Objective To investigate the effects of acupuncture at points Baihui (GV20), Yintang (Ex-HN3) and Sishencong (Ex-HN1) on spatial learning and memory abilities and hippocampal expression of brain-derived neurotrophic factor (BDNF) in a rat model of post-stroke depression (PSD). Method Thirty male adult SD rats were allocated, using a random number table, tocontrol, acupuncture and sham operation groups, 10 rats each. A rat model of PSD was made by classical middle cerebral artery occlusion (MCAO) using an intraluminal thread technique, chronic unpredictable mild stress (CUMS) and solitary feeding. At the fourteenth day after the beginning of stress, the acupuncture group received acupuncture intervention for 14 consecutive days. At one week after the end of stress, the Morris water maze test was conducted to assess rat spatial learning and memory abilities. After the completion of the water maze test, hippocampal CA3 regional BDNF expression was determined by immunohistochemical method. Result Rat learning and memory abilities decreased significantly in the control group compared with the sham operation group (P<0.05). Space exploration time and the number of crossing platform reflecting memory abilities decreased significantly in the control group (22.36±6.20 s and 3.72±0.90 times, respectively) compared with the sham operation group (48.73±5.40 s and 8.60±1.10 times, respectively); there were statistically significant differences (P<0.01). Learning abilities, space exploration time, the number of crossing platform and BDNF expression increased significantly in PSD rats treated with acupuncture compared with the control group; there were statistically significant differences (P<0.01). Conclusion Acupuncture can improve spatial learning and memory abilities and raise hippocampal regional BDNF levels in rats with post-stroke depression.

A quantitative analysis method of multi-components with a single marker (QAMS) for simultaneous determination of six marker compounds (one from phenolic acids and five from phthalides) in Chuanxiong Rhizoma was established by applying HPLC and using butylidenephthalide as the internal reference substance. And also the feasibility and accuracy of the established method for quality evaluation and application of Chuanxiong Rhizoma were investigated and validated. The analysis was performed with the mobile phase consisting of acetonitrile - 0.2% aqueous formic acid. The flow rate was 1.0 mL . min-1 and the column temperature was maintained at 30 °C. The detection wavelengths were set at 252 nm (for ferulic acid, Z-ligustilide, and butylidenephthalide) and 266 nm (for senkyunolide I, senkyunolide A, and coniferyl ferulate), separately, and 20 µL was injected for analysis with gradient elution. The results showed that there were no significant differences observed between the HPLC-QAMS method and the external standard method (RSD <5%). The relative correction factors were credible (RSD < 5%) in changed chromatographic conditions. The established HPLC-QAMS method can be accurately used for simultaneously evaluating and controlling the quality of Chuanxiong Rhizoma with multi-components.

Objective To investigate the relationship between the changes in intracranial excitatory amino acid(EAA) levels and the incidence of postoperative cognitive dysfunction (POCD) in eldedy patients after general anesthesia. Methods Forty ASA Ⅰ-Ⅲ patients of both sexes aged 65-79 yr weighing 48-76 kg undergoing elective non-cardiac surgery under general anesthesia were studied.Anesthesia was induced with midazolam, fentanyl, etomidate and succinylcholine and maintained with continuous iv infusion of propofol, remifentanil, isoflurane inhalation and intermittent iv boluses of vecuronium. The right internal jugular vein was cannulated with a catheter which was advanced cephalad until jugular bulb. The jugular bulb venous blood samples were taken before (T0,baseline),at the end of (T1) and at 24,48 and 72h(T2-4) after operation for measurement of plasma concentrations of glutamate (Glu),aspartate (Asp) and glycine (Gly) by RP-HPLC.The cognitive function was evaluated by mini-mental state examination (MMSE) at T0 and T4.Results Eleven of the 40 patients developed POCD (28%).The plasma Gh,Asp and Gly concentrations were significantly increased after operation as compared with the baseline values in POCD patients and were significantly higher than in non-POCD patients. The plasma concentrations of Glu and Asp were negatively correlated with MMSE score(Glu:r=-0.86.P<0.01;Asp:r=-0.99,P<0.01).Conclusion Elderly patients may develop POCD after operation performed under general anesthesia through increase in intracranial EAA levels.
