Objective To evaluate the immune effect of the diphtheria toxoid(DT)vaccine using chitosan as adjuvant on mice,so as to provide an experimental basis for the preparation of novel adjuvant vaccines of DT.Methods A total of 30 male C57BL/6 mice were divided into mucosal immunization group and humoral immunization group,and each group was randomly divided into 6 groups:negative control group(PBS),positive control group(DT),aluminum adjuvant group,chitosan adjuvant group(final solution with 0.5% chitosan),aluminum adjuvant vaccine group and chitosan adjuvant vaccine group,with 5 mice in each group. Mucosal immunization group and humoral immunization group were inoculated with 500 μL PBS buffer,10 μg DT,500 μg aluminum adjuvant and 500 μL chitosan adjuvant per mouse by nasal drip and intraperitoneal injection respectively. The mice were observed for the status,collected for the eyeball serum before inoculation(0 d)and 7,21,35 d after inoculation,and collected for the nasal lotion simultaneously. The levels of IgG antibody in serum and sIgA antibody in nasal lotion were detected by ELISA.Results After intraperitoneal injection and nasal drip of chitosan adjuvant vaccine,the mental and behavioral state of mice was normal. Chitosan adjuvant vaccine effectively induced the IgG antibody against diphtheria in mice with not significant difference,compared with the classic aluminum adjuvant vaccine group(F = 127.926 > F_(0.05(1,8)),P > 0.05);It also induced the production of sIgA in mice,which was significantly higher than that in the classical aluminum adjuvant vaccine group 21 d after immunization(F =127.926 > F_(0.05(1,8)),P < 0.05).Conclusion Chitosan adjuvant DT vaccine can effectively stimulate the mucosal immunity and humoral immunity in mice with good safety,and has the potential to be an alternative to aluminum adjuvant for vaccine preparation,which can be considered as nasal drop vaccines.

Objective:To investigate the compensation of the anterior alveolar bone thickness in skeletal class Ⅲ patients treated with orthodontic-surgical treatment.Methods:The samples consisted of 54 ske-letal classⅢpatients treated with orthodontic-surgical treatment.Lateral cephalograms were taken before treatment.Descriptive statistics were calculated for corresponding variables,and the differences between the samples and the norms from Peking University normal occlusion sample library were assessed by inde-pendent-sample t test.Correlation analyses were performed to find associations between skeletal charac-teristics and anterior alveolar bone thickness.According to skeletal anteroposterior discrepancy/vertical type (ANB,criteria=-4°;SN-MP,criteria=37.7°),the samples were allocated into group A (severe anteroposterior discrepancy/hypodivergent vertical type,n=1 1 ),group B (moderate anteroposterior dis-crepancy/hypodivergent vertical type,n=1 6),group C (severe anteroposterior discrepancy/hyperdiver-gent vertical type,n=1 4),and group D (moderate anteroposterior discrepancy/hyperdivergent vertical type,n=1 3),and one-way ANOVA with SNK multiple comparison test were performed.Results:The anterior alveolar bone thickness of the skeletal class Ⅲ patients were thinner compared with norm values (P<0.05 ).Correlational analyses showed that,both the upper and lower anterior alveolar bone thick-ness was correlated with the skeletal vertical discrepancy (P<0.05 ),but the anteroposterior type was only correlated with the lower anterior alveolar bone thickness (P<0.05).For the 4 groups according to skeletal anteroposterior discrepancy/vertical type,hypodivergent vertical type had thinner lower ligual and total bone thickness (LP,LW,P<0.05).while for upper alveolar bone thickness (UW),group C and group B exhibited the thinnest and thickest (the values of UW were 7.86 mm and 9.05 mm).Conclusion:The upper and lower anterior alveolar bone thickness of skeletal classⅢpatients are thinner compared with normal occlusion.Different skeletal anteroposterior discrepancy/vertical type results in differences in the anterior alveolar bone thickness,so decompensation should be treated differently and carefully.

