Background and purpose:Tumor necrosis factor-related apoptosis inducing ligand(TRAIL)is a successful clinical anti-tumor agent.However,not all tumor cells are sensitive to TRAIL-mediated apoptosis.In this study,we examined the TRAIL-induced apoptosis after pretreatment with pentamidine in K562 cells.Methods:When K562 cells were pretreated with pentamidine followed by TRAIL,apoptosis of cells was analyzed by cellular morphology via viewing live cells under a light microscope and annexin V-FITC/propidium iodine FACS.Western blot was performed to examine the cleavage of caspase-3,-8 and poly(ADP-ribose)polymerase(PARR)and protein levels of X-linked inhibitor of apoptosis protein(XIAP).Results:Apoptotic cell death occurred and cleavage of caspase-3,-8 and PARR appeared when K562 cells were pretreated with 10?g/ml of pentamidine followed by 200 ng/ml of TRAIL.In addition,pentamidine decreased protein levels of XIAP.Conclusions:Combined treatment with pentamidine and TRAIL may become a new strategy for cancer therapy.

In National Comprehensive Cancer Network guideline, patients with the clinical stages of Ⅰ B-ⅢA esophageal neoplasms should be given radical PF regimen (cisplatin + 5-fluorouracil) concurrent chemoradiotherapy.The majorization PF and radiation dosage are studied by some scholars in China and abroad, and the local control rate and survival rate are improved, adverse reaction is within a controllable range.In recent years, some new drugs such as capecitabine, paclitaxel, docetaxel, pemetrexed and carboplatin also show their advantages in radical concurrent chemoradiotherapy.

For the purpose of diagnosis of sudden manhood death syndrome, immu- nohistochemical study of ANP was performed in right atria of 10 cases of sudden manhood death syndrome (SMDS) and 10 cases of noncardiac death controls with LSAB-method. It was found that the ANP granulus in right atria of SMDS were obviously depleted, compared with that in the control groups. The results showed that depletion of ANP granules in atria plays an important role in the causes of death of SMDS. This experiment also provides a new approach for studying the causes of SMDS.

Objective To explore the effect of empathy and forgiveness on urban residents' life satisfaction.Methods Interpersonal Reactivity Index(IRI),Trait Forgivingness Scale(TFS) and the Satisfaction with Life Scale(SWLS) were administrated to 3537 participants living in several provinces in China.Results ①The variable of perspective-taking had significant main effect on both forgiveness and life satisfaction(β=0.409,P ＜0.01 ; β=0.145,P ＜0.01),and particularly personal distress had significant negative effect on both forgiveness and life satisfaction (β=-0.291,P ＜ 0.01 ; β=-0.181,P ＜ 0.01)②Forgiveneas had significant main effect on life satisfaction(β=0.142,P＜0.01).③The relationship between perspective-taking and life satisfaction was partially mediated by forgiveness(P＜0.01).The relationship between personal distress and life satisfaction was also partially mediated by forgiveness (P ＜ 0.01).Conclusion Perspective-taking and personal distress has different predicting effect on life satisfaction,and forgiveness plays an important mediating role in the relationship between them.

Objective To study genotyping and molecular epidemiology distribution of GBS pathogenic strains of GBS positive pregnant women in Guangzhou,for GBS pathogenic strains of rapid molecular diagnosis and epidemiological surveillance pro-vide certain theoretical basis and method.Methods In the Guangzhou area,used multi stage stratified sampling method col-lecting GBS positive pregnant women’s reproductive tract specimens from January to December 2015,drug sensitivity quality control standard strains:Streptococcus pneumoniae (ATCC49619)and Staphylococcus aureus (ATCC25923),took culture of bacterial,strain,identification,DNA extraction,PCR,gene detection method,through the relevant software for data analy-sis,analyzed GBS strains of gene and molecular epidemiology.Results In the study,collected 2 812 samples of secretions,af-ter identification of strains isolated from 178 strains of pathogenic GBS strains,the detection rate was 6.33%.GBS patho-genic strains to linezolid vancomycin,penicillin,nitrfurantion and other antimicrobial drug resistance rate was 0,GBS parho-genic strains to ampicillin,ciprfloxacin moxifloxacin and levofloxacintesistant parts,the restance rates were 1.1%,16.9%, 18.0% and 22.5%,but GBS pathogenic strains to erythromycin,clindamycin tetracydine antibiotics showed a high resistance rate,the resistance rates were 50.6%,47.8%(of which 20 cases of erythromycin induced clindamycin resistance accouted for 23.5%)and 73.0%.Among them,65 strains of GBS detected the mreA gene,56 strains of GBS detected the ermB gene,36 strains of GBS detected the mefA gene,28 strains of GBS detected the mefE gene,5 strains of GBS detected the ermA gene, ermC gene was not detected in the gene.Among them,carried five multidrug resistance gene of 3 strains (1.6 9%)and 4 kinds of resistant gene carried with 15 strains (8.43%),carried three resistance genes of 19 strains (10.67%),2 kinds of resistant gene carrying a 25 strains (14.04%),carried the resistance gene of 5 strains (2.81%),did not carry resistance gene of 1 strain (0.56%).The nucleotide sequences of the five drug resistance genes were 100%,and no gene mutation oc-curred.Conclusion The main GBS disease resistant gene was mreA,ermA,ermB,mrfA,mefE and its nucleotide sequence homology was 100%.The clinical need to strengthen the detection of resistant gene and molecular level and guide clinical more scientific and rational drug use.

