Aim Toinvestigatetheinducementeffect of isatin on apoptosis of breast cancer cell line MCF-7 , andexploreitsdetailedmechanism. Methods MCF-7 cell lines were exposed to isatin at different concentra-tions(0,50,100,200 μmol·L-1 )for 48 h.Apop-totic features were demonstrated by nuclei staining with Hoechst 33258.Bcl-2,Bax and p53 mRNA were ana-lyzed by RT-PCR.Caspase-9 activation and mitochon-drial depolarization were assayed by flow cytometry. Bcl-2,Bax,p53 and cytochrome c proteins were ana-lyzedbyWesternblot.Results Isatininducesapopto-sis of MCF-7 cells.Furthermore,Bcl-2 expression was decreased and the ratio of Bcl-2 to Bax was significant-ly decreased by isatin.While,p53 expression relative-ly decreased. The mitochondrial transmembrane poten-tial was markedly reduced and the release of cyto-chrome c into the cytosol was increased after treatment with isatin.Simultaneously,caspase-9 was activated. Conclusions Isatinsignificantlyinducedtheapopto-sis of MCF-7 cells in vitro. These results strongly sug-gest that the p53 dependent mitochondrial pathway is involved in apoptosis.

Objective:To observe the clinical effect of wind-expelling method of acupuncture and Chinese herbs for sudden deafness.
Methods:A total of 90 cases with sudden deafness were randomly divided into three groups based upon their visit order and random digital table, an acupuncture plus Chinese herbs (APCH) group, an acupuncture group, and a Western medication (WM) group, 30 cases in each group. The APCH group was treated by puncturing Fengchi (GB 20), Fengfu (GV 16) and Fengmen (BL 12), without retaining the needles, and then by puncturing Zhongwan (CV 12), Guanyuan (CV 4), Qihai (CV 6) and Tianshu (ST 25) with reinforcing technique after arrival of the needling sensation, and the needles were retained for 30 min, plus oral administration of wind-expelling Chinese herbs. The acupuncture group was treated with the acupoints selected by syndrome differentiation in reference to the textbook of Acupuncture and Moxibustion Science. The WM group was treated by intravenous infusion of Alprostadil injection, Adenosine Disodium Triphosphate and Coenzyme A and muscular injection of Vitamin B12 and Vitamin B1. The decibel values of hearing loss before and after treatment were respectively recorded and analyzed comparatively.
Results:In the intra-group comparison after treatment among the three groups, the decibel values of hearing loss all declined, with statistically significant differences (allP<0.05). After treatment, the decibel value of hearing loss in the APCH group was better than those in the acupuncture group (P<0.05) and the WM group (P<0.05). The total effective rate was respectively 83.3% in the APCH group, 60.0% in the acupuncture group and 56.7% in the WM group. The total effective rate in the APCH group was better than those in other two groups (bothP<0.05).
Conclusion:The therapeutic effect was better for sudden deafness by wind-expelling method of acupuncture and Chinese herbs than by acupuncture alone or by Western medication.

BACKGROUND: Heart valve tissue engineering is aimed to construct heart valve grafts with the physiological function and biological activity by using engineering and life science principles and methods, but still in the animal experiment stage.OBJECTIVE: To summarize the commonly used tissue engineered heart valve, and to evaluate the reliability of different types of heart valve biomaterials.METHODS: Using "biological materials, heart valve, scaffolds, reviews, tissue engineering" in Chinese as the key words, a computer retrieval was performed for articles published from January 2000 to December 2010. Articles regarding the biomaterials in tissue engineered heart valve were included; the duplicated research or meta-analysis were excluded.RESULTS AND CONCLUSION: A total of 20 papers about the biomaterials and tissue engineering heart valve were screened out. Due to the superior biocompatibility and three-dimensional conformation, natural scaffold materials exhibit unparalleled bionic property compared with other materials. Synthetic biodegradable polymer materials with good mechanical properties and controllability has thus been highly favored by researchers, while the composite scaffold materials of natural materials and polymer materials provides a new strategy and direction for the investigations of tissue engineered heart valve, and has broad application prospects.

Objective To establish and validate a standard method using flow cytometry (FCM)in human ABO blood group antibody (Ab) detection. Methods Sera samples from 52 blood donors were incubated with standardize red blood cells (RBCs), and anti-A/B antibody (IgM/IgG) was measured by indirect flow cytometry after adding secondary isotype-specific fluoresce labeled Ab. The results were compared with those by using ELISA reader. Results The changes of anti-A/B IgM measured by both methods correlated well and an overall correlation coefficiency of 0.730 for IgM was obtained by Spearman's rank testing (P<0.05). FCM showed better specificity, sensitivity and reproducibility over ELISA reader. In blood group A samples (n = 16) or B samples (n = 16) ,the antirespectively. IgG blood group Ab could only be detected in blood group O sera. Conclusion Compared with traditional methods,FCM is a more objective method to offer accurate detection of natural ABO blood group Ab (IgM/IgG) and allowed semi-quantitative measurement of antibody levels.

