Objective To predict as well as bioinforrmatically analyze the target genes of has-miR-451.Methods miRBase, miRanda, TargetScan and PicTar were used to predict the target genes of hsa-miRNA-451.The functions of the target genes were demonstrated by Gene Ontology and pathway enrichment analysis.P<0.05 was set as statistically significant. Results 18 target spots of hsa-miRNA-451 were predicted by 3 databases or prediction software at least. The functions of the target genes were enriched in proliferation and development of epithelial cells and regulation of kinase activity (P< 0.05).Pathway analysis showed that transforming growth factor-beta signaling pathway, mitogen-activated protein kinase signaling pathway, epidermal growth factor signaling pathway, Wnt signaling pathway and mammalian target of rapamycin signaling pathway were significantly enriched (P<0.05).Conclusion hsa-miRNA-451 might be involved in various signaling pathways related to proliferation and development of epithelial cells.

Objective To compare the axial length (AL) measured by Lenstar and contact A-Scan in the patients with idiopathic macular hole and study the correlation between the difference of the two measurements and the foveal thickness measured by optical coherence tomography (OCT).Methods Twenty-seven eyes of 26 idiopathic macular hole patients (IMH group) and 27 eyes of 25 patients with mild cataract (control group) were enrolled in this study.Foveal thickness was measured with 3D OCT.The AL was measured by Lenstar and contact A-Scan,and the consistency of the two measurements was determined by Bland Altman analysis. The correlation between the difference of the two measurements and foveal thickness was analyzed by Pearson correlation analysis. Results Mean foveal thickness of IMH and control eyes were (372.85±60.02) μm and (243.44±22.50) μm,respectively. The difference between the foveal thickness of the two groups was highly significant (t=-10.490,P<0.001).In the IMH group, the AL measured by Lenstar and contact A-Scan were (23.20± 1.12) mm and (23.18± 1.13) mm,respectively, the difference between the two measurements was not statistically significant (t =-0.549,P =0.588),whereas in the control group, the AL was (23.41 ± 0.72) mm by Lenstar and (23.33 ± 0.74) mm by contact A-Scan,the two measurements were significantly different (t=-4.832,P<0.001).However,no correlation was found by Pearson correlation analysis between the difference of the two measurements and the foveal thickness in either IMH or control group (r=0.181,-0.141; P>0.05).Conclusions Although there is no difference of axial length measurements using Lenstar and contact A-Scan in IMH eyes,in clinical measurements the results of two instruments should be taken into comprehensive consideration.

Objective To explore the consistency and significance of optical coherence tomography (OCT) and clinical and histopathological findings in adoptively transferred uveitis in mice. Methods The adoptively transferred experimental autoimmune uveitis (EAU) model was established by intraperitoneal injection of antigen-specific T cells in C57BL/6 mice.Since 9 days after transferred, inflammation of eyes was observed by indirect ophthalmoscope with +90D lens and record clinical scores every 3 days. The disease was divided into 6 phases including onset phase, early phase,pre-peak phase, peak phase, resolution phase and late phase of EAU,which respectively corresponding to clinical score 0.5,1.0,1.5-2.0,2.5-3.0,1.0-2.0 and less than 1.0.Since 9 days after transferred, the retina and retinal thickness (RT) was measured by spectralis OCT about 1 disc from the disc edge in 10 time points including 9,11,16,21,25,30,35,40,50 and 60 days after transferred. The OCT score was recorded as from 0.0 to 4.0.After transferred 9,21 and 60 days, the mice were killed and eye balls were examined in histology.OCT score, clinical score and histology in the mouse were compared and analyzed. Results The disease was divided into onset phase, early phase,pre-peak phase and peak phase of EAU,which respectively corresponding to 9,16,21 and 26 days after transferred. In four phases,OCT score were 0.5,1.0,2.0 and 4.0 respectively. After transferred 30 days, which was in resolution phase of EAU,the inflammation cells in vitreous were decreased and OCT score was 3.0.After transferred 60 days, which was in late phase of EAU,inflammation cells in vitreous were disappeared and retina was atrophic topically. The histology showed the vitreous has slight inflammation cells and retinal structure was normal at onset of EAU.The vitreous has massive inflammation cells and retina structure was disorder at pre-peak of EAU.And in resolution phase of EAU,the inflammation cells in vitreous were slightly and retina was atrophic and thinned. The data in this study demonstrated that OCT score was well correlated with clinical score in EAU (r=0.957 9,P<0.000 1).Conclusion OCT and clinical and histopathological findings in adoptively transferred uveitis in mice were consistency and OCT is contribute to evaluate the disease dynamically and quantifiably.

Objective To compare the differences of atria and atrial myocyte structure in patients with atrial fibrillation (AF) and sinus rhythm(SR) .To evaluate the influence of AF on cardiac function .Methods 79 patients without heart failure undergoing car-diopulmonary bypass surgery were divided into the AF group (n=39) and the SR group(n=40) .Echocardiography was performed for analysis of left ventricular end diastolic dimension(LVDd) ,left ventricular end-systolic dimension(LVDs) ,left ventricular poste-rior wall(LVPW) ,interventricular septum(IVS) and left atrial diameter(LAD) .Part of left atrial appendages was taken freshly for HE staining in order to observe atrial tissue structure .Results LVDd ,LVDs ,LVPW and IVS of the AF group were lower than that of the SR group .But LAD of the AF group was higher than the SR group .There was statistical significance in IVS and LAD between the two groups(P<0 .05) ,and there were no significant differences in other aspects (P>0 .05) .Compared with the SR group ,the AF group had thinner myocardial atrophy ,more obvious fibrosis ,smaller nucleus and darker HE staining .Conclusion The incidence of AF was mainly in rheumatic heart disease .The development of AF was mostly accompanied with the enlargement of LAD and the change of atrial tissue structure which showed that AF can reduce cardiac function .

Objective To investigate the clinical biomarkers of acute lung injury(ALI) after the Stanford A aortic dissection. Methods Thirty patients underwent Stanford A aoatic dissection were selected as subjects, who hospitalized from January 2006 to March 2013.Of which,21 patients underwent total arch replacement with stented elephant trunk procedure and 9 patients underwent triple-branched stent graft placement. The general information of patients, preoperation echocardiogram data, and arterial partial pressure of oxygen (PaO2),partial pressure of carbon dioxide (PaCO2) and fraction of inspired oxygen(FiO2) were recorded before, after the operation and entering ICU.Alveolar-arterial oxygen difference (A-aDO2),oxygenation index (OI) were calculated. Results A-aDO2 and OI at preoperation, postoperative and entering ICU point were (112.47 ±41.06) mmHg,(136.13 ± 29.51) mmHg and (141.37 ± 25.94) mmHg; (535.23 ± 70.15) mmHg; (491.50 ± 73.12) mmHg and (387.33 ± 91.32) mmHg respectively, and the differences were significant (F=35.926,323.742;P =0.000).The levels of A-aDO2 and OI at entering ICU were significant different from that of pre-operation and post-operation (P < 0.01,P < 0.05).Conclusion Early postoperative oxygenation and switching functions of patients with Stanford A aortic dissection are subject to damage to some degree. The A-aDO2 and OI might be sensitive biomarkers of the diagnosis for early acute lung injury of aortic dissection patients.
