2.Clinical features of cryptogenic stroke accompanied with patent foramen ovale

Objective To investigate the clinical features of cryptogenic stroke accompanied with patent foramen ovale ( PFO) .Methods The clinical data of 8 cryptogenic stroke patients accompanied with PFO were analyzed retrospectively.Results Eight patients aged 15-55 years old, average age was 39.75 years old.They were acute onset.The clinical symptoms were limb weakness in 5 cases, speech disorder in 2 cases, walking instability, weakness of lower limbs, epilepsy, acmesthesia decrement of one side in 1 case retrospectively. Doppler echocardiography showed PFO was in all the patients.Brain MRI showed frontal lobe, parietal lobe, temporal lobe, occipital lobe, insular cortex, periventricular body, hind limb of the internal capsule, basal ganglia, cerebellum had low T1 single, and high T2 single.Four cases were unilateral involvement, and 4 cases were bilateral involvement;6 cases were multiple lesions, 2 cases were single lesion;involved in anterior circun systemlatio system was in 4 cases, involved in posterior circulation system was in 1 case, and involved in anterior and posterior circulation system was in 3 cases.All cases had no recurrence after treatment.Conclusions The onset age of cryptogenic stroke patients accompanied with PFO is relatively young.Brain MRI shows multiple lesions, it can involed in bilateral, anterior and posterior circulation system.Prognosis of cryptogenic strok accompanied with PFO is good.
3.Meta-analysis of Effectiveness and Safety of Butylphthalide Capsules Combined with Xueshuantong Injec-tion in the Treatment of Acute Cerebral Infarction

Objective:To evaluate the effectiveness and safety of butyl phthalide capsules combined with Xueshuantong injection in the treatment of acute cerebral infarction (ACI). Methods: Cochrane Library, Medline, Embase, SCI, CBM, CNKI, VIP and Wanfang databases were searched from the building time to May 2016. The enrolled randomized controlled trails were studied by using Cochrane system evaluation methods to perform methodological quality assessment, and RevMan 5. 2 software was used to carry out Me-ta-analysis. Results:A total of 7 randomized controlled trails including 640 patients were enrolled. The results of Meta-analysis dem-onstrated that the effective rate of butylphthalide capsules combined with Xueshuantong injection was superior to that of Xueshuantong injection (RR=1. 33, 95%CI:1. 16-1. 52, P<0. 0001), butylphthalide (RR=1. 45, 95%CI:1. 15-1. 83, P=0. 002) and blank control (RR=1. 31, 95%CI:1. 08-1. 57, P=0. 005). NHISS of butylphthalide capsules combined with Xueshuantong injection was higher than that of Xueshuantong injection (MD=4. 63, 95%CI:3. 38-5. 87, P<0. 00001) and blank control (MD=6. 85, 95%CI:4. 90-8. 80, P<0. 00001). There was no significant difference in adverse drug reactions. Conclusion: Butylphthalide capsules combined with Xueshuantong injection is effective for the therapy of ACI. However, due to the limited quantity and quality of the in-cluded studies, larger scale trials are needed.
5.Influence of RNA interference in p53 gene on the expressions of genes involved in ultraviolet B-induced premature senescence and photocarcinogenesis in human skin fibroblasts

Objective To evaluate the effect of RNA interference in p53 gene on the expressions of genes involved in ultraviolet B (UVB)-induced premature senescence and photocarcinogenesis in human skin fibroblasts (HSFs).Methods A previously established HSF cell clone with repressed expression of p53,which was named as HSF-p53,was cultured and irradiated with a subcytotoxic dose (10 mJ/cm2) of UVB once a day for five consecutive days.The HSFs with normal expression of p53 served as the control.Subsequently,β-galactosidase (SA-β-gal)-staining was performed to estimate the degree of senescence,quantitative real-time PCR array was performed to determine the mRNA expressions of photocarcinogenesis-and senescence-associated genes,including p53,p21,p19,p16,pRb,fibronectin,osteonectin,smooth muscle 22 (SM22),bax,bcl-2,hypoxia-inducible factor-1 α (HIF-1α),vascular endothelial growth factor(VEGF),and human double minute-2 (hdm2).Statistical analysis was carried out by Student's t test using the software SPSS 10.0.Results The percentage of SA-β-gal-positive cells in irradiated HSF-p53 was 19.70% ± 0.85%,significantly higher than that in unirradiated HSF-p53 (12.77% ± 0.81%,t =6.45,P < 0.05),but lower than that in irradiated control HSFs (50.48% ± 5.30%,t =7.86,P < 0.05),and similar to that in unirradiated control HSFs (18.50% ± 0.45%,t =2.57,P > 0.05).Compared with the control HSFs,the HSF-p53 showed decreased expressions of p21,p19,fibronectin,osteonectin,SM22 and bax genes (all P < 0.05),but increased expressions of bcl-2,HIF-1α,VEGF and hdm2 genes (all P < 0.05),and a similar expression of p16 gene (P > 0.05); the repeated UVB radiation significantly promoted the expressions of p16 and pRb genes (both P < 0.05),but had no obvious effect on the expressions of the other genes in HSF-p53 compared with unirradiated HSF-p53 (all P > 0.05).Conclusions The inhibition of p53 expression may decelerate the UVB-induced premature senescence in HSFs,which may be involved in the p53-dependent tumor suppression.
6.Electromyographical study on muscle fatigue in repetitive forearm tasks.

