Objective:To investigate the protective effects of neferine on lipopolysaccharide ( LPS)-induced human umbilical vein endothelial cells ( HUVECs) injury. Methods:The optimum inducing concentration of LPS was screened out through pretests and used for the model establishment of HUVECs damage. CCK8 was used to detect the influence of neferine at different concentrations on LPS-induced human umbilical vein endothelial cells ( HUVECs) injury. Nitric oxide ( NO) content was measured by the Griess Reagent method. The nitric-oxide synthase (NOS) activity was assessed by the commercially available kits. Results:The inhibitory rate of HU-VECs was 54. 50% detected by CCK8, which induced by LPS at the concentration of 100 μg·ml-1(P<0. 01). Neferine at the con-centrations of 0. 3-5. 0μmol·L-1 could increase the cell viability in a concentration-dependent manner, while it inhibited the cell pro-liferation at the concentration of 10 μmol·L-1(P<0.05). Neferine could reverse the situation, and the NO release was increased and the tNOS/iNOS activity was increased induced by LPS (P<0. 05). The results shown by the inverted microscope suggested that the floating dead cells were decreased, the cell shape was basically sound and tightly packed with the concentration increase of neferine (0. 3-5. 0μmol·L-1) in a concentration-dependent manner. Conclusion:The results show that neferine has protective effects on HU-VECs injury induced by LPS, and the mechanism may be related with the decrease of intracellular levels of NO and NOS activity.

OBJECTIVE: Study on the correlation between chronic sinusitis with nasal septum deviation. METHOD: Randomly selected 722 patients with coronal sinuses CT, statistics the number of cases of nasal septum deviation, cases of nasal septum deviation with chronic sinusitis, the wide and narrow side cases of nasal septum deviation complicated with sinusitis. The number of sinusitis without deviation, and paired test. RESULT: The incidence of sinusitis between deviation of nasal septum and non deviation were 54. 13% and 44. 66%, the difference between two groups was statistically significant (P<. 05), the incidence of sinusitis with nasal septum deviation of wide and narrow side were 31. 65% and 32. 12%, no significant difference between the two groups (P>0. 01). The incidence of sinusitis high deviation and non high deviation were 59. 54% and 46. 97%, the difference between the two groups was statistically significant (P<0. 05). The number of wide side of upper nasal septum deviation with sinusitis was 54, the narrow side was 66, there is no significant difference between the two groups (P>0. 05). CONCLUSION The deviation of nasal septum is associated with the formation of chronic sinusitis, the high deviation is more prone to sinusitis, The incidence of sinusitis and nasal septum deviation on both sides was no different.

Objective:To compare the inhibitory effect of neferine and isoliensinine on 5α-reductase to provide reference for the development of 5α-reductase inhibitors. Methods: Different reaction tubes and control tubes were prepared, liver was homogenated, and reducing coenzyme II ( NADPH) , testosterone, pending test sample, the positive drug and buffer was respectively added into 96-well plates. The change in the absorbance of NADPH at 340nm in 1h was determined by a microplate reader. Compared the experimen-tal group with the blank control group, the inhibition rate ( I%) of the test drugs against 5α-reductase was calculated. Results:As for the six concentration gradients (2-40 mg·ml-1 ) in the experiments, the best inhibitory concentration of neferine and isoliensinine was 10 mg·ml-1(I% =25.00 ±1.030% and 29.90 ±2.410%, respectively). Compared with the control group, neferine and isoliensi-nine showed significant inhibition against 5α-reductase (P<0. 05). Compared with the positive group at the same concentration (10 mg ·ml-1), the inhibition of neferine and isoliensinine was significantly lower (P<0. 05). The inhibition effect of isoliensinine was rel-atively better than that of neferine (P<0. 05). Conclusion:Neferine and isoliensininein have notable inhibitory effect on 5α-reduc-tase, which show certain application prospect in the treatment of prostatic hyperplasia in clinics.

Nuciferine is a major aporphine alkaloid in the leaves of Nelumbo nueifera. It has great effects such as lipid-lowering,anti-oxidation,antimicrobial activitity and anti-cancer. The review focused on its pharmacological effects and pharmacokinetic characters reported in recent years,which can be helpful to the further researches on nuciferine.

Objective To investigate the clinical application value of endotoxin and (1-3)-β-D glucan detection in the patients with acute pancreatitis(AP).Methods The MB-80 microbiology rapid dynamic detection system was applied to detect the concentration of plasma LPS and (1-3)-β-D glucan in the patients with AP.Then the results were analyzed.Results Among 184 cases of AP complicating peripancreatic abscess and bilateral lung infection,endotoxin G-lipopolysaccharides positive was in 23 cases,the average value was 394.07 pg/mL,the positive rate was 10.87%,and 15 cases of Gram-negative bacterial infection were verified by the microorganism culture.Among 175 cases of AP complicating peripancreatic abscess and bilateral lung infection,67 cases were(1-3)-β-D glucan positive,the average value was 93.22 pg/mL,the positive rate was 38.29%,which were with the positive rate of serum(1-3)-β-D glucan in the non-AP group,the difference was statistically significant(P<0.05),the microorganism culture or smear verified that 22 cases were fungal infection.Conclusion Plasma endotoxin and (1-3)-β-D glucan detection provides the laboratory biological indicator and is conducive to the assessment of AP infection severity and clinical medication.

