-level, each tested nurse filled out marital quality inventory, general well-being scale,social support scale and Eysenck personality questionnaire(EPQ), and withdrew 416 copies(effective rate 90.83%) of ef-fective questionnaire after filling independently. Results (1)There were significant difference in the var-ious scores of marital quality between the nurses and the norm. (2) The marital quality was correlated with general well-being, social support and psychoticism, neuroticism and lying degree of EPQ.(3) Multivariable linear regression which took marital quality as dependent variable ,the variables which entered the equation were age of marriage, social support, psychoticism,subjective well- being,lying degree and objective support. Conclusions (1) The marital quality of night-shift nurses was low. (2)Marital quality was positively cor-relat-ed with subjective well-being, social support and lying degree, negatively correlated with neuroticism and psychoticism. (3) Marital quality were influenced by time of marriage, social support, psycholepsy, sub-jec-tive well-being, lying degree and objective support, among which social support has a great influence.

Objective To evaluate the value of procalcitonin(PCT) and endotoxin(ET) in early diagnosis and prognosis in critically illness patients with sepsis.Methods A total of 104 cases with suspected sepsis were prospectively collected in this study from February 2015 to October 2016 in ICU.There were 31 cases of systemic inflammatory response syndrome (SIRS group),30 cases of sepsis 24 cases of severe sepsis and 19 cases of septic shock.Prognosis was observed in patients with sepsis discharged,57 cases were survival (survival group) while 16 cases were dead (death group).The level of PCT and ET and the clinical characteristics were recorded,The the ROC curve was applied to evaluate the discriminative power of variables.Results The levels of PCT,APACHE-Ⅱ score,ET of cases with sepsis,severe sepsis and septic shock were significantly higher than those in SIRS group(P<0.05).The PCT and APACHE-Ⅱ scores were elevated by the severities of disease (P<0.05).ROC curve analysis showed that,the sensitivity of PCT value was 82.2% and specificity was 87.5%.PCT and APACHE-Ⅱ scores were higher in the death group (P<0.05),whereas there was no significant difference on ET between the two groups (P>0.05).ROC curve showed that AUC of PCT was 0.867,which was significantly higher than that for APACHE-Ⅱ score (AUC=0.762) (P>0.05).Conclusion Compared with APACHE-Ⅱ score and ET,PCT can help clinicians to early diagnose and treat cases with sepsis.

Objective To establish a double TaqMan real-time fluorescence nested PCR method for rapid detection of α-thalassemia SEA deletion.Methods One hundred blood samples for thalassemia screening were collected from May to July of 2010 in the Tianhe Maternal and Child Health Hospital of Guangzhou.Seven fetal specimens for prenatal diagnosis were collected from December 2010 to February 2011 in Dongguan TungWah Hospital(2 villi and 5 amniotic fluid specimens).Fifty samples of α-thalassemia SEA deletion with genotyping results were selected from the sample bank of our laboratory.The double TaqMan real-time fluorescence nested PCR was applied to detect the truncated sequence of SEA deletion and the normal sequence within deletion range simultaneously for all these samples with the same detecting system.The genotype of α-thalassemia SEA deletion was accurately acquired according to the positive result of fluorescent PCR combined with the Ct value difference.Meanwhile,the accuracy and feasibility of this method were verified and analyzed by parallely detecting these samples with routine gapPCR for α-thalassemia SEA deletion.The genotype could be obtained according to PCR amplification and agarose gel electrophoresis.Results Two amplification efficiencies of the optimized dual TaqMan real-time fluorescence nested PCR system established in this study were both close to 100% with the slops of -3.153 and -3.182,respectively.The results of 50 samples of α-thalassemia SEA deletion with genotyping results showed that this method could not only realize rapid diagnosis,but also effectively avoid false negative or false-positive misdiagnosis by accurately determine the external contamination in the sample.Among 100 blood samples,eleven samples with SEA deletion were detected respectively and the diagnosis coincidence rate of these two methods was 100%,3 samples with SEA deletion were detected by gap-PCR,but 2 samples with SEA deletion and 1 villi sample with normal genotype but contaminated by SEA were detected by this method among 7 fetal samples.Conclusions A double TaqMan real-time fluorescence nested PCR method for α-thalassemia SEA deletion was developed.The method is a rapid and reliable test with simple operation,and is suitable for large-scale population screening and routine molecular diagnosis.

Objective To explore the effect of gene mutations of epidermal growth factor receptor ( EGFR) on targeting therapy of advanced non-small cell lung cancer (NSCLC). Methods The 139 hospitalized patients who had been treated at least once with platinum-based chemotherapy and had tumor progression or recurrence after the last chemotherapy between December 2005 and December 2009, underwent EGFR gene test extracted from the pathological tissues. Based on the results of the test, the patients were divided into three groups: EGFR mutation per os (p.o.)Gefitinib (MPG) group, wild-type EGFR per os (p. o. ) Gefitinib (WpG) group and wild-type EGFR post-docetaxel chemotherapy (WpD) group. Clinical characteristics, pathology, treatment efficacy,survival time, performance status (PS) score, adverse reaction and quality of life of patients in the three groups were assessed. Results The EGFR mutation rate were higher in female, patients with adenocarcinoma and non-smokers than in male, smokers and those without adenocarcinoma. There were significant differences in median progression-free survival and median survival time among the three groups, which were 2.8 and 8. 9 months in MpG group, 2.0 and 7.1 months in WpG group,2.5 and 7. 8 months in WpD group(H=11. 198, 16. 991 ,all P＜0.01). The changes of PS score were significantly different between MpG group and WpG group (96. 8% vs. 62. 0%, x2 = 12. 583 ,P＜0. 01 ). However, there was no difference in changes of PS score between WpG group and WpD group (62. 0% vs. 66. 0%, x2 =0. 878,P＞0. 05). Conclusions The gene mutation of epidermal growth factor receptor may be served as an important indicator of advanced non-small cell cancer therapy.