OBJECTIVE:To study the pathogenic organism of sepsis in children with tumor and its drug sensitivity for references of clinical medication.METHODS:The drug sensitivity of hemoculture samples of children with neoplastic hema-tologic disorder was detected by paper disk method.RESULTS:Of the total109pathogenic bacteria isolated from hemoculture samples,60(55.0%)were gram-positive(G + )bacteria,47(43.1%)were gram-negative(G - )bacteria and2(1.8%)were fungi.Vancomycin,teicoplanin and aminoglycosides were more sensitive to G + bacteria,while carbopenems and cefoperazone/sulbactam were more sensitive to G - bacteria.CONCLUSIONS:In treating sepsis in children with tumor,the choice of drugs should be based on the drug sensitivity test,the possibility of G + and G - bacteria infections should be considered in empiric treatment and the possibility of mycotic infection should also be considered after long-term administration of antibacterials.

Extranodal natural killer/T cell lymphoma (ENKTL) is an EBV-associated and highly aggressive subtype of non-Hodgkin lymphoma. Most patients are distributed in Asia and South America, and advanced-stage patients have poor prognosis. Reports on research progress of ENKTL in the 58th American Society of Hematology Annual Meeting covered multiple respects which range from basic research to clinical prognosis and treatment: further study on pathogenesis of ENKTL is developing based on high-throughput sequence, and new prognostic index system can help to stratify patients more exactly; what's more, novel regimens make more benefits to the clinical efficacy and safety of ENKTL patients.

OBJECTIVE:To explore the role of clinical pharmacists in the treatment of critical patients at perinatal stage. METHODS:During the treatment process of 4 critical patients at perinatal stage,according to the patients'special pathophysiolog-ic status (such as urea cycle disorder,dyslipidemia,severe infection,can not recover circulating blood volume rapidly,etc.), weighing the order of priority,based on relevant guidelines and literatures,clinical pharmacists provided individual medication ad-vices in respects of drug selection,medication timing,treatment course,drug combination and dosage adjustment,such as using cefuroxime,reduced glutathione,ursodesoxycholic acid,etc.,stopping drugs timely or adjusting antibiotics therapy plan. They al-so provided pharmaceutical care as lab index determination and ADR monitoring. RESULTS:Physicians adopted clinical pharma-cists'advices,and then 4 critical patients were all recovered. CONCLUSIONS:During the treatment course of critical patients at perinatal stage,clinical pharmacists should select safe and effective drugs as much as possible and adjust therapy plan in time ac-cording to clinical symptoms. When the security runs counter to effectiveness,clinical pharmacists should fully weigh the advantag-es and disadvantages and guarantee the maximum safety of pregnant or delivery women and fetuses.

Objective： To study the role of mechanical stress on osteoblast-like characteristics expressed by human periodontal ligament cells in vitro. Methods： Human periodontal ligament cells (hPDLC) were cultured by explant method. We investigated the gene expression for osteoblast-like marker proteins in periodontal ligament stimulated by cyclical mechanical force in vitro by in situ hybridization with non-radioisotopic oligonucleotide probe for alkline phosphatase (ALP) and osteocalcin (OCN). Results： Human periodontal ligament cells exhibited a stronger signal for ALP mRNA stimulated by cyclical mechanical force, and exhibited a moderately intense positive signal for OCN mRNA—OCN was a marker of differentiated osteoblast-like cells. Conclusion： These findings suggest that mechanical force plays role in differentiation from hPDLC to osteoblast-like cells.

Objective: TO study the change of substance P(SP) level in Plasma and its relationship with ankylosing spondylitis(AS).Method: Radioimmunoassay. Results: The author observed that the level of SP in Plasma of AS is singificantly higher than that of healthy controls. SP level in AS patients during active stage is higher than in those during inactive stage. Conclusion: Increased SP may play an importantrole in the pathogenesis Of AS.

