1.Comparison of improvement of two kinds of preventative administration of inter-leukin-27 on airway inflammation and impaired phosphorylation of STAT1 in asthma mice

Objective:Animal models were set up to explore the best preventative intra-nasal administration of interleukin 27 (IL-27)to diminish allergic airway inflammation of asthma and the related molecular mechanisms. Methods:Ninety-six female C57/6J mice were randomly divided into four groups,a group of the control group,a group of asthma group,and two groups of the prevention group. Based on being sensitizing and challenging with Ovalbumin ( OVA ) in the asthma model, two kinds of IL-27 administration asthma animal models were set up,one of which was low-dose-multiple preventive administration before OVA sensitization,one was low-dose-multiple preventive administration after OVA sensitization but before OVA challenge. Sacrificed the mice after challenging and analyzed the IL-5 and IL-13 levels in supernatant of Broncho alveolar lavage fluid( BAL) using ELISA;and HE stain and inflammation score were done for the lungs. Sacrificed the mice before challenging and used the lungs to analyze the level of total signal transducer and activator of transcription-1(STAT1) protein and phos-STAT1 based on the method of Western blot. Results: In low-dose-multiple administration before sensitization preventions group,IL-27 inhibits the secretion of IL-5 and IL-13(P<0. 05) and inflammation around bronchial and vascular obviously, and the inflammation score was lower than asthma group ( P<0. 05 ) , while another group has no significant effects ( P>0. 05 ) . The phosphorylation of STAT1 was impaired in mice after OVA sensitization, and preventative administration of IL-27 before sensitization could reverse the impairment of STAT1, whereas another group had no obviously changes. Conclusion:Preventative administration of IL-27 before sensitization can attenuate the airway inflammation in the mouse asthma model via reversing the phosphorylation of STAT1,while the mice which has been sensitized resisting the inhibition of IL-27 due to the impairment of the phosphorylation of STAT1 and already committed Th2-CD4+T cells existed in sensitization-mice airway might be the reason for such IL-27 resistance.
2.Protective effect of tiopronin supplemented with chemotherapy in treating advanced breast cancer

Objective To observe the efficacy and safety of tiopronin supplemented with chemotherapy in treating advanced breast cancer.Methods Sixty patients with advanced breast cancer were randomly divided into two groups:treatment group(n = 28) and control group (n = 32).Two groups were treated the same of NVB + DDP,the treatment group was supplemented with tiopronin,given for 10 days.Efficacy,toxicity in two groups were compared.Results The effective rate in the treatment group and the control group were 46.4% and 46.9% respectively, with no significant difference between the two groups ,P > 0.05.But the improved quality of life of patients in the treatment group was higher than that in the control group, P < 0.05.The rate of adverse reaction in liver function damaged (9.4%)and leucocyte lassitude(46.4%) were apparent lower than those in the control group(31.2% ,81.2% ),with significant difference between the two groups(P < 0.05, P < 0.01).Conclusion Tiopronin supplemented with chemotherapy show apparent effect in decreasing the adverse reaction of chemotherapy,improving the quality of life and not influence efficacy in advanced breast cancer.So tiopronln may act as protective drug for chemotherapy and deserve further testing in the clinic.
3.Diagnosis and treatment of post-transplant lymphoproliferative disease after hematopoietic stem cell transplantation: Report of one case and literature review

Objective To investigate the diagnosis and treatment of post-transplant lymphoproliferative disease (PTLD) after hematopoietic stem cell transplantation (HSCT).Methods Clinical data of one case of PTLD,including clinical manifestations,diagnosis and treatment,were retrospectively analyzed.And the related literatures were reviewed.Results From January 2007 to November 2012,one case developed PTLD among 196 patients who were followed up after HSCT in the First Affiliated Hospital of Chongqing Medical University.The incidence of PTLD was 0.5%.Clinical manifestations of the patient were not typical,including frequent fever,multiple lymphadenopathy and multiple pulmonary nodules.The patient was diagnosed as diffuse large B-cell lymphoma (B-DLCL) by pathological examination of the swollen lymph node.After withdrawal of immunosuppressants,the temperature returned to normal,and lymphadenopathy and pulmonary nodules disappeared completely.Conclusion PTLD is a severe complication of HSCT with distinctive morphologic and clinical characteristics.The origin of this disease may be associated with Epstein-Barr virus infection and imrnunosuppressive therapy.It needs the pathologic detection to make a definite diagnosis.Several different treatment strategies have been employed,and reduction of immunosuppressive therapy may lead to regression of PTLD.
4.Exploration of the training form of cardiopulmonary resuscitation popularization among agricultural residence registration population in towns and villages