The purpose of this study was to examine whether repetitive muscle tasks in low weight load might influence the fatigue of forearm muscles, and to identify ergonomic risk factors of forearm muscle fatigue in these tasks. Sixteen healthy male volunteers performed eight wrist extensions in different frequency, weight and angle loads while being instructed to keep a dominant upper limb posture as constant as possible. Surface electromyograph (sEMG) was recorded from right extensors digitorium (ED), flexor carpi radialis (FCR), flexor carpi ulnaris (FCU) and extensor carpi ulnaris (ECU) during the task performance. Our results showed that mean power frequency (MPF) and median frequency (MF) values of ED, FCR and FCU were significantly lower (P<0.05) at high frequency load level than at low load level. However, MPF and MF values of ED were significantly lower (P<0.01) in higher load groups of frequency, angle and weight than in lower load groups. These results indicated that the fatigue of muscles varied in the same task, and the number-one risk factor of ECU, ED and FCR was angle load.

7.Clinical significance of a new alternatively spliced variant of prostate specific membrane antigen

AIM: To discuss the relationship between prostate specific membrane antigen(PSMA) and prostate cancer and to seek a target for diagnosis and therapy of prostate cancer.METHODS: A pair of primers was designed according to the published PSMA mRNA sequence.Total RNA was extracted from prostate cancer tissues and was reversely transcribed into cDNA,which was used as a template for PCR to amplify the PSMA gene.The recombinant was sequenced and the result was analyzed by BLAST.The PSMA5 gene specific primers were designed to identify its expression in different cells and prostate tissues.RESULTS: A new alternatively spliced variant of PSMA named PSMA5 was discovered when sequencing the recombinant.PSMA5 showed well pathological tissue-specificity,and its expression rate in prostate cancer,benign prostatic hyperplasia of prostate,and normal prostate tissue were 92.6%,78.8% and 10.0%,respectively.It expressed specifically in Pca cell line LNCaP,not in cell lines of PC3,bladder carcinoma,renal carcinoma,or hepatoma.CONCLUSION: A new alternative spliced variant of PSMA named PSMA5 was discovered,which was well correlated with prostate cancer and benign prostatic hyperplasia.This finding may give a new clue to the evolution of prostate cancer and may provide a target for the diagnosis and therapy of prostate cancer.
8.Analysis of gene structure and diversity of new alternative spliced variants of prostate specific membrane antigen

AIM: To find out the gene structure and diversity of protate specific membrane antigen(PSMA) alternative spliced variants, and probe into the pathogenesis of prostate cancer.METHODS: 5'-RACE and 3'-RACE methods were used to amplify the 5' and 3'end of alternative spliced variant and then those viariants were sequenced for analyzing the gene stucture and diversity of PSMA alternative spliced variants of prostate cancer tissues.RESULTS: Four new alternative spliced variants of PSMA were discovered from prostate cancer tissues.Compared with reported PSMA alternative spliced variants,different insertions and deletions existed in different sites of those new variants.CONCLUSION: The discovery of the new variants confirms the diversity of PSMA spliced variants and provides the clues for seeking the target of diagnosis and therapy of prostate cancer.
9.Construction and sequence analysis of eukaryotic expression vector of Chinese prostate-specific membrane antigen

AIM: To obtain eukaryotic expression vector of Chinese prostate-specific membrane antigen. METHODS: Chinese prostate-specific membrane antigen (PSMA) cDNA was amplified by RT-PCR from prostate cancer tissues, then cloned into eukaryotic expression vector pcDNA3 0 and sequenced. RESULTS: Seven bases in Chinese PSMA cDNA sequence were found different from those reported by Israeli, which lead to two different amino acids. CONCLUSION: We have obtained the PSMA cDNA, and the recombinant eukaryotic expression vector was successfully constructed. The study lays foundation for DCs vaccine modified by PSMA gene for the treatment of prostate neoplasms.
10.Liquid chromatography-tandem mass spectrometry simultaneous determination of repaglinide and metformin in human plasma and its application to bioequivalence study.

A simple, sensitive, selective, and reproducible liquid chromatography-tandem mass spectrometric method was developed for the simultaneous determination of repaglinide and metformin in human plasma using d5-repaglinide and d6-metformin as internal standards (ISs). After a simple protein precipitation using acetonitrile as the precipitation solvent, both analytes and ISs were separated on a Venusil ASB C 18 (150 mm x 4.6 mm, 5 microm) via gradient elution using acetonitrile--10 mmol x L(-1) ammonium acetate as the mobile phase. A chromatographic total run time of 7.5 min was achieved. Mass spectrometric detection was conducted with atmospheric pressure chemical ionization under positive-ion and multiple-reaction monitoring modes. The method was linear over the 0.2 to 60.0 ng x mL(-1) concentration range for repaglinide and over the 4 to 1 000 ng x mL(-1) range for metformin. For both analytes, the intra- and inter-accuracies and precisions were within the +/- 15% acceptable limit across all concentrations. The validated method was successfully applied to a clinical bioequivalence study.