Objective To investigate the relationship between miR-411 and breast cancer,and the expression of miR-411 and its effects and mechanism research on proliferation and metastasis in breast cancer.Methods Real-time polymerase chain reaction (RT-PCR) was used to detect the expression of miR411 in breast cancer cells and tissues.Methyl thiazolyl tetrazolium (MTT),clone formation assay and Transwell assay were used to detect the effect of miR-411 expression on the proliferation,migration and invasion in breast cancer cells.The effect of miR-411 on growth factor receptor-bound protein 2 (GRB2) expression in breast cancer cells was detected by RT-PCR and Western blot.The direct effect of miR-411 target on GRB2 was detected by dual luciferase reporter assay.MTT,clone formation assay and Transwell assay detect the effect of GRB2 expression on the proliferation and invasion in breast cancer cells.Detection the effect high expression of miR-411 on GRB2 downstream signaling pathway related molecules expression in breast cancer cell with Western blot.Results The expression of miR-411 in breast cancer tissues was significantly lower than that in adjacent non-cancerous tissues (P ＜ 0.05).The expression of miR-411 in breast cancer cells SK-BR-3,BT-549 and MDA-MB-231 was significantly lower (P ＜ 0.05).Compared to the negative control group,the transfection of miR-411 mimic inhibited the proliferation,migration and invasion of MDA-MB-231 breast cancer cells (P ＜ 0.05).Targetscan showed that miR-411 could bind to GRB2 3'UTR at position 741-747.Compared with the negative control group,GRB2 3'UTR wild-type plasmid and miR-411 co-transfection reduced the fluorescence activity (P ＜ 0.05).Transfection of siGRB2 significantly reduced the expression of GRB2 protein in MDA-MB-231 breast cancer cells (P ＜ 0.05).Compared to the negative control group,the inhibition of GRB2 expression reduced the proliferation and the number of colony formation of MDA-MB-231 breast cancer cells (P ＜ 0.05).Transwell assay showed that transfection of siGRB2 significantly reduced the number of invasive cells in MDA-MB-231 breast cancer cells (P ＜ 0.05).Conclusions miR-411 is related closely to the occurrence and development of breast cancer,miR-411-GRB2-Ras axis is expected to become a new target for biological treatment of breast cancer.

Objective To explore the relationship among absolute income,big five personality and life satisfaction,and the mediating effect of relative income.Methods 5927 citizens from different cities were assessed by NEO five-factor inventory (NEO-FFI) and satisfaction with life scale (SWLS).Results (1) The mean score of life satisfaction was 19.61 ± 5.99.Female scored higher than male,and there were statistically significant differences between them (19.98 ± 5.92 vs 19.14 ± 5.95,t =-5.08,P ＜ 0.01).(2) The absolute income,extraversion and conscientiousness had a significant main effect on life satisfaction (β =0.13 ～ 0.19,P ＜ 0.01),neuroticism,openness and agreeableness had a significant main negative effect on life satisfaction (β =-0.04--0.15,P＜0.01).(3)The relative income had a significant main effect on life satisfaction (β =0.35,P ＜0.01).(4)Relative income had a mediating effect on the relation between absolute income,neuroticism,extraversion,agreeableness,conscientiousness and life satisfaction (P ＜ 0.01).Conclusion Relative income has a mediating effect on the relation between absolute income,personality and life satisfaction.