Objective To explore the short and long-term effect of cardiopulmonary resuscitation (CPR) training for agricultural residence registration population with the language education and practice education,in order to establish a more effective method of first-aid training for agriculture population.Methods 989 agricultural residence registration people were divided into group A (721 people)and group B (268 people) randomly.Group A received CPR training with language education and group B adopted practice education.On the time before and after training immediatelythe 3rd and 6th month after training,we checked the training results and analyzed the data with statistical methods.Results The short- term effect of two methods was satisfying (accuracy more then 90% ) and without statistical significance.The effect of group B was better than group A in the 3rd and 6th month after training.Two groups showed higher forgetting rate and some accuracy were less than 50%.Conclusions Agriculture population is in urgent need of first-aid.It may be a more effective method of first-aid training for agriculture population to transform gradually from language education to practice education.At the same time,we may use 4 hours as CPR practice education training time and 6 months as re-training period.
5.Yang-warming and Spleen-strengthening Therapy Combined with Antihyperglycemic Drugs for the Treatment of Insulin Resistance in Type 2 Diabetes with Yang Deficiency:An Observation of 70 Cases

0.05),but the decrease of 2-hour postprandial blood glucose level and the increase of ISI in group A were superior to those in group B (P0.05).For the therapeutic effect,11 patients were cured,24 markedly effective,22 effective,13 ineffective and the total effective rate was 81.43% in group A;5 patients were cured,6 markedly effective,11 effective,13 ineffective and the total effective rate was 62.86% in group B.No obvious toxic and side effects were observed in the two groups during the treatment.【Conclusion】Yang-warming and spleen-strengthening therapy combined with antihyperglycemic drugs exerts a better effect on the treatment of insulin resistance in type 2 diabetes.
6.Correlation between upper airway MRI parameters and the severity of patients with obstructive sleep apnea hypopnea syndrome

[ABSTRACT]OBJECTIVETo investigate the relationship between upper airway MRI parameters and the severity of patients with obstructive sleep apnea hypopnea syndrome (OSAHS). Then the values of upper airway MRI parameters in waking state were assessed.METHODS The MRI upper airway parameters of 37 OSAHS patients and 18 nonsnoring normal subjects were measured and compared. Then data analysis were carried out to find out significant correlations between the MRI upper airway parameters and the severity of patients (the apnea hypopnea index, AHI; the lowest oxygen saturation, LSaO2; the oxygen desaturation index, ODI) with OSAHS.RESULTSThe correlation between cross-sectional areas and left-right diameter in the narrowest retropalatal region, the thickness of lateral pharyngeal wall, the length of the tongue, the length and thickness of the soft palate and the cross-sectional areas of the soft palate on midsagittal planes and severity of OSAHS were found (P<0.05).CONCLUSIONThe MRI upper airway parameters can be used in evaluating the severity of OSAHS in some ways, locating the obstruction sites and choosing the best treatment planning.
7.Prognostic value of prognostic nutritional index in patients with non small cell lung cancer

Objective To investigate the prognostic value of prognostic nutritional index (PNI) in patients with non-small cell lung cancer.Methods A total of 179 patients with non small cell lung cancer was enrolled in our hospital from November 2010 to January 2014.All patients were pathologically confirmed to be non small cell lung cancer.The clinical data of patients were collected,and the PNI values of each patient were calculated.The patients were divided into 4 groups according to the patients in the PNI of the patients in the treatment group (PNIQ0-25 group,PNIQ25-50 group,PNIQ50-75 group and PNIQ75-100 group).The survival curve was drawn by Kaplan-Meier method,and the difference of progression free survival (DFS) and overall survival (OS) of each group was compared by Log-rank method.Results (1) compared with PNIQ0-25 group,PNIQ25-50 group,PNIQ50-75 group and PNIQ75-100 group,there was signifi cant difference in age,smoking and KPS score (P < 0.05).(2) the PNIQ0-25 group had a median overall survival of 11.5 months (95% CI:6.6 ~ 15.4),the 3 year survival rate was 6.7%;PNIQ25-50 group had a median overall survival of 12.2 months (95% CI:9.1 ~ 18),the 3 year survival rate was 6.4% in PNIQ50-75 group;the median overall survival was 14.1 the month of (95% CI:8.7 ~ 13.3),the 3 year survival rate was 11.4%,the PNIQ75-100 group had a median overall survival of 15 months (95% CI:12.3 ~ 17.8),the 3 year survival rate was 11.6%.The Log-rank test,the four groups of patients with a significant difference in overall survival (x2 =15.6,P =0.001).(3) the PNIQ0-25 group had a median progres sion free survival was 5 months for (95% CI:4.3 ~5.6),the 3 year progression free survival rate was 4.4%;PNIQ25-50 group had a median progression free survival was 6.4 months for (95% CI:4.7 ~8.1),the 3 year progression free survival rate was 4.3% in the PNIQ50-75 group;the median progression free survival was 7.4 months (95% CI:6 ~ 8.7),the 3 year progression free survival rate was 9.1% in PNIQ75-100 group,the median progression free survival was 8.9 months for (95% CI:6.4 ~ 10.8),the 3 year progression free survival rate was 9.3% by Log-rank test,survival was statistically significant was no difference between the four groups (x2 =26.7,P =0.000).Conclusions PNI has a good application value in the prognosis of patients with non-small cell lung cancer.
8.Analysis of hierarchical training for specialized nurses in department of chilidren's hepatobiliary surgery