Chinese Journal of Postgraduates of Medicine 2018;41(7):585-589
Objective To observe the effect of different doses of mivacurium chloride on muscle relaxation time-course and hemodynamics in children with different ages. Methods One hundred children of selective inguinal hernia repair under general anesthesia with endotracheal intubation from January 2016 to January 2017 were enrolled, and the age was 0.5 to 6.0 years. The children were divided into low age group (0.5 to 3.0 years) and high age group (3.1 to 6.0 years) according to the age, then the children were divided into low dose group (mivacurium chloride 0.20 mg/kg) and high dose group (mivacurium chloride 0.25 mg/kg) according to the doses of mivacurium chloride. Therefore, the children were divided into low age low dose group, low age high dose group, high age low dose group and high age high dose group with 25 cases each. The mean arterial pressure (MAP) and heart rates before anesthesia (T0) and 1 min (T1), 3 min (T2), 5 min (T3), 10 min (T4) after intravenous injection of mivacurium chloride were recorded. The times of first intravenous injection of mivacurium chloride to neuromuscular block 75% (ThD75), 90% (ThD90) and maximum (ThDmax) were recorded. The recovery index (RI) was recorded. The times of last intravenous injection of mivacurium chloride to onset of muscle convulsions (Th) and muscle convulsions recovery 10% (ThR10), 25% (ThR25), 75% (ThR75), 90% (ThR90) were recorded. The times of ratio of the fourth muscle twitch to the first muscle twitch (TOFR) recovery 75% (TOFR75) and 90% (TOFR90) were recorded. Results There were no statistical difference in MAP and heart rate among 4 groups (P>0.05). The ThD75, ThD90 and ThDmax in low age low dose group were (126 ± 40), (163 ± 59) and (192 ± 49) s, those in low age high dose group were (73 ± 15), (115 ± 41) and (142 ± 37) s, those in high age low dose group were (149 ± 38), (193 ± 44) and (221 ± 47)s, and those in high age high dose group were (105 ± 32), (138 ± 35) and (167 ± 44)s. The ThD75, ThD90 and ThDmax in low age high dose group were significantly shorter than those in low age low dose group, those in high age high dose group were significantly shorter than those in high age low dose group, those in high age low dose group were significantly longer than those in low age low dose group, those in high age high dose group were significantly longer than those in low age high dose group, and there were statistical differences (P<0.05). There were no statistical differences in Th, ThR10, ThR25, ThD75, ThD90, RI, TOFR75 and TOFR90 among low age low dose group and low age high dose group, high age low dose group and high age high dose group (P>0.05). Conclusions In the children of 0.5 to 3.0 years, the effect of mivacurium chloride is significantly faster than that in the children of aged 3.1 to 6.0 years. Compared with 0.20 mg/kg of mivacurium chloride, 0.25 mg/kg of mivacurium chloride has less time to display muscle relaxation effect. The recovery time is not affected by age and induction dose. Mivacurium chloride has no significant effect on hemodynamics.
8.Mechanism of Psoralea corylifolia Linn on liver injury based on bioinformatics
Journal of Pharmaceutical Practice 2022;40(3):254-258
Objective To explore the mechanism of Psoralea corylifolia Linn (PCL) on liver injury by establishing the biological function and pathway network of PCL components, targets and protein interactions based on bioinformatics. Methods The components of PCL and potential liver-injury related targets were collected from TCMIP database. The targets of PCL were predicted by the reverse pharmacophore matching method. Cytoscape software was applied for the construction of active components-targets network map. Protein-protein interaction network was constructed by STRING database. Gene ontology functional enrichment analysis and KEGG pathway enrichment analysis were conducted to predict the liver injury mechanism of PCL. Results 22 components were identified from PCL with the corresponding 31 potential liver injury targets, mainly on serum albumin (ALB), glutathione S-transferase P (GSTP1), transthyretin (TTR) and peroxisome proliferator activated receptor gamma (PPARG) by PPI network analysis. The chemical carcinogenesis, adenosine 5 '- monophosphate activated protein kinase (AMPK) signal, PPAR signal, liver enzyme P450 and its harmful substance metabolism, glutathione metabolism and other signaling pathways were selected by KEGG analysis. Conclusion The active components of PCL may target on ALB, GSTP1, TTR and PPARG to regulate AMPK and PPAR signaling pathways, leading to liver injury.

Objective:To discuss the prediction efficacy of Down′s screening serological indicators in the second trimester of gestational hypertension (GH).Methods:64 849 pregnant women who had undergone regular check-ups at the Beijing Obstetrics and Gynecology Hospital from June 2013 to June 2019 and finally gave birth were the subjects of the study. There were 3 808 cases of the GH group (including patients with GH, preeclampsia, and eclampsia diagnosed) and 61 041 cases of non-GH group respectively. 3 ml of fasting venous blood was drawn during 14-19 weeks of pregnancy, and body weight, serum β-human chorionic gonadotropin (β-HCG), free estriol (uE3) and alpha fetal protein (AFP) were obtained through physical check and laboratory testing. β-HCG, AFP, uE3 were analyzed by using the corresponding median times of gestational age (MOM value), and multivariate logistic regression model and receiver operating characteristic curve (ROC curve) were used to analyze the predictive efficiency of serological indicators. Results:Among 64 849 research subjects, the GH group (3 808 cases) and the non-GH group (61 041) were (29.51±2.72), (29.38±2.68) years old, the weights were (65.46±12.17), (58.73±9.13) kg, gestational age of blood collection were (16.53±0.89) and (16.58±0.90) weeks respectively. The analysis of multivariate logistic regression model showed that the risk of GH increased with the increase of body weight, β-HCG and AFP concentration, OR (95% CI) values were 1.059 (1.056-1.062), 1.329 (1.188-1.487) and 1.195 (1.125-1.269); so did when the concentration of uE3 decrease, OR (95% CI) value was 0.527 (0.464-0.599). ROC curve analysis results show that age, weight, serum AFP, β-HCG, uE3 and multivariate diagnosis all have predictive efficiency for GH ( P values<0.05); combined diagnosis of multiple indicators and weight AUC (95% CI) were 0.684 (0.675-0.693) and 0.673 (0.663-0.682), but the Youden index, sensitivity, and specificity of the two were relatively small. Conclusion:The β-HCG, AFP and uE3 of Down′s screening in the second trimester of pregnancy were relatively low in predicting GH.