Objective:To study the possible role of LPS and IL-13 on IL-12 production of mesangial cells in vitro.Methods:The production of IL-12 in mesangial cells was detected with ELISA kit under following different treatments:①Mesangial cells were cultured with LPS(20 ?g/ml)under different time;②The cells were cultured with different concentrations of LPS for 24 h.For observation of the effect of IL-13 on IL-12 production by mesangial cells,three groups were designed:control group,LPS(10 ?g/ml)group and different concentrations of IL-13 with LPS (10 ?g/ml).ELISA was applied to detect IL-12 in supernatants fluid from mesangial cells cultured for 24 h.The expression of IL-12p40 mRNA by mesangial cells cultured for 20 h was evaluated by RT-PCR.Results:Under basal conditions (no LPS) the production of IL-12 was hardly detected.The levels of IL-12 stimulated by LPS was significantly increased in certain doses,but the levels of IL-12 reduced accompanied with the doses and time increasing of LPS stimulantion.IL-13(1-100 ng/ml) inhibited the protein and mRNA expression of IL-12 in a dose-dependent manner(P

Objective:To isolate and culture the pulp cells from human young permanent teeth (pDPC),and to observe their biological characteristics and the expression of some specific markers,and to induce these pulp cells to differentiate into osteoblast, adipocyte, neuron and chondrocyte lineages. Methods:Pulp cells were isolated and cultured from orthodontic extracted premolars of children. The attached cells after at least 3 passages were used for the following experiments:1. Morphology and ultrastructure analysis; 2. Cell cycle and phenotype were analyzed by flowcytometry; 3. Growth curve were recorded;4. pDPC were induced to differentiate into osteoblast,adipocyte,neuron in vitro,and were identified by histochemical methods and RT-PCR. Results: 1. Attached pDPCs were fibrablast-like cells,which were distinguished from BMSC. 2. The cell organs in dDPCs were well developed. 3. pDPCs were highly positive for CD90, CD44, CD147,which are mesenchymal stem-cell markers,but were negative for other markers including CD34, CD38, CD45, HLA-DR. 4. pDPCs showed high growth rate. 5. pDPCs could be induced to differentiate into osteoblast, adipocyte, and neuron lineages,but not chondrocyte lineages. Conclusion:pDPCs were characterized by their ability to proliferate with high growth rate in vitro. The expression of some BMSC markers in these cells were observed. They showed the potential to differentiate into multiple mesenchymal lineages such as osteoblst,adipocyte, neuron lineages under specific conditions in vitro.

OBJECTIVE To explore cell death type of lateral line hair cellsinduced by cisplatin in zebrafish. METHODS Zebrafish larva were incubated in 1mM cisplatin solution for 6 hrs to induce about 90%lateral line hair cells loss. Time lapse imaging was used to detect the morphology of cisplatin-incubated hair cells in wildtypezebrafish pre-labelled by live dyes Bodipy TR C5-ceramide and Sytogreen 24. TUNEL assay and In situ anti-active Caspase-3 antibody staining were performed to detect nuclei fragmentation and Caspase-3 activity respectively. RESULTS Compared to control group, hair cells condensationand nuclei fragmentation (P<0.05) were detected in cisplatin-incubated group, and active Caspase-3 activity was also observed after cisplatin addition. CONCLUSION Cisplatinmay induced zebrafish lateral line hair cells loss by Caspase-3-dependent apoptotic pathway.

Objective To investigate the effects of nephroblastoma overexpressed (NOV) on proliferation,adhesion,migration and invasion of remal cell curcinomai (RCC) cells. Methods We constructed a NOV expression plasmid and transfected the plasmid into RCC cell line 786-O and analyzed the effects of NOV expression on proliferation,adhesion,migration and invasion of RCC cells by growth curve assay,WST-1 assay,cell adhesion assay,matrigel invasion assay and transwell migration assay. Results The stable NOV transfected 786-O cells (786-O-NOV) showed decreased growth rate,at 48 h and 72 h,the proliferation activities of 786-O-NOV cells were inhibited by 29.14％ and 32.46％ the proliferation activities of empty vector cells were inhibited by 9.25％ and - 8.16％,respectively,compared to 786-O cells (P ＜0.05); while the 786-O cells transfected with empty vector (786-O-mock) had no difference with 786-Ocells.Adhesion assay indicated significantly increased adhesion of 786-O-NOV cells to fibronectin (0.26 ±0.03) and laminin (0.28 ±0.04),compared to 786-O cells (0.15 ±0.01,0.12±0.10) and 786-O-mock cells (0.14 ±0.02,0.13 ± 0.08).Invasion assay displayed that the numbers of cells penetrated through matrigel membrane were significantly higher in 786-O-NOV cells (240.25 ± 23.12) compared to 786-O cells ( 56.16 ± 6.25 ) and 786-O-mock cells ( 50.28 ± 7.13 ).Migration assay displayed that the numbers of cells passed through polycarbonate filters were significantly higher in 786-O-NOV cells (267.25 ± 20.94) compared to 786-O cells ( 66.10 ± 5.68 ) and 786-O-mock cells ( 56.28 ± 4.11 ).Conclusion NOV exhibits anti-proliferative effects on RCC cells; however,it promotes adhesion,migration and invasion of RCC cells.