Improving the special nursing ability is an important guarantee to carry out the clinical work for specialized nurse, so it is necessary to receive special hierarchical training.According to the length of employment and position, all nurse staff in the Department of Hepatobiliary Surgery ,Children's Hospital of Chongqing Medical University were classified, and the training schedule was made according to the level of personnel development to implement specialist training and operational skills training and assessment.And then examination was administrated;individualized guidance was got across to everyone according to the outcome of examination.Before and after the hierarchical training,analysis of doctors' and patients' satisfaction for nursing work, nursing theory, nursing service quality was carried out.The aim of this study is to verify the positive role of hierarchical training mode in promoting individual nursing ability and improve the level of nursing team, to provide experience and reference for nursing managers.
9.NaF· EDTA-K2 negatively bias blood lipids and lipoproteins measurement in pregnant women

Objective To explore the impact of NaF · EDTA-K2 on blood lipids measurement in pregnant women , determine whetherthe empty-stomach plasma for OGTT could be used as substitute for serumin blood lipids analyses.Methods Fastingplasma with NaF · EDTA-K2 and serumfrom 100 pregnant women werecollected,andconcentration of CHO,TG, HDL,LDL,apoA1 and apoB were tested with Hitachi 7 600 automatic biochemical analyzer.Data were analyzed by SPSS 19.0 software.Results The concentrations oflipids and lipoproteins with NaF · EDTA-K2 were slightly lower than those of serum.There were statistical significances between the means of CHO , HDL, LDL, apoA1 and apoBof two groups ( P<0.05) except TG.Correlation coefficients of the six analyteswere CHO 0.968, TG 0.995, HDL 0.979, LDL 0.991, apoA1 0.692, apoB 0.846respectively.Standard Error of Estimate for plasma were: CHO 0.281, TG 0.094, HDL 0.077, LDL 0.112, apoA1 0.230, apoB 0.111 respectively.The modified coefficients were 1.08 for CHO, HDL and LDL, 1.14 for apoA1 and 1.07 for apoB.Conclusion NaF· EDTA-K2 negatively biased the blood lipidsmeasurement of CHO , HDL, LDL, apoA1 and apoB in pregnant women,yet lipids concentrations in plasma with NaF · EDTA-K2 were closely related to those in serum , which may be used to predict the bloodlipidslevel of pregnant women afterajusted.
10.The Activity Study of Pancreatic Cancer Inhibited by Guttiferone K both in vitro and in vivo

This study aimed at exploring the effects of Guttiferone K (GUTK),a compound isolated from G.yunnanensis,on inhibiting the proliferation of pancreatic cancer cells both in vitro and in vivo.MTT assay was used to detect the inhibitory effects of GUTK on the proliferation of five human pancreatic cancer cell lines.Western blot was adopted to detect the apoptosis-related protein expressions of Caspase-3,poly adenosinediphosphate-ribose polymerase (PARP) and Bcl-xL.For in vivo study,the human pancreatic cancer cell MIA PaCa-2 was orthotopically injected into the pancreatic tail of the orthotopic mice.One week later,GUTK was administered by intraperitoneal (i.p.) injection every other day for 4 weeks.The volume and weight of the tumor tissue were measured.The protein expression level of cleaved caspase-3 in tumor tissue of all the groups was quantified by immunohistochemistry.As a result,it was found that GUTK effectively inhibited the proliferation of the five human pancreatic cancer cell lines at a low concentration.GUTK induced caspase-related apoptosis by triggering a series of events in MIA PaCa-2 cells including cleaved Caspase-3 and PARP activation,Bcl-xL down-regulation,and eventually cell death in a time and dose dependent manner.Furthermore,in vivo study revealed that intraperitoneal injection of GUTK significantly suppressed the growth of pancreatic cancer cells in the orthotopic mouse models,and the protein level of cleaved caspase-3 was increased in the GUTK and gemcitabine treated groups.It was concluded that GUTK induced apoptosis in human pancreatic cancer both in vitro and in vivo,and was potential to develop into a clinical anticancer agent.