Objective To establish the acute hind limb ischemia on Wistar rat with diabetes mellitus.MethodsThirty diabetes rats were induced by intraperitoneal STZ (50 mg/kg) injection, as well as the blood glucose level tested over 16.8 mmol/L. Objective: To prepare the sustained release system of icariin (ICA) @ gelatin nanoparticles (GNPs)-polyactic-co-glycolic acid (PLGA) (ICA @ GNPs-PLGA), and to optimize the conditions. Methods: ICA@GNPs-PLGA sustained release system was prepared using two-step desolvation method and S/O/W emulsion solvent-evaporation technique. The effects of different conditions, such as the PLGA: GNPs mass ratio and the total quality of ICA added on the entrapment efficiency (EE) of ICA® GNPs-PLGA composite microspheres were detected to optimize the preparation process. The surface morphology of GNPs and ICA @ GNPs-PLGA composite microspheres were observed by SEM. The EE and the release results of ICA@GNPs-PLGA in the sample were determined with HPLC Results: The prepared composite microspheres and nanocomplex were were white powder. The SEM results showed that the composite microspheres and nanocomplexs were spherical, the surfaces were smoothy, and the particle size distribution range was 4-12 μm and 150-200 nm, respectively, relatively uniform At a GNPs mass fraction of 6 mg, the critical concentration of PLGA in DCM ranged within 0.5%-1.0%. At a GNPs mass fraction of 12 mg, the critical concentration of PLGA in DCM ranged within 1.0%-2.0%. However, at a critical PLGA mass fraction lower than 0.25%, no fully formed composite microspheres were observed. Within the critical concentration, the average EE of ICA@ GNPs-PLGA microspheres was higher than (62.00 ± 1.25)%. In addition, the EE of ICA in the microspheres was negatively correlated with the quality of ICA added. The accumulative release rate was less than in 24 h and it was 65.21% in 40 d. Conclusion: The ICA@GNPs-PLGA microspheres with homogeneous particle size distribution, high EE, low initial burst and without agglomeration can be acquired under the optimized conditions.

OBJECTIVE: To evaluate the protective effects of oral administration of D-methionine tablets on noise-induced hearing impairment. METHOD: Two hundred and three volunteers were recruited and randomly divided into two groups: experimental group (n= 113) and control group (n=90). The subjects received oral administration of D-methionine tablets in the experimental group and placebo in the control group before noise exposure. Routine audiometric evaluation and ABR testing were performed and recorded before and after noise exposure. The statistical analysis was carried out with SPSS 11.0. RESULT: There were no differences in auditory threshold and I-V interpeak latency pre-noise exposure (P>0.05), however, there were statistically significant differences between two groups 1 d, 7 d after noise exposure (P<0.01 and P<0.05). In-respective-group, there were statistically significant differences between before and 1 d after noise exposure (P<0.05 and P<0.01), however, no difference was noted between before and 7 d after noise exposure in the experimental group (P>0.05). CONCLUSION The protective effects of D-methionine tablets were prominent on the noise-induced hearing impairment. Further studies regarding the optimal time of D-methionine administration will be recommended in the future.

A high performance liquid chromatographic assay has been developed for the simultaneous determination of four water soluble vitamins,thia min, ribo-flavin, uiacin and pyridoxine, in cereal products and fresh vegetables. For hydrolyzing sample in 0.4 mol/L HCL, autoclaving at 120℃, 15 psi for 30 minutes, a ?Bondapak column (300mm ? 3.9mm, Waters Co), a mobile phase of methanol-water (30:70) (0.005 mol/L heptanesulfonic acid) and a flow rate of 1.0ml/min gave the most satisfactory separation of the four vitamins. A double channel detector was used: three vitamins (B1, B2 and B5) were detected by UV spectrophotometry (254 nm) first, pyridoxine (B6) was detected by fluorometry (EX 290 nm, EM 390 nm) afterwards. Detection limits were 2, 5, 5 and 2ng, linear ranges were 5-40 ng, 5-50 ng, 5-40 ng and 5-50 ng for B1, B2, B5 and Be, respectively. Recoveries were 91.3%-102% (B1), 103%-105% (B2), 92%-100% (B5) and 99%-100% (B6), comparing with reference method and checking with food composition tables, very satisfactory results were obtained by this method.