Objective To evaluate the efficacy and safety of platinum-based chemotherapy in elderly patients with advanced non-small cell lung cancer(NSCLC). Methods Clinical data of forty-one elderly patients (≥70 years) with advanced NSCLC who were treated by platinum-based chemotherapy were analyzed retrospectively. Results Among the 41 patients, 18 (43.9%) patientswere treated with vinorelbine, 9 (22.0%) patients with gemcitabine, 7 (17.1%) patients with paclitaxel and docetaxel respectively. The overall response rate was 19.5%. The median time to progression (MTTP) was 5.8 months, and 1-year survival rate was 65.8% with a median survival time (MST) of 14.2 months. No significant statistical differences were found among the four regimens. The major side effect was bone marrow suppression. Three patients withdrew chemotherapy after one cycle due to grade Ⅲ-Ⅳ myelosuppression. There was no death associated with chemotherapy. Conclusions The platinum-based combined chemotherapy is effective and well-tolerated in elderly patients with advanced NSCLC.

Objective:To evaluate the role of mitochondrial calcium homeostasis in hyperoxia-induced injury to alveolar epithelial cells typeⅡ(AECⅡ) in rats.Methods:Rat AECⅡcell lines (RLE-6TN) were randomly divided into 3 groups ( n=18 each) using a random number table method: nomoxia group (group C), hyperoxia group (group H), and hyperoxia plus ruthenium red group (group HR). The cell lines were cultured in a conventional incubator for 4 h in group C. The cell lines were incubated in an incubator with 90% O 2 for 4 h in group H. In group HR, ruthenium red 2 μmol/L was added first, and then the cell lines were incubated in an incubator with 90% O 2 for 4 h. After the treatment, the Ca 2+ concentrations in mitochondria were measured using the mitochondrial Ca 2+ specific fluorescent probe Rhod-2-AM, the ROS level was measured using the fluorescent probe DCFH-DA, and the cell morphology and mitochondrial ultrastructure were observed. Results:Compared with group C, the Ca 2+ concentrations in mitochondria and ROS level were significantly increased in H and HR groups ( P<0.05). Compared with group H, the Ca 2+ concentrations in mitochondria and ROS level were significantly decreased in group HR ( P<0.05). Compared group C, the cell volume was reduced, the morphology was shrunken round, the cell arrangement was loose, the mitochondrial double-layer membrane structure was broken, and the mitochondrial cristal fragments were observed in group H. Compared with group H, the cell morphology was significantly improved, and the slight damage to mitochondrial double-layer membrane structure was found, and the structure of mitochondrial cristae was normal in group HR. Conclusion:Mitochondrial calcium homeostasis is involved in the pathophysiological process of hyperoxia-induced injury to AECⅡ in rats.

OBJECTIVE:To prepare Carbinoxamine maleate sustained-release suspension,and evaluate its quality. METH-ODS:Using carbinoxamine maleate as raw material,drug-loaded resin was prepared by cation exchange resin;surface coating method was used to finally prepare sustained-release suspension,using Eudragit RS100 as sustained-release coating material to pre-pare sustained-release microparticles. HPLC was conducted to determine the content of carbinoxamine maleate,release degree of original preparations and self-made suspensions was compared,drug-loading capacity was calculated. RESULTS:The drug amount in preparing drug-loaded resin was 2%,reaction temperature was 25 ℃,and reaction time was 4 h;the drug-loading capacity in surface coating was 35%,amount of coating material was 10%,and reaction temperature was 40 ℃. The drug-loading capacities of sustained particles before and after coating were 35.23%,32.72%,respectively;the yield was 96.82%. The carbinoxamine ma-leate in prepared sustained-release suspension accounted for 98.76% of the labeled amount;release degree in 10 h reached about 80%,f2 was 65.73. CONCLUSIONS:Carbinoxamine maleate sustained-release suspension is prepared successfully,and its release is similar to the original preparation.
